1GPS-constrained inversion of present-day slip rates along major faults of the Sichuan-Yunnan region, China显示文摘A linked-fault-element model is employed to invert for contemporary slip rates along major active faults in the Sichuan-Yunnan region (96°-108°E, 21°-35°N) using the least squares method. The model is based on known fault geometry, and constrained by a GPS-derived horizontal velocity field. Our results support a model attributing the eastward extrusion of the Tibetan Plateau driven mainly by the north-northeastward indentation of the Indian plate into Tibet and the gravitational collapse of the plateau. Resisted by a relatively stable south China block, materials of the Sichuan-Yunnan region rotate clockwise around the eastern Himalayan tectonic syntaxis. During the process the Garzê-Yushu, Xianshuihe, Anninghe, Zemuhe, Daliangshan, and Xiaojiang faults, the southwest extension of the Xiaojiang fault, and the Daluo-Jinghong and Mae Chan faults constitute the northeast and east boundaries of the eastward extrusion, with their left slip rates being 0.3-14.7, 8.9-17.1, 5.1 ± 2.5, 2.8 ± 2.3, 7.1 ± 2.1, 9.4 ± 1.2, 10.1 ± 2.0, 7.3 ± 2.6, and 4.9 ± 3.0 mm/a respectively. The southwestern boundary consists of a widely distributed dextral transpressional zone other than a single fault. Right slip rates of 4.2 ± 1.3, 4.3 ± 1.1, and 8.5 ± 1.7 mm/a are detected across the Nanhua-Chuxiong-Jianshui, Wuliangshan, and Longling-Lancang faults. Crustal deformation across the Longmenshan fault is weak, with short-ening rates of 1.4 ± 1.0 and 1.6 ± 1.3 mm/a across the Baoxing-Beichuan and Beichuan-Qingchuan segments. Northwest of the Longmenshan fault lies an active deformation zone (the Longriba fault) with 5.1±1.2 mm/a right slip across. Relatively large slip rates are detected across a few faults within the Sichuan-Yunnan block: 4.4±1.3 mm/a left slip and 2.7±1.1 mm/a shortening across the Litang fault, and 2.7±2.3 mm/a right-lateral shearing and 6.7±2.3 mm/a shortening across the Yunongxi fault and its surrounding regions. In conclusion, we find that the Sichuan-Yunnan region is divided into more than a dozen active micro-blocks by a large number of faults with relatively slow slip rates. 3Sliding Mode Control with Perturbation Estimation and Hysteresis Compensator Based on Bouc-Wen Model in Tackling Fast-Varying Sinusoidal Position Control of a Piezoelectric Actuator显示文摘In this paper,a sliding mode control with perturbation estimation(SMCPE) coupled with an inverse hysteresis compensator is proposed for the motion tracking control of a microposition system with piezoelectric actuation.The inverse hysteresis compensator is employed to cancel the hysteresis nonlinearity,thus reducing the nonlinear system to a linear system with an inversion error.Then,a SMCPE controller is adopted to deal with all the unmodeled dynamics and disturbances,aiming at improving the dynamic performance and the robustness of system.An experiment of a piezoelectric actuator is presented to demonstrate the feasibility and effectiveness of the proposed control scheme.The result shows that for a fast-rate control input,the proposed method is capable of leading to a good performance of system behavior.GAN Minggang QIAO Zhi LI Yanlong 2016Journal of Systems Science & Complexity2016,29,2:5

6High-throughput screening for amyloid-β binding natural small-molecules based on the combinational use of biolayer interferometry and UHPLC−DAD-Q/TOF-MS/MS显示文摘Discovery of drugs rapidly and effectively is an important aspect for Alzheimer's disease(AD).In this study,a novel high-throughput screening(HTS)method aims at screening the small-molecules with amyloid-β(Aβ)binding affinity from natural medicines,based on the combinational use of biolayer interferometry(BLI)and ultra-high-performance liquid chromatography coupled with diode-array detector and quadrupole/time-of-flight tandem mass spectrometry(UHPLC−DAD-Q/TOF-MS/MS)has been firstly developed.Briefly,the components in natural medicines disassociated from biotinylated Aβwere collected to analyze their potential Aβbinding affinity by UHPLC−DAD-Q/TOF-MS/MS.Here,baicalein was confirmed to exhibit the highest binding affinity with Aβin Scutellaria baicalensis.Moreover,polyporenic acid C(PPAC),dehydrotumulosic acid(DTA),and tumulosic acid(TA)in Kai-Xin-San(KXS)were also identified as potent Aβinhibitors.Further bioactivity validations indicated that these compounds could inhibit Aβfibrillation,improve the viability in Aβ-induced PC-12 cells,and decrease the Aβcontent and improve the behavioral ability in Caenorhabditis elegans.The molecular docking results confirmed that PPAC,DTA,and TA possessed good binding properties with Aβ.Collectively,the present study has provided a novel and effective HTS method for the identification of natural inhibitors on Aβfibrillation,which may accelerate the process on anti-AD drugs discovery and development.Minsong Guo Fengdan Zhu Wenqiao Qiu Gan Qiao Betty Yuen-Kwan Law Lu Yu Jianming Wu Yong Tang Chonglin Yu Dalian Qin Xiaogang Zhou Anguo Wu 2022Acta Pharmaceutica Sinica B2022,12,4:3
