2Therapeutic temperature modulation is associated with pulmonary complications in patients with severe traumatic brain injury显示文摘AIM: To examine complications associated with the use of therapeutic temperature modulation(mild hypothermia and normothermia) in patients with severe traumatic brain injury(TBI). METHODS: One hundred and fourteen charts were reviewed. Inclusion criteria were: severe TBI with Glasgow Coma Scale(GCS) < 9, intensive care unit(ICU) stay > 24 h and non-penetrating TBI. Patients were divided into two cohorts: the treatment group received therapeutic temperature modulation(TTM) with continuous surface cooling and indwelling bladder temperature probes. The control group received standard treatment with intermittent acetaminophen for fever. Information regarding complications during the time in the ICU was collected as follows: Pneumonia was identified using a combination of clinical and laboratory data. Pulmonary embolism, pneumothorax and deep venous thrombosis were identified based onimaging results. Cardiac arrhythmias and renal failure were extracted from the clinical documentation. acute respiratory distress syndrome and acute lung injury were determined based on chest imaging and arterial blood gas results. A logistic regression was conducted to predict hospital mortality and a multiple regression was used to assess number and type of clinical complications. RESULTS: One hundred and fourteen patients were included in the analysis(mean age = 41.4, SD = 19.1, 93 males), admitted to the Jackson Memorial Hospital Neuroscience ICU and Ryder Trauma Center(mean GCS = 4.67, range 3-9), were identified and included in the analysis. Method of injury included motor vehicle accident(n = 29), motor cycle crash(n = 220), blunt head trauma(n = 212), fall(n = 229), pedestrian hit by car(n = 216), and gunshot wound to the head(n = 27). Ethnicity was primarily Caucasian(n = 260), as well as Hispanic(n = 227) and African American(n = 223); four patients had unknown ethnicity. Patients received either TTM(43) or standard therapy(71). Within the TTM group eight patients were treated with normothermia after TBI and 35 patients were treated with hypothermia. A logistic regression predicting in hospital mortality with age, GCS, and TM demonstrated that GCS(Beta = 0.572, P < 0.01) and age(Beta =-0.029) but not temperature modulation(Beta = 0.797, ns) were significant predictors of in-hospital mortality [χ2(3) = 22.27, P < 0.01] A multiple regression predicting number of complications demonstrated that receiving TTM was the main contributor and was associated with a higher number of pulmonary complications(t =-3.425, P = 0.001). CONCLUSION: Exposure to TTM is associated with an increase in pulmonary complications. 3北极虾在不同冷藏条件下的质量变化与质量指标(英文)显示文摘采用感官评估、物理、化学、微生物分析方法,对贮藏在不同冷藏条件下的北极虾的质量变化与质量指标进行了研究。保藏方法分别为传统冰块冷却法、液态冰冷却法、盐水-冰混合冷却剂法,贮藏环境温度为-1.5℃或1.5℃。研究结果表明,液态冰冷藏法对延缓虾腐败变质的效果比传统冰块冷藏法、盐水-冰混合冷却剂冷藏法好。经过1d的贮藏后发现,无论在-1.5℃还是1.5℃的环境温度下,保藏在液态冰中虾的挥发性总氮(TVB N)都有所下降,并且在-1.5℃的环境温度下,其TVB N值在3d后才回升到刚开始贮藏时的水平,而在其它保藏方法中,虾的TVB N值始终随贮藏时间的延长而增加;除保藏在液态冰-1.5℃环境中虾的三甲胺(TMA)在贮藏的第1天有所降低以外,其它各试验组虾的TMA值均随贮藏时间的延长而增加。检测结果还表明,保藏在传统冰块及盐水-冰混合冷却剂中虾的细菌总数(TVC)增加最快,其次是保藏在液态冰中1.5℃的环境下。而保藏在液态冰中-1.5℃的环境下,虾的细菌繁殖最慢,并且在贮藏初期,产生了明显的细菌总数降低及细菌繁殖被抑制的现象。采用主要因子分析(PCA)和方差分析(ANOVA)发现,质量指标如TVB N、TMA、TVC、pH、NH3(电子鼻测量值)和感官评估结果等相互之间存在着良好的相关性。曾庆祝 Kristin Anna Thorarinsdottir Guerun Olafsdottir 2005水产学报2005,29,1:15
4Connections Between Insomnia and Cognitive Aging显示文摘Insomnia is a common sleep disorder among older adults,and a risk factor for poor physical and mental health.However,the relationship between insomnia and cognitive health is not well understood.Here,we review observational studies that have investigated whether insomnia is associated with deficits in objective cognitive performance and an increased risk of dementia,magnetic resonance imaging studies that have assessed grey matter volumes and white matter microstructure,and interventional studies that have explored whether the treatment of insomnia can improve cognitive outcomes.There are inconsistent findings regarding impaired performance in objective cognitive tests and reduced grey matter volumes,and limited,emerging,evidence that suggests that insomnia is associated with an increased risk of dementia and reduced white matter integrity.Although the interventional literature is still in its infancy,there is some indication that treatment may have an impact on vigilance.Well-powered studies examining sources of heterogeneity are warranted.Claire E.Sexton Konstantina Sykara Elissaios Karageorgiou Jenny Zitser Talita Rosa Kristine Yaffe Yue Leng 2020Neuroscience Bulletin2020,36,1:13

