3Structures of EV71 RNA-dependent RNA polymerase in complex with substrate and analogue provide a drug target against the hand-foot-and-mouth disease pandemic in China显示文摘Enterovirus 71(EV71),one of the major causative agents for hand-foot-and-mouth disease(HFMD),has caused more than 100 deaths among Chinese children since March 2008.The EV71 genome encodes an RNAdependent RNA polymerase(RdRp),denoted 3D^(pol),which is central for viral genome replication and is a key target for the discovery of specific antiviral therapeutics.Here we report the crystal structures of EV71 RdRp(3D^(pol))and in complex with substrate guanosine-5'-triphosphate and analog 5-bromouridine-5'-triphosphate best to 2.4Åresolution.The structure of EV71 RdRp(3D^(pol))has a wider open thumb domain compared with the most closely related crystal structure of poliovirus RdRp.And the EV71 RdRp(3D^(pol))complex with GTP or Br-UTP bounded shows two distinct movements of the polymerase by substrate or analogue binding.The model of the complex with the template:primer derived by superimposition with foot-and-mouth disease virus(FMDV)3D/RNA complex reveals the likely recognition and binding of template:primer RNA by the polymerase.These results together provide a molecular basis for EV71 RNA replication and reveal a potential target for anti-EV71 drug discovery.Yang Wu Zhiyong Lou Yi Miao Yue Yu Hui Dong Wei Peng Mark Bartlam Xuemei Li Zihe Rao 2010Protein & Cell2010,1,5:30

5Effect and Safety of Guanxinning Tablet(冠心宁片) for Stable Angina Pectoris Patients with Xin(Heart)-Blood Stagnation Syndrome:A Randomized,Multicenter,Placebo-Controlled Trial显示文摘Objective: To investigate the effect and safety of Guanxinning Tablet(冠心宁片, GXN) for the treatment of stable angina pectoris patients with Xin(Heart)-blood stagnation syndrome(XBSS). Methods: One hundred and sixty stable angina pectoris patients with XBSS were randomly assigned to receive GXN(80 cases) or placebo(80 cases, Guanxinning simulation tablets, mainly composed of lactose), 4 tablets(0.38 g/tablet), thrice daily for 12 weeks. After treatment, an exercise stress test(treadmill protocol), Chinese medicine(CM) syndrome score, electrocardiogram(ECG), and nitroglycerin withdrawal rate were evaluated and compared in the patients between the two groups. Meanwhile, adverse events(AEs) were evaluated during the whole clinical trial. Results: Compared with the control group, the time extension of exercise duration in the GXN group increased 29.28±17.67 s after treatment(P>0.05);moreover, the change of exercise duration in the GXN group increased 63.10±96.96 s in subgroup analysis(P<0.05). The effective rates of angina pectoris, CM syndrome and ECG as well as nitroglycerin withdrawal rate were 81.33%, 90.67%, 45.76%, and 70.73%, respectively in the GXN group, which were all significantly higher than those in the control group(40.58%, 75.36%, 26.92%, 28.21%, respectively, P<0.05). 8Impact of Cardiovascular Disease Deaths on Life Expectancy in Chinese Population显示文摘Objective We aimed to analyze the impact of cardiovascular disease(CVD) deaths on life expectancy(LE) in Chinese population and estimate the percentage reduction in CVD mortality needed to increase LE by 1 year from the current level, a national target of health improvement. Methods We used life tables, cause-elimination life tables, and age decomposition of LE with corrected mortality data from the National Disease Surveillance System in 2010. Results LE at birth of Chinese people was 73.24 years in 2010. Women had a longer LE than men, and urban population had a longer LE than rural population. CVD deaths resulted in a 4.79-year LE loss and premature deaths in people aged 25 to 64 years were responsible for a substantial part of LE loss from CVD. Death from ischemic heart disease and cerebrovascular diseases accounted for 69.2% of LE loss from CVD deaths and death from cerebrovascular diseases was the largest contributor. In rural men, 51.1% LE loss from CVD deaths was caused by cerebrovascular diseases. If there were no changes in mortality rates for all other diseases, a 27.4% reduction in CVD mortality would increase LE by 1 year in Chinese population. Conclusion There is a considerable impact of CVD deaths on LE. A 1-year LE increase in the future requires at least a 27.4% reduction in CVD mortality from the current level. 12Prevalence and Genotypes of Rotavirus A and Human Adenovirus among Hospitalized Children with Acute Gastroenteritis in Fujian, China, 2009-2017显示文摘Gastroenteritis is an infectious diarrhea that has been considered as an important cause of hospitalizations and death in children aged < 5 years, particularly in developing countries. Unsanitary water, contaminated food, poor hygiene, and inadequate disposal of waste and feces are all risk factors for gastroenteritis, resulting in the higher incidence in developing countries. Gastroenteritis is generally caused by viral infections, among which rotavirus (RV) infections have been reported to be the most common, especially among young children aged < 5 years with acute gastroenteritis in Asia and Africa[1]. Other viruses associated with acute gastroenteritis include human Adenovirus (HAdV), Norovirus, Sapovirus (SaV), human Astrovirus (HAstV), and Aichi virus. Recent research has reported that adenovirus types 40 and 41, belonging to species F, cause gastroenteritis and were therefore termed as enteric adenoviruses. 