| 4 | Some Suggested Future Directions ofQuantitative Resource Assessments显示文摘Future quantitative assessments will be expected to estimate quantities, values, and locations of undiscovered mineral resources in a form that conveys both economic viability and uncertainty associated with the resources. Historically, declining metal prices point to the need for larger deposits over time. Sensitivity analysis demonstrates that the greatest opportunity for reducing uncertainty in assessments lies in lowering uncertainty associated with tonnage estimates. Of all errors possible in assessments, these affecting tonnage estimates are by far the most important. Selecting the correct deposit model is the most important way of controlling errors because of the dominance of tonnage-deposit models are the best known predictors of tonnage. Much of the surface is covered with apparently barren rocks and sediments in many large regions. Because many exposed mineral deposits are believed to have been found, a prime concern is the presence of possible mineralized rock under cover. Assessments of areas with resources under cover must rely on extrapolation from surrounding areas, new geologic maps of rocks under cover, or analogy with other well-explored areas that can be considered training tracts. Cover has a profound effect on uncertainty and on methods and procedures of assessments because geology is seldom known and geophysical methods typically have attenuated responses. Many earlier assessment methods were based on relationships of geochemical and geophysical variables to deposits learned from deposits exposed on the surface-these will need to be relearned based on covered deposits. Mineral-deposit models are important in quantitative resource assessments for two reasons: (1) grades and tonnages of most deposit types are significantly different, and (2) deposit types are present in different geologic settings that can be identified from geologic maps. Mineral-deposit models are the keystone in combining the diverse geoscience information on geology, mineral occurrences, geophysics, and geochemistry used in resource assessments and mineral exploration. | 6 | High level of preoperative carbohydrate antigen 19-9 is a poor survival predictor in gastric cancer显示文摘AIM:To assess the clinical significance and the prognostic value of preoperative serum carbohydrate antigen 19-9(CA 19-9)level in gastric cancer.METHODS:Between January 2005 and December2006,1960 patients underwent surgery for histologically confirmed gastric cancer.Of these,163 patients had elevated serum levels of CA 19-9 preoperatively,and1628 patients had normal serum levels of CA 19-9 preoperatively.For this study,325 patients were selected from the group of 1628 patients by age,sex,and cancer stage to serve as controls.Statistically significant differences in survival rates were calculated using the log-rank test.A P value less than 0.05 was considered statistically significant and was determined using SAS software.RESULTS:The baseline characteristics showed some differences between the two groups with regard to histology.Overall survival(OS)in the elevated and nonelevated group was 37.90 and 68.67 mo,respectively(P<0.001).N stage(P=0.001)was a significant predictor of disease-free survival by multivariate analysis.Also,N stage(P<0.001),and the presence of peritoneal metastasis(P<0.001)remained independent factors in predicting OS by multivariate analysis.Additionally,preoperative serum CA 19-9 levels were significantly associated with OS in univariate(P=0.009)and multivariate(P=0.021)analyses.CONCLUSION:Serum CA 19-9 can be considered an independent prognostic factor in predicting OS in patients anticipating surgery for gastric cancer. | A Ra Choi Jun Chul Park Jie-Hyun Kim Sung Kwan Shin Sang Kil Lee Yong Chan Lee Jae Bock Chung | 2013 | World Journal of Gastroenterology2013，19，32： | 8 |
| 7 | Dual probiotic strains suppress high fructose-induced metabolic syndrome显示文摘AIM:To investigate the effect of novel probiotics on the clinical characteristics of high-fructose induced metabolic syndrome.METHODS:Male Wistar rats aged 4 wk were fed a 70% w/w high-fructose diet(n = 27) or chow diet(n = 9) for 3 wk to induce metabolic syndrome,the rats were then randomized into groups and administered probiotic [Lactobacillus curvatus(L.curvatus) HY7601 and Lactobacillus plantarum(L.plantarum) KY1032] at 109 cfu/d or 1010 cfu/d or placebo by oral gavage for 3 wk.Food intake and body weight were measured once a week.After 6 wk,the rats were fasted for 12 h,then anesthetized with diethyl ether and sacrificed.Blood samples were taken from the inferior vena cava for plasma analysis of glucose,insulin,C-peptide,totalcholesterol,triglycerides and thiobarbituric acid-reacting substances.Real-time polymerase chain reaction was performed using mouse-specific Taqman probe sets to assess genes related to fatty acid β-oxidation,lipogenesis and cholesterol metabolism