6Prescribing physical activity to prevent and manage gestational diabetes显示文摘Gestational diabetes mellitus(GDM)is the most prevalent metabolic disorder during pregnancy.Women diagnosed with GDM have a substantially greater risk of developing type 2 diabetes within 5-10 years after delivery,and the risk is increased by excess body weight.Uncontrolled hyperglycemia during pregnancy is potentially harmful to both mother and fetus,resulting in a greater need for Caesarian-section deliveries,delivery of larger infants with more excess body fat,a greater risk of infant death and stillbirth,and an elevated risk of infant hypoglycemia immediately after birth.Fortunately,engaging in physical activity prior to and during pregnancy may lower the risk of developing GDM.Pregnant women should also be advised how to safely increase their physical activity during pregnancy and the postpartum period.An initial approach to becoming more physically active can simply be to encourage women to incorporate more unstructured physical activity into daily living,both before and during pregnancy.Giving women an appropriate exercise prescription can encourage them to participate in physical activity safely and effectively throughout pregnancy to prevent and/or manage GDM.Engaging in 30 min of moderate intensity physical activity on most,if not all,days of the week has been adopted as a recommendation for all pregnant women.Sheri R Colberg Kristin Castorino Lois Jovanovi 2013World Journal of Diabetes2013,4,6:12
7Effective mass and Fermi surface complexity factor from ab initio band structure calculations显示文摘The effective mass is a convenient descriptor of the electronic band structure used to characterize the density of states and electron transport based on a free electron model.While effective mass is an excellent first-order descriptor in real systems,the exact value can have several definitions,each of which describe a different aspect of electron transport.Here we use Boltzmann transport calculations applied to ab initio band structures to extract a density-of-states effective mass from the Seebeck Coefficient and an inertial mass from the electrical conductivity to characterize the band structure irrespective of the exact scattering mechanism.We identify a Fermi Surface Complexity Factor:N_(v)^(*)K^(*) from the ratio of these two masses,which in simple cases depends on the number of Fermi surface pockets eN_(v)^(*) T and their anisotropy K^(*),both of which are beneficial to high thermoelectric performance as exemplified by the high values found in PbTe.The Fermi Surface Complexity factor can be used in high-throughput search of promising thermoelectric materials.Zachary M.Gibbs Francesco Ricci Guodong Li Hong Zhu Kristin Persson Gerbrand Ceder Geoffroy Hautier Anubhav Jain G.Jeffrey Snyder 2017npj Computational Materials2017,,1:11
8Complex role for the immune system in initiation and progression of pancreatic cancer显示文摘The immune system plays a complex role in the development and progression of pancreatic cancer. Inflammation can promote the formation of premalignant lesions and accelerate pancreatic cancer development. Conversely, pancreatic cancer is characterized by an immunosuppressive environment, which is thought to promote tumor progression and invasion. Here we review the current literature describing the role of the immune response in the progressive development of pancreatic cancer, with a focus on the mechanisms that drive recruitment and activation of immune cells at the tumor site, and our current understanding of the function of the immune cell types at the tumor. Recent clinical and preclinical data are reviewed, detailing the involvement of the immune response in pancreatitis and pancreatic cancer, including the role of specific cytokines and implications for disease outcome. Acute pancreatitis is characterized by a predominantly innate immune response, while chronic pancreatitis elicits an immune response that involves both innate and adaptive immune cells, and often results in profound sys-temic immune-suppression. Pancreatic adenocarcinoma is characterized by marked immune dysfunction driven by immunosuppressive cell types, tumor-promoting immune cells, and defective or absent inflammatory cells. Recent studies reveal that immune cells interact with cancer stem cells and tumor stromal cells, and these interactions have an impact on development and progression of pancreatic ductal adenocarcinoma(PDAC). 