1Cytokines in adipose-derived mesenchymal stem cells promote the healing of liver disease显示文摘Adipose-derived mesenchymal stem cells(ADSCs) are a treatment cell source for patients with chronic liver injury. ADSCs are characterized by being harvested from the patient's own subcutaneous adipose tissue, a high cell yield(i.e., reduced immune rejection response), accumulation at a disease nidus, suppression of excessive immune response, production of various growth factors and cytokines, angiogenic effects, antiapoptotic effects, and control of immune cells via cellcell interaction. We previously showed that conditioned medium of ADSCs promoted hepatocyte proliferation and improved the liver function in a mouse model of acute liver failure. Furthermore, as found by many other groups, the administration of ADSCs improved liver tissue fibrosis in a mouse model of liver cirrhosis. A comprehensive protein expression analysis by liquid chromatography with tandem mass spectrometry showed that the various cytokines and chemokines produced by ADSCs promote the healing of liver disease. In this review, we examine the ability of expressed protein components of ADSCs to promote healing in cell therapy for liver disease. Previous studies demonstrated that ADSCs are a treatment cell source for patients with chronic liver injury. 2Mitochondrial DNA mutations-candidate biomarkers for breast cancer diagnosis in Bangladesh显示文摘Breast cancer is a major health problem that affects more than 24% of women in Bangladesh. Furthermore, among low-income countries including Bangladesh, individuals have a high risk for developing breast cancer. This study aimed to identify candidate mitochondrial DNA (mtDNA) biomarkers for breast cancer diagnosis in Bangladeshi women to be used as a preventive approach. We screened the blood samples from 24 breast cancer patients and 20 healthy controls to detect polymorphisms in the D-loop and the ND3-and ND4-coding regions of mtDNA by direct sequencing. Among 14 distinct mutations, 10 polymorphisms were found in the D-loop, 3 were found in the ND3-coding region, and 1 was found in the ND4-coding region. The frequency of two novel polymorphisms in the D-loop, one at position 16290 (Tins) and the other at position 16293 (A-del), was higher in breast cancer patients than in control subjects (position 16290: odds ratio = 6.011, 95% confidence interval = 1.2482 to 28.8411, P = 0.002; position 16293: odds ratio = 5.6028, 95% confidence interval = 1.4357 to 21.8925, P = 0.010). We also observed one novel mutation in the ND3-coding region at position 10316 (A > G) in 69% of breast cancer patients but not in control subjects. 3Anti-endothelial cell antibody rich sera from rheumatic heart disease patients induces proinflammatory phenotype and methylation alteration in endothelial cells显示文摘Rheumatic heart disease(RHD)is a major cause of cardiovascular morbidity and mortality in developing nations like India.RHD commonly affects the mitral valve which is lined by a single layer of endothelial cells(ECs).The role of ECs in mitral valve damage during RHD is not well elucidated.In here,anti-endothelial cell antibody from RHD patients has been used to stimulate the ECs(HUVECs and HMVECs).ECs proinflammatory phenotype with increased expression of TNFa,IL-6,IL-8,IFNg,IL-1b,ICAM1,VCAM1,E-selectin,laminin B,and vimentin was documented in both ECs.The promoter hypomethylation of various key inflammatory cytokines(TNFa,IL-6,and IL-8),integrin(ICAM1)associated with leukocyte transendothelial migration,and extracellular matrix genes(vimentin,and laminin)were also observed.Further,the in-vitro data was in accordance with ex-vivo observations which correlated significantly with the etiological factors such as smoking,socioeconomic status,and housing.Thus,the study sheds light on the role of ECs in RHD which is a step forward in the elucidation of disease pathogenesis.Mukul Rastogi Subendu Sarkar Ankita Makol Rana Sandip Singh Uma Nahar Saikia Dibyajyoti Banerjee Seema Chopra Anuradha Chakraborti 2018Genes & Diseases2018,5,3:5
