| 2 | 胸苷酸合成酶和拓扑异构酶-1及Ki-67对伊立替康联合5-氟脲嘧啶治疗晚期大肠癌的预测价值显示文摘目的探讨胸苷酸合成酶(TS)、拓扑异构酶1(Topo1)和肿瘤增殖指标Ki67对伊立替康(CPT11)联合5氟脲嘧啶(5Fu)治疗晚期大肠癌患者的临床疗效和预后的预测价值。方法采用免疫组化方法检测了CPT11+5Fu一线治疗78例大肠癌患者的TS、Topo1和Ki67的表达,并与化疗疗效和患者的临床预后进行了分析。结果各检测指标的表达水平与临床疗效之间无关,但是临床预后不同。其中,TS低表达患者表现明显长的肿瘤进展时间(TTP,P<0.05)和存活期(OS,P<0.05);Ki67低表达也预示长的OS(P<0.05)。与单个指标相比较,两个指标的联合也不能预测临床疗效,但是对预后的判断作用明显提高。TS低表达、Ki67低表达以及TS和Ki67均低表达患者的中位TTP分别为9,8和17个月(P=0.02);TS低表达、Topo1低表达以及TS和Topo1均低表达患者的中位TTP分别为9,9和13个月(P=0.03);而Topo1低表达、Ki67低表达、Ki67和Topo1均低表达患者的中位TTP分别为8,9和11个月(P=0.05)。其中任何两个指标均低表达肿瘤的TTP和OS均明显长于高表达者(P<0.05)。结论TS、Topo1和Ki67不能预测大肠癌患者CPT11+5Fu的疗效,但对其临床预后有一定的预测作用。其中任何两个指标的联合,比单一指标对患者预后的预测价值明显提高。 | 徐建明 朱步东 Mangia Anita Simone Gianni Montemurro Severino Giuliani Francesco Maiello Evaristo Colucci Giuseppe Paradiso Angelo | 2005 | 中华肿瘤杂志2005,27,5: | 9 |
| 3 | Multiple responses optimization in the development of a headspace gas chromatography method for the determination of residual solvents in pharmaceuticals显示文摘An efficient generic static headspace gas chromatography(HSGC) method was developed,optimized and validated for the routine determination of several residual solvents(RS) in drug substance,using a strategy with two sets of calibration.Dimethylsulfoxide(DMSO) was selected as the sample diluent and internal standards were used to minimize signal variations due to the preparative step.A gas chromatograph from Agilent Model 6890 equipped with flame ionization detector(FID) and a DB-624(30 m × 0.53 mm i.d.,3.00 μm film thickness) column was used.The inlet split ratio was 5:1.The influencing factors in the chromatographic separation of the analytes were determined through a fractional factorial experimental design.Significant variables:the initial temperature(IT),the final temperature(FT) of the oven and the carrier gas flow rate(F) were optimized using a central composite design.Response transformation and desirability function were applied to find out the optimal combination of the chromatographic variables to achieve an adequate resolution of the analytes and short analysis time.These conditions were 30 ℃ for IT,158 ℃ for FT and 1.90 mL/min for F.The method was proven to be accurate,linear in a wide range and very sensitive for the analyzed solvents through a comprehensive validation according to the ICH guidelines. | Carla M.Teglia Milagros Montemurro María M.De Zan María S.Cmara | 2015 | Journal of Pharmaceutical Analysis2015,5,5: | 8 |
| 4 | H pylori status and angiogenesis factors in human gastric carcinoma显示文摘AIM: To investigate H pylori expression in gastric cancer patients in relation to primary tumor angiogenic markers, such as microvessel density (MVD), thymidine phospho- rylase (TP), vascular endothelial growth factor receptor-1 (VEGF-R1), p53 and circulating VEGF levels. METHODS: Angiogenic markers were analyzed immu- nohistochemically in 56 primary gastric cancers. H pylori cytotoxin (vacA) and the cytotoxin-associated gene (cagA) amplification were evaluated using PCR assay. Serum H pylori IgG antibodies and serum/plasma circulating VEGF levels were detected in 39 and 38 patients by ELI- SA, respectively. RESULTS: A total of 69% of patients were positive for circulating IgG antibodies against H pylori. cagA-positive H pylori strains were found in 41% of gastric patients. vacA was found in 50% of patients; s1 strains were more highly expressed among vacA-positive patients. The presence of the s1 strain was signifi cantly associ- ated with cagA (P = 0.0001). MVD was signifi cantly cor- related with both tumor VEGF expression (r = 0.361, P = 0.009) and serum VEGF levels (r = -0.347, P = 0.041).Conversely, neither VEGF-R1 expression nor MVD was related to p53 expression. However, H pylori was not related to any angiogenic markers except for the plasma VEGF level (P = 0.026). CONCLUSION: H pylori antigen is related to higher plasma VEGF levels, but not to angiogenic character- istics. | 5 | Functional conservation of the grapevine candidate gene INNER NO OUTER for ovule development and seed formation显示文摘Seedlessness represents a highly appreciated trait in table grapes.Based on an interesting case of seedless fruit production described in the crop species Annona squamosa,we focused on the Vitis vinifera INNER NO OUTER(INO)gene as a candidate.This gene encodes a transcription factor belonging to the YABBY family involved in the determination of abaxial identity in several organs.In Arabidopsis thaliana,this gene was shown to be essential for the formation and asymmetric growth of the ovule outer integument and its mutation leads to a phenotypic defect of ovules and failure in seed formation.In this study,we identified in silico the V.vinifera orthologue and investigated its phylogenetic relationship to INO genes from other species and its expression in different organs in seeded and seedless varieties.Applying cross-species complementation,we have tested its functionality in the Arabidopsis ino-1 mutant.We show that the V.vinifera INO successfully rescues the ovule outer integument growth and seeds set and also partially complements the outer integument asymmetric growth in the Arabidopsis mutant,differently from orthologues from other species.These data demonstrate that VviINO retains similar activity and protein targets in grapevine as in Arabidopsis.Potential implications for grapevine breeding are discussed. | Valentina di Rienzo Zahra Imanifard Isabella Mascio Charles SGasser Debra JSkinner Ciro Leonardo Pierri Martina Marini Valentina Fanelli Wilma Sabetta Cinzia Montemurro Diana Bellin | 2021 | Horticulture Research2021,8,1: | 2 |
