1Analysis of risk factors for polypoid lesions of gallbladder among health examinees显示文摘AIM: To investigate the prevalence and risk factors of polypoid lesions of gallbladder (PLG) among the health examinees in the Shanghai region, China. METHODS: A total of 11 816 subjects who underwent health examinations in our hospital between August 2010 and February 2011 were analyzed retrospectively. Among them, there were 7174 men and 4642 women. PLG was diagnosed by the real-time ultrasonography. Those with the body mass index (BMI) ≥ 28 were considered to be obese. Blood biochemical indices were detected with the fully automatic biochemical analyzer and hepatitis B surface antigen (HBsAg) was tested by the automated enzyme immunoassay. The correlations between the prevalence of PLG and age, sex, BMI, serum cholesterol (T-Cho), triglycerides (TG),blood sugar, HBsAg, high-density lipoprotein (HDL-C), low-density lipoprotein (LDL-C), gallstone and fatty liver were investigated. After univariate analysis of 11 variables, stepwise logistic regression analysis was performed to explore the risk factors of PLG. RESULTS: There was a significant difference in sex, T-Cho, HBsAg, HDL-C, LDL-C and fatty liver between the PLG-positive group and the PLG-negative group (332/163 vs 6842/4479, P = 0.003; 22/473 vs 295/11 026, P =0.013; 92/403 vs 993/10 328, P = 0.001; 47/448 vs 332/10 989, P = 0.001; 32/463 vs 381/10 940, P = 0.001; 83/412 vs 3260/8061, P = 0.001). No significant difference was found in the age, BMI, TG, blood sugar and gallstone between the two groups (47.3 ± 26 vs 45.1 ± 33, P = 0.173; 59/436 vs 1097/10 224, P = 0.102; 52/443 vs 982/10 339, P = 0.158; 17/478 vs 295/11 026, P = 0.26; 24/471 vs 395/10 926, P = 0.109). Logistic regression analysis showed that the sex, HBsAg and HDL-C were independent risk factors for the development of PLG in a descending order of HDL-C > HBsAg > sex. 2Upreguiation of human telomerase reverse transcriptase mRNA expression by in vitro transfection of hepatitis B virus X gene into human hepatocarcinoma and cholangiocarcinoma cells显示文摘AIM: To study the changes of human telomerase reverse transcriptase (hTERT) mRNA expression in human hepatocarcinoma cell lines (HepG2) and cholangiocarcinoma cell lines (QBC939) after HBx gene transfection and to illustrate the significance of transcriptional regulation of hTERT gene by HBx gene in the carcinogenesis.METHODS: HepG2 and QBC939 cell lines were cultured and co-transfected with eukaryotic expression vector containing the HBx coding region and cloning vector containing enhanced green fluorescent protein (EGFP) coding sequence using lipid-mediated gene transduction technique. Thirty-six hours after transfection, EGFP expression in cells was used as the indicator of successful transfection. Flow cytometry was performed to determine the transfection efficiency.Cells were harvested and total RNA was extracted using TRIzol() reagent. The expression of hTERT mRNA in HepG2and QBC939 cell lines was assayed by reverse transcriptionpolymerase chain reaction. The expression of HBx protein in both cell lines was detected by immunocytochemical staining and Western blotting.RESULTS: Flow cytometry showed that the transfection efficiency was 46.4% in HepG2 cells and 29.6% in QBC939cells for both HBx gene expression vector and blank vector. The expression of hTERT mRNA was meaningfully increased in HepG2 and QBC939 cell lines when transfected with HBx gene expression vector compared to those transfected with OPTI-MEM medium and blank vector.Immunocytochemical staining and Western blotting revealed HBx protein expression in HepG2 and QBC939cells only when transfected with HBx gene.CONCLUSION: HBx gene transfection can upregulate the transcriptional expression of hTERT mRNA. 3Expediting redox kinetics of sulfur species by atomic-scale electrocatalysts in lithium–sulfur batteries显示文摘Lithium–sulfur(Li–S)batteries have extremely high theoretical energy density that make them as promising systems toward vast practical applications.Expediting redox kinetics