| 1 | 尿激酶治疗急性心肌梗塞多中心临床试验1406例总结显示文摘为观察尿激酶天普洛欣(UKTP)经静脉溶栓治疗急性心肌梗塞(AMI)的临床有效性及安全性。收集协作组148家医院1994年11月至1996年4月经静脉UKTP溶栓治疗AMI患者1406例,观察临床疗效、副作用及病死率等。其中124例行90分钟冠状动脉造影评价梗塞血管开通情况。结果:梗塞血管临床再灌注率为73.5%,90分钟冠状动脉造影血管开通率为72.6%,5周总病死率为7.8%(109/1406),轻度出血10.2%(143/1406),中重度出血0.43%(6/1406),脑出血0.50%(7/1406)。老年(>65岁)甚至高龄(>75岁)患者溶栓及距发病超过6小时者,其用药仍然安全有效,UKTP合适的用药剂量可能为150万U左右。结果提示UKTP治疗AMI安全有效。 | The collaborative study group for national multicenter clinical trial of urokinase thrombolytic therapy (Correspondence: Hu Dayi, Xu Zhimin. | 2 | Mechanism for the formation of the January 2013 heavy haze pollution episode over central and eastern China显示文摘In January 2013,a long-lasting episode of severe haze occurred in central and eastern China,and it attracted attention from all sectors of society.The process and evolution of haze pollution episodes were observed by the'Forming Mechanism and Control Strategies of Haze in China'group using an intensive aerosol and trace gases campaign that simultaneously obtained data at 11 ground-based observing sites in the CARE-China network.The characteristics and formation mechanism of haze pollution episodes were discussed.Five haze pollution episodes were identified in the Beijing-Tianjin-Hebei(Jing-Jin-Ji)area;the two most severe episodes occurred during 9–15 January and 25–31 January.During these two haze pollution episodes,the maximum hourly PM2.5mass concentrations in Beijing were 680 and 530μg m 3,respectively.The process and evolution of haze pollution episodes in other major cities in the Jing-Jin-Ji area,such as Shijiazhuang and Tianjin were almost the same as those observed in Beijing.The external cause of the severe haze episodes was the unusual atmospheric circulation,the depression of strong cold air activities and the very unfavorable dispersion due to geographical and meteorological conditions.However,the internal cause was the quick secondary transformation of primary gaseous pollutants to secondary aerosols,which contributed to the'explosive growth'and'sustained growth'of PM2.5.Particularly,the abnormally high amount of nitric oxide(NOx)in the haze episodes,produced by fossil fuel combustion and vehicle emissions,played a direct or indirect role in the quick secondary transformation of coal-burning sulphur dioxide(SO2)to sulphate aerosols.Furthermore,gaseous pollutants were transformed into secondary aerosols through heterogeneous reactions on the surface of fine particles,which can change the particle’s size and chemical composition.Consequently,the proportion of secondary inorganic ions,such as sulphate and nitrate,gradually increased,which enhances particle hygroscopicity and thereby accelerating formation of the haze pollution. | WANG YueSi YAO Li WANG LiLi LIU ZiRui JI DongSheng TANG GuiQian ZHANG JunKe SUN Yang HU Bo XIN JinYuan | 2014 | Science China Earth Sciences2014,57,1: | 189 |
| 3 | A rapid advice guideline for the diagnosis and treatment of 2019 novel coronavirus(2019-nCoV) infected pneumonia(standard version)显示文摘In December 2019, a new type viral pneumonia cases occurred in Wuhan, Hubei Province;and then named '2019 novel coronavirus(2019-nCoV)' by the World Health Organization(WHO) on 12 January 2020. For it is a never been experienced respiratory disease before and with infection ability widely and quickly, it attracted the world’s attention but without treatment and control manual. For the request from frontline clinicians and public health professionals of 2019-nCoV infected pneumonia management, an evidence-based guideline urgently needs to be developed. Therefore, we drafted this guideline according to the rapid advice guidelines methodology and general rules of WHO guideline development;we also added the first-hand management data of Zhongnan Hospital of Wuhan University. This guideline includes the guideline methodology, epidemiological characteristics, disease screening and population prevention, diagnosis, treatment and control(including traditional Chinese Medicine), nosocomial infection prevention and control, and disease nursing of the 2019-nCoV. Moreover, we also provide a whole process of a successful treatment case of the severe 2019-nCoV infected pneumonia and experience and lessons of hospital rescue for 2019-nCoV infections. | 4 | Preventive effect of glutamine on intestinal barrier dysfunction induced by severe trauma显示文摘AIM: To investigate the mechanism underlying intestinalbarrier function damage after severe trauma and thetherapeutic