| 1 | WNT/β-catenin signaling in urothelial carcinoma of bladder显示文摘Urothelial carcinoma of bladder is the second most prevalent genitourinary disease.It is a highly heterogeneous disease as it represents a spectrum of neoplasms,including non-muscle invasive bladder cancer(NMIBC),muscle invasive bladder cancer(MIBC)and metastatic lesions.Genome-wide approaches and candidate gene analysis suggest that malignant transformation of the bladder is multifactorial and a multitude of genes are involved in the development of MIBC or NMIBC phenotypes.Wnt signaling is being examined to control and maintain balance between stemness and differentiation in adult stem cell niches.Owing to its participation in urothelial development and maintenance of adult urothelial tissue homeostasis,the components of Wnt signaling are reported as an important diagnostic and prognostic markers as well as novel therapeutic targets.Mutations/epigenetic alterations in the key molecules of Wnt/β-catenin canonical pathway have been linked with tumorigenesis,development of drug resistance and enhanced survival.Present review extends our understanding on the functions of key regulatory molecules of canonical Wnt/β-catenin pathway in urothelial tumorigenesis by inducing cancer stem cell phenotype(UCSCs).UCSCs may be responsible for tumor heterogeneity,high recurrence rates and complex biological behavior of bladder cancer.Therefore,understanding the role of UCSCs and the regulatory mechanisms that are responsible for high relapse rates and metastasis could help to develop pathway inhibitors and augment current therapies.Potential implications in the treatment of urothelial carcinoma of bladder by targeting this pathway primarily in UCSCs as well as in bulk tumor population that are responsible for high relapse rates and metastasis may facilitate potential therapeutic avenues and better prognosis. | Minal Garg Niharika Maurya | 2019 | World Journal of Nephrology2019,8,5: | 11 |

| 3 | MicroRNA-99a mimics inhibit M1 macrophage phenotype and adipose tissue inflammation by targeting TNFα显示文摘In human adipose tissue and obesity,miR-99a expression is negatively correlated with inflammation.Therefore,the present study investigated the role of miR-99a in macrophage phenotype activation and adipose tissue inflammation.M2 BMDMs showed a significant increase in miR-99a expression when compared to the M0 and M1 phenotypes.Phenotype-switching experiments established an association between upregulated miR-99a expression and the M2 phenotype.Overexpression of miR-99a prevented M1 phenotype activation and attenuated bactericidal activity.Likewise,knockdown of miR-99a abolished M2 phenotype activation.By means of in silico target prediction tools and a luciferase reporter assay,TNFαwas identified as a direct target of miR-99a.Knockdown of TNFαrecapitulated the effect of miR-99a overexpression in M1 BMDMs.In a db/db mice model,miR-99a expression was reduced in eWAT and F4/80+ATMs.Systemic overexpression of miR-99a in db/db mice attenuated adipocyte hypertrophy with increased CD301 and reduced CD86 immunostaining.Flow cytometry analysis also showed an increased M2 and a reduced M1 macrophage population.Mimics of miR-99a also improved the diabetic dyslipidemia and insulin signaling in eWAT and liver,with an attenuated expression of gluconeogenesis and cholesterol metabolism genes in the liver.Furthermore,adoptive transfer of miR-99a-overexpressing macrophages in the db/db mice recapitulated in vivo miR-99a mimic effects with increased M2 and reduced M1 macrophage populations and improved systemic glucose,insulin sensitivity,and insulin signaling in the eWAT and liver.The present study demonstrates that miR-99a mimics can regulate macrophage M1 phenotype activation by targeting TNFα.miR-99a therapeutics in diabetic mice reduces the adipose tissue inflammation and improves insulin sensitivity. | Anant Jaiswal Sukka Santosh Reddy Mohita Maurya Preeti Maurya Manoj Kumar Barthwal | 2019 | Cellular & Molecular Immunology2019,16,5: | 7 |

| 5 | Mechanical, tribological and anti-corrosive properties of polyaniline/graphene coated Mg-9Li-7Al-1Sn and Mg-9Li-5Al-3Sn-1Zn alloys显示文摘The mechanical,tribological and corrosion protection offered to Mg-9Li-7Al-1Sn and Mg-9Li-5Al-3Sn-1Zn alloys by the epoxy coating containing polyaniline/graphene(PANI/Gr)pigments is undertaken in the current work.PANI/Gr containing coatings were observed to be strongly adherent with a higher scratch hardness(Hs)and plowing hardness(Hp),i.e.Hsof 0.43 GPa,and Hpof 0.61 GPa,respectively when compared to that of neat epoxy coating(Hsof 0.17 GPa,and Hpof 0.40 GPa,respectively).Due to their higher Hsand Hpvalues,PANI/Gr based coatings displayed an enhanced wear resistance(Wear volume,Wv=4.53×10^-3 m^3)than that of neat epoxy coating(Wv=5.15×10^-3 m^3).The corrosion protection efficiency in corrosive environment of 3.5 wt%NaCl solution was obtained to be>99% for PANI/Gr containing coatings when compared to that of neat epoxy coating.The charge-transfer resistance(R(ct))of the PANI/Gr containing coatings were estimated to be>10^6 cm^2,which indicates their highly protective nature when compared to that of neat epoxy coating(R(ct)10^5 Ω cm^2).Hence,PANI/Gr containing coatings can be potentially used for wear resistance and corrosion protection applications in marine environments. | Rita Maurya Abdul Rahim Siddiqui Prvan Kumar Katiyar Kantesh Balani | 2019 | Journal of Materials Science & Technology2019,35,8: | 4 |