of sulfur species is a decisive task to break the kinetic limitation of insulating lithium sulfide/disulfide precipitation/dissolution.Herein,we proposed a porphyrinderived atomic electrocatalyst to exert atomic-efficient electrocatalytic effects on polysulfide intermediates.Quantifying electrocatalytic efficiency of liquid/solid conversion through a potentiostatic intermittent titration technique measurement presents a kinetic understanding of specific phase evolutions imparted by the atomic electrocatalyst.Benefiting from atomically dispersed“lithiophilic”and“sulfiphilic”sites on conductive substrates,the finely designed atomic electrocatalyst endows Li–S cells with remarkable cycling stablity(cyclic decay rate of 0.10%in 300 cycles),excellent rate capability(1035 mAh g−1 at 2 C),and impressive areal capacity(10.9 mAh cm−2 at a sulfur loading of 11.3 mg cm−2).The present work expands atomic electrocatalysts to the Li–S chemistry,deepens kinetic understanding of sulfur species evolution,and encourages application of emerging electrocatalysis in other multielectron/multiphase reaction energy systems.Bo-Quan Li Long Kong Chang-Xin Zhao Qi Jin Xiao Chen Hong-Jie Peng Jin-Lei Qin Jin-Xiu Chen Hong Yuan Qiang Zhang Jia-Qi Huang 2019InfoMat2019,1,4:18

6Transjugular intrahepatic portosystemic shunt combined with esophagogastric variceal embolization in the treatment of a large gastrorenal shunt显示文摘AIM: To evaluate the efficacy and safety of transjugular intrahepatic portosystemic shunt(TIPS) combined with stomach and esophageal variceal embolization(SEVE) in cirrhotic patients with a large gastrorenal vessel shunt(GRVS).METHODS: Eighty-one cirrhotic patients with gastric variceal bleeding(GVB) associated with a GRVS were enrolled in the study and accepted TIPS combined with SEVE(TIPS + SEVE), by which portosystemic pressuregradient(PPG), biochemical, TIPS-related complications, shunt dysfunction, rebleeding, and death were evaluated. RESULTS: The PPGs before TIPS were greater than 12 mmH g in 81 patients. TIPS + SEVE treatment caused a significant decrease in PPG(from 37.97 ± 6.36 mmH g to 28.15 ± 6.52 mm Hg, t = 19.22, P < 0.001). The percentage of reduction in PPG was greater than 20%from baseline. There were no significant differences in albumin, alanine aminotransferase, aspartate aminotransferase, bilirubin, prothrombin time, or Child-Pugh score before and after operation. In all patients, rebleeding rates were 3%, 6%, 12%, 18%, and 18% at 1,3, 6, 12, and 18 mo, respectively. Five patients(6.2%)were diagnosed as having hepatic encephalopathy. The rates of shunt dysfunction were 0%, 4%, 9%, 26%,and 26%, at 1, 3, 6, 12, and 18 mo, respectively. 8Psychological status and diabetes-related distress of Chinese type 1 diabetes patients in Jiangsu province, China显示文摘We sought to assess the psychological status and diabetes-related distress of Chinese type 1 diabetes patients in Jiangsu province,China.According to the World Health Organization criteria,42 patients with type 1 diabetes were enrolled in the study and assessed with the Self-Rating Anxiety Scale(SAS),Self-Rating Depression Scale(SDS)and Diabetes Distress Scale(DDS).All data were tabulated and statistical analyses were performed.The study summarized cases of 42 patients with type 1 diabetes,including 17 males and 25 females with a mean age of 23±12 years and a mean duration of diabetes of 7±6 years.Compared to the Chinese normative data,the SAS standard score was significantly higher,whereas SDS standard score had no statistical significance.The SAS standard score was most highly correlated with diabetes duration(γ=0.547,P=0.011).Additionally,19.5%of the patients had moderate or even severe diabetes-related distress and 21.4%had moderate or even severe emotional burden while 26.2%had regimen-related distress.Multiple stepwise regression analysis showed that the mean correlation between DDS and the four domains was high,particularly the emotional burden domain(estimatedβ=0.363,P<0.001)and regimen-related distress domain(estimatedβ=0.356,P<0.001).The correlation between SAS and DDS was positive(estimatedβ=0.039,P=0.027).In conclusion,the results showed the importance of psychological aspects in Chinese individuals with type 1 diabetes.Screening and treatment of psychological aspects may result in better adherence and increased quality of life for patients with diabetes.Lulu Kong Yun Cai Gang Mei Rong Gu Xiaojiao Zhang Yao Qin Ya Cai Yan Li Hongwen Zhou Mei Zhang Tao Yang 2013The Journal of Biomedical Research2013,27,5:9
