Hypothetical mode of action of earthworm extract with hepatoprotective and antioxidant properties显示文摘The hepatoprotective potential of earthworm extract (EE) (Lampito mauritii, Kinberg) was evaluated against paracetamol-induced liver injury in Wistar albino rat, in comparison with silymarin, the standard hepatoprotective drug. We observed a reduction in liver antioxidants, such as glutathione (GSH), superoxide dismutase (SOD), glutathione peroxidase (GPx), and catalase (CAT) and in serum total protein, and an increase in serum alkaline phosphatase (ALP), serum aspertate aminotranferase (AST), serum alanine aminotranferase (ALT), bilirubin and liver thiobarbituric acid reactive substances (TBARS) due to liver injury in the paracetamol-administered rats (2 g/kg). On the contrary, increased activities of liver GSH, SOD, GPx, CAT and serum total protein level, and decrease in the contents of serum ALP, AST, ALT, bilirubin and liver TBARS were observed in rats administered with different doses of EE (100, 200 and 300 mg/kg), which are similar to the activities of hepato-protective drug silymarin (150 mg/kg). The mode of action of EE as evidenced by the above parameters may suggest that EE, on the one hand, prevents the formation of the reactive oxygen groups, or scavenges these groups, thereby preventing the damage on the hepatic cells, and, on the other hand, modulates the genes responsible for synthesis of antioxidant enzymes such as GPx, CAT and SOD in liver tissue and decreases the serum enzymatic activities such as ALP, AST and ALT. | Mariappan BALAMURUGAN Kasi PARTHASARATHI Lalpet Souri RANGANATHAN Edwin L. | 2 | Preliminary investigation on the antibacterial activity of mango(Mangifera indica L:Anacardiaceae) seed kernel显示文摘Objective:To evaluate the antibacterial activity of the methanolic extract of mango(Mangifera indica L) seed kernel. Methods:Chokanan mango seed kernel and seed kernels from assorted mango varieties were collected, cleaned, dried and powered.Crude methanolic extracts of mango seed kernel were analyzed for the phytochemical constituents. The free radical scavenging activity was determined by 2,2-Diphenyl-1-picrylhydrazyl(DPPH) assay.Antibacterial activity was evaluated by disc diffusion assay with three medically important bacterial pathogens such as methicillin resistant Staphylococcus aureus(S.aureus)(MRSA),Escherichia coli(E.coli) and Vibrio vulnificus(V.vulnificus).Results:Qualitative phytochemical analysis indicated the presence of important phytochemical compounds such as glycosides, saponins, flavanoids, tannins and alkaloids. There was no significant difference in the phytochemical content between the single and assorted mango seed kernels. However, the free radical scavenging study indicated that the assorted mango kernels showed slighdy higher activity than the single species(P<0.05). The crude methanolic extract of mango seed kernel at a concentration of 100 mg/mL is found to have potential antimicrobial activity against MRSA and E.coli compared to V.vulnificus. Study on the antibacterial activity also indicated that there was no significant difference in the antibacterial activity of the single and assorted mango seed kernel extracts. Conclusions:The present study conclusively demonstrates the free radical scavenging activity and antibacterial activities of mango seed kernel. In addition, the results also indicated that there is no significant difference in the phytochemical content and biological activity of mango kernels from single and assorted mango varieties. | Jasminder Kaur Xavier Rathinam Marimuthu Kasi Khoo Miew Leng Rajasekaran Ayyalu Kathiresan S Sreeramanan Subramaniam | 2010 | Asian Pacific Journal of Tropical Medicine2010,3,9: | 11
A comparative study on the antioxidant activity of methanolic leaf extracts of Ficus religiosa L,Chromolaena odorata(L.) King & Rabinson,Cynodon dactylon(L.) Pers.and Tridax procumbens L.