| 8 | Effect of sintering temperature on phase evolution of Al86Ni6Y4.5Co2La1.5 bulk amorphous composites synthesized via mechanical alloying and spark plasma sintering显示文摘Al86Ni6Y4.5Co2La1.5 amorphous powders were synthesized by mechanical alloying for 200 h. Subsequent consolidation was performed via spark plasma sintering in the temperature range of 250 ℃ to 500 ℃ at the pressure of 500 MPa. The role of viscous flow on densification was investigated by studying the viscosity change of the amorphous phase at different consolidation temperatures. The decrease in viscosity at higher sintering temperatures resulted in better particle bonding and densification of consolidated samples. The formation of only FCC Al was observed in the consolidated samples at sintering temperatures ≤ 300 ℃ and the intermetallic phases formed at temperatures ≥ 400 ℃. The mechanical properties of the bulk samples were measured by Vickers microhardness and nanoindentation tests. The testing results showed that the average values of microhardness, nanohardness and elastic modulus of the sample consolidated at 500 ℃ were 3.06 ± 0.14 GPa,4.85 ± 1.14 GPa and 89.53 ± 9.25 GPa, respectively. | 9 | Clinicohistopathological implications of phosphoserine 9 glycogen synthase kinase-3β/β-catenin in urinary bladder cancer patients显示文摘BACKGROUND Aberrant activation of phosphorylated form of glycogen synthase kinase-3β[pS9 GSK-3β(Serine 9 phosphorylation)] is known to trigger Wnt/β-catenin signal cascade but its clinicohistopathological implications in bladder carcinogenesis remain unknown.AIM To investigate the diagnostic and prognostic relevance of expressions of pS9 GSK-3β, β-catenin and its target genes in the pathobiology of bladder cancer.METHODS Bladder tumor tissues from ninety patients were analyzed for quantitative expression and cellular localization of pS9 GSK-3β by immunohistochemical(IHC) staining. Real time-quantitative polymerase chain reaction and IHC were done to check the expression of β-catenin, Cyclin D1, Snail and Slug at transcriptome and protein level respectively. Clinicohistopathological variables were obtained from histology reports, follow up and OPD visits of patients.Expressions of the markers were statistically correlated with these variables to determine their significance in clinical setting. Results were analysed using SPSS20.0 software.RESULTS Aberrant(low or no membranous/high nuclear/high cytoplasmic) expression of pS9 GSK-3β was noted in 51% patients and found to be significantly associated with tumor stage and tumor grade(P = 0.01 and 0.04; Mann Whitney U test).Thirty one percent tumors exhibited aberrant co-expression of pS9 GSK-3β andβ–catenin proteins and showed strong statistical association with tumor stage,tumor type, smoking/tobacco chewing status(P = 0.01, 0.02 and 0.04, MannWhitney U test) and shorter overall survival probabilities of patients(P = 0.02;Kaplan Meier test). Nuclear immunostaining of Cyclin D1 in tumors with altered pS9 GSK-3β/β–catenin showed relevance with tumor stage, grade and type.CONCLUSIONβ–catenin and pS9 GSK-3β proteins are identified as markers of diagnostic/prognostic significance in disease pathogenesis. | 10 | Dynamic parameters of vehicles under heterogeneous traffic stream with non-lane discipline:An experimental study显示文摘On the heterogeneous and no lane disciplined traffic, the abreast maneuver of vehicles depends upon the driver behavior, vehicle type and most importantly the traffic parameters such as vehicle speed and acceleration. Hence, the drivers have two basic tasks while driving, first is to control the vehicle's position along longitudinal direction of motion and second is to control the vehicle's position along lateral direction i.e. the width of the roadway. The relation between the dynamic parameters(speed and lateral/longitudinal acceleration), can very well represent the driving behavior of vehicles in mixed with weak or no lane disciplined traffic. Hence, the study of longitudinal and lateral control of vehicles in a comprehensive manner is necessary. In the present paper, driving behavior of vehicles are studied by analyzing longitudinal and lateral acceleration/deceleration(A/D) with operating speed of vehicles on different straight roadway sections. Data are collected from five major cities of India using GPS based instrument(Video-VBOX) mounted on five different type of vehicles. The probability distribution of longitudinal A/D and the lateral acceleration are analysed, and their relationship with operating speed of vehicles are studied on roads with a different number of lanes for different vehicle types. | 12 | GDF15 promotes prostate cancer bone metastasis and colonization through osteoblastic CCL2 and RANKL activation显示文摘Bone metastases occur in patients with advanced-stage prostate cancer(PCa). The cell-cell interaction between PCa and the bone microenvironment forms a vicious cycle that modulates the bone microenvironment, increases bone deformities, and drives tumor growth in the bone. However, the molecular mechanisms of PCa-mediated modulation of the bone microenvironment are complex and remain poorly defined. Here, we evaluated growth differentiation factor-15(GDF15) function using in vivo preclinical PCa-bone metastasis mouse models and an in vitro bone cell coculture system. Our results suggest that PCa-secreted GDF15 promotes bone metastases and induces bone microarchitectural alterations in a preclinical xenograft model. Mechanistic studies revealed that GDF15 increases osteoblast function and facilitates the growth of PCa in bone by activating osteoclastogenesis through osteoblastic production of CCL2 and RANKL and recruitment of osteomacs.
