Alterations in metastatic properties of hepatocellular carcinoma cell following H-ras oncogene transfection显示文摘AIM To demonstrate the relationship betweenH-ras oncogene and hepatocellular carcinoma(HCC) metastasis.METHODS Activated H-ras oncogene wastransfected into SMMC 7721, a cell line derivedfrom human HCC, by calcium phosphatetransfection method. Some metastasis-relatedparameters were detected in vitro, includingadhesion assay, migration assay, expression ofcollagenase ⅣV (c ⅣV ase) and epidermal growthfactor receptor (EGFR).RESULTS The abilities of H-ras-transfected cellclones in adhesion to laminin (LN) or fibronectin(FN), migration, c Ⅳ ase secretion increasedmarkedly, and the expression of EGFR elevatedmoderately. More importantly, these alterationswere consistent positively with the expressionof p21, the protein product of H-ras oncogene.CONCLUSION H-ras oncogene could inducethe metastatic phenotype of HCC cell in vitro toraise its metastatic potential. | Qing Wang~1 Zhi Ying Lin~2 Xiao Li Feng~3 ~1Department of Microbiology,Medical Center of Fudan University.the former Shanghai Medical University,Shanghai 200032,China ~2Liver Cancer Institute,Zhongshan Hospital,Shanghai 200032,China ~3Shanghai Institute of Biochemistry,Academy Sinica,Shanghai 200031,ChinaQing Wang earned master degree from Shanghai Medical University in 1996,now a senior lecturer of microbiology, specialized in the role of oncogcncs on tumor metastasis, having 8 papers published. | 2001 | World Journal of Gastroenterology2001,7,3: | 48
Draining Tangjiashan Barrier Lake after Wenchuan Earthquake and the flood propagation after the dam break显示文摘Tangjiashan Barrier Lake is one of the largest barrier lakes caused by the Wenchuan Earthquake. Its risk analysis, emergency plan and effect of the emergency plan are introduced in this paper. The dam height of Tangjiashan Barrier Dam is about 105 m, and the reservoir storage capacity is 3.2×108 m3. When the dam broke the flood peak were estimated to be larger than 48000 m3/s, which might cause a enormous disaster to the downstream cities and residents. A discharge channel with 13 m deep and 8 m wide was drug, so that the water may flow out of the lake before the dam breaks. As a result, the drainage and risk mitigation project are successful. During the drainage process, the flood peak was about 6500 m3/s, and about 1.6×108 m3 of water was drained off and the residual reservoir capacity was only 8.97×107 m3. A new channel with average width 100 m was formed, which can bear floods of 200 years frequency. The successful experience and the collected data can be used to deal with the similar natural disasters in future. | LIU Ning1, ZHANG JianXin2, LIN Wei3, CHENG WuYI4 & CHEN ZuYu5 1 Ministry of Water Resources of the People’s Republic of China, Beijing 100053, China 2 Bureau of Hydrology of the Ministry of Water Resources of the People’s Republic of China, Beijing 100053, China 3 Sichuan Hydrographic and Water Resources Survey Bureau Chengdu 610031, China 4 HydroChina Chengdu Engineering Corporation, Chengdu 610072, China 5 China Institute of Water Resources and Hydropower Research, Beijing 100044, China | 2009 | Science China(Technological Sciences)2009,52,4: | 29
Cytochrome P450 2E1 genetic polymorphism and gastric cancer in Changle,Fujian Province显示文摘AIM: Genetic polymorphism in enzymes of carcinogen metabolism has been found to have the influence on the susceptibility to cancer. Cytochrome P450 2E1 ( CYP2 E1) is considered to play an important role in the metabolic activation of procarcinogens such as N-nitroscoamines and Iow molecular weight organic compounds. The purpose of this study is to determine whether CYP450 2Elpolymorphisms are associated with risk s of gastric cancer.METHODS: We conducted a population based case-control study in Changle county, Fujian Province, a high-risk region of gastric cancer in China. Ninety-one incident gastric cancer patients and ninety-four healthy controls were included in our study. Datas including dsmographic characteristcs, diet intake, and alcohol and tobacco consumption of indivduals in our study were completed by a standardized questionnaire. PCR-RFLP revealed three genotypes: heterozygote (C1/C2) and two homozygotes (C1/C1 and C2/C2) in CYP2E1.RESULTS: The frequency of variant genotypes (C1/C2 and C2/C2) in gastric cancer cases and controls was 36.3% and 24.5%, respectively. The rare homozygous C2/C2 genotype was found in 6 indivduals in gastric cancer group(6.6%),whereas