| 1 | AMV和WCMV在转基因红三叶中的基因累集(英文)显示文摘随着转基因技术的发展及其在植物育种中的应用,出现了一大批表现优良、经济效益高的转基因植物,但生产实践要求一种植物同时具有多个优良的转基因性状。目前解决这一问题的方法主要是基因累集(Genepyramiding),就是将多个基因集中到同一植株,可以将已经转入1个基因的植株做原材料,用农杆菌介导或基因枪等方法将另一基因转进去。也可以采用人工杂交,在不同转基因植株之间进行人工授粉,从后代中筛选出所需要的基因型。本研究对抗苜蓿花叶病毒(AMV)的转基因红三叶和抗白三叶花叶病毒(WCMV)的转基因红三叶进行了人工杂交,以期获得兼有2个基因、同时抵抗2种病毒的新植株,为下一步的育种打好基础。几乎所有的杂交组合都结了种子,杂交一代的发芽率随杂交亲本转基因拷贝数的不同而有所变化,只有1个转基因拷贝的亲本其杂种发芽率最高。对所有幼苗先用PCR进行检测,从中挑出较理想的基因型作进一步的分子生物学分析。Southern和Northern印迹分析结果表明,有些杂交一代的转入基因多于1个拷贝,有一部分同时兼有2种抗病基因,并且得到了表达。为进一步检测其抗病性,对它们进行了温室接种和田间评估,结果显示,尽管有的个体Northern分析呈阳性,但在田间仍然对病毒敏感,不过其感病程度都比对照有显著降低。 | 赵桂琴 Roger KALLA | 2003 | 草业学报2003，12，2： | 11 |
| 2 | 铁苋菜叶提取物抑制大鼠实验性心肌缺血的生物化学和组织病理学研究(英文)显示文摘目的:研究富含类黄酮的热带铁苋菜叶的甲醇提取物抗心肌缺血的作用。方法:用异丙肾上腺素诱导大鼠心肌缺血。成年Wistar白化大鼠经热带铁苋菜提取物(100和200mg/kg大鼠体质量,口服)30d预处理后连续2d注射异丙肾上腺素(85mg/kg大鼠体质量,皮下注射)。第2次注射异丙肾上腺素后12h,肌肉注射氯胺酮(24mg/kg大鼠体质量)处死大鼠。检测血浆总胆固醇、三酰甘油、高密度脂蛋白胆固醇、低密度脂蛋白胆固醇、极低密度脂蛋白胆固醇、游离脂肪酸、磷脂质以及心肌脂质、肝脏脂质的含量,以衡量热带铁苋菜提取物的抗心肌缺血作用,同时观察大鼠心脏组织的组织病理学变化。结果:与模型组大鼠相比,所有热带铁苋菜提取物给药组(100和200mg/kg)大鼠的血浆脂质水平改善,且与正常组相比无明显变化。模型组大鼠心脏组织病理学结果显示心肌纤维缺失,广泛性心内膜下坏死,炎症细胞浸润,心肌细胞明显水肿,空泡变性。而经200mg/kg热带铁苋菜提取物预处理的大鼠心脏组织观察到较完整的心肌结构,心肌纤维可见,没有炎症细胞浸润、细胞水肿和坏死现象。结论:本研究的生化指标及组织学结果表明热带铁苋菜可抑制异丙肾上腺素诱导的大鼠心肌缺血。 | Kalla C.Mouli Tartte Vijaya Sirpurkar Dattatreya Rao | 2012 | 中西医结合学报2012，10，7： | 3 |
