1Anti-SARS-CoV-2 activities in vitro of Shuanghuanglian preparations and bioactive ingredients显示文摘Human infection with severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)causes coronavirus disease 2019(COVID-19)and there is no cure currently.The 3CL protease(3CLpro)is a highly conserved protease which is indispensable for CoVs replication,and is a promising target for development of broad-spectrum antiviral drugs.In this study we investigated the anti-SARS-CoV-2 potential of Shuanghuanglian preparation,a Chinese traditional patent medicine with a long history for treating respiratory tract infection in China.We showed that either the oral liquid of Shuanghuanglian,the lyophilized powder of Shuanghuanglian for injection or their bioactive components dose-dependently inhibited SARS-CoV-23CLpro as well as the replication of SARS-CoV-2 in Vero E6 cells.Baicalin and baicalein,two ingredients of Shuanghuanglian,were characterized as the first noncovalent,nonpeptidomimetic inhibitors of SARS-CoV-23CLpro and exhibited potent antiviral activities in a cell-based system.Remarkably,the binding mode of baicalein with SARS-CoV-23CLpro determined by X-ray protein crystallography was distinctly different from those of known 3CLpro inhibitors.Baicalein was productively ensconced in the core of the substrate-binding pocket by interacting with two catalytic residues,the crucial S1/S2 subsites and the oxyanion loop,acting as a“shield”in front of the catalytic dyad to effectively prevent substrate access to the catalytic dyad within the active site.Overall,this study provides an example for exploring the in vitro potency of Chinese traditional patent medicines and effectively identifying bioactive ingredients toward a specific target,and gains evidence supporting the in vivo studies of Shuanghuanglian oral liquid as well as two natural products for COVID-19 treatment.Hai-xia Su Sheng Yao Wen-feng Zhao Min-jun Li Jia Liu Wei-juan Shang Hang Xie Chang-qiang Ke Hang-chen Hu Mei-na Gao Kun-qian Yu Hong Liu Jing-shan Shen Wei Tang Lei-ke Zhang Geng-fu Xiao Li Ni Dao-wen Wang Jian-ping Zuo Hua-liang Jiang Fang Bai Yan Wu Yang Ye Ye-chun Xu 2020Acta Pharmacologica Sinica2020,41,9:32
2Isolation and characterization of dengue virus serotype 2 from the large dengue outbreak in Guangdong, China in 2014显示文摘Dengue has been well recognized as a global public health threat,but only sporadic epidemics and imported cases were reported in recent decades in China.Since July 2014,an unexpected large dengue outbreak has occurred in Guangdong province,China,resulting in more than 40000 patients including six deaths.To clarify and characterize the causative agent of this outbreak,the acute phase serum from a patient diagnosed with severe dengue was subjected to virus isolation and high-throughput sequencing(HTS).Traditional real-time RT-PCR and HTS with Ion Torrent PGM detected the presence of dengue virus serotype 2(DENV-2).A clinical DENV-2 isolate GZ05/2014 was obtained by culturing the patient serum in mosquito C6/36 cells.The complete genome of GZ05/2014 was determined and deposited in Gen Bank under the access number KP012546.Phylogenetic analysis based on the complete envelope gene showed that the newly DENV-2 isolate belonged to Cosmopolitan genotype and clustered closely with other Guangdong strains isolated in the past decade.No amino acid mutations that are obviously known to increase virulence or replication were identified throughout the genome of GZ05/2014.The high homology of Guangdong DENV-2 strains indicated the possibility of establishment of local DENV-2 circulation in Guangdong,China.These results help clarify the origin of this epidemic and predict the future status of dengue in China.ZHAO Hui ZHAO Ling Zhai JIANG Tao LI Xiao Feng FAN Hang HONG Wen Xin ZHANG Yu ZHU Qin YE Qing TONG Yi Gang CAO Wu Chun ZHANG Fu Chun QIN Cheng Feng 2014Science China(Life Sciences)2014,57,12:21
