| 1 | Assessing inflammation in Chinese subjects with subtypes of heart failure: an observational study of the Chinese PLA Hospital Heart Failure Registry显示文摘Background Inflammation is an important element of the pathophysiological process of heart failure (HF) and is correlated with subtypes of HF. The association between multiple biomarkers of inflammation and HF subtypes in Chinese subjects remains unclear. This study aimed to compare the differences in inflammation biomarkers among Chinese patients with different subtypes of HF who have been identified to date. Methods We included 413 consecutive patients with HF,including 262 with preserved ejection fraction (HFpEF),55 with middle-ranged ejection fraction (HFmrEF) and 96 with reduced ejection fraction (HFrEF). Ten inflammation biomarkers were analyzed and compared according to the HF subtypes. One hundred contemporary non-HF subjects were also recruited as the control group. Moreover,the correlations between the inflammatory biomarkers and left ventricular ejection fraction of the HF subtypes were assessed. Results The mean age of the HF patients was 65.0 ± 12.0 years,65.8% were male. Distinct subtypes of HF demonstrated different inflammation biomarker panels. IL-6,PTX-3,ANGPTL-4 and TNF-α were correlated with HFrEF;IL-1β and PTX-3 were correlated with HFmrEF;and IL-1β and IL-6 were correlated with HFpEF. The multivariable logistic regression showed that IL-1β[relative ratio (RR)= 1.08,95% CI: 1.02–1.15,P = 0.010],IL-6 (RR = 1.03,95% CI: 1.01–1.06,P = 0.016),PTX-3 (RR = 1.31,95% CI: 1.11–1.55,P = 0.001),and ANGPTL-4 (RR = 1.05,95% CI: 1.02–1.07,P < 0.001) were independently associated with HF,while IL-6 (RR = 1.03,95% CI: 1.01–1.04,P = 0.019),PTX-3 (RR = 1.23,95% CI: 1.06–1.43,P = 0.007),and ANGPTL-4 (RR = 1.03,95% CI: 1.01–1.06,P = 0.005) were independently associated with the HF subtype. Conclusions Diverse inflammation biomarkers have multifaceted presentations according to the subtype of HF,which may illustrate the diverse mechanisms of inflammation in Chinese HF patients. | 3 | Automatic analytical approach for the determination of 12 illicit drugs and nicotine metabolites in wastewater using on-line SPE-UHPLC-MS/MS显示文摘In this study,we developed a novel on-line solid phase extraction(SPE)-ultra-high-performance liquid chromatography tandem mass spectrometry(UHPLC-MS/MS)-based analytical method for simultaneously quantifying 12 illicit drugs and metabolites(methamphetamine,amphetamine,morphine,codeine,6-monoacetylmorphine,benzoylecgonine,3,4-methylenedioxymethamphetamine,3,4-methylenedioxyamphetamine,cocaine,ketamine,norketamine,and methcathinone)and cotinine(COT)in wastewater samples.The analysis was performed by loading 2 m L of the sample onto an Oasis hydrophilic-lipophilic balance cartridge and using a cleanup step(5%methanol)to eliminate interference with a total run time of 13 min.The isotope-labeled internal standard method was used to quantify the target substances and correct for unavoidable losses and matrix effects during the on-line SPE process.Typical analytical characteristics used for method validation were sensitivity,linearity,precision,repeatability,recovery,and matrix effects.The limit of detection(LOD)and limit of quantification(LOQ)of each target were set at 0.20 ng/L and 0.50 ng/L,respectively.The linearity was between 0.5 ng/L and250 ng/L,except for that of COT.The intra-and inter-day precisions were<10.45%and 25.64%,respectively,and the relative recovery ranged from 83.74%to 162.26%.The method was used to analyze various wastewater samples from 33 cities in China,and the results were compared with the experimental results of identical samples analyzed using off-line SPE.The difference rate was between 19.91%and-20.44%,and the error range could be considered acceptable.These findings showed that on-line SPE is a suitable alternative to off-line SPE for the analysis of illicit drugs in samples. | Jingyuan Wang Likai Qia Chenzhi Hou Tingting Zhang Mengyi Chen Haitao Meng Mengxiang Su Hui Xu Zhendong Hua Youmei Wang Bin Di | 2021 | Journal of Pharmaceutical Analysis2021,11,6: | 5 |
| 4 | Efficacy and safety of tacrolimus and low-dose prednisone in Chinese children with steroid-resistant nephrotic syndrome显示文摘Background Tacrolimus,a calcineurin inhibitor,is recommended by the recent guidelines from the Kidney Disease Improving Global Outcomes Group as the first-line treatment for steroid-resistant nephrotic syndrome(SRNS),but its clinical application in China is still limited.We investigated the efficacy and safety of tacrolimus combined with low-dose corticosteroids in a population of Chinese children with SRNS.Methods In this prospective non-randomized,non-controlled study,Chinese children with SRNS who failed the previous full-dose prednisone treatment were given tacrolimus(0.1 mg/kg/day)and low-dose prednisone(0.25-0.50 mg/kg/day).We compared the overall remission rate(ORR)and