1Characteristics and factors that influenced damage to dams in the M_s 8.0 Wenchuan earthquake显示文摘Based on raw data from dams damaged in the Wenchuan earthquake, including many that were severely damaged, characteristics and factors that influenced the damage are discussed in this paper. Findings from this study include: severely damaged dams were densely distributed along the seismologic fault; small dams, especially small earth-rock dams, had the most serious damage that was caused by a variety of factors; the most serious damage was caused by seismic waves; damage was aggregated by aftershocks; and the extent of the damage patterns increased with the seismic intensity. Damage patterns varied in different intensity zones and cracking was the most common type of damage. Most of the dams had a good base with relatively high bearing capacity, and the walls of the earth-rock dams were mostly of clay soil. This type of base and body material mitigated some of the damage to dams. Reservoir maintenance and other factors also have a significant impact on the seismic safety of the dam. 2A SEMI-ANALYTICAL METHOD FOR THE VIBRATION OF AND SOUND RADIATION FROM A TWO-DIMENTIONAL BEAM-STIFFENED PLATE显示文摘A semi-analytical method based on space harmonics to investigate the vibration of and sound radiation from an infinite,fluid-loaded plate is presented.The plate is reinforced with two sets of orthogonally and equally spaced beam stiffeners,which are assumed to be line forces.The response of the stiffened plate to a convected harmonic pressure in the wave-number space is obtained by adopting the Green's function and Fourier transform methods.Using the boundary conditions and space harmonic method,we establish the relationship between the stiffener forces and the vibration displacement of the plate.In this paper,the stiffener forces are expressed in terms of harmonic amplitudes of the plate displacement,which are calculated by using a numerical reduction technique.Finally,the Fourier inverse transform is employed to find expressions of the vibration and sound radiation in physical space.Agreements with existing results prove the validity of this approach and more numerical results are presented to show that this method converges rapidly.Haian Zhou Xiaomlng Wang Yulin Mei 2011Acta Mechanica Solida Sinica2011,24,3:6
3Room temperature ferromagnetism of Ni, (Ni, Li), (Ni, N)-doped ZnO thin films显示文摘Ni-doped ZnO thin films (Ni concentration up to 10 mol%) were generated on Si (100) substrates by a sol-gel technique. The films showed wurtzite structure and no other phase was found. The chemical state of Ni was found to be bivalent by X-ray photoelectron spectroscopy. The results of magnetic measurements at room temperature indicated that the films were ferromagnetic, and magnetic moment decreased with rise of Ni concentration. 4NSD3S stabilizes MYC through hindering its interaction with FBXW7显示文摘The MYC transcription factor plays a key role in cell growth control. Enhanced MYC protein stability has been found to promote tumorigenesis. Thus, understanding how MYC stability is controlled may have significant implications for revealing MYC-driven growth regulatory mechanisms in physiological and pathological processes. Our previous work identified the histone lysine methyltransferase nuclear receptor binding SET domain protein 3 (NSD3) as a MYC modulator. NSD3S, a noncatalytic isoform of NSD3 with oncogenic activity, appears to bind, stabilize, and activate the transcriptional activity of MYC. However, the mechanism by which NSD3S stabilizes MYC remains to be elucidated. To uncover the nature of the interaction and the underlying mechanism of MYC regulation by NSD3S, we characterized the binding interface between both proteins by narrowing the interface to a 15-amino acid region in NSD3S that is partially required for MYC regulation. Mechanistically, NSD3S binds to MYC and reduces the association of F-box and WD repeat domain containing 7 (FBXW7) with MYC, which results in suppression of FBXW7-mediated proteasomal degradation of MYC and an increase in MYC protein half-life. 6Large tumor suppressor 2,LATS2,activates iNK in kinase-independent mechanism through ASK1显示文摘Apoptosis signal-regulating kinase 1 (ASK1)is an important mediator of the cell stress response pathways.Because of its central role in regulating cell death,the activity of ASK1 is tightly regulated by protein-protein interactions and post-translational modifications.Deregulation of ASK1 activity has been linked to human diseases,such as neurological disorders and cancer.Here we describe the identification and characterization of large tumor suppressor 2 (LATS2)as a novel binding partner for ASK1.LATS2 is a core kinase in the Hippo signaling pathway and is commonly downregulated in cancer.We found that LATS2 interacts with ASK1 and increases ASK1-mediated signaUng to promote apoptosis and activate the iNK mitogen-activated protein kinase (MAPK).This change in MAPK signaling is dependent on the catalytic activity of ASK1 but does not require LATS2 kinase activity. 