3Geochronology and Petrogenesis of Triassic High-K Calc-Alkaline Granodiorites in the East Kunlun Orogen,West China:Juvenile Lower Crustal Melting during Post-Collisional Extension显示文摘This study reports zircon U-Pb and Hf isotopes and whole-rock elemental data for granodiorites from the East Kunlun orogen. The zircon U-Pb dating defines their crystallization age of 235 Ma. The rocks are characterized by high-K calc-alkaline, magnesian and metaluminous with(K_2O+Na_2O)=6.38 wt.%–7.01 wt.%, Mg#=42–50 [Mg#=100×molar Mg/(Mg+Fe OT)], A/CNK=0.92–0.98, coupled with high εHf(t) values from-0.65 to-1.80. The rocks were derived from partial melting of a juvenile mafic crustal source within normal crust thickness. The juvenile lower crust was generated by mixing lithospheric mantle-derived melt(55%–60%) and supracrustal melt(40%–45%) during the seafloor subduction. 6A hybrid invisibility cloak based on integration of transparent metasurfaces and zero-index materials显示文摘The invisibility cloak,a long-standing fantastic dream for humans,has become more tangible with the development of metamaterials.Recently,metasurface-based invisibility cloaks have been proposed and realized with significantly reduced thickness and complexity of the cloaking shell.However,the previous scheme is based on reflection-type metasurfaces and is thus limited to reflection geometry.In this work,by integrating the wavefront tailoring functionality of transparent metasurfaces and the wave tunneling functionality of zero-index materials,we have realized a unique type of hybrid invisibility cloak that functions in transmission geometry.The principle is general and applicable to arbitrary shapes.For experimental demonstration,we constructed a rhombic double-layer cloaking shell composed of a highly transparent metasurface and a double-zero medium consisting of dielectric photonic crystals with Dirac cone dispersions.The cloaking effect is verified by both full-wave simulations and microwave experimental results.The principle also reveals exciting possibilities for realizing skin-thick ultrathin cloaking shells in transmission geometry,which can eliminate the need for spatially varying extreme parameters.Our work paves a path for novel optical and electromagnetic devices based on the integration of metasurfaces and metamaterials.Hongchen Chu Qi Li Bingbing Liu Jie Luo Shulin Sun Zhi Hong Hang Lei Zhou Yun Lai 2018Light(Science & Applications)2018,7,1:13
7Effects of different mixing ratios on emissions from passenger cars fueled with methanol/gasoline blends显示文摘Regulated and unregulated emissions from four passenger cars fueled with methanol/gasoline blends at different mixing ratios (M15,M20,M30,M50,M85 and M100) were tested over the New European Driving Cycle (NEDC).Volatile organic compounds (VOCs) were sampled by Tenax TA and analyzed by thermal desorption-gas chromatograph/mass spectrometer (TD-GC/MS).Carbonyls were trapped on dinitrophenylhydrazine (DNPH) cartridges and analyzed by high performance liquid chromatography (HPLC).The results showed that total emissions of VOCs and BTEX (benzene,toluene,ethylbenzene,p,m,o-xylene) from all vehicles fueled with methanol/gasoline blends were lower than those from vehicles fueled with only gasoline.Compared to the baseline,the use of M85 decreased BTEX emissions by 97.4%,while the use of M15 decreased it by 19.7%.At low-to-middle mixing ratios (M15,M20,M30 and M50),formaldehyde emissions showed a slight increase while those of high mixing ratios (M85 and M100) were three times compared with the baseline gasoline only.When the vehicles were retrofitted with new three-way catalytic converters (TWC),emissions of carbon monoxide (CO),total hydrocarbon (THC),and nitrogen oxides (NOx) were decreased by 24%-50%,10%-35%,and 24%-58% respectively,compared with the cars using the original equipment manufacture (OEM) TWC.Using the new TWC,emissions of formaldehyde and BTEX were decreased,while those of other carbonyl increased.It is necessary that vehicles fueled with methanol/gasoline blends be retrofitted with a new TWC.In addition,the specific reactivity of emissions of vehicles fueled with M15 and retrofitted with the new TWC was reduced from 4.51 to 4.08 compared to the baseline vehicle.This indicates that the use of methanol/gasoline blend at a low mixing ratio may have lower effect on environment than gasoline.Hong Zhao Yunshan Ge Jianwei Tan Hang Yin Jiadong Guo Wei Zhao Peipei Dai 2011Journal of Environmental Sciences2011,23,11:12