effect of glutamine.METHODS: Burned patients, and animal models of severstrauma replicated by hemorrhagic shock combined withendotoxin infusion and burn injury, were included in a serialexperiment. Effects of oral glutamine on intestinal barrierfunction were observed in scalded rets. Parametersmeasured in these experiments were as follows: plasmalevels of diamine oxidase (DAO), tumor necrosis factor(TNFα), endotoxin (LPS), and lactate as well as D-lactateby biochemical methods, lactose/mannitol (L/M) ratio inurine by SP-3400, and pathological examination of intestinalmucosa under light microscopy.RESULTS: Plasma DAO activity was significantly increasedafter injury. | 5 | Moxibustion inhibits interleukin-12 and tumor necrosis factor alpha and modulates intestinal flora in rat with ulcerative colitis显示文摘AIM: To investigate the effect of moxibustion on intestinal flora and release of interleukin-12 (IL-12) and tumor necrosis factor-α (TNF-α) from the colon in rat with ulcerative colitis (UC). METHODS: A rat model of UC was established by local stimulation of the intestine with supernatant from colonic contents harvested from human UC patients. A total of 40 male Sprague-Dawley rats were randomly divided into the following groups: normal (sham), model (UC), herb-partition moxibustion (HPM-treated), and positive control sulfasalazine (SA-treated). Rats treated with HPM received HPM at acupuncture points ST25 and RN6, once a day for 15 min, for a total of 8 d. Rats in the SA group were perfused with SA twice a day for 8 d. The colonic histopathology was observed by hematoxylin-eosin. The levels of intestinal flora, including Bifidobacterium, Lactobacillus, Escherichia coli (E. coli), and Bacteroides fragilis (B. fragilis), were tested by real-time quantitative polymerase chain reaction to detect bacterial 16S rRNA/DNA in order to determine DNA copy numbers of each specific species. Immunohistochemical assays were used to observe the expression of TNF-α and IL-12 in the rat colons. RESULTS: HPM treatment inhibited immunopathology in colonic tissues of UC rats; the general morphological score and the immunopathological score were significantly decreased in the HPM and SA groups compared with the model group [3.5 (2.0-4.0), 3.0 (1.5-3.5) vs 6.0 (5.5-7.0), P < 0.05 for the general morphological score, and 3.00 (2.00-3.50), 3.00 (2.50-3.50) vs 5.00 (4.50-5.50), P < 0.01 for the immunopathological score]. As measured by DNA copy number, we found that Bifidobacterium and Lactobacillus, which are associated with a healthy colon, were significantly higher in the HPM and SA groups than in the model group (1.395 ± 1.339, 1.461 ± 1.152 vs 0.045 ± 0.036, P < 0.01 for Bifidobacterium, and 0.395 ± 0.325, 0.851 ± 0.651 vs 0.0015 ± 0.0014, P < 0.01 for Lactobacillus). On the other hand, E. coli and B. fragilis, which are associated with an inflamed colon, were significantly lower in the HPM and SA groups than in the model group (0.244 ± 0.107, 0.628 ± 0.257 vs 1.691 ± 0.683, P < 0.01 for E. coli, and 0.351 ± 0.181, 0.416 ± 0.329 vs 1.285 ± 1.039, P < 0.01 for B. fragilis). The expression of TNF-α and IL-12 was decreased after HPM and SA treatment as compared to UC model alone (4970.81 ± 959.78, 6635.45 ± 1135.16 vs 12333.81 ± 680.79, P < 0.01 for TNF-α, and 5528.75 ± 1245.72, 7477.38 ± 1259.16 vs 12550.29 ± 1973.30, P < 0.01 for IL-12). | 6 | Autophagy and multidrug resistance in cancer显示文摘Multidrug resistance(MDR) occurs frequently after long-term chemotherapy, resulting in refractory cancer and tumor recurrence.Therefore, combatting MDR is an important issue. Autophagy, a self-degradative system, universally arises during the treatment of sensitive and MDR cancer. Autophagy can be a double-edged sword for MDR tumors: it participates in the development of MDR and protects cancer cells from chemotherapeutics but can also kill MDR cancer cells in which apoptosis pathways are inactive. Autophagy induced by anticancer drugs could also activate apoptosis signaling pathways in MDR cells, facilitating MDR reversal. Therefore, research on the regulation of autophagy to combat MDR is expanding and is becoming increasingly important. | 8 | Deferoxamine promotes recovery of traumatic spinal cord injury by inhibiting ferroptosis显示文摘Ferroptosis is an iron-dependent novel cell death pathway. Deferoxamine, a ferroptosis inhibitor, has been reported to promote spinal cord injury repair. It has yet to be clarified whether ferroptosis inhibition represents the mechanism of action of Deferoxamine on spinal cord injury recovery. A rat model of Deferoxamine at thoracic 10 segment was established using a modified Allen's method. Ninety 8-week-old female Wistar rats were used. Rats in the Deferoxamine group were intraperitoneally injected with 100 mg/kg Deferoxamine 30 minutes before injury. Simultaneously, the Sham and Deferoxamine groups served as controls. Drug administration was conducted for 7 consecutive days. The results were as follows:(1) Electron microscopy revealed shrunken mitochondria in the spinal cord injury group.