there was only one in the control group (1.1%).However, there was no statistically significan difference between the two groups (two-tailed Fisher′s exact test, P =0.066). Indivduals in gastric cancer group were more likely to carry genotype C1/C2 (odds ratio, OR = 1.50) and C2/C2(OR = 7.34) than indivduals in control group (X2 = 4.597, for trend P=0.032). The frequencies of genofypes with the C2allele ( C1/C2 and C2/C2 genotypes) were compared with those of genotypes without C2 allele ( C1/C1 genotype )among indivduals in gastric cancer group and control group according to the pattern of gastric cancer risk factors. The results show that indivduals who exposed to these gastric cancer risk factors and carry the C2 allele seemed to have a higher risk of developing gastric cancer.CONCLUSION: Polymorphism of CYP2E1 gene may have some effct in the development of gastric cancer in Changle county, Fujian Province. | Lin Cai~1 Shun-Zhang Yu~2 Zuo-Feng Zhang~3 1 Department of Epidemiology,Fujian Medical University,Fuzhou 350004,Fujian Province,China2 Department of Epidemiology,Shanghai Medical University,Shanghai 200032,China3 Department of Epidemiology,UCLA School of Public Health,Los Angeles California,USA | 2001 | World Journal of Gastroenterology2001,7,6: | 25
Efficient production of omega-3 fatty acid desaturase (sFat-1)-transgenic pigs by somatic cell nuclear transfer显示文摘Omega-3(ω-3) fatty acid desaturase transgenic pigs may improve carcass fatty acid composition. The use of transgenic pigs is also an excellent large animal model for studying the role of ω-3 fatty acids in the prevention and treatment of coronary heart disease and cancer. Transgenic pigs carrying synthesized fatty acid desaturase-1 gene (sFat-1) from Caenorhabditis briggsae by somatic cell nuclear transfer (SCNT) were produced for the first time in China. Porcine fetal fibroblast cells were transfected with a sFat-1 expression cassette by the liposome-mediated method. Transgenic embryos were reconstructed by nuclear transfer of positive cells into enucleated in vitro matured oocytes. A total of 1889 reconstructed embryos were transferred into 10 naturally cycling gilts. Nine early pregnancies were established, 7 of which went to term. Twenty-one piglets were born. The cloning efficiency was 1.1% (born piglets/transferred embryos). The integration of the sFat-1 gene was confirmed in 15 live cloned piglets by PCR and Southern blot except for 2 piglets. Expression of the sFat-1 gene in 12 of 13 piglets was detected with RT-PCR. The data demonstrates that an efficient system for sFat-1 transgenic cloned pigs was developed, which led to the successful production of piglets expressing the sFat-1 gene. | PAN DengKe1, ZHANG Li1, ZHOU YanRong2, FENG Chong1, LONG Chuan1, LIU Xiao1, WAN Rong3, ZHANG Jian3, LIN AiXing3, DONG EnQiu4, WANG ShuChen4, XU HouGang4 & CHEN HongXing2 1 Institute of Animal Sciences, Chinese Academy of Agricultural Sciences, Beijing 100193, China 2 Beijing Institute of Biotechnology, Beijing 100071, China 3 China Agricultural University, Beijing 100193, China 4 Yutian Breeding Swine Farms, Yutian 064104, China | 2010 | Science China(Life Sciences)2010,53,4: | 21
Forecasting the summer rainfall in North China using the year-to-year increment approach显示文摘A new approach to forecasting the year-to-year increment of rainfall in North China in July-August (JA) is proposed. DY is defined as the difference of a variable between the current year and the preceding year (year-to-year increment). NR denotes the seasonal mean precipitation rate over North China in JA. After analyzing the atmospheric circulation anomalies associated with the DY of NR, five key predictors for the DY of NR have been identified. The prediction model for the DY of NR is established by using multi-linear regression method and the NR is obtained (the current forecasted DY of NR added to the preceding observed NR). The prediction model shows a high correlation coefficient (0.8) between the simulated and the observed DY of NR throughout period 1965-1999, with an average relative root mean square error of 19% for the percentage of precipitation rate anomaly over North China. The prediction model makes a hindcast for 2000-2007, with an average relative root mean square error of 21% for the percentage of precipitation rate anomaly over North China. The model reproduces the downward trend of the percentage of precipitation rate anomaly over North China during 1965-2006. Because the current operational