8Activation of mTORC1 in subchondral bone preosteoblasts promotes osteoarthritis by stimulating bone sclerosis and secretion of CXCL12显示文摘Increasing evidences show that aberrant subchondral bone remodeling plays an important role in the development of osteoarthritis(OA).However,how subchondral bone formation is activated and the mechanism by which increased subchondral bone turnover promotes cartilage degeneration during OA remains unclear.Here,we show that the mechanistic target of rapamycin complex 1(mTORC1)pathway is activated in subchondral bone preosteoblasts(Osterix+)from OA patients and mice.Constitutive activation of mTORC1 in preosteoblasts by deletion of the mTORC1 upstream inhibitor,tuberous sclerosis 1,induced aberrant subchondral bone formation,and sclerosis with little-to-no effects on articular cartilage integrity,but accelerated posttraumatic OA development in mice.In contrast,inhibition of mTORC1 in preosteoblasts by disruption of Raptor(mTORC1-specific component)reduced subchondral bone formation and cartilage degeneration,and attenuated post-traumatic OA in mice.Mechanistically,mTORC1 activation promoted preosteoblast expansion and Cxcl12 secretion,which induced subchondral bone remodeling and cartilage degeneration during OA.A Cxcl12-neutralizing antibody reduced cartilage degeneration and alleviated OA in mice.Altogether,these findings demonstrate that mTORC1 activation in subchondral preosteoblasts is not sufficient to induce OA,but can induce aberrant subchondral bone formation and secrete of Cxcl12 to accelerate disease progression following surgical destabilization of the joint.Pharmaceutical inhibition of the pathway presents a promising therapeutic approach for OA treatment.Chuangxin Lin Liangliang Liu Chun Zeng Zhong-Kai Cui Yuhui Chen Pinling Lai Hong Wang Yan Shao Haiyan Zhang Rongkai Zhang Chang Zhao Hang Fang Daozhang Cai Xiaochun Bai 2019Bone Research2019,7,1:12

11Alterations in enterocyte mitochondrial respiratory function and enzyme activities in gastrointestinal dysfunction following brain injury显示文摘AIM:To determine the alterations in rat enterocyte mitochondrial respiratory function and enzyme activities following traumatic brain injury(TBI).METHODS:Fifty-six male SD rats were randomly divided into seven groups(8 rats in each group):a control group(rats with sham operation)and traumatic brain injury groups at 6,12,24 h,days 2,3,and 7after operation.TBI models were induced by Feendy’s free-falling method.Mitochondrial respiratory function(respiratory control ratio and ADP/O ratio)was measured with a Clark oxygen electrode.The activities of respiratory chain complexⅠ-Ⅳand related enzymes were determined by spectrophotometry.RESULTS:Compared with the control group,the mitochondrial respiratory control ratio(RCR)declined at 6 h and remained at a low level until day 7 after TBI(control,5.42±0.46;6 h,5.20±0.18;12 h,4.55±0.35;24 h,3.75±0.22;2 d,4.12±0.53;3 d,3.45±0.41;7 d,5.23±0.24,P<0.01).The value of phosphate-to-oxygen(P/O)significantly decreased at12,24 h,day 2 and day 3,respectively(12 h,3.30±0.10;24 h,2.61±0.21;2 d,2.95±0.18;3 d,2.76±0.09,P<0.01)compared with the control group(3.46±0.12).Two troughs of mitochondrial respiratory function were seen at 24 h and day 3 after TBI.The activities of mitochondrial complex Ⅰ (6 h:110±10,12 h:115±12,24 h:85±9,day 2:80±15,day 3:65±16,P<0.01)and complexⅡ(6 h:105±8,12 h:110±92,24 h:80±10,day 2:76±8,day 3:68±12,P<0.01)were increased at 6 h and 12 h following TBI,and then significantly decreased at 24 h,day 2 and day3,respectively.However,there were no differences in complex Ⅰ andⅡactivities between the control and TBI