| 1 | Pokemon reduces Bcl-2 expression through NF-κ Bp65:a possible mechanism of hepatocellular carcinoma显示文摘Objective:To investigate the relationship among Pokemon,NF-κB p65 and Bcl-2 in hepatoma cells.Methods:HCC cell HepG2,SMMC7721 and human fetal liver cell line L02 cells were used,and expression of Pokemon,NF-κB p65 and Bcl-2 in three cells were detected by realtime PCR and western blot.Then siRNA of Pokemon was applied to inhibit the expression of Pokemon and NF-κB p65 and apoptotic rate was determined by flow cytometric analysis. Results:Expressions of Pokemon,NF-κB p65 and Bcl-2 in human hepatoma cell HepG2, SMMC7721 expression were significantly higher than those in human embryonic stem cells L02. siRNA of Pokemon inhibited the expression of Pokemon,NF-κB p65 and Bcl-2 in liver cancer cells,and significantly increased apoptosis of liver cells.While siRNA of NF-κB n65 inhibited the expression of NF-κB p65 and Bcl-2,but Pokemon expression in hepatoma cells had no significant change.Conclusions:The proto-oncogene Pokemon can inhibit PI4ARF by specific transcription regulation of cell cycle and can induce tumors.In addition,Pokemon can regulate NF-κB p65 through the expression of apoptosis repressor,and promote the development of liver cancer.It suggests signal network in the liver include the regulation of new non-classical NF-κB regulatory pathway. | Xinkai Zhao Qiaoming Ning Xiaoning Sun De'an Tian | 2011 | Asian Pacific Journal of Tropical Medicine2011，4，6： | 11 |

| 3 | Streptozotocin-induced expression of Ngn3 and Pax4 in neonatal rat pancreatic α-cells显示文摘AIM:To investigate the mechanism behind β-cell regeneration in neonatal rat pancreas treated with streptozotocin (STZ).METHODS:Neonatal Sprague Dawley rats were intraperitoneally injected with 70mg/kg STZ.Body weight,pancreas weight and blood glucose were recorded every two days after the treatment.To identify the expression and location of transcription factors in the rat pancreas,double immunofluorescent staining was performed using antibodies to specific cell markers and transcription factors.RESULTS:Expression of Neurogenin 3 (Ngn3),a marker for endocrine precursor cells,was observed by immunofluorescence in a few β-cells and many α-cells.The expression reached a peak 12 d after treatment.Pax4,a transcription factor that lies downstream of Ngn3 and plays an important role in β-cell differentiation,was also expressed in the α-cells of STZ-treated rats.We did not observe significant changes in Nkx6.1,which is essential for β-cell maturation in the treated rats.CONCLUSION:α-cells dedifferentiated into endocrine precursor cells and acquired the ability to dedifferentiate in the neonatal rat pancreas after STZ treatment. | Xiao-Di Liang Yuan-Yuan Guo Ming Sun Ying Ding Ning Wang Li Yuan Wei De | 2011 | World Journal of Gastroenterology2011，17，23： | 5 |
| 4 | Engineering osteoarthritic cartilage model through differentiating senescent human mesenchymal stem cells for testing disease-modifying drugs显示文摘Significant cellular senescence has been observed in cartilage harvested from patients with osteoarthritis(OA).In this study,we aim to develop a senescence-relevant OA-like cartilage model for developing disease-modifying OA drugs(DMOADs).Spe-cifically,human bone marrow-derived mesenchymal stromal cells(MSCs)were expanded in vitro up to passage 10(P10-MSCs).Following their senescent phenotype formation,P10-MSCs were subjected to pellet culture in chondrogenic medium.Results from qRT-PCR,histology,and immunostaining indicated that cartilage generated from P10-MSCs displayed both senescent and OA-like phenotypes without using other OA-inducing agents,when compared to that from normal passage 4(P4)-MSCs.Interestingly,the same gene expression differences observed between P4-MSCs and P10-MSC-derived cartilage tissues were also observed between the preserved and damaged OA cartilage regions taken from human samples,as demonstrated by RNA sequencing data and other analysis methods.Lastly,the utility of this senescence-initiated OA-like cartilage model in drug development was assessed by testing several potential DMOADs and senolytics.The results suggest that pre-existing cellular senescence can induce the generation of OA-like changes in cartilage.The P4-and P10-MSCs derived cartilage models also represent a novel platform for predicting the efficacy and toxicity of potential DMOADs on both preserved and damaged cartilage in humans. | Ning Wang Yuchen He Silvia Liu Meagan J.Makarcyzk Guanghua Lei Alexander Chang Peter G Alexander Tingjun Hao Anne-Marie Padget Nuria de Pedro Tsapekos Menelaos Hang Lin | 2022 | Science China(Life Sciences)2022，65，2： | 5 |

A total of 45 elements, including Li, Be, Sc, Ti, V, Cr, Mn, Co, Ni, Cu, Zn, Ga, Rb, Sr, Y, Zr, Nb, Mo, Cd, In, Sb, Cs, Ba, La, Ce, Pr, Nd, Sm, Eu, Gd, Tb, Dy, Ho, Er, Tm, Yb, Lu, Hf, Ta, W, Tl, Pb, Bi, Th and U, were measured by inductively coupled plasma mass spectrometry (ICP-MS). Condi- tions and sample experiments showed that this procedure defines a good experimental method which has the advantages of clear interference, easy operation and reliable results.