prediction models of the summer precipitation have average forecast scores of 60%-70%, it has been more difficult to forecast the summer rainfall over North China. Thus this new approach for predicting the year-to-year increment of the summer precipitation (and hence the summer precipitation itself) has the potential to significantly improve operational forecasting skill for summer precipitation. | FAN Ke1, LIN MeiJing1,2 & GAO YuZhong3 1 Nansen-Zhu International Research Center, Institute of Atmospheric Physics, Chinese Academy of Sciences, Beijing 100029, China 2 Graduate University of the Chinese Academy of Sciences, Beijing 100049, China 3 Heilongjiang Meteorological Administration, Harbin 150030, China | 2009 | Science China Earth Sciences2009,52,4: | 18
Interactions between hotspots and the Southwest Indian Ridge during the last 90 Ma:Implications on the formation of oceanic plateaus and intra-plate seamounts显示文摘This study investigates the relationship between the hotspot-ridge interaction and the formation of oceanic plateaus and seamounts in the Southwest Indian Ocean. We first calculated the relative distance between the Southwest Indian Ridge (SWIR) and relevant hotspots on the basis of models of plate reconstruction, and then calculated the corresponding excess magmatic anomalies of the hotspots on the basis of residual bathymetry and Airy isostasy. The results reveal that the activities of the Marion hotspot can be divided into three main phases:interaction with the paleo-Rodrigues triple junction (73.6-68.5 Ma),interaction with the SWIR (68.5-42.7 Ma),and intra-plate volcanism (42.7-0 Ma).These three phases correspond to the formation of the eastern, central, and western parts of the Del Cano Rise, respectively. The magnitude and apparent periodicity of the magmatic volume flux of the Marion hotspot appear to be dominated by the hotspot-ridge distance. The periodicity of the Marion hotspot is about 25 Ma,which is much longer than that of the Hawaii and Iceland hotspots (about 15 Ma). | ZHANG Tao 1,2,3,4,LIN Jian 5 & GAO JinYao 3,4 1 Institute of Geodesy and Geophysics,Chinese Academy of Sciences,Wuhan 430077,China 2 Graduate University of Chinese Academy of Sciences,Beijing 100049,China 3 The Second Institute of Oceanography,State Oceanic Administrator,Hangzhou 310012,China 4 Key Laboratory of Submarine Geosciences,State Oceanic Administrator,Hangzhou 310012,China 5 Department of Geology and Geophysics,Woods Hole Oceanographic Institution,Woods Hole,MA 02543,USA | 2011 | Science China Earth Sciences2011,54,8: | 16
Rb1 protects endothelial cells from hydrogen peroxide-induced cell senescence by modulating redox status显示文摘Objectives Endothelial senescence has been proposed to be involved in endothelial dysfunction and atherogenesis. This study investigates the effects of ginsenoside Rbl, a major constituent of ginseng,on H2O2-induced endothelial senescence. Methods Primary human umbilical vein endothelial cells(HUVECs) senescence was induced by H2O2 as judged by senescence-associated P-galactosidase assay (SA-P-gal).Fntracellur superoxide dismutase(S0D1) activity and malondialdehyde(MDA) level were determined by commercial kit.S0D1 mRNA and protein expression were analyzed by real time PCR and Western blot.Reactive oxygen species(ROS) were determined by flow cytometry. Results Rb1 was found to reverse endothelial senescence,as witnessed by a significant decrease of senescent cell numbers. Rbl could markedly increase intracellular SOD activity, decrease the MDA level, and suppress the generation of intracellular ROS in H2O2-treated HUVECs.Consistent with these findings,Rbl can effectively restore SOD1 mRNA and protein expression which decreased in H2O2 treated cells. Conclusions Our report demonstrates thatRbl can exert reversal effects on H2O2-induced cellular senescence through modulating cellular redox status. | LIU Ding-hui~1,CHEN Yan-ming~2,LIU Yong~1,HAO Bao-shun~1, ZHOU Bin~1,WU Lin~1,WANG Min~1,CHEN Lin~1,WU Wei-kang~3,QIAN Xiao-xian~1 (1.Department of Cardiology,The Third Affiliated Hospital of Sun Yat-sen University,Guagnzhou 510630 2.Department of Endocrinology,The Third Affiliated Hospital of Sun Yat-sen, Guagnzhou 510630 3.Institute Integrated Traditional Chinese and Western Medicine,Sun Yat-sen University,Guagnzhou 510630) | 2011 | 岭南心血管病杂志2011,17,S1: | 15