4Non-invasive diagnosis of H pylori infection: Evaluation of serological tests with and without current infection marker CIM显示文摘AIM:To evaluate the performance of commercially available immunochromatographic (ICT) and immunoblot tests covering the current infection marker CIM and conventional ELISA for the diagnosis of H pylori infection in adult dyspeptic patients. METHODS:Consecutive non-treated dyspeptic patients undergoing diagnostic endoscopy were tested for H pylori infection by culture, rapid urease test, and histology of gastric biopsy specimens. Serum from 61 H pylori infected and 21 non-infected patients were tested for anti-H pylori IgG antibodies by commercial ELISA (AccuBindTM ELISA, Monobind, USA), ICT (Assure H pylori Rapid Test, Genelabs Diagnostics, Singapore), and immunoblot (Helico Blot 2.1, Genelabs Diagnostics, Singapore) assays. ICT and immunoblot kits cover CIM among other parameters and their performance with and without CIM was evaluated separately. RESULTS:Sensitivity, specificity, positive predictive value (PPV), negative predictive value (NPV), and accuracy of ELISA were 96.7%, 42.8%, 83.1%, 81.8%, and 82.9%, of ICT were 90.1%, 80.9%, 93.2%, 73.9%, and 87.8%, of ICT with CIM were 88.5%, 90.4%, 96.4%, 73.0%, and 89.0%, of immunoblot were 98.3%, 80.9%, 93.7%, 94.4%, and 93.9%, and of immunoblot with CIM were 98.3%, 90.4%, 96.7%, 95.0%, and 96.3%, respectively. CONCLUSION:Immunoblot with CIM had the best performance. 5Evaluation of a multiplex PCR assay for detection of cytomegalovirus in stool samples from patients with ulcerative colitis显示文摘AIM: To evaluate a multiplex PCR assay for the detection of bacterial and viral enteropathogens in stool samples from patients with ulcerative colitis(UC). METHODS: We prospectively analyzed 300 individuals, including immunocompetent patients, immunocompromised patients, and patients with UC. Stool samples were collected from the recto-sigmoid region of the colon by endoscopy. The samples were qualitatively analyzed for bacterial and viral enteropathogens with a multiplex PCR assay using a Seeplex® Kit. Additional clinical and laboratory data were collected from the medical records. RESULTS: A multiplex PCR assay detected 397 pathogens(191 bacteria and 206 viruses) in 215 samples(71.7%). The most frequently detected bacteria were Escherichia coli H7, 85(28.3%); followed by Aeromonas spp., 43(14.3%); and Clostridium perfringens, 36(12.0%) samples. The most prevalent viruses were Epstein-Barr virus(EBV), 90(30.0%); followed by human herpes virus-6(HHV-6), 53(17.7%); and cytomegalovirus(CMV), 37(12.3%) samples. The prevalence rate of CMV infection was significantly higher in the immunocompromised group than in the immunocompetent group(P < 0.01). CMV infection was more common in patients with UC(26/71; 36.6%)than in the immunocompetent patients excluding UC(6/188; 3.2%)(P < 0.01). CMV infection was more prevalent in UC active patients(25/58; 43.1%) than in UC inactive patients(1/13; 7.7%)(P < 0.05). Among 4 groups which defined by the UC activity and immunosuppressive drugs, the prevalence rate of CMV infection was highest in the UC active patients with immunosuppressive drugs(19/34; 55.8%). EpsteinBarr virus(EBV) infection was more common in the immunocompromised patients excluding UC(18/41; 43.9%) than in the immunocompetent patients excluding UC(47/188; 25.0%)(P < 0.05). The simultaneous presence of CMV and EBV and/or HHV6 in UC active patients(14/58; 24.1%) was greater than in immunocompromised patients excluding UC(5/41; 12.2%)(P < 0.05). 6肾透明细胞癌CT增强程度与微血管及成熟血管多参数的关系研究显示文摘目的 探讨肾脏透明细胞癌(RCC)CT增强程度与微血管及成熟血管多种血管特征之间的关系.方法 选择在本院行平扫及三期增强CT扫描并经手术病理证实的RCC患者37例(37个病灶)进行回顾性分析,测量动脉期、静脉期和延迟期RCC的绝对增强值(增强CT值-同部位平扫CT值)和相对增强值(绝对增强值÷同期正常肾实质增强值),同时将病灶分为富血管和乏血管组.CD34和a-SMA免疫组化染色分别测量微血管和成熟血管的数目、平均周长、面积及直径、形态指数、总面积等.对各CT测量参数与病理学指标之间的关系进行分析.结果 在动脉、静脉和延迟期,RCC成熟血管和微血管的数目及总面积均与其绝对强化值正相关(P〈0.05),成熟血管及微血管的数目均与相对强化值正相关(P〈0.05);在静脉和延迟期,成熟血管的平均面积、平均周长、平均直径则均与相对强化值负相关(P〈0.05),而其余血管参数与绝对和相对强化值之间未见相关性(P〉0.05).在富血供和乏血管RCC之间,成熟血管和微血管的数目均具有统计学差异(P〈0.05).结论 RCC的绝对增强值和相对增强值与微血管和成熟血管的一些特定指标密切相关,利用CT增强参数可对这些病理学血管指标进行大致评估,同时也拓展了RCC复杂CT增强表现的理论基础.Ara Shamim 杨传彬 方家杨 Nahar Nur Mohammad Noor 于德新 马祥兴 2017实用放射学杂志2017,33,7:3
7Antioxidant Protection Mechanisms Reveal Significant Response in Drought-Induced Oxidative Stress in Some Traditional Rice of Assam, India显示文摘Drought tolerance levels and antioxidant protection mechanisms were evaluated for 21 traditional rice varieties of Assam, India, along with Sahbhagi Dhan(drought tolerant) and IR64(drought sensitive) as controls. Drought was imposed in hydroponic culture with polyethylene glycol 6000(PEG6000) that was initially standardized with different concentrations. All the rice varieties showed apparent decreases in growth characteristics under drought stress(initially at 15% for 7 d followed by 20% PEG6000 for 7 d in Yoshida medium). On the basis of standard evaluation score(SES), eight rice varieties showed high drought tolerance which were carried forward for further biochemical analyses. Based on different morphophysiological parameters, SN03(Bora), SN04(Prosad Bhog), SN05(Kola Joha), SN06(Helash Bora), SN08(Salihoi Bao), SN12(Kola Amona), SN20(Ronga Bora) and SN21(Sok-Bonglong) were identified as promising drought tolerant varieties. The non-enzymatic antioxidants activities viz., glutathione, ascorbate and enzymatic antioxidant activities such as superoxide dismutase(SOD), catalase(CAT), guaiacol peroxidase(GPX), ascorbate peroxidase(APX), glutathione reductase(GR) in shoots and roots of all the selected varieties revealed significant level of protection mechanisms as compared with controls. 8Effects of variation in seed sources and pre-sowing treatments on seed germination of Tamarindus indica: a multi-purpose tree species in Bangladesh显示文摘We investigated the morphology and germination of Tamarindus indica seeds in order to discover the effects of variation in seed sources and pre-sowing treatments on germination. The experiment was carried out in the nursery of the Forestry and Wood Technology Discipline, Khulna University, Bangladesh. Seeds were collected from different but healthy trees from various locations in Bangladesh and subjected to four pre-sowing treatments, i.e., control, immersion in cold water (4°C for 24 h), immersion in hot water (80°C for 10 min) and scarification with sand paper. Seed germination was carried out in poly-bags with a mixture of topsoil and cow dung in the ratio of 3:1. The average length of the fresh seeds was 1.35±0.26 cm, their width 1.07 ±0.20 cm and thickness 0.69±0.11 cm. The results revealed that pre-sowing treatments affected the germination process of seeds, which significantly increased the germination percentage with the cold water (81.67%) and scarification with sand paper (82.33%) treatments, compared with those in the control (58.33%) and hot water treatment (59.00%). In all treatments germination started on average within 5 to 7 days after the start of the treatments and were completed from 13 to 19 days later. The greatest success in germination (82.33%) was found in scarification with sand paper, followed with 81.67% in the cold water treatment. The results of analyses of variance (ANOVA) showed significant differences (p<0.05) in germination closing dates, seed germination periods and germination percentages among the treatments but no significant difference among seed sources. However, the effect of the interaction between seed source variation and pre-treatment differed significantly in seed germination closing dates and germination percentages. We recommend cold water treatment (submersion of seed at a temperature of 4°C for a 24-h period) for seed germination of T. indica in rural Bangladesh.Md. Salim AZAD Nurun NAHAR Md. 10孟加拉国Bhanugach西部保育林用地类型变化(英文)显示文摘应用遥感和地理信息系统调查了孟加拉国Sylhet林业分局所辖的Bhanugach西部保育林从1988年到2006年间用地类型的变化。应用了监督和非监督两种方法,用全球定位系统搜集了地面控制点。地理信息系统软件制成地图。结果表明,植被覆盖从1988年的1826hm2大幅地降低到1996年的1714.85hm2,随着协同管理措施的实施,植被覆盖自1996年后逐年增长到2006年的1847.83hm2。在没有其他类型的用地转变成裸地的情况下,裸地变化与植被覆盖间成反比。18年间水体面积从1988年的307.67hm2增加到1996年379.53hm2,96-97年间略有下降,1997年以后直到2006年基本保持不变。本文还提出了将遥感技术和地理信息系统应用于用地类型变化及地理信息系统数据库创建的一些建议。Md. 11Study of cyanide removal from contaminated water using zinc peroxide nanomaterial显示文摘The present study highlights the potential application of zinc peroxide(ZnO_2)nanomaterial as an efficient material for the decontamination of cyanide from contaminated water. A process patent for ZnO_2 synthesis has been granted in United States of America(US Patent number 8,715,612; May 2014),South Africa,Bangladesh,and India. The ZnO_2 nanomaterial was capped with polyvinylpyrrolidone(PVP)to control the particle size. The PVP capped ZnO_2nanomaterial(PVP-ZnO_2)before and after adsorption of cyanide was characterized by scanning electron microscope,transmission electron microscope,X-ray diffractometer,Fourier transform infrared spectroscopy and time of flight-secondary ion mass spectrometry. The remaining concentration of cyanide after adsorption by PVP-ZnO_2 was determined using ion chromatograph. The adsorption of cyanide over PVP-ZnO_2 was also studied as a function of p H,adsorbent dose,time and concentration of cyanide. The maximum removal of cyanide was observed in p H range 5.8–7.8 within 15 min. The adsorption data was fitted to Langmuir and Fruendlich isotherm and it has been observed that data follows both the isotherms and also follows second order kinetics.Himani Uppal S. 12Potassium-Induced Regulation of Cellular Antioxidant Defense and Improvement of Physiological Processes in Wheat under Water Deficit Condition显示文摘Drought is the most common form of abiotic stress that reduces plant growth and productivity.It causes plant injuries through elevated production of reactive oxygen species(ROS).Potassium(K)is a vital plant nutrient that notably ameliorates the detrimental effect of drought stress in the plant.A pot experiment was conducted at the Laboratory of Plant Stress Responses,Faculty of Agriculture,Kagawa University,Japan,under controlled environment of green house to explore the role of K in mitigating drought severity in wheat(Triticum asevitum L.)seedlings.Three days after germination,seedlings were exposed to three water regimes viz.,100,50,and 20%field capacity(FC)for 21 days.Potassium was adjusted in Hoagland nutrient solution at 0,6 and 12 mM concentration and applied to pot instead of normal water.Results show that,water deficit stress notably reduced plant growth,biomass accumulation,leaf relative water content(RWC)along with reduced photosynthetic pigments.Increased amount of biochemical stress markers viz.,malondialdehyde(MDA),hydrogen peroxide(H_(2)O_(2)),methylglyoxal(MG),proline(Pro)as well as an impaired antioxidant defense system were observed in drought affected wheat plants.On the contrary,K supplementation resulted in improvement of biochemical and physiological parameters that worked behind in improving growth and development of the wheat plants.In addition,enzymes of ascorbateglutathione(AsA-GSH)cycle were also enhanced by supplemented K that accelerated the ROS detoxification process in plant.Although glyoxalse system did not performed well till MG was detoxified might following another short stepped pathways.Our results revealed that drought stressed plants showed better performances in terms of biochemical and physiological attributes,antioxidant defense and glyoxalase system,as well as ROS detoxification due to K supplementation with better performance at 12 mM K added in 50%FC growing condition.Abdul Awal Chowdhury Masud MdFazlul Karim M.H.M.Borhannuddin Bhuyan Jubayer Al Mahmud Kamrun Nahar Masayuki Fujita Mirza Hasanuzzaman 2021Phyton-International Journal of Experimental Botany2021,90,2:2
13Comparative study of leaching of silver nanoparticles from fabric and effective effluent treatment显示文摘Nano silver (Agn) is employed as an active antimicrobial agent, but the environmental impact of Agn released from commercial products is unknown. The quantity of nanomaterial released from consumer products during use should be determined to assess the environmental risks of advancement of nanotechnology. This work investigated the amount of silver released from three different types of fabric into water during washing. Three different types of fabric were loaded with chemically synthesized Ag nanoparticles and washed repeatedly under simulated washing conditions. Variable leaching rates among fabric types suggest that the manufacturing process may control the release of silver reaching the waste water treatment plants. In an attempt to recover the Agn for reutilization and to save it from polluting water, the effluents from the wash were efficiently treated with bacterial strains. This treatment was based on biosorption and was very efficient for the elimination of silver nanoparticles in the wash water.