11Active learning for accelerated design of layered materials显示文摘Hetero-structures made from vertically stacked monolayers of transition metal dichalcogenides hold great potential for optoelectronic and thermoelectric devices.Discovery of the optimal layered material for specific applications necessitates the estimation of key material properties,such as electronic band structure and thermal transport coefficients.However,screening of material properties via brute force ab initio calculations of the entire material structure space exceeds the limits of current computing resources.Moreover,the functional dependence of material properties on the structures is often complicated,making simplistic statistical procedures for prediction difficult to employ without large amounts of data collection.Here,we present a Gaussian process regression model,which predicts material properties of an input hetero-structure,as well as an active learning model based on Bayesian optimization,which can efficiently discover the optimal hetero-structure using a minimal number of ab initio calculations.The electronic band gap,conduction/valence band dispersions,and thermoelectric performance are used as representative material properties for prediction and optimization.The Materials Project platform is used for electronic structure computation,while the BoltzTraP code is used to compute thermoelectric properties.Bayesian optimization is shown to significantly reduce the computational cost of discovering the optimal structure when compared with finding an optimal structure by building a regression model to predict material properties.The models can be used for predictions with respect to any material property and our software,including data preparation code based on the Python Materials Genomics(PyMatGen)library as well as python-based machine learning code,is available open source.Lindsay Bassman Pankaj Rajak Rajiv K.Kalia Aiichiro Nakano Fei Sha Jifeng Sun David J.Singh Muratahan Aykol Patrick Huck Kristin Persson Priya Vashishta 2018npj Computational Materials2018,,1:10

13A high-throughput framework for determining adsorption energies on solid surfaces显示文摘In this work,we present a high-throughput workflow for calculation of adsorption energies on solid surfaces using density functional theory.Using open-source computational tools from the Materials Project infrastructure,we automate the procedure of constructing symmetrically distinct adsorbate configurations for arbitrary slabs.These algorithms are further used to construct and run workflows in a standard,automated way such that user intervention in the simulation procedure is minimal.To validate our approach,we compare results from our workflow to previous experimental and theoretical benchmarks from the CE27 database of chemisorption energies on solid surfaces.These benchmarks also illustrate how the task of performing and managing over 200 individual density functional theory calculations may be reduced to a single submission procedure and subsequent analysis.By enabling more efficient high-throughput computations of adsorption energies,these tools will accelerate theory-guided discovery of advanced materials for applications in catalysis and surface science.Joseph H.Montoya Kristin A.Persson 2017npj Computational Materials2017,,1:7