9Effects of Accelerated Aging on Physiological and Biochemical Characteristics of Waxy and Non-waxy Wheat Seeds显示文摘Experiments were conducted to evaluate the physiological and the biochemical characteristics of waxy wheat seeds under accelerated aging conditions. Five waxy wheat lines, which were Waxy 1, Waxy 4, Waxy 8, Waxy 9, and Waxy 15; and five non-waxy wheat lines: S-39, 04J89, Jan-81, Ⅲ42-4, and Ⅲ10 were studied. The seeds were subjected to accelerated aging at 40℃, 45℃, 50℃, 55℃, and 60℃, and 90% relative humidity for 0, 2, 4, 6, and 8 days, respectively. The results showed a gradual increase in conductivity and decrease in germination rate during accelerated aging. SOD, POD and CAT activities increased at low-grade treatment, but decreased at severe treatment. On the other hand, the soluble protein content decreased at 45℃, but successively increased, then decreased 50℃. 10Ketogenic diet alleviates colitis by reduction of colonic group 3 innate lymphoid cells through altering gut microbiome显示文摘Accumulating evidence suggests that ketogenic diets(KDs)mediate the rise of circulating ketone bodies and exert a potential antiinflammatory effect;however,the consequences of this unique diet on colitis remain unknown.We performed a series of systematic studies using a dextran sulfate sodium(DSS)animal model of inflammatory colitis.Animals were fed with a KD,low-carbohydrate diet(LCD),or normal diet(ND).Germ-free mice were utilized in validation experiments.Colon tissues were analyzed by transcriptome sequencing,RT2 profiler PCR array,histopathology,and immunofluorescence.Serum samples were analyzed by metabolic assay kit.Fecal samples were analyzed by 16S rRNA gene sequencing,liquid chromatography-mass spectrometry and gas chromatography-mass spectrometry.We observed that KD alleviated colitis by altering the gut microbiota and metabolites in a manner distinct from LCD.Quantitative diet experiments confirmed the unique impact of KD relative to LCD with a reproducible increase in Akkermansia,whereas the opposite was observed for Escherichia/Shigella.After colitis induction,the KD protected intestinal barrier function,and reduced the production of R0Ryt+CD3_group 3 innate lymphoid cells(ILC3s)and related inflammatory cytokines(IL-17a,IL-18,IL-22,Ccl4).Finally,fecal microbiota transplantation into germ-free mice revealed that the KD-mediated colitis inhibition and ILC3 regulation were dependent on the modification of gut microbiota.Taken together,our study presents a global view of microbiome-metabolomics changes that occur during KD colitis treatment,and identifies the regulation of gut microbiome and ILC3s as novel targets involving in IBD dietary therapy.Cheng Kong Xuebing Yan Yongqiang Liu Linsheng Huang Yefei Zhu Jide He Renyuan Gao Matthew F.Kalady Ajay Goel Huanlong Qin Yanlei Ma 2021Signal Transduction and Targeted Therapy2021,6,5:8

12Alterations in intestinal microbiota of colorectal cancer patients receiving radical surgery combined with adjuvant CapeOx therapy显示文摘An intricate relationship exists and interactions occur between gut microbiota and colorectal cancer(CRC).Radical surgery combined with adjuvant chemotherapy(AC) serves as the mainstream therapeutic scheme for most CRC patients.The current research was conducted to assess the effect of surgery or chemotherapy on gut microbiota.Forty-three CRC patients who received radical surgery and AC were enrolled.Fecal samples were collected preoperatively,postoperatively,and after the first to fifth cycles of postoperative chemotherapy.The microbial community of each