adverse events in the follow-up period with this therapeutic regimen.Results A total of 76 children were enrolled into the study with an average follow-up period of 18±6 months(maximum 36 months).ORR achieved by the first,third,and sixth months was 94.7%,94.7%,and 96.0%,respectively.All patients who attained an initial tacrolimus trough concentration(FK506C0)>6 ng/mL(60.3%)achieved remission.The relative risk of relapse at FK506C0<3 ng/mL compared to 3-6 ng/mL,6-9 ng/mL,and 9-12 ng/mL was 2.3,3.2,and 16.9,respectively.During the follow-up period,adverse effects that had been previously reported were rare.Conclusions Combination of tacrolimus and low-dose prednisone was safe and effective for the treatment of children with SRNS,with high remission rates observed as early as the first month.Relapses were infrequent,but tended to increase significantly with decreases in FK506C0. | Hai-Xia Chen Qia Cheng Fang Li Qing-Nan He Yan Cao Zhu-Wen Yi Xiao-Chuan Wu | 2020 | World Journal of Pediatrics2020,16,2: | 4 |
| 5 | Investigation on the Influence of Refrigerant Charge Amount on the Cooling Performance of Air Conditioning Heat Pump System for Electric Vehicles显示文摘The application of air conditioning heat pump(ACHP) in electric vehicles could lead to significant electrical power saving effect. As for an air conditioning heat pump system for electric vehicles, the influence of refrigerant charge amount should be investigated during the design phase. In this study, experimental method was employed to investigate the influence of the refrigerant charge amount on the performance of the ACHP system. The results showed that the refrigerant charge amount had different influence on the refrigerant properties at various locations within the system. The coefficient of performance and pressure-enthalpy diagram were calculated, and showed a close relationship with refrigerant charge amount under different compressor speeds. The degree of subcooling and the degree of superheating were recorded and the critical charge amount was determined by a typical practical test method. | 6 | Amyloid β-protein differentially affects NMDA receptor-and GABA_A receptor-mediated currents in rat hippocampal CA1 neurons显示文摘Although the aggregated amyloid b-protein (Ab) in senile plaques is one of the key neuropathological features of Alzheimer’s disease (AD), soluble forms of Ab also interfere with synaptic plasticity at the early stage of AD. The suppressive action of acute application of Ab on hippocampal long-term potentiation (LTP) has been reported widely, whereas the mechanism underlying the effects of Ab is still mostly unknown. The present study, using the whole-cell patch clamp technique, investigated the effects of Ab fragments (Ab25–35 and Ab31–35) on the LTP induction-related postsynaptic ligand-gated channel currents in isolated hippocampal CA1 neurons. The results showed a rapid but opposite action of both peptides on excitatory and inhibitory receptor currents. Glutamate application-induced currents weresuppressed by Ab25–35in a dose-dependent manner,andfurtherN-methyl-D-aspartate(NMDA)receptor-mediated currents were selectively inhibited. In contrast, pretreatment with Ab fragments potentiated c-aminobutyric acid (GABA)-induced whole-cell currents. As a control, Ab35–31, the reversed sequence of Ab31–35, showed no effect on the currents induced by glutamate, NMDA or GABA. | 20 | A new diagnostic model of primary open angle glaucoma based on FD-OCT parameters显示文摘AIM: To build a clinical diagnostic model of primary open angle glaucoma(POAG) using the normal probability chart of frequency-domain optical coherence tomography(FD-OCT).METHODS: This is a cross-sectional study. Total 133 eyes from 133 healthy subjects and 99 eyes from 99 early POAG patients were included in the study. The retinal nerve fibre layer(RNFL) thickness parameters of optic nerve head(ONH) and RNFL3.45 scan were measured in one randomly selected eye of each subject using RTVue-100 FD-OCT. Then, we used these parameters to establish the diagnostic models. Four different diagnostic models based on two different area partition strategies on ONH and RNFL3.45 parameters, including ONH traditional area partition model(ONH-T), ONH new area partition model(ONH-N), RNFL3.45 traditional area partition model(RNFL3.45-T) and RNFL3.45 new area partition model(RNFL3.45-N), were built and tested by cross-validation.RESULTS: The new area partition models had higher area under the receiver operating characteristic(AROC; ONH-N:0.990; RNFL3.45-N: 0.939) than corresponding traditional area partition models(ONH-T: 0.979; RNFL3.45-T: 0.881).There was no statistical difference among AROC of ONH-T,ONH-N, and RNFL3.45-N. Nevertheless, ONH-N was the simplest model.CONCLUSION: The new area partition models had higher diagnostic accuracy than corresponding traditional area partition models, which can improve the diagnostic ability of early POAG.