10Acquisition of taxane resistance by p53 inactivation in ovarian cancer cells显示文摘Ovarian cancer is one of the most common gynecologic malignancies in women and has a poor prognosis.Taxanes are a class of standard first-line chemotherapeutic agents for the treatment of ovarian cancer.However,tumor-intrinsic and acquired resistance to taxanes poses major challenges to improving clinical outcomes.Hence,there is an urgent clinical need to understand the mechanisms of resistance in order to discover potential biomarkers and therapeutic strategies to increase taxane sensitivity in ovarian cancer.Here,we report the identification of an association between the TP53 status and taxane sensitivity in ovarian cancer cells through complementary experimental and informatics approaches.We found that TP53 inactivation is associated with taxane resistance in ovarian cancer cells,supported by the evidence from(i)drug sensitivity profiling with bioinformatic analysis of large-scale cancer therapeutic response and genomic datasets and(ii)gene signature identification based on experimental isogenic cell line models.Further,our studies revealed TP53-dependent gene expression patterns,such as overexpression of ACSM3,as potential predictive biomarkers of taxane resistance in ovarian cancer.The TP53-dependent hyperactivation of the WNT/β-catenin pathway discovered herein revealed a potential vulnerability to exploit in developing combination therapeutic strategies.Identification of this genotype-phenotype relationship between the TP53 status and taxane sensitivity sheds light on TP53-directed patient stratification and therapeutic discoveries for ovarian cancer treatment.Changfa Shu Xi Zheng Alafate Wuhafu Danielle Cicka Sean Doyle Qiankun Niu Dacheng Fan Kun Qian Andrey AIvanov Yuhong Du Xiulei Mo Haian Fu 2022Acta Pharmacologica Sinica2022,43,9:1

14Development of a miniaturized 3D organoid culture platform for ultra-high-throughput screening显示文摘The recent advent of robust methods to grow human tissues as 3D organoids allows us to recapitulate the 3D architecture of tumors in an in vitro setting and offers a new orthogonal approach for drug discovery.However,organoid culturing with extracellular matrix to support 3D architecture has been challenging for high-throughput screening(HTS)-based drug discovery due to technical difficulties.Using genetically engineered human colon organoids as a model system,here we report our effort to miniaturize such 3D organoid culture with extracellular matrix support in high-density plates to enable HTS.We first established organoid culturing in a 384-well plate format and validated its application in a cell viability HTS assay by screening a 2036-compound library.We further miniaturized the 3D organoid culturing in a 1536-well ultra-HTS format and demonstrated its robust performance for large-scale primary compound screening.Our miniaturized organoid culturing method may be adapted to other types of organoids.By leveraging the power of 3D organoid culture in a high-density plate format,we provide a physiologically relevant screening platform to model tumors to accelerate organoid-based research and drug discovery.Yuhong Du Xingnan Li Qiankun Niu Xiulei Mo Min Qui Tingxuan Ma Calvin J.Kuo Haian Fu 2020Journal of Molecular Cell Biology2020,12,8:1

19The vibroacoustic response and sound absorption performance of multilayer, microperforated rib-stiffened plates显示文摘The vibroacoustic response and sound absorption performance of a structure composed of multilayer plates and one rigid back wall are theoretically analyzed. In this structure, all plates are two-dimensional, microperforated, and periodically rib-stiffened. To investigate such a structural system, semianalytical models of one-layer and multilayer plate structures considering the vibration effects are first developed. Then approaches of the space harmonic method and Fourier transforms are applied to a one-layer plate, and finally the cascade connection method is utilized for a multilayer plate structure. Based on fundamental acoustic formulas,the vibroacoustic responses of microperforated stiffened plates are expressed as functions of a series of harmonic amplitudes of plate displacement, which are then solved by employing the numerical truncation method. Applying the inverse Fourier transform, wave propagation, and linear addition properties, the equations of the sound pressures and absorption coefficients for the one-layer and multilayer stiffened plates in physical space are finally derived. Using numerical examples, the effects of the most important physical parameters—for example, the perforation ratio of the plate, sound incident angles, and periodical rib spacing—on sound absorption performance are examined. 20Efficient semi-analytical methods for the vibration response of and acoustic radiation from a periodical orthogonally rib-stiffened plate显示文摘In order to theoretically predict and analyze the vibration response and acoustic radiation characteristics of a periodical orthogonally rib-stiffened plate,its vibro-acoustic equations of an underwater infinite model are established.The rib-stiffened plate is stimulated by a harmonic plane pressure.By using the Fourier transforms,Poisson’s summation formula and space harmonic method,the structural vibration response and acoustic radiation pressure are expressed as functions of displacement harmonic components.Efficient semi-analytical methods are proposed in this work,and then approximate solutions for finite terms of the harmonic components are obtained by employing the truncation technique.Effects of the vibration response,rib spacing and torsional moment of the ribs on the radiation pressure are examined,and the validity of the present methods is also verified.Theoretical results show that the torsional moment of the ribs affects the modal frequencies of the stiffened plate,which should not be neglected in engineering applications with high precision requirement.With attachment of the ribs to the thin plate,its far field radiation pressure can be reduced in the low frequency range by adjusting rib spacing and cross sectional size of the ribs.ZHOU Haian WANG Xiaoming WU Huayong MENG Jianbing LI Lijun 2019Chinese Journal of Acoustics2019,38,3:0