sample was analyzed using high throughput 16S rRNA amplicon sequencing.Compared with preoperative samples,fecal samples collected postoperatively exhibited a significant decrease of obligate anaerobes,tumor-related bacteria,and butyric acid-producing bacteria.However,a significant increase of some conditional pathogens was observed.In addition,the AC regimen(CapeOx) was found to alter intestinal microbiota dramatically.In particular,several changes were observed after chemotherapy including an increase of pathogenic bacteria,the 'rebound effect' of chemotherapy-adapted bacteria,the shift of lactate-utilizing microbiota from Veillonella to Butyricimonas and Butyricicoccus,as well as the decrease of probiotics.Both radical surgery and CapeOx chemotherapy exert a non-negligible effect on the gut microbiota of CRC patients.Microbiota-based intervention may be beneficial for patients during postoperative clinical management.Cheng Kong Renyuan Gao Xuebing Yan Linsheng Huang Jide He Hao Li Jie You Huanlong Qin 2019Science China(Life Sciences)2019,62,9:7
13Pathogenic effects of biofilm with chronic pseudomonas aeruginosa lung infection in rats显示文摘Objective:To establish an animal model of P.aeruginosa biofilm associated with chronic pulmonary infection and investigate the pathogenic effects of biofilm.Methods:Experiments in vitro,measuring the MICS,MBCS of levofloxacin(LFX),ceftazidime(CAZ)in PAO579 in alginate beads and planktonic PAO579.Rats were challenged with 0.1 ml of PAO579(109CFU/ml)in alginate beads or 0.1 ml of planktonic PAO579(109 CFU/ml),3,7,14 days after challenging,bacteriological,pathological features were observed.Results:The MICS,MBCS of LFX,CAZ in PAO579 in alginate beads were higher than those in planktonic PAO579 in vitro.CFU/lung in alginate beads group was significantly higher than that in planktonic bacteria group(P = 0.002,P = 0.004,P = 0.002,respectively);macroscopic lung pathology and the inflammation in alginate beads group were significantly more severe compared to those in planktonic bacteria group in vivo.Conclusion:P.aeruginosa biofilm protected bacterium from killing of antibiotics and might mediate the host immune damage in the lung tissue and made bacterium evade the host immune defense.Ping Yan Yiqiang Chen Zhijun Song Hong Wu Jinliang Kong Xuejun Qin 2008Journal of Nanjing Medical University2008,22,1:7
14Genome editing mediated by SpCas9 variants with broad non-canonical PAM compatibility in plants显示文摘Streptococcus pyogenes Cas9(SpCas9)is the most widely used genome editing tool in plants.The editing induced by SpCas9 strictly requires a canonical NGG protospacer-adjacent motif(PAM),significantly limiting its scope of application.Recently,five SpCas9 variants,SpCas9-NRRH,SpCas9-NRCH,SpCas9-NRTH,SpG,and SPRY,were developed to recognize non-canonical PAMs in human cells.In this study,these variants were engineered for plant genome editing,and their targeted mutagenesis capabilities were comprehensively examined at various canonical and non-canonical PAM sites in rice(Oryza sativa)by stable transformation.Moreover,both cytosine base editors using a rat APOBEC1 or a human APO-BEC3a and adenine base editors using a directly evolved highly compatible TadA*-8e deaminase were developed from these SpCas9 variants.Our results demonstrated that the developed SpCas9 variantsbased base editors readily generated conversions between C.G and T.A in the target sites with noncanonical PAMs in transgenic rice lines.Collectively,the toolbox developed in this study substantially expands the scope of SpCas9-mediated genome editing and will greatly facilitate gene disruption and precise editing in plants.Juan Li Rongfang Xu Ruiying Qin Xiaoshuang Liu Fanna Kong Pengcheng Wei 2021Molecular Plant2021,14,2:6