in the liver.Target gene expression was normalized to the housekeeping gene,glyceraldehyde-3-phosphate dehydrogenase.RESULTS:Rodents fed a high-fructose diet developed clinical characteristics of the metabolic syndrome including increased plasma glucose,insulin,triglycerides,total cholesterol and oxidative stress levels,as well as increased liver mass and liver lipids compared to chow fed controls.Probiotic treatment(L.curvatus HY7601 and L.plantarum KY1032) at high(1010 cfu/d) or low dosage(109 cfu/d) lowered plasma glucose,insulin,triglycerides and oxidative stress levels.Only high-dose probiotic treatment reduced liver mass and liver cholesterol.Probiotic treatment reduced lipogenesis via downregulation of SREBP1,FAS and SCD1 mRNA levels and increased β-oxidation via up-regulation of PPARα and CPT2 mRNA levels.CONCLUSION:Probiotic L.curvatus HY7601 and L.plantarum KY1032 combined suppressed the clinical characteristics of high-fructose-induced metabolic syndrome,therefore,may provide a natural alternative for the treatment of diet-induced metabolic syndrome. | Do-Young Park Young-Tae Ahn Chul-Sung Huh Robin A McGregor Myung-Sook Choi | 2013 | World Journal of Gastroenterology2013，19，2： | 5 |

| 10 | Stress and sleep quality in doctors working on-call shifts are associated with functional gastrointestinal disorders显示文摘AIM To investigate the role of sleep quality and psychosocial problems as predictors of functional gastrointestinal disorders(FGIDs) in doctors that work 24 hour-on-call shifts.METHODS In this cross-sectional observation study, using the Rome Ⅲ Questionnaire and Pittsburgh Sleep Quality Index(PSQI), we analyzed 170 doctors with 24 houron-call shifts. RESULTS Among the participants that had experienced a 24 hour-on-call shift within the last 6 mo, 48(28.2%) had FGIDs. Overall prevalence of irritable bowel syndrome(IBS) and functional dyspepsia(FD) were 16.5% and 17.1%, respectively, with 5.3% exhibiting both. Sleep scores(PSQI)(8.79 ± 2.71 vs 7.30 ± 3.43, P = 0.008), the presence of serious psychosocial alarm(83.3%vs 56.6%, P = 0.004), and the proportion of doctors who experienced over two months of recent on-call work(81.2% vs 68.9%, P = 0.044) were significantly different between individuals with or without FGIDs. Multivariate analysis revealed that presenting serious psychosocial alarm was an independent risk factor for prevalence of FD(OR = 5.47, 95%CI: 1.06-28.15, P = 0.042) and poor sleep quality(PSQI ≥ 6) was a predictor of IBS(OR = 4.17, 95%CI: 1.92-19.02, P = 0.016). | 13 | Cancer-associated fibroblast-derived secreted phosphoprotein 1 contributes to resistance of hepatocellular carcinoma to sorafenib and lenvatinib显示文摘Background:Cancer-associated fibroblasts(CAFs)play an important role in the induction of chemo-resistance.This study aimed to clarify the mechanism underlying CAF-mediated resistance to two tyrosine kinase inhibitors(TKIs),sorafenib and lenvatinib,and to identify a novel therapeutic target for overcoming TKI resistance in hepatocellular carcinoma(HCC).Methods:We performed a systematic integrative analysis of publicly available gene expression datasets and whole-transcriptome sequencing data from 9 pairs of CAFs and para-cancer fibroblasts isolated from human HCC and para-tumor tissues,respectively,to identify key molecules that might induce resistance to TKIs.We then performed in vitro and in vivo experiments to validate selected targets and related mechanisms.The associations of plasma secreted phosphoprotein 1(SPP1)expression levels before sorafenib/lenvatinib treatment with progression-free survival(PFS)and overall survival(OS)of 54 patients with advanced HCC were evaluated using Kaplan-Meier and Cox regression analysis.Results:Bioinformatic analysis identified CAF-derived SPP1 as a candidate molecule driving TKI resistance.SPP1 inhibitors reversed CAF-induced TKI resistance in vitro and in vivo.CAF-derived SPP1 activated rapidly accelerated fibrosarcoma(RAF)/mitogen-activated protein kinase(MAPK)and phosphatidylinositol 3-kinase(PI3K)/protein kinase B(AKT)/mammalian target of rapamycin(mTOR)through the integrin-protein kinase C-alpha(PKCα)signaling pathway and promoted epithelial-to-mesenchymal transition(EMT).A high plasma SPP1 level before TKI treatment was identified as an independent predictor of poor PFS(P=0.026)and OS(P=0.047)in patients with advanced HCC after TKI treatment.Conclusions:CAF-derived SPP1 enhances TKI resistance in HCC via bypass activation of oncogenic signals and EMT promotion.Its inhibition represents a promising therapeutic strategy against TKI resistance inHCC.Moreover,plasma SPP1 level before TKI treatment represents a potential biomarker for treatment response prediction. | JungWoo Eun Jung Hwan Yoon Hye Ri Ahn Seokhwi Kim Young Bae Kim Su Bin Lim Won Park TaeWook Kang Geum Ok Baek Moon Gyeong Yoon Ju A Son Ji HyangWeon Soon Sun Kim Hyo Jung Cho Jae Youn Cheong | 2023 | Cancer Communications2023，43，4： | 2 |