groups.Furthermore,pyruvate dehydrogenase(PDH)activity was significantly decreased at 6 h and continued up to 7 d after TBI compared with the control group(6 h:90±8,12 h:85±10,24 h:65±12,day 2:60±9,day 3:55±6,day 7:88±11,P<0.01).The changes inα-ketoglutaric dehydrogenase(KGDH)activity were similar to PDH,except that the decrease in KGDH activity began at 12 h after TBI(12 h:90±12,24 h:80±9,day 2:76±15,day 3:68±7,day7:90±13,P<0.01).No significant change in malate dehydrogenase(MDH)activity was observed.CONCLUSION:Rat enterocyte mitochondrial respiratory function and enzyme activities are inhibited following TBI.Mitochondrial dysfunction may play an important role in TBI-induced gastrointestinal dysfunction.Ke-Jun Zhu Hong Huang Hui Chu Hang Yu Shi-Ming Zhang 2014World Journal of Gastroenterology2014,20,28:10

13Two case reports and literature review for hepatic epithelioid angiomyolipoma: Pitfall of misdiagnosis显示文摘BACKGROUND Hepatic epithelioid angiomyolipoma(HEAML) is a rare liver disease and is easily misdiagnosed. Enhanced recognition of HEAML is beneficial to the differential diagnosis of rare liver diseases.CASE SUMMARY We presented two cases of HEAML in Changzheng Hospital, Naval Medical University, and then collected and analyzed all reports about HEAML recorded in PubMed, MEDLINE, China Science Periodical Database, and VIP database from January 2000 to March 2018. A total of 409 cases of HEAML in 97 reports were collected, with a ratio of men to women of 1:4.84 and an age range from 12 years to 80 years(median 44 years). Among the patients with clinical symptoms mentioned, 61.93%(205/331) were asymptomatic, 34.74%(115/331) showed upper or right upper quadrant abdomen discomfort, while a few of them showed abdominal mass, gastrointestinal symptoms, low fever, or weight loss. The misdiagnosis rate of HEAML was as high as 40.34%(165/409) due to its nonspecific imaging findings. Most of the tumors were solitary and round in morphology, with clear boundaries. Ultrasound scan indicated low echo with internal nonuniformity and rich blood supply in most cases. Computer tomography/magnetic resonance imaging enhanced scan showed varied characteristics. The ratio of fast wash-in and fast wash-out, fast wash-in and slow wash-out, and delayed enhancement was roughly 4:5:1. A definite diagnosis of HEAML depended on the pathological findings of the epithelioid cells in lesions and the expression of human melanoma black 45, smooth muscle actin,melanoma antigen, and actin by immunohistochemical staining. HEAML had a relatively low malignant rate of 3.91%. 15Phase 1 studies comparing safety, tolerability, pharmacokinetics and pharmacodynamics of HLX01(a rituximab biosimilar) to reference rituximab in Chinese patients with CD20-positive B-cell lymphoma显示文摘Objective: This study aimed to compare the pharmacokinetic, pharmacodynamic and safety profiles of HLX01(a rituximab biosimilar) and reference rituximab sourced from China(Mab Thera?;rituximab-CN).Methods: Here we report the results of two phase 1 studies. In the phase 1 a, open-label, dose-escalation study(NCT03218072, CTR20140400), eligible patients received 250, 375 and 500 mg/m^(2) HLX01 sequentially at 7-day intervals, after confirming no dose-limiting toxicity(DLT). In the phase 1 b, double-blind study(NCT02584920,CTR20140764), eligible patients were given a single dose of 375 mg/m^(2) HLX01 or rituximab-CN. The primary endpoints included safety and tolerability parameters for the phase 1 a and the area under the plasma concentrationtime curve from time zero to day 91(AUC0-91 d) for the phase 1 b study. Equivalence was concluded if 90%confidence interval(90% CI) for the geometric least squares mean ratio(GLSMR) fell in the pre-specified equivalence criteria(80%-125%).Results: Between June 20, 2014 and January 5, 2015, 12 patients were enrolled in the phase 1 a study. The pharmacokinetics of HLX01 showed dose proportionality and accumulation to steady state. HLX01 was well tolerated, with no serious adverse events(AEs), discontinuations or DLTs. Between November 8, 2014 and August13, 2015, 87 eligible patients were enrolled in the phase 1 b study, including 43 who received HLX01 and 44 who were treated with rituximab-CN. The equivalence endpoint was met with GLSMR for AUC0-91 d being 89.6%(90% CI: 80.4%-99.8%). 