显示文摘Objective:To compare the antioxidative effects of the methanolic leaf extracts of Ficus religiosa (F.religiosa),Chromolaena odorata(C.odorata),Cynodon dactylon(C.dactylon) and Trida,x procumbens(T.procumbens) as well as the contents of antioxidants in the extracts. Methods: Total phenol and total flavanoid contents were measured according to the standard procedures. The total antioxidant capacity was determined using the phosphomolybdenum method.Reducing power was determined by the potassium ferrieyanide reducing method. The free radical scavenging activity was measured by 2,2’-Diphenyl-l-picrylhydrazyl(DPPH) assay. Results: Quantitative phytochemical analysis of total phenol content showed that C.odorata had the highest content of phenolic compounds significantly followed by F.religiosa,T.procumbens and C.dactylon.As for the total flavanoids content,F.religiosa had the highest content, followed by C.odorata,T.procumbens and C.daxtylon.Study on the total antioxidant capacity revealed that F.religiosa,C.dactylon and C.odorata showed higher total antioxidant capacity.T.procumbens showed the lowest capacity.Meanwhile,T.procumbens and C.odorata have the highest reducing power activity followed by F.religiosa and C.dactylon.The results of DPPH radical scavenging activity indicated that T.procumbens induced the largest elevation as the concentration of its extract increased, followed by C.odorata and F.religiosa and C.dactylon.Conclusions:The present study demonstrates the antioxidative capacity of all the four plant species.Of all the plants,C.odorata,a perennial weed plant showed potentially a high antioxidant activity, with higher phenolic and flavonoids contents. The data suggest that C.odorata can be best utilized in developing bioantioxidants. | Melinda Krishanti P Xavier Rathinam Marimuthu Kasi Diwakar Ayyalu Ramanathan Surash Kathiresan Sadasivam Sreeramanan Subramaniam | 2010 | Asian Pacific Journal of Tropical Medicine2010,3,5: | 6
In vivo imaging of structural, metabolic and functional brain changes in glaucoma显示文摘Glaucoma, the world's leading cause of irreversible blindness, is a condition for which elevated intraocular pressure is currently the only modifiable risk factor. However, the disorder can continue to progress even at reduced intraocular pressure. This indicates additional key factors that contribute to the etiopathogenesis. There has been a growing amount of literature suggesting glaucoma as a neurodegenerative disease of the visual system. However, it remains debatable whether the observed pathophysiological conditions are causes or consequences. This review summarizes recent in vivo imaging studies that helped advance the understanding of early glaucoma involvements and disease progression in the brains of humans and experimental animal models. In particular, we focused on the non-invasive detection of early structural and functional brain changes before substantial clinical visual field loss in glaucoma patients; the eye-brain interactions across disease severity; the metabolic changes occurring in the brain's visual system in glaucoma; and, the widespread brain involvements beyond the visual pathway as well as the potential behavioral relevance. If the mechanisms of glaucomatous brain changes are reliably identified, novel neurotherapeutics that target parameters beyond intraocular pressure lowering can be the promise of the near future, which would lead to reduced prevalence of this irreversible but preventable disease. | Anisha Kasi Muneeb A.Faiq Kevin C.Chan | 2019 | Neural Regeneration Research2019,14,3: | 3
Isolation and identification of Salmonella from curry samples and its sensitivity to commercial antibiotics and aqueous extracts of Camelia sinensis(L.) and Trachyspermum ammi(L.)显示文摘Objective:To isolate Salmonella from curry samples and to evaluate the drug sensitivity of the food-borne Salmonella and its susceptibility to specific plant extracts. Methods:Salmonella was isolated from the curry samples by standard microbiological methods and was confirmed by biochemical tests. The antibiotic susceptibility test was conducted by disc diffusion method using commercially available antibiotics such as ampicillin, tetracycline, chloramphenicol, kanamycin, and penicillin. In addition, the susceptibility of the food-borne Salmonella was also evaluated against the aqueous extracts of Camelia sinensis(L.) Theaceae(tea leaves) and the Trachyspermum ammi(L.) Apiaceae(ajwain or omum seeds). Results:Out of fifty curry samples, only seven samples were identified to have Salmonella contamination. The Salmonella isolates showed a significant drug resistance pattern except for kanamycin. The plant extracts showed a considerable antibacterial activity against the isolates, indicating the presence of antimicrobial principle which can be exploited after complete pharmacological investigations. Conclusions:The present study demonstrates the occurrence of Salmonella in the curry samples, and shows significant drug resistance against most of the commercially available antibiotics, except kanamycin.Antimicrobial effect of the plant extracts against the food-bone Salmonella suggests that dietary including medicinal herbs would be one strategy to manage food borne pathogens. | Thanes Gunasegaran Xavier Rathinam Marimuthu Kasi Kathiresan Sathasivam Sasidharan Sreenivasan Sreeramanan Subramaniam | 2011 | Asian Pacific Journal of Tropical Biomedicine2011,1,4: | 3