(2) The Basso, Beattie and Bresnahan locomotor rating score showed that recovery of the hindlimb was remarkably better in the Deferoxamine group than in the spinal cord injury group.(3) The iron concentration was lower in the Deferoxamine group than in the spinal cord injury group after injury.(4) Western blot assay revealed that, compared with the spinal cord injury group, GPX4, xCT, and glutathione expression was markedly increased in the Deferoxamine group.(5) Real-time polymerase chain reaction revealed that, compared with the Deferoxamine group, mRNA levels of ferroptosis-related genes Acyl-CoA synthetase family member 2(ACSF2) and iron-responsive element-binding protein 2(IREB2) were up-regulated in the Deferoxamine group.(6) Deferoxamine increased survival of neurons and inhibited gliosis. These findings confirm that Deferoxamine can repair spinal cord injury by inhibiting ferroptosis. | 9 | Anti-SARS-CoV-2 activities in vitro of Shuanghuanglian preparations and bioactive ingredients显示文摘Human infection with severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)causes coronavirus disease 2019(COVID-19)and there is no cure currently.The 3CL protease(3CLpro)is a highly conserved protease which is indispensable for CoVs replication,and is a promising target for development of broad-spectrum antiviral drugs.In this study we investigated the anti-SARS-CoV-2 potential of Shuanghuanglian preparation,a Chinese traditional patent medicine with a long history for treating respiratory tract infection in China.We showed that either the oral liquid of Shuanghuanglian,the lyophilized powder of Shuanghuanglian for injection or their bioactive components dose-dependently inhibited SARS-CoV-23CLpro as well as the replication of SARS-CoV-2 in Vero E6 cells.Baicalin and baicalein,two ingredients of Shuanghuanglian,were characterized as the first noncovalent,nonpeptidomimetic inhibitors of SARS-CoV-23CLpro and exhibited potent antiviral activities in a cell-based system.Remarkably,the binding mode of baicalein with SARS-CoV-23CLpro determined by X-ray protein crystallography was distinctly different from those of known 3CLpro inhibitors.Baicalein was productively ensconced in the core of the substrate-binding pocket by interacting with two catalytic residues,the crucial S1/S2 subsites and the oxyanion loop,acting as a“shield”in front of the catalytic dyad to effectively prevent substrate access to the catalytic dyad within the active site.Overall,this study provides an example for exploring the in vitro potency of Chinese traditional patent medicines and effectively identifying bioactive ingredients toward a specific target,and gains evidence supporting the in vivo studies of Shuanghuanglian oral liquid as well as two natural products for COVID-19 treatment. | Hai-xia Su Sheng Yao Wen-feng Zhao Min-jun Li Jia Liu Wei-juan Shang Hang Xie Chang-qiang Ke Hang-chen Hu Mei-na Gao Kun-qian Yu Hong Liu Jing-shan Shen Wei Tang Lei-ke Zhang Geng-fu Xiao Li Ni Dao-wen Wang Jian-ping Zuo Hua-liang Jiang Fang Bai Yan Wu Yang Ye Ye-chun Xu | 2020 | Acta Pharmacologica Sinica2020,41,9: | 32 |

| 11 | An atomic force microscopy study of coal nanopore structure显示文摘Coal nanopore structure is an important factor in understanding the storage and migration of absorbed gas in coal. A new method for studying coal nanopore structures is proposed. This idea is based on the nano-level resolution of atomic force microscopy, which can be employed to observe the structural features of coal nanopores clearly, conduct quantitative three-dimensional measurements and obtain structural parameters. Analysis results show that coal nanopores are mainly metamorphic pores and intermolecular pores. The metamorphic pores are commonly rounded and elliptical, increasing quantitatively with the coalification degree. The forms of intermolecular pores change markedly. The