14Architecture design for reliable and reconfigurable FPGA-based GNC computer for deep space exploration显示文摘SRAM(static random access memory)-based FPGA(field programmable gate array), owing to its large capacity, high performance, and dynamical reconfiguration, has become an attractive platform for So PC(system on programmable chip) development. However, as the configuration memory and logic memory of the SRAM-based FPGA are highly susceptible to SEUs(single-event upsets) in deep space, it is a challenge to design and implement a highly reliable FPGA-based system for spacecraft, and no practical architecture has been proposed. In this paper, a new architecture for a reliable and reconfigurable FPGAbased computer in a highly critical GNC(guidance navigation and control) system is proposed. To mitigate the effect of an SEU on the system, multi-layer reconfiguration and multi-layer TMR(triple module redundancy) techniques are proposed, with a reliable reconfigurable real-time operating system(Space OS) managing the system level fault tolerance of the computer in the architecture. The proposed architecture for the reconfigurable FPGA-based computer has been implemented with COTS(commercial off the shelf) FPGA and has firstly been applied to the GNC system of a circumlunar return and reentry flight vehicle. 15Ginsenoside Rb1 Attenuates Isoflurane/surgery-induced Cognitive Dysfunction via Inhibiting Neuroinflammation and Oxidative Stress显示文摘Objective Anesthetic isoflurane plus surgery has been reported to induce cognitive impairment. The underlying mechanism and targeted intervention remain largely to be determined. Ginsenoside Rb1 was reported to be neuroprotective. We therefore set out to determine whether ginsenoside Rb1 can attenuate isoflurane/surgery-induced cognitive dysfunction via inhibiting neuroinflammation and oxidative stress. Methods Five-months-old C57BL/6J female mice were treated with 1.4% isoflurane plus abdominal surgery for two hours. Sixty mg/kg ginsenoside Rb1 were given intraperitoneally from 7 days before surgery. Cognition of the mice were assessed by Barnes Maze. Levels of postsynaptic density-95 and synaptophysin in mice hippocampus were measured by Western blot. Levels of reactive oxygen species, tumor necrosis factor-α and interleukin-6 in mice hippocampus were measured by ELISA. Results Here we show for the first time that the ginsenoside Rb1 treatment attenuated the isoflurane/surgery-induced cognitive impairment. Moreover, ginsenoside Rb1 attenuated the isoflurane/surgery-induced synapse dysfunction. Finally, ginsenoside Rb1 mitigated the isoflurane/surgery-induced elevation levels of reactive oxygen species, tumor necrosis factor-α and interleukin-6 in the mice hippocampus. 16Nia1 and Nia2 are Involved in Exogenous Salicylic Acid-induced Nitric Oxide Generation and Stomatal Closure in Arabidopsis显示文摘Phytohormone salicylic acid (SA) plays important roles in plant responses to environmental stress. However, knowledge about the molecular mechanisms for SA affecting the stomatal movements is limited. In this paper, we demonstrated that exogenous SA significantly induced stomatal closure and nitric oxide (NO) generation in Arabidopsis guard cells based on genetic and physiological data. These effects were significantly inhibited by the NO scavenger c-PTIO, NO synthase (NOS) inhibitor L-NAME or nitrate reductase suppressor tungstate respectively, implying that NOS and nitrate reductase (NR) participate in SA-evoked stomatal closing. Furthermore, the effects of SA promotion of stomatal closure and NO synthesis are significantly suppressed in NR single mutants of nia1, nia2 or double mutant nia1/nia2, compared with the wild type plants. This suggests that both Nia1 and Nia2 are involved in SA-stimulated stomatal closure. 17Application of quantification theory in risk assessment of mine flooding显示文摘Hundreds of mine flooding accidents have occurred in China since the 1950s. These flooding accidents result in submerged working faces, even entire coal mines, leading to tremendous economic losses. It is reported that among 601 state-owned mines in China, 285 mines are exposed to water-inrush risks. The water pressure is becoming larger and larger with the increase of mining depth, leading to an increase of water-inrush hazards. Only when the risk of mine flooding is predicted in a reasonable manner, can we take timely and effective measures to prevent mine flooding from taking place. In our investigation quantifica- tion(II) theory is used to study the risk prediction problem about mine flooding. By investigating the main factors which affect mine flooding, eight risk assessment items have been identified. The extent of risk is classified into 4 grades. Given the data from different periods in the Feicheng mining area, a prediction model for the risk of mine flooding is established. The test analysis indicates a model correlation coefficient of 0.97 and the incidence of discrimination is as high as 97.37%, which implies that the effect of the model is quite satisfactory. 