15Characteristics of vertical ozone distribution in the lower troposphere in the Yangtze River Delta at Lin'an in the spring of 2001显示文摘We analyzed vertical distributions of ozone (O3) in the lower troposphere (< 5 km above ground) at Lin’an (119.75°E, 30.30°N), Zhejiang Province using electrochemical concen- tration cell (ECC) ozonesonde data obtained from February 21 to April 13, 2001. The results showed that the vertical O3 distributions are controlled by metrological conditions and the char- acteristics of O3 profiles are related to those of wet bulb potential temperature and wind field. O3 below 2 km showed that the strongest variability and enhanced O3 mixing ratios were associated with easterly winds that blow pollutants from the upwind source region of the Yangtze River Delta (YRD) region. Vertical O3 profiles below 5 km can be grouped into 5 categories: (1) peak mixing ratio type, (2) well-mixed type, (3) layered-structure type, (4) episodic pollution type and (5) alti- tudinal increasing type. Vertical distributions of O3 affected by regional transport of polluted air masses were investigated. Transport of polluted air from high latitudes of northern China, ac- companying subsiding motion of air and stagnant atmospheric conditions are important factors that lead to high mixing ratios of O3 at Lin’an. The stagnant atmospheric conditions associated with a continental high pressure system and pollution plume transported from the YRD and cen- tral-eastern China also lead to regional accumulation of O3 and high O3 mixing ratio at Lin’an. 16Sediment transportation and bed morphology reshaping in Yellow River Delta显示文摘The Yellow River Delta supports the ecological function as a typical estuarine foreshore wetland. The wetland area is changing greatly every year because of sediment deposition and erosion, which influences the wetland function tremendously. Application of environmental fluid dynamics code (EFDC) to the Yellow River Delta is on the basis of the mobile bed dynamic model and wetting-drying process. Careful calibration is carried out for the numerical model which is set up for the Yellow River Delta, the sediment transport process of the model is compatible to the Yellow River situation. The simulated bed elevation by considering the sediment deposition in the Mouth is particularly focused on, the numerical results are in agreement with the measured bed morphology within 1992 2000. Simulation in this paper indicates that most of the sediment deposited just out of the Mouth which makes the mouth move forward into the sea 2.5 km per year. 17Centro-parietal N200: An event-related potential component specific to Chinese visual word recognition显示文摘Almost all written languages currently used in the world are alphabetic or phonologically-based scripts (e.g., English) whose word recognition involves discrimination of one-dimensional linear combinations of several dozens of letters or phonological units, except written Chinese whose vocabulary consists of a large number of block characters, the analysis of which relies on the extraction of 2-dimensional form information at multiple levels including radical, character, and multiple-character word. Whether the dramatic contrast between the 2 types of orthography leads to distinctive mental processes is a fundamental question in psycholinguistic research unanswered in the past three decades. Using event-related potentials and visual lexical decision tasks, we asked native Chinese speakers to discriminate between real Chinese words and pseudoor non-words and observed a widespread negative deflection with centro-parietal focus elicited 200 ms post-stimulus onset. This N200 response showed a clear and large amplitude enhancement upon word repetition and seems to be specific to Chinese as no similar effects had been reported in word recognition studies involving alphabetic scripts under similar experimental conditions. Further evidence showed that this N200 could not be attributed to non-linguistic sensori-perceptual processes, nor phonological or semantic processes, but likely reflects very early identification of the orthography of individual words involving extensive and higher-level visual analysis. Recently the first author proposed a meaning-spelling theory of written Chinese vocabulary proposing that the Chinese and alphabetic scripts are the only 2 possible logical types of mature human writing systems, and that the former is more thoroughly a visual language compared with the latter and thus shall emphasize more of visual processing. 