14Stem cell application for osteoarthritis in the knee joint: A minireview显示文摘Knee osteoarthritis is a chronic, indolent disease that will affect an ever increasing number of patients, especially the elderly and the obese. It is characterized by degeneration of the cartilage substance inside the knee which leads to pain, stiffness and tenderness. By some estimations in 2030, only in the United States, this medical condition will burden 67 million people. While conventional treatments like physiotherapy or drugs offer temporary relief of clinical symptoms, restoration of normal cartilage function has been difficult to achieve. Moreover, in severe cases of knee osteoarthritis total knee replacement may be required. Total knee replacements come together with high effort and costs and are not always successful. The aim of this review is to outline the latest advances in stem cell therapy for knee osteoarthritis as well as highlight some of the advantages of stem cell therapy over traditional approaches aimed at restoration of cartilage function in the knee. In addition to the latest advances in the field, challenges associated with stem cell therapy regarding knee cartilage regeneration and chondrogenesis in vitro and in vivo are also outlined and analyzed. 15Predicting defect behavior in B2 intermetallics by merging ab initio modeling and machine learning显示文摘We present a combination of machine learning and high throughput calculations to predict the points defects behavior in binary intermetallic(A–B)compounds,using as an example systems with the cubic B2 crystal structure(with equiatomic AB stoichiometry).To the best of our knowledge,this work is the first application of machine learning-models for point defect properties.High throughput first principles density functional calculations have been employed to compute intrinsic point defect energies in 100 B2 intermetallic compounds.The systems are classified into two groups:(i)those for which the intrinsic defects are antisites for both A and B rich compositions,and(ii)those for which vacancies are the dominant defect for either or both composition ranges.The data was analyzed by machine learning-techniques using decision tree,and full and reduced multiple additive regression tree(MART)models.Among these three schemes,a reduced MART(r-MART)model using six descriptors(formation energy,minimum and difference of electron densities at the Wigner–Seitz cell boundary,atomic radius difference,maximal atomic number and maximal electronegativity)presents the highest fit(98%)and predictive(75%)accuracy.This model is used to predict the defect behavior of other B2 compounds,and it is found that 45%of the compounds considered feature vacancies as dominant defects for either A or B rich compositions(or both).The ability to predict dominant defect types is important for the modeling of thermodynamic and kinetic properties of intermetallic compounds,and the present results illustrate how this information can be derived using modern tools combining high throughput calculations and data analytics.Bharat Medasani Anthony Gamst Hong Ding Wei Chen Kristin A Persson Mark Asta Andrew Canning Maciej Haranczyk 2016npj Computational Materials2016,,1:6
16Animal models for the study of hepatitis C virus infection and replication显示文摘Hepatitis C virus (HCV) hepatitis, initially termed non-A, non-B hepatitis, has become one of the leading causes of cirrhosis and hepatocellular carcinoma worldwide. With the help of animal models, our understanding of the virus has grown substantially from the time of initial discovery. There is a paucity of available animal models for the study of HCV, mainly because of the selective susceptibility limited to humans and primates. Recent work has focused modification of animals to permit HCV entry, replication and transmission. In this review, we highlight the currently available models for the study of HCV including chimpanzees, tupaia, mouse and rat models. Discussion will include methods of model design as well as the advantages and disadvantages of each model. Particular focus is dedicated to knowledge of pathophysiologic mechanisms of HCV infection that have been elucidated through animal studies. 17Effects of cereal fiber on bowel function: A systematic review of intervention trials显示文摘AIM: To comprehensively review and quantitatively summarize results from intervention studies that examined the effects of intact cereal dietary fiber on parameters of bowel function. METHODS: A systematic literature search was conducted using Pub Med and EMBASE. Supplementary literature searches included screening reference lists from relevant studies and reviews. Eligible outcomes were stool wet and dry weight, percentage water in stools, stool frequency and consistency, and total transit time. Weighted regression analyses generated mean change(± SD) in these measures per g/d of dietary fiber. RESULTS: Sixty-five intervention studies among generally healthy populations were identified. A quantitative examination of the effects of non-wheat sources of intact cereal dietary fibers was not possible due to an insufficient number of studies. Weighted regression analyses demonstrated that each extra g/d of wheat fiber increased total stool weight by 3.7 ± 0.09 g/d(P < 0.0001; 95%CI: 3.50-3.84), dry stool weight by 0.75 ± 0.03 g/d(P < 0.0001; 95%CI: 0.69-0.82), and stool frequency by 0.004 ± 0.002 times/d(P = 0.0346; 95%CI: 0.0003-0.0078). 