18Model based on γ-glutamyltransferase and alkaline phosphatase for hepatocellular carcinoma prognosis显示文摘AIM: To determine the prognostic value of alkaline phosphatase(ALP) and γ-glutamyltransferase(GGT) for hepatocellular carcinoma(HCC).METHODS: We analyzed the outcome of 172 HCC patients who underwent liver resection. Receiver operating characteristic(ROC) curve analysis was performed to determine the cut-off value of ALP and GGT. Then, preoperative risk factors for survival were evaluated by multivariate analysis. Based on the significant factors, aprognostic score model was established.RESULTS: By ROC curve analysis, ALP > 120 U/L and GGT > 115 U/L were considered elevated. Overall survival(OS) and tumor-free survival(TFS) for patients with elevated ALP and GGT were significantly worse than for patients with ALP and GGT within the normal range. Multivariate analysis showed that the elevated levels of ALP, GGT and tumor size were independent prognostic factors. Giving each positive factor as a score of 1, we established a preoperative prognostic score model. The 5-year OS for patients with a score of 0, 1, 2 and 3 were 84.0%, 45.9%, 44.1% and 0%, respectively, while the TFS was 80.6%, 40.0%, 38.8% and 0%, respectively. When combining patients with scores of 1 and 2 into the middle risk group, and patients with scores of 0 and 3 into the low-risk and highrisk groups, respectively, different outcomes would be significantly distinguished by the risk groups.CONCLUSION: Elevated ALP and GGT levels were risk predictors in HCC patients. 19Comparative analysis between endometrial proteomes of pregnant and non-pregnant ewes during the peri-implantation period显示文摘Background:Early pregnancy failure has a profound impact on both human reproductive health and animal production.2/3 pregnancy failures occur during the peri-implantation period;however,the underlying mechanism(s)remains unclear.Well-organized modification of the endometrium to a receptive state is critical to establish pregnancy.Aberrant endometrial modification during implantation is thought to be largely responsible for early pregnancy loss.Result:In this study,using well-managed recipient ewes that received embryo transfer as model,we compared the endometrial proteome between pregnant and non-pregnant ewes during implantation period.After embryo transfer,recipients were assigned as pregnant or non-pregnant ewes according to the presence or absence of an elongated conceptus at Day 17 of pregnancy.By comparing the endometrial proteomic profiles between pregnant and non-pregnant ewes,we identified 94 and 257 differentially expressed proteins(DEPs) in the endometrial caruncular and intercaruncular areas,respectively.Functional analysis showed that the DEPs were mainly associated with immune response,nutrient transport and utilization,as well as proteasome-mediated proteolysis.Conclusion:These analysis imply that dysfunction of these biological processes or pathways of DEP in the endometrium is highly associated with early pregnancy loss.In addition,many proteins that are essential for the establishment of pregnancy showed dysregulation in the endometrium of non-pregnant ewes.These proteins,as potential candidates,may contribute to early pregnancy loss.Haichao Zhao Linlin Sui Kai Miao Lei An Dong Wang Zhuocheng Hou Rui Wang Min Guo Zhilong Wang Jiqiang Xu Zhonghong Wu Jianhui Tian 2015Journal of Animal Science and Biotechnology2015,6,3:8
20Sensitivity-enhanced surface plasmon resonance sensor utilizing a tungsten disulfide (WS_2) nanosheets overlayer显示文摘Tungsten disulfide(WS_2), as a representative layered transition metal dichalcogenide(TMDC) material, possesses important potential for applications in highly sensitive sensors. Here, a sensitivity-enhanced surface plasmon resonance(SPR) sensor with a metal film modified by an overlayer of WS_2 nanosheets is proposed and demonstrated. The SPR sensitivity is related to the thickness of the WS_2 overlayer, which can be tailored by coating a WS_2 ethanol suspension with different concentrations or by the number of times of repeated post-coating.Benefitting from its large surface area, high refractive index, and unique optoelectronic properties, the WS_2 nanosheet overlayer coated on the gold film significantly improves the sensing sensitivity. The highest sensitivity(up to 2459.3 nm∕RIU) in the experiment is achieved by coating the WS_2 suspension once. Compared to the case without a WS_2 overlayer, this result shows a sensitivity enhancement of 26.6%. The influence of the WS_2 nanosheet overlayer on the sensing performance improvement is analyzed and discussed. Moreover, the proposed WS_2 SPR sensor has a linear correlation coefficient of 99.76% in refractive index range of 1.333 to 1.360.