18PREDICTION OF PROJECTILE PENETRATION AND PERFORATION BY FINITE CAVITY EXPANSION METHOD WITH THE FREE-SURFACE EFFECT显示文摘With a target treated as the incompressible Tresca and Mohr-Coulomb material,by assuming that cavity expansion produces plastic-elastic and plastic-cracked-elastic response region, the decay function for the free-surface effect is constructed for metal and geological targets, respectively. The forcing function for oblique penetration and perforation is obtained by multiplying the forcing function derived on the basis of infinite target assumption with the decay function. Then the projectile is modeled with an explicit transient dynamic finite element code and the target is represented by the forcing function as the pressure boundary condition.This methodology eliminates discretizing the target as well as the need for a complex contact algorithm and is implemented in ABAQUS explicit solver via the user subroutine VDLOAD. It is found that the free-surface effect must be considered in terms of the projectile deformation,residual velocity, projectile trajectory, ricochet limits and critical reverse velocity. 19Development and experimental validation of a novel hybrid powertrain with dual planetary gear sets for transit bus applications显示文摘Given that energy conservation and environmental protection are two important goals for the automotive industry, the application of a hybrid electric powertrain can improve vehicle energy efficiency while decreasing fuel consumption and engine emissions. Planetary gear-based power-split hybrid powertrains have become widely used in passenger vehicles, but remain rarely employed on transit buses. This study proposes a novel hybrid powertrain based on two planetary gear sets(CHS) and presents its operating principles along with development of a control strategy for the powertrain. The CHS hybrid powertrain operates in electric mode when the driving power demand is low, and changes to a hybrid electric mode according to the power-split principle of the planetary gear set. To validate the feasibility of the designed CHS hybrid powertrain, a prototype transit bus equipped with the designed hybrid powertrain system was built, and the operating characteristics of the system were analyzed through a performance test conducted on a chassis dynamometer. Compared with a conventional powertrain, the CHS hybrid powertrain can reduce fuel consumption by 39%. 20Characterization of T.aestivum-H.californicum chromosome addition lines DA2H and MA5H显示文摘In order to transfer useful genes of Hordeum californicum into common wheat(Triticum aestivum L.),the T.aestivum c.v.Chinese Spring(CS)-H.californicum amphiploid was crossed to CS,and its backcrossing and self-fertilized progenies were analyzed by morphological observation,cytological,biochemical and molecular marker techniques.Alien addition lines with two H.californicum chromosomes were identified and their genetic constitution was characterized.STS-PCR analysis using chromosome 2B specific markers indicated that chromosome H3 of H.californicum belongs to homoeologous group 2,and was thus designated 2H.SDS-PAGE showed that chromosome H2 of H.californicum belongs to homoeologous group 5,and was designated 5H.The CS-H.californicum amphiploid and the chromosome addition lines(DA2H and MA5H) identified were evaluated for powdery mildew(Erysiphe graminis f.sp.triticii) resistance in field.The preliminary results indicated that the amphiploid showed higher powdery mildew resistance than CS.However,chromosome addition lines DA2H and MA5H were highly susceptible to powdery mildew,indicating that major powdery mildew resistant genes of H.californicum should be located on chromosomes other than 2H and 5H.Fang Kong Haiyan Wang Aizhong Cao Bi Qin Jianhui Ji Suling Wang Xiu-E Wang 2008Journal of Genetics and Genomics2008,35,11:5