Safety, immunogenicity, and protection provided by unadjuvanted and adjuvanted formulations of a recombinant plant-derived virus-like particle vaccine candidate for COVID-19 in nonhuman primates显示文摘Although antivirals are important tools to control severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)infection,effective vaccines are essential to control the current coronavirus disease 2019(COVID-19)pandemic.Plant-derived virus-like particle(VLP)vaccine candidates have previously demonstrated immunogenicity and efficacy against influenza. Here,we report the immunogenicity and protection induced in rhesus macaques by intramuscular injections of a VLP bearing a SARS-CoV-2 spike protein(CoVLP)vaccine candidate formulated with or without Adjuvant System 03(AS03)or cytidine-phospho-guanosine(CpG)1018.Although a single dose of the unadjuvanted CoVLP vaccine candidate stimulated humoral and cell-mediated immune responses, booster immunization(at 28 days after priming)and adjuvant administration significantly improved both responses, with higher immunogenicity and protection provided by the AS03-adjuvanted CoVLP.Fifteen micrograms of CoVLP adjuvanted with AS03 induced a polyfunctional interleukin-2(IL-2)-driven response and IL-4 expression in CD4 T cells.Animals were challenged by multiple routes(i.e.,intratracheal,intranasal,and ocular)with a total viral dose of 106 plaque-forming units of SARS-CoV-2.Lower viral replication in nasal swabs and bronchoalveolar lavage fluid(BALF)as well as fewer SARS-CoV-2-infected cells and immune cell infiltrates in the lungs concomitant with reduced levels of proinflammatory cytokines and chemotactic factors in the BALF were observed in animals immunized with the CoVLP adjuvanted with AS03.No clinical, pathologic,or virologic evidence of vaccineassociated enhanced disease was observed in vaccinated animals. The CoVLP adjuvanted with AS03 was therefore selected for vaccine development and clinical trials. | Stéphane Pillet Prabhu SArunachalam Guadalupe Andreani Nadia Golden Jane Fontenot Pyone Pyone Aye Katharina Röltgen Gabrielle Lehmicke Philipe Gobeil Charlotte Dubé Sonia Trépanier Nathalie Charland Marc-AndréD’Aoust Kasi Russell-Lodrigue Christopher Monjure Robert V.Blair Scott D.Boyd Rudolf P.Bohm Jay Rappaport François Villinger Nathalie Landry Bali Pulendran Brian J.Ward | 2022 | Cellular & Molecular Immunology2022,19,2: | 2

Alleviation of nickel toxicity in finger millet(Eleusine coracana L.)germinating seedlings by exogenous application of salicylic acid and nitric oxide显示文摘This study investigated the effect of salicylic acid(SA) and sodium nitroprusside(SNP; NO donor) on nickel(Ni) toxicity in germinating finger millet seedlings. Fourteen-day-old finger millet plants were subjected to 0.5 mmol L^(-1) Ni overload and treated with 0.2 mmol L^(-1)salicylic acid and 0.2 mmol L^(-1)sodium nitroprusside to lessen the toxic effect of Ni.The Ni overload led to high accumulation in the roots of growing plants compared to shoots, causing oxidative stress. It further reduced root and shoot length, dry mass, total chlorophyll, and mineral content. Exogenous addition of either 0.2 mmol L^(-1)SA or0.2 mmol L^(-1)SNP reduced the toxic effect of Ni, and supplementation with both SA and SNP significantly reduced the toxic effect of Ni and increased root and shoot length, chlorophyll content, dry mass, and mineral concentration in Ni-treated plants. The results show that oxidative stress can be triggered in finger millet plants by Ni stress by induction of lipoxygenase activity, increase in levels of proline, O_2^(·-) radical, MDA, and H_2O_2, and reduction in the activity of antioxidant enzymes such as CAT, SOD, and APX in shoots and roots. Exogenous application of SA or SNP, specifically the combination of SA + SNP,protects finger millet plants from oxidative stress observed under Ni treatment. | Kasi Viswanath Kotapati Bhagath Kumar Palaka Dinakara Rao Ampasala | 2017 | The Crop Journal2017,5,3: | 1