High-resolution climate variability of southwest China during 57―70 ka reflected in a stalagmite δ^(18)O record from Xinya Cave显示文摘A 26-cm-long stalagmite (XY2) from Xinya Cave in northeastern Chongqing of China has been ICP-MS 230Th/U dated, showing a depositional hiatus at 2.3 cm depth from the top. The growth of the 2.3―26 cm interval determined by four dates was between 57 ka and 70 ka, with a linear growth rate of 0.023 mm/a. We have analyzed 190 samples for δ 18O and δ 13C, mostly in the 2.3―26 cm part. The δ 18O and δ 13C values between 57 ka and 70 ka reveal decadal-to-centennial climatic variability during the glacial in-terval of Marine Isotope Stage 4 (MIS4), exhibiting much higher resolution than that of the published Hulu and Dongge records during this interval. Speleothem δ 18O in eastern China, including our study area can be used as a proxy of summer monsoon strength, with lighter values pointing to stronger summer monsoon and higher precipitation, and vice versa. Two decreases in the δ 18O signature of XY2 record around 59.5 and 64.5 Ka are argued to correspond to the Dansgaard-Oeschger (D-O) events 17 and 18 respectively. The Heinrich event 6 (H6) can be identified in the record as a heavy δ 18O peak around 60 ka, indicating significant weakening of the monsoon in Chongqing during the cold period. The XY2 δ 18O record shows very rapid change toward to the interstadial condition of the D-O event, but more gradual change toward to the cold stadial condition. This phenomenon found in the Greenland ice core records is rarely observed so clearly in previously published speleothem records. According to SPECMAP δ 18O record, the glacial maximum of MIS 4 was around 64.5 ka with the boundary of MIS 3/4 around 60 ka. Unlike the marine record, the speleothem record of XY2, China, exhibits much high fre-quency variations without an apparent glacial maximum during MIS 4. However, the timing of MIS 3/4 boundary seems to be around 60 ka when the H6 terminated, in agreement with the marine chronology. The growth period of sample XY2 during glacial times probably reflects a local karstic routing of water, rather than having climatic significance. | LI TingYong1, YUAN DaoXian1,2, LI HongChun1,3 ,YANG Yan2, WANG JianLi1, WANG XinYa1, LI JunYun1, QIN JiaMing2, ZHANG MeiLiang2 & LIN YuShi2 1 College of Geographical Science, Southwest University, Chongqing 400715, China 2 Karst Dynamics Laboratory, Institute of Karst Geology, Chinese Academy of Geological Sciences, Guilin 541004, China 3 Department of Earth Sciences, Cheng-Kung University, Tainan 70101, Taiwan, China | 2007 | Science China Earth Sciences2007,50,8: | 14
Enrichment mechanisms of rare earth elements in marine hydrogenic ferromanganese crusts显示文摘A series of selective extraction experiments were conducted on marine hydrogenic ferromanganese crusts from the seamount near the Line Island to investigate the distribution of rare earth elements(REEs) and the mechanisms of combination between REEs and the hydrogenic ferromanganese crusts. The enrichment of REEs in hydrogenic ferromanganese crusts is attributed mainly to the mineral or oxide/hydroxide of the crust rather than the growth environment. The REEs are present mainly in amorphous ferric oxide/hydroxide rather than in δ-MnO2.Amorphous ferric oxide/hydroxide preferentially combines free REE3+,while δ-MnO2 combines the REE-carbonate complexes in modern seawater. The REEs in seawater are combined more strongly with amorphous ferric oxide/hydroxides than with δ-MnO2.The positive Ce anomaly in hydrogenic ferromanganese crusts results from the oxidation of soluble Ce3+ to insoluble Ce4+ by the actively soluble oxygen of the bottom current rather than by the oxidation of δ-MnO2. | JIANG XueJun1,2,LIN XueHui1,2,YAO De3,4 & GUO WeiDong5 1 Key Laboratory of Marine Oil and Gas Resources and Marine Environment,Ministry of Land and Resources,Qingdao 266071,China 2 Qingdao Institute of Marine Geology,China Geological Survey,Qingdao 266071,China 3 Institute of Resources and Environment,Shandong University of Technology,Zibo 255049,China 4 Institute of Oceanology,Chinese Academy of Sciences,Qingdao 266071,China 5 Department of Oceanography,Xiamen University,Xiamen 361005,China | 2011 | Science China Earth Sciences2011,54,2: | 14