7Coordinated transcription of ANRIL and P16 genes is silenced by P16 DNA methylation显示文摘Objective: To investigate the relationship between the transcription of ANRIL, P15, P14 and P16 at the same locus and the regulation mechanism of ANRIL.Methods: Publicly available database of Cancer Cell Line Encyclopedia(CCLE) was used in bioinformatic analyses. Methylation of Cp G islands was detected by denaturing high performance liquid chromatography(DHPLC). Gene transcript levels were determined using quantitative real-time polymerase chain reaction(q RTPCR) assays. An engineered P16-specific transcription factor and DNA methyltransferase were used to induce P16-specific DNA demethylation and methylation.Results: The expression level of ANRIL was positively and significantly correlated with that of P16 but not with that of P15 in the CCLE database. This was confirmed in human cell lines and patient colon tissue samples. In addition, ANRIL was significantly upregulated in colon cancer tissues. Transcription of ANRIL and P16 was observed only in cell lines in which the P16 alleles were unmethylated and not in cell lines with fully methylated P16 alleles.Notably, P16-specific methylation significantly decreased transcription of P16 and ANRIL in BGC823 and GES1 cells. 8Factors Affecting Patient Satisfaction with Community Health Service under the Gatekeeper System:A Cross-sectional Study in Nanjing,China显示文摘The gatekeeper policy has been implemented for approximately ten years on a pilot population in China. It is necessary to assess the satisfaction of patients utilizing community health service(CHS) under the gatekeeper system. Our study showed that the cognition of gatekeeper policy was associated with four dimensions including doctor‐patient relationships, information and support, organization of care, and accessibility(P < 0.001). One or more factors such as gender and self‐perceived health scores also affected their satisfaction. General practitioners must be prepared to focus on these aspects of information and support, organization of care, and accessibility as indicators of potential opportunities for improvement. 9Granulin A synergizes with cisplatin to inhibit the growth of human hepatocellular carcinoma显示文摘OBJECTIVE Granulin A(GRN A),a cytokinesis protein,is derived from proteolysis of progranulin.The previous study in our laboratory has shown that GRN A is able to inhibit cancer cell growth significantly.This study aimed to investigate the effect of combination of GRN A and cisplatin on in vitro and in vivo on the growth of hepatocellular carcinoma.METHODS The in vitro and in vivo antitumor effects of combination of GRN A and Cisplatin were evaluated with MTS assay and subcuta.neous transplantation tumor model.Chou-Talalay method was used to calculate the combination index(CI).Colony formation assay and flow cytometry were used to detect the effects of GRN A on apoptosis.The expression of apoptosis-related proteins were detected by Western blot.RESULTS MTS assay showed that GRN A significantly inhibit hepatocellular carcinoma cells growth with the IC50 of 5.6 μmol·L^(-1),and GRN A combined with cisplatin synergistically inhibit hepatocellular carcinoma proliferation,with the CI<1.The colony-formation assay showed that GRN A significantly enhanced the inhibitory effects of cisplatin on cellular anchorage-independent growth.Flow cytometry showed that GRN A combined with cisplatin synergistically induced apoptosis,with the apoptotic rates of 5.87%,32.74%,35.67% and 67.15% in control,GRN A,Cisplatin,and combination of GRN A and Cisplatin groups,respectively.Western blot confirmed that the two drugs synergistically changed the expressions of proteins related to apoptosis.In vivo experiment indicated that combination of GRN A and cisplatin significantly suppressed tumor growth compared with single drug treatment groups.CONCLUSION The combination of GRN A and cisplatin resulted in synergistic antitumor effects against hepatocellular carcinoma both in vitro and in vivo.Gan QIAO Huan-li XU Ye TIAN Cong LI Xiao LI Xiao-hui LIU Xiu-kun LIN 2018中国药理学与毒理学杂志2018,32,4:2

16Zn-doped CuO nanocomposites inhibit in vitro and in vivo growth of pancreatic cancer by inducing autophagy through AMPK/mTOR pathway显示文摘OBJECTIVE Zn-doped CuO nanocomposites(Zn-CuO NPs) are novel nanoparticles synthesized by our research group.In this study,we assessed the in vitro and in vivo antitumor effects of Zn-CuO NPS on pancreatic cancer cells,as well as the potential mechanisms.METHODS MTS assay was used to detect the effects of Zn-CuO NPS on proliferation pancreatic cancer cells(Panc-mia and Aspc-1).The in vivo antitumor effects of Zn-CuO NPs were detected by xenografts model in nude mice.The effects of Zn-CuO NPS on autophagy were detected bytransmission electron microscopy(TEM) andflow cytometry.Autophagy related proteins were detected by Western blotting.RESULTS Zn-CuO NPS significantly inhibited the proliferation of Panc-mia cells and Aspc-1 cells.In vivo experi.ments showed that Zn-CuO NPS significantly inhibited the tumor growth in nude mice without affecting the body weight of the mice.TEM and flow cytometry showed that Zn-CuO NPS induced autophagy,and significantly increased the number of autophagosome.Western Blot showed that Zn-CuO NPS alterd the expression of autophagy related proteins,such as AMPK,mTORand Beclin-1.Also,AMPK inhibitor could significantly reduce Zn-CuO NPS-induced autophagy pathwayas analyzed byWestern blotting.CONCLUSION The findings suggested that Zn-doped CuO nanocomposites inhibited the in vitro and in vivo growth of pancreatic cancer by inducing autophagy through AMPK/mTOR pathway.Xiao LI Huan-li XU Ye TIAN Gan QIAO Cong LI Xiao-hui LIU Xiu-kun LIN 2018中国药理学与毒理学杂志2018,32,4:1