16Chemical formation and source apportionment of PM_(2.5) at an urban site at the southern foot of the Taihang mountains显示文摘The region along the Taihang Mountains in the North China Plain(NCP) is characterized by serious fine particle pollution. To clarify the formation mechanism and controlling factors, an observational study was conducted to investigate the physical and chemical properties of the fine particulate matter in Jiaozuo city, China. Mass concentrations of the water-soluble ions(WSIs) in PM_(2.5) and gaseous pollutant precursors were measured on an hourly basis from December 1, 2017, to February 27, 2018. The positive matrix factorization(PMF) method and the FLEXible PARTicle(FLEXPART) model were employed to identify the sources of PM_(2.5). The results showed that the average mass concentration of PM_(2.5) was 111 μg/m^(3) during the observation period. Among the major WSIs, sulfate, nitrate, and ammonium(SNA) constituted 62% of the total PM_(2.5) mass, and NO_(3)^(-) ranked the highest with an average contribution of 24.6%. NH_(4)^(+) was abundant in most cases in Jiaozuo. According to chemical balance analysis, SO_(2)-4, NO_(3)^(-), and Cl^(-) might be present in the form of(NH_4)_(2)SO_4, NH_4NO_3, NH_4Cl, and KCl. The liquid-phase oxidation of SO_(2) and NO_(2) was severe during the haze period. The relative humidity and pH were the key factors influencing SO_(4)^(2-) formation. We found that NO_(3)^(-) mainly stemmed from homogeneous gas-phase reactions in the daytime and originated from the hydrolysis of N_(2)O_5 in the nighttime, which was inconsistent with previous studies. 17Room-temperature-densified H_(3)BO_(3) microwave dielectric ceramics with ultra-low permittivity and ultra-high Qf value显示文摘With the rapid development of mobile communication technology towards 5G and 6G,the microwave dielectric materials with ultra-low permittivity and ultra-high Qf value are urgently demanded.Here,the excellent microwave dielectric properties are reported in H3BO3 ceramics with the molecular crystal structure,whose permittivity(2.84)and density(1.46 g/cm^(3))are record-low among the low-loss ceramics.The ultra-high Qf value of 146,000 GHz(or the ultra-low dielectric loss of 1.03×10^(-4) at 15 GHz)is also distinguished.Besides,the H_(3)BO_(3) ceramics can be densified at room temperature by a simple cold sintering process in a short time of 10 min,and this brings many advantages for the integration with microwave circuits.The large molecule volume originating from the molecular crystal structure and the low dielectric polarizabilities of H^(+) and B^(3+) are responsible for the ultra-low permittivity of H_(3)BO_(3) ceramics,and more microwave dielectric materials with ultra-low permittivity and ultra-high Qf value are expected to be explored in the molecular crystals.Wen Bin Hong Lei Li Han Yan Shu Ya Wu Hang Sheng Yang Xiang Ming Chen 2020Journal of Materiomics2020,6,2:3

19Anti-inflammatory effects of fatty acids isolated from Chromolaena odorata显示文摘Objective:To identify inhibitors of nitric oxide production and NF-κB activity from Chromolaena odorata(C.odorata).Methods:The compounds isolated from the aerial parts of C.odorata by bioassay-guided fractionation were investigated for their inhibitory effects on the NO production and NF-κB activity in LPS-stimulated RAW264.7 cells.Results:Six fatty acids (S)-coriolic acid(1),(S)-coriolic acid methyl ester(2),(5)-15,16-didehydrocoriolic acid(3),(S)- 15,16-didehydrocoriolic acid methyl ester(4),linoleamide(5) and linolenamide(6) were isolated. All compounds inhibited the NO production at concentrations consistent with those required for NF-κB inhibition.Compound 2 was the most active with the IC50 values of 5.22 and 5.73μM.