Spatial analysis and data management are the advantages of GIS, which can define distribution trend and spatial relations of environmental factors, and show ecological security pattern graphically. Spatial differences of ecological security assessment based on GIS are discussed in this paper, of which the middle and lower reaches of the Liaohe River is taken as a study case. First, to work out pressure-state-response (P-S-R) assessment indicators system, and investigate in person and gather information; second, to digitize the watershed; third, to quantize and calculate by the fuzzy method; last, to construct GIS grid database, and expound spatial differences of ecological security by GIS interpolation and assembly analysis. | Wang Geng 1,2 , Nie Baochi 3, Wang Lin 1 & Wu Wei 1 1. | 14 | Performance-based methodology for assessing seismic vulnerability and capacity of buildings显示文摘This paper presents a performance-based methodology for the assessment of seismic vulnerability and capacity of buildings. The vulnerability assessment methodology is based on the HAZUS methodology and the improved capacity- demand-diagram method. The spectral displacement (Sd) of performance points on a capacity curve is used to estimate the damage level of a building. The relationship between Sd and peak ground acceleration (PGA) is established, and then a new vulnerability function is expressed in terms of PGA. Furthermore, the expected value of the seismic capacity index (SCev) is provided to estimate the seismic capacity of buildings based on the probability distribution of damage levels and the corresponding seismic capacity index. The results indicate that the proposed vulnerability methodology is able to assess seismic damage of a large number of building stock directly and quickly following an earthquake. The SCev provides an effective index to measure the seismic capacity of buildings and illustrate the relationship between the seismic capacity of buildings and seismic action. The estimated result is compared with damage surveys of the cities of Dujiangyan and Jiangyou in the M8.0 Wenchuan earthquake, revealing that the methodology is acceptable for seismic risk assessment and decision making. The primary reasons for discrepancies between the estimated results and the damage surveys are discussed. | Lin Shibin1, Xie Lili1, 2,Gong Maosheng1,2 and Li Ming1,3 1.Institute of Engineering Mechanics,China Earthquake Administration, Harbin 150080, China 2.School of Civil Engineering,Harbin Institute of Technology,Harbin 150090, China 3.School of Civil Engineering,Shenyang Jianzhu University,Shenyang 11016, China | 2010 | Earthquake Engineering and Engineering Vibration2010,9,2: | 12 |

People's mental states and behaviors rely on their holding beliefs for self and others. To investigate the neural substrates of false belief reasoning, the 32 channels event-related potentials (ERP) of 14 normal adults were measured while they understood false-belief and true belief used de-ceptive appearance task. After onset of the false-belief or true-belief questions, N100, P200 and late negative component (LNC) were elicited at centro-frontal sites. Compared with true belief, false belief reasoning elicited significant declined LNC in the time window from 400 to 800 ms. | 20 | Alterations in DNA methylation and genome structure in two rice mutant lines induced by high pressure显示文摘By using high-pressure treatment, two mutant lines were obtained from a genetically stable japonica rice cultivar Bijing38. Genomic DNA of the mutant lines, together with the original line (Bijing38), was either undigested or digested by Hpa II/Msp I, and then subjected to molecular analy-sis using two markers, ISSR and RAPD. Results indicated that changes in the PCR amplification pro-files of both markers are apparent in the two mutant lines compared with the original rice cultivar, suggesting that there had been both sequence changes and DNA methylation modifications in the mutant lines. Southern blot analysis using diverse sequences, including two cellular genes (S2 and S3), a set of retrotransposons (Osr7, Osr36, Tos19 and more), and a MITE transposon family (mPing and Pong), confirmed the results, and indicated that changes in DNA methylation pattern, genomic structure, and possible activation of some transposons indeed occurred in the mutant lines. Moreover, these changes are stably maintained through selfed generations and in different organs. Thus, our results indicate that it is possible to obtain stable mutants in rice by high pressure treatments, and the molecular basis of the mutants may include both genetic and epigenetic changes. Therefore, high hydrostatic pressure seems a promising approach for plant mutagenesis. | SHEN Sile1,3, WANG Zhenwei3, SHAN Xiaohui2, WANG Hua3, LI Ling3, LIN Xuyun2, LONG Likun2, WENG Kenan4, LIU Bao2 & ZOU Guangtian1 1.