average pore size of low-rank coal is bigger than high-rank coal, and the number of intermolecular pores decreases as the coal rank increases. Section analysis effectively characterizes the coal pore microstructure, bearing analysis is a vital approach to measure microporosity, and grain analysis can be employed to study the pore size distribution. | 12 | Puerarin protects brain tissue against cerebral ischemia/reperfusion injury by inhibiting the inflammatory response显示文摘Puerarin, a traditional Chinese medicine, exerts a powerful neuroprotective effect in cerebral ischemia/reperfusion injury, but its mechanism is unknown. Here, we established rat models of middle cerebral artery ischemia/reperfusion injury using the suture method. Puerarin(100 mg/kg) was administered intraperitoneally 30 minutes before middle cerebral artery occlusion and 8 hours after reperfusion. Twenty-four hours after reperfusion, we found that puerarin significantly improved neurological deficit, reduced infarct size and brain water content, and notably diminished the expression of Toll-like receptor-4, myeloid differentiation factor 88, nuclear factor kappa B and tumor necrosis factor-α in the ischemic region. | 15 | Chinese Trauma Surgeon Association for management guidelines of vacuum sealing drainage application in abdominal surgeries-Update and systematic review显示文摘Vacuum sealing drainage (VSD) is frequently used in abdominal surgeries. However, relevant guidelines are rare. Chin ese Trauma Surge on Associati on orga nized a committee composed of 28 experts across China in July 2017, aiming to provide an evidence-based recommendation for the application of VSD in abdominal surgeries.Eleven questions regarding the use of VSD in abdominal surgeries were addressed:(1) which type of materials should be respectively chosen for the intraperitoneal cavity, retroperitoneal cavity and superficial incisions?(2) Can VSD be preventively used for a high-risk abdominal incision w让h primary suture?(3) Can VSD be used in severely contaminated/infected abdominal surgical sites?(4) Can VSD be used for temporary abdominal cavity closure under some special conditions such as severe abdominal trauma, infection, liver transplantation and intra-abdominal volume increment in abdominal compartment syndrome?(5) Can VSD be used in abdominal organ inflammation, injury, or postoperative drainage?(6) Can VSD be used in the treatment of intestinal fistula and pancreatic fistula?(7) Can VSD be used in the treatment of intra-abdominal and extra-peritoneal abscess?(8) Can VSD be used in the treatment of abdominal wall wounds, wound cavity, and defects?(9) Does VSD in crease the risk of bleeding?(10) Does VSD increase the risk of intestinal wail injury?(11) Does VSD increase the risk of peritoneal adhesion? Focusing on these questions, evidence-based recommendations were given accordingly. VSD was strongly recommended regarding the questions 2-4. Weak recommendations were made regarding questions 1 and 5-11. | 20 | An elastic-viscous-plastic model for overconsolidated clays显示文摘The influences of time on clays are discussed first,and the concept of the instant normal compression line is proposed by analyzing the existing theories and experimental results.Based on the creep law,the relationship between the aging time and the overconsolidation parameter is built.With the reloading equation of the UH model(unified hardening model for overconsolidated clays) used to calculate the instant compression deformation,a one-dimensional stress-strain-time relationship is proposed.Furthermore,the evolution of this relationship is analyzed,and the characteristic rate that is a function of the overconsolidation parameter is defined.Then a three-dimensional elastic-viscous-plastic constitutive model is suggested by incorporating equivalent time into the current yield function of the UH model.The new model can describe not only creep,rate effect and other viscous phenomena,but also shear dilatancy,strain softening and other behaviors of overconsolidated clays.Besides,compared with the modified Cam-clay model it requires only one additional parameter(the coefficient of secondary compression) to consider the creep law.Finally,because the proposed model can be changed into the UH model under instantaneous loading,the elastic-plastic and elastic-viscous-plastic frameworks are unified. | YAO YangPing KONG LingMing HU Jing | 2013 | Science China(Technological Sciences)2013,56,2: | 19 |
