2Incretins and selective renal sodium-glucose co-transporter 2 inhibitors in hypertension and coronary heart disease显示文摘Hyperglycemia is associated with an increased risk of cardiovascular disease,and the consequences ofintensive therapy may depend on the mechanism of the anti-diabetic agent(s)used to achieve a tight control.In animal models,stable analogues of glucagon-like peptide-1(GLP-1)were able to reduce body weight and blood pressure and also had favorable effects on ischemia following coronary reperfusion.In a similar way,dipeptidyl peptidase IV(DPPIV)showed to have favorable effects in animal models of ischemia/reperfusion.This could be due to the fact that DPPIV inhibitors were able to prevent the breakdown of GLP-1 and glucose-dependent insulinotropic polypeptide,but they also decreased the degradation of several vasoactive peptides.Preclinical data for GLP-1,its derivatives and inhibitors of the DPPIV enzyme degradation suggests that these agents may be able to,besides controlling glycaemia,induce cardio-protective and vasodilator effects.Notwithstanding the many favorable cardiovascular effects of GLP-1/incretins reported in different studies,many questions remain unanswered due the limited number of studies in human beings that aim to examine the effects of GLP-1 on cardiovascular endpoints.For this reason,long-term trials searching for positive cardiovascular effects are now in process,such as the CAROLINA and CARMELINA trials,which are supported by small pilot studies performed in humans(and many more animal studies)with incretin-based therapies.On the other hand,selective renal sodium-glucose co-transporter 2 inhibitors were also evaluated in the prevention of cardiovascular outcomes in type 2 diabetes.However,it is quite early to draw conclusions,since data on cardiovascular outcomes and cardiovascular death are limited and long-term studies are still ongoing.In this review,we will analyze the mechanisms underlying the cardiovascular effects of incretins and,at the same time,we will present a critical position about the real value of these compounds in the cardiovascular system and its protection.Ramiro A Sanchez Hugo Sanabria Cecilia de los Santos Agustin J Ramirez 2015World Journal of Diabetes2015,6,11:5
3Clinical-pathological and molecular characterization of long-term survivors with advanced non-small cell lung cancer显示文摘Objective:Long-term survivors(LS)of non-small cell lung cancer(NSCLC)without driver alterations,displaying an overall survival(OS)of more than 3 years,comprise around 10%of cases in several series treated with chemotherapy.There are classical prognosis factors for these cases[stage,Eastern Cooperative Oncology Group(ECOG),etc.],but more data are required in the literature.In this multi-center study,we focused on LS of advanced NSCLC with OS above 36 months to perform a clinical-pathological and molecular characterization.Methods:In the first step,we conducted a clinical-pathological characterization of the patients.Afterwards,we carried out a genetic analysis by comparing LS to a sample of short-term survivors(SS)(with an OS less than 9 months).We initially used whole-genome RNA-seq to identify differentiating profiles of LS and SS,and later confirmed these with reverse transcription-polymerase chain reaction(RT-PCR)for the rest of the samples.Results:A total of 94 patients were included,who were mainly men,former smokers,having adenocarcinoma(AC)-type NSCLC with an ECOG of 0-1.We obtained an initial differential transcriptome expression,displaying 5 over-and 33 under-expressed genes involved in different pathways:namely,the secretin receptor,surfactant protein,trefoil factor 1(T FF1),serpin,Ca-channels,and Tolllike receptor(TLRs)families.Finally,RT-PCR analysis of 40(20 LS/20 SS)samples confirmed that four genes(surfactant proteins and SFTP)were significantly down-regulated in SS compared to LS by using an analysis of covariance(ANCOVA)model:SFTPA1(P=0.023),SFTPA2(P=0.027),SFTPB{P=0.02),and SFT PC(P=0.047).Conclusions:We present a sequential genetic analysis of a sample of NSCLCLS with no driver alterations,obtaining a differential RNA-seq/RT-PCR profile showing an abnormal expression of SF genes.Juan Moreno-Rubio Santiago Ponce Rosa Alvarez Maria Eugenia Olmedo Sandra Falagan Xabier Mielgo Fatima Navarro Patricia Cruz Luis Cabezon-Gutierrez Carlos Aguado Gonzalo Colmenarejo Marta Munoz-Fernandez de Leglaria Ana Belen Enguita Maria Cebollero Amparo Benito Isabel Alemany Carolina del Castillo Ricardo Ramos Ana Ramirez de Molina Enrique Casado Maria Sereno 2020Cancer Biology & Medicine2020,17,2:4
4Human pluripotent stem cells:From biology to cell therapy显示文摘Human pluripotent stem cells(PSCs),encompassing embryonic stem cells and induced pluripotent stem cells,proliferate extensively and differentiate into virtually any desired cell type.PSCs endow regenerative medicine with an unlimited source of replacement cells suitable for human therapy.Several hurdles must be carefully addressