1Erianin,a novel dibenzyl compound in Dendrobium extract,inhibits lung cancer cell growth and migration via calcium/calmodulin-dependent ferroptosis显示文摘Ferroptosis,a novel form of programmed cell death,is characterized by iron-dependent lipid peroxidation and has been shown to be involved in multiple diseases,including cancer.Stimulating ferroptosis in cancer cells may be a potential strategy for cancer therapy.Therefore,ferroptosis-inducing drugs are attracting more attention for cancer treatment.Here,we showed that erianin,a natural product isolated from Dendrobium chrysotoxum Lindl,exerted its anticancer activity by inducing cell death and inhibiting cell migration in lung cancer cells.Subsequently,we demonstrated for the first time that erianin induced ferroptotic cell death in lung cancer cells,which was accompanied by ROS accumulation,lipid peroxidation,and GSH depletion.The ferroptosis inhibitors Fer-1 and Lip-1 but not Z-VAD-FMK,CQ,or necrostatin-1 rescued erianin-induced cell death,indicating that ferroptosis contributed to erianin-induced cell death.Furthermore,we demonstrated that Ca^(2+)/CaM signaling was a critical mediator of erianin-induced ferroptosis and that blockade of this signaling significantly rescued cell death induced by erianin treatment by suppressing ferroptosis.Taken together,our data suggest that the natural product erianin exerts its anticancer effects by inducing Ca^(2+)/CaMdependent ferroptosis and inhibiting cell migration,and erianin will hopefully serve as a prospective compound for lung cancer treatment.Peng Chen Qibiao Wu Jiao Feng Lili Yan Yitian Sun Shuiping Liu Yu Xiang Mingming Zhang Ting Pan Xiaying Chen Ting Duan Lijuan Zhai Bingtao Zhai Wengang Wang Ruonan Zhang Bi Chen Xuemeng Han Yicong Li Liuxi Chen Ying Liu Xingxing Huang Ting Jin Wenzheng Zhang Hong Luo Xiaohui Chen Yongqiang Li Qiujie Li Guohua Li Qin Zhang Lvjia Zhuo Zuyi Yang Huifen Tang Tian Xie Xiaoping Ouyang Xinbing Sui 2020Signal Transduction and Targeted Therapy2020,5,1:42
2Chinese Trauma Surgeon Association for management guidelines of vacuum sealing drainage application in abdominal surgeries-Update and systematic review显示文摘Vacuum sealing drainage (VSD) is frequently used in abdominal surgeries. However, relevant guidelines are rare. Chin ese Trauma Surge on Associati on orga nized a committee composed of 28 experts across China in July 2017, aiming to provide an evidence-based recommendation for the application of VSD in abdominal surgeries.Eleven questions regarding the use of VSD in abdominal surgeries were addressed:(1) which type of materials should be respectively chosen for the intraperitoneal cavity, retroperitoneal cavity and superficial incisions?(2) Can VSD be preventively used for a high-risk abdominal incision w让h primary suture?(3) Can VSD be used in severely contaminated/infected abdominal surgical sites?(4) Can VSD be used for temporary abdominal cavity closure under some special conditions such as severe abdominal trauma, infection, liver transplantation and intra-abdominal volume increment in abdominal compartment syndrome?(5) Can VSD be used in abdominal organ inflammation, injury, or postoperative drainage?(6) Can VSD be used in the treatment of intestinal fistula and pancreatic fistula?(7) Can VSD be used in the treatment of intra-abdominal and extra-peritoneal abscess?(8) Can VSD be used in the treatment of abdominal wall wounds, wound cavity, and defects?(9) Does VSD in crease the risk of bleeding?(10) Does VSD increase the risk of intestinal wail injury?(11) Does VSD increase the risk of peritoneal adhesion? Focusing on these questions, evidence-based recommendations were given accordingly. VSD was strongly recommended regarding the questions 2-4. Weak recommendations were made regarding questions 1 and 5-11. 4Serum levels of mi RNA in patients with hepatitis B virus-associated acute-on-chronic liver failure显示文摘Background: Hepatitis B virus(HBV)-associated acute-on-chronic liver failure(HBV-ACLF) is a lifethreatening condition and its exact pathophysiology and progression remain unclear. The present study aimed to assess the role of serum mi RNAs in the evaluation of HBV-ACLF and to develop a model to predict the outcomes for ACLF.Methods: Serum was collected from 41 chronic hepatitis B and 55 HBV-ACLF patients in addition to30 chronic asymptomatic HBV carriers as controls. The mi RNAs expressions were measured by real-time quantitative PCR(q-PCR). Statistical analyses were conducted to assess the ability of differentially expressed mi RNAs and other prognostic factors in identifying ACLF prognosis and to develop a new predictive model.Results: Real-time q-PCR indicated that serum miR-146 a-5 p, mi R-122-3 p and mi R-328-3 p levels were significantly upregulated in ACLF patients compared to chronic hepatitis B and chronic asymptomatic HBV carriers patients. In addition, multivariate regression analyses indicated that Na+, INR, gastrointestinal bleeding and mi R-122-3 p are all independent factors that are reliable and sensitive to the prognosis of HBV-ACLF. Therefore, we developed a new model for the prediction of HBV-ACLF disease state: Y = 0.402 × Na+-1.72 × INR-4.963 × gastrointestinal bleeding(Yes = 0; No = 1)-0.278 ×(mi R-122-3 p) + 50.449. 6Anti-loosening performance of coatings on fasteners subjected to dynamic shear load显示文摘This paper investigates the self-loosening of threaded fasteners subjected to dynamic shear load. Three kinds of typical coatings, PTFE, MoS_2, and TiN applied to bolts and nuts, are tested in this investigation. The study experimentally examines the loosening mechanisms of fasteners and assesses the anti-loosening performance of the three tested coatings based on their tightening characteristics, loosening curves, and the damage of thread surface. Additionally, the anti-loosening performance of the three coatings is compared under different load forms. The results indicate that the PTFE and MoS_2 coatings have significant anti-loosening effect, whereas the anti-loosening performance of Ti N coating is not satisfactory. It is also found that an appropriate increase of the initial tightening torque can significantly improve the anti-loosening effect. 7Pharmacokinetics of the prototype and hydrolyzed carboxylic forms of ginkgolides A, B, and K administered as a ginkgo diterpene lactones meglumine injection in beagle dogs显示文摘Ginkgo diterpene lactones meglumine injection(GDLI)is a commercially available product used for neuroprotection.However,the pharmacokinetic properties of the prototypes and hydrolyzed carboxylic forms of the primary components in GDLI,i.e.,ginkgolide A(GA),ginkgolide B(GB),and ginkgolide K(GK),have never been fully evaluated in beagle dogs.In this work,a simple,sensitive,and reliable method based on ultra-fast liquid chromatography-tandem mass spectrometry(UFLC-MS/MS)was developed,and the prototypes and total amounts of GA,GB,and GK were determined in beagle dog plasma.The plasma concentrations of the hydrolyzed carboxylic forms were calculated by subtracting the prototype concentrations from the total lactone concentrations.For the first time,the pharmacokinetics of GA,GB,and GK were fully assessed in three forms,i.e.,the prototypes,the hydrolyzed carboxylic forms,and the total amounts,after intravenous administration of GDLI in beagle dogs.It was shown that ginkgolides primarily existed in the hydrolyzed form in plasma,and the ratio of hydrolysates to prototype forms of GA and GB decreased gradually to a homeostatic ratio.All of the three forms of the three ginkgolides showed linear exposure of AUC to the dosages.GA,GB,and GK showed a constant half-life approximately 2.7,3.4,and 1.2 h,respectively,which were consistent for the forms at three dose levels(0.3,1.0,and 3.0 mg·kg^(-1))and after a consecutive injection of GDLI for 7 days(1.0 mg·kg^(-1)).WANG Shu-Yao A Ji-Ye FEI Fei GENG Jian-Liang PENG Ying OUYANG Bing-Chen WANG Pei JIN Xiao-Liang ZHAO Yu-Qing WANG Jian-Kun GENG Ting LI Yan-Jing HUA NG Wen-Zhe WANG Zhen-Zhong XIAO Wei WANG Guang-Ji 2017Chinese Journal of Natural Medicines2017,15,10:9

9The Effects of Different Sex-Linked Dwarf Variations on Chinese Native Chickens显示文摘Variants in chicken growth hormone receptor (GHR) gene lead to sex-linked dwarf (SLD) chickens, but effects of different variants are distinct. In this study, 11 SLD chicken breeds or strains including 3 Chinese native breeds and 8 breeding strains were studied in order to investigate the effects of different sex-linked dwarf variations on growth performance. The results showed that there were three reasons which could lead to dwarfism in the 11 breeds or strains. Firstly, an about 1.7 kb deletion of growth hormone receptor (GHR) gene leads to dwarfism in Jiangxi dwarf chicken, strains GF24, GF26, N308, N309, and N310. Secondly, a T354C mutation in exon 5 of the GHR gene leads to dwarfism in strains N301 and N305. Thirdly, an unknown variant leads to dwarfism in Guizhou Yellow Dwarf chicken and Yixing Bantam chicken. In addition, all individuals of N303 had the 1.7 kb deletion of the GHR gene, and additionally, some of them also carried the T354C mutation. As far as the performance of individuals were compared among T354C homozygote, deletion homozygote, and heterozygote carrying both T354C and deletion, it was found that the T354C's impacts on body weight of Chinese chickens were maximum, the body weight of chickens with homozygote T354C was 92.12% of those with heterozygote, and the difference of the body weight between deletion homozygote and heterozygote was not significant. 11A New Fire Detection Method Using a Multi-Expert System Based on Color Dispersion, Similarity and Centroid Motion in Indoor Environment显示文摘In this paper, a video fire detection method is proposed, which demonstrated good performance in indoor environment. Three main novel ideas have been introduced. Firstly, a flame color model in RGB and HIS color space is used to extract pre-detected regions instead of traditional motion differential method, as it’s more suitable for fire detection in indoor environment. Secondly, according to the flicker characteristic of the flame, similarity and two main values of centroid motion are proposed. At the same time, a simple but effective method for tracking the same regions in consecutive frames is established. Thirdly,a multi-expert system consisting of color component dispersion,similarity and centroid motion is established to identify flames.The proposed method has been tested on a very large dataset of fire videos acquired both in real indoor environment tests and from the Internet. 15The role of tocopherol cyclase in salt stress tolerance of rice(Oryza sativa)显示文摘Tocopherols synthesized exclusively by photosynthetic organisms are major antioxidants in biomembranes.In plants,tocopherol cyclase(TC/VTE1) catalyzes the conversion of 2,3-dimethyl-5-phytyl-1,4-benzoquinone(DMPBQ) to γ-tocopherol.In the present study,OsVTE1,which encodes a rice tocopherol cyclase ortholog,was cloned and characterized.OsVTE1 was induced significantly by abiotic stresses such as high salt,H2O2,drought,cold and by the plant hormones ABA and salicylic acid.The tissue-specific expression pattern and OsVTE1-promoter GUS activity assay showed that OsVTE1 was mainly expressed in the leaf,and also could be detected in the root,stem and panicle.Compared with control plants,transgenic plants with OsVTE1 RNA interference(OsVTE1-RNAi) were more sensitive to salt stress whereas,in contrast,transgenic plants overexpressing OsVTE1(OsVTE1-OX) showed higher tolerance to salt stress.The DAB in vivo staining showed that OsVTE1-OX plants accumulated less H2O2 than did control plants.OUYANG ShouQiang HE SiJie LIU Peng ZHANG WanKe ZHANG JinSong CHEN ShouYi 2011Science China(Life Sciences)2011,54,2:6

17Progress review of US-China joint research on advanced technologies for plug-in electric vehicles显示文摘The United States and China are the world's largest automobile markets and oil consumers, and both face a severe challenge to conserve energy and reduce tailpipe emissions. Thus, both countries urgently need to transform conventional internal combustion engines to electrified powertrains. Targeting the advanced core technologies of plug-in electric vehicles(PEVs), a joint research collaboration between China and the US, called the 'Clean Vehicle Consortium'(CVC), was set up in 2010. Six years of collaboration on PEV technologies has resulted in significant progress in three technical areas. Based on CVC publications,we review herein the progress made by the CVC research efforts on three key advanced PEV technologies. This includes the development of a safe battery with an energy density of 260 W h kg^(-1) and a systematic method for designing safe traction battery systems. Thus, a breakthrough in high power density and efficient traction motor systems has occurred. In addition to discussing advanced electric-drive powertrains, we also discuss global energy management strategies that aim to improve PEV energy efficiency. 20Long-term adenosine A1 receptor activation-induced sortilin expression promotes α-synuclein upregulation in dopaminergic neurons显示文摘Prolonged activation of adenosine A1 receptor likely leads to damage of dopaminergic neurons and subsequent development of neurodegenerative diseases.However,the pathogenesis underlying long-term adenosine A1 receptor activation-induced neurodegeneration remains unclear.In this study,rats were intraperitoneally injected with 5 mg/kg of the adenosine A1 receptor agonist N6-cyclopentyladenosine(CPA)for five weeks.The mobility of rats was evaluated by forced swimming test,while their cognitive capabilities were evaluated by Y-maze test.Expression of sortilin,α-synuclein,p-JUN,and c-JUN proteins in the substantia nigra were detected by western blot analysis.In addition,immunofluorescence staining of sortilin andα-synuclein was performed to detect expression in the substantia nigra.The results showed that,compared with adenosine A1 receptor antagonist 8-cyclopentyl-1,3-dipropylxanthine(5 mg/kg)+CPA co-treated rats,motor and memory abilities were reduced,surface expression of sortin andα-synuclein in dopaminergic neurons was reduced,and total sortilin and totalα-synuclein were increased in CPA-treated rats.MN9D cells were incubated with 500 nM CPA alone or in combination with 10μM SP600125(JNK inhibitor)for 48 hours.Quantitative real-time polymerase chain reaction analysis of sortilin andα-synuclein mRNA levels in MN9D cells revealed upregulated sortilin expression in MN9D cells cultured with CPA alone,but the combination of CPA and SP600125 could inhibit this expression.Predictions made using Jasper,PROMO,and Alibaba online databases identified a highly conserved sequence in the sortilin promoter that was predicted to bind JUN in both humans and rodents.A luciferase reporter assay of sortilin promoter plasmid-transfected HEK293T cells confirmed this prediction.After sortilin expression was inhibited by sh-SORT1,expression of p-JUN and c-JUN was detected by western blot analysis.Long-term adenosine A1 receptor activation levels upregulatedα-synuclein expression at the post-transcriptional level by affecting sortilin expression.The online tool Raptor-X-Binding and Discovery Studio 4.5 prediction software predicted that sortilin can bind toα-synuclein.Co-immunoprecipitation revealed an interaction between sortilin andα-synuclein in MN9D cells.Our findings indicate that suppression of prolonged adenosine A1 receptor activation potently inhibited sortilin expression andα-synuclein accumulation,and dramatically improved host cognition and kineticism.This study was approved by the University Committee of Animal Care and Supply at the University of Saskatchewan(approval No.AUP#20070090)in March 2007 and the Animals Ethics Committee of University of South China(approval No.LL0387-USC)in June 2017.Yun-Cheng Lv An-Bo Gao Jing Yang Li-Yuan Zhong Bo Jia Shu-Hui Ouyang Le Gui Tian-Hong Peng Sha Sun Francisco S.Cayabyab 2020Neural Regeneration Research2020,15,4:4