18Model combining pre-transplant tumor biomarkers and tumor size shows more utility in predicting hepatocellular carcinoma recurrence and survival than the BALAD models显示文摘AIM To assess the performance of BALAD, BALAD-2 and their component biomarkers in predicting outcome of hepatocellular carcinoma(HCC) patients after liver transplant.METHODS BALAD score and BALAD-2 class are derived from bilirubin, albumin, alpha-fetoprotein(AFP), Lens culinaris agglutinin-reactive AFP(AFP-L3), and des-gammacarboxyprothrombin(DCP). Pre-transplant AFP, AFP-L3 and DCP were measured in 113 patients transplanted for HCC from 2000 to 2008. Hazard ratios(HR) for recurrence and death were calculated. Univariate and multivariate regression analyses were conducted. C-statistics were used to compare biomarker-based to predictive models. RESULTS During a median follow-up of 12.2 years, 38 patients recurred and 87 died. The HRs for recurrence in patients with elevated AFP, AFP-L3, and DCP defined by BALAD cut-off values were 2.42(1.18-5.00), 1.86(0.98-3.52), and 2.83(1.42-5.61), respectively. For BALAD, the HRs for recurrence and death per unit increased score were 1.48(1.15-1.91) and 1.59(1.28-1.97). For BALAD-2, the HRs for recurrence and death per unit increased class were 1.45(1.06-1.98) and 1.38(1.09-1.76). For recurrence prediction, the combination of three biomarkers had the highest c-statistic of 0.66 vs. 0.64, 0.61, 0.53, and 0.53 for BALAD, BALAD-2, Milan, and UCSF, respectively. Similarly, for death prediction, the combination of three biomarkers had the highest c-statistic of 0.66 vs 0.65,0.61, 0.52, and 0.50 for BALAD, BALAD-2, Milan, and UCSF. A new model combining biomarkers with tumor size at the time of transplant(S-LAD) demonstrated the highest predictive capability with c-statistics of 0.71 and 0.69 for recurrence and death. 19IS6110限制性片段多态性分析标准方法的建立及其在结核分枝杆菌分子分型中的应用显示文摘目的建立IS6110限制性片段多态性分析(IS6110一RFLP)标准方法并评价该方法的分型能力。方法采用核酸提取、PCR、限制性内切酶分析、Southern杂交、琼脂糖凝胶电泳等技术,结合Gel—Proanalyzer3.1和BioNumerics(Version5.0)软件,对78株结核分枝杆菌插入序列IS6110-RFLP进行分析。结果确定标准化的IS6110-RFLP技术,包括核酸提取、PCR、限制性内切酶分析、Southern杂交、琼脂糖凝胶电泳等实验步骤及标化参数的相关数据分析软件的使用;采用该技术,将78株结核分枝杆菌分为75个不同的基因型,分别归属于11个基因簇,其中有52株归属于同一个基因簇,占菌株总数的66.7%(52/78)。结论建立标准化的IS6110-RFLP技术方案,该方法具有很强的基因分型和株水平鉴定能力,可用于结核病的病原学监测。刘敬华 Kristin Kremer Christine Pourcel Arnout Mulder 刘志广 赵秀芹 万康林 2008中华流行病学杂志2008,29,8:5
20Expression and Chromosomal Mapping of Mouse Gpx2 Gene Encoding the Gastrointestinal Form of Glutathione Peroxidase, GPX-GI显示文摘GPX-GI is a cytosolic tetrameric Se-dependent glutathione peroxidase, similar in properties to GPX-1. Unlike the almost ubiquitous GPX-1, GPX-GI is mainly expressed in the epithelium of gastrointestinal tract. GPX-GI contributes to at least fifty percent of GPX activity in rodent small intestmal epithelium. The total GPX activity consists of at least 70% of selenium-dependent GPX activity in this compartment.By analyzing a panel of mouse mterspecies DNA from the Jackson Laboratory’s backcross resource,we mapped Gpx2 gene to mouse chromosome 12 between D12Mit4 and D12Mit5, near the Ccs1 locus which contains a colon cancer susceptibility gene. A pseudogene, Gpx2-ps is mapped to mouse chromosome 7.Comparison of Gpx2 gene expression in three pairs of C57BL/6Ha and ICR/Ha mice which are respectively resistant and sensitive to dimethylhydrazine-induced colon cancer, we found a higher Gpx2 mRNA level in C57BL/6Ha colon than ICR/Ha colon. Interestingly, a lower level of GPX activity is found in the resistant strain of mice. Because GPX-1 has three times higher specific activity than GPX GI, our data suggest that the decreased GPX activity may result from a higher level of Gpx2 gene expression in those cells co-express GPx1FONG-FONG CHU R.
