| 1 | Label-free visualization of cholesteatoma in the mastoid and tympanic membrane using CARS microscopy显示文摘Objective:The present study aimed to evaluate the possibility of using coherent anti-Stokes Raman spectroscopy(CARS) microscopy to determine the specific molecular morphology of cholesteatoma by detecting the natural vibrational contrast of the chemical bonds without any staining.Materials and methods:Specimens from the mastoid and tympanic membrane with and without cholesteatoma were analyzed using CARS microscopy,two-photon excited fluorescence(TPEF) microscopy,and the second harmonic generation(SHG) microscopy.Results:In cholesteatoma tissues from the mastoid,a strong resonant signal at 2845 cm^(-1) was observed by CARS,which indicated the detection of the CH_2 hydro-carbon lipid bonds that do not generate visible signals at 2940 cm^(-1) suggestive of CH_3 bonds in amino acids.A strong resonant signal at 2940 cm^(-1) appeared in an area of the same specimen,which also generated abundant signals by TPEF and SHG microscopy at 817 nm,which was suggestive of collagen.In the tympanic membrane specimen with cholesteatoma,a strong resonant signal with corrugated morphology was detected,which indicated the presence of lipids.A strong signal was detected in the tympanic membrane with chronic otitis media using TPEF/SHG at 817 nm,which indicated collagen enrichment.The CARS and TPEF/SHG images were in accordance with the histology results.Conclusion:These results suggest the need to develop a novel CARS microendoscope that can be used in combination with TPEF/SHG to distinguish cholesteatoma from inflammatory tissues. | Jing Zou Antti Isomaki Timo Hirvonen Antti Aarnisalo Jussi Jero Ilmari Pyykko | 2016 | Journal of Otology2016,11,3: | 4 |
| 2 | Human adipose-derived mesenchymal stem cells accelerate decellularized neobladder regeneration显示文摘Decellularized natural bladder matrices(neobladders)represent an exciting means to regenerate the bladder following bladder cancer-associated cystectomy.In this study,we compare the evolution of decellularized matrices with recellularized matrices by seeding it with human adiposederived mesenchymal stem cells(ADSC)after implantation following partial cystectomy in rats.We discovered significant anatomical differences since 10 days after neobladder implantation with the ADSC-containing matrices promoting a significant recovery of mature p63-and cytokeratin 7-positive urothelium.We also discovered significantly induced expression of the vimentin mesoderm marker in the submucosal layer in ADSC-seeded matrices.Interestingly,we found a higher expression of smooth muscle actin in transversal and longitudinal smooth muscle layers with ADSC-seeded matrices.Furthermore,ADSC also showed increased vascularization and nerve innervation of the neobladder as determined by the distribution of CD31 and S100b reactivity,respectively.We believe that ADSC and their paracrine-acting pro-regenerative secretome within decellularized matrices represent an efficient bladder substitution strategy;however,we require a fuller understanding of the mechanisms involved before clinical studies can begin. | Victoria Moreno-Manzano Maravillas Mellado-Lo´pez Maria Jose Morera-Esteve Ana Alastrue-Agudo Viviana Bisbal-Velasco Jero´nimo Forteza-Vila A´ngel Serrano-Aroca Ce´sar David Vera-Donoso | 2020 | Regenerative Biomaterials2020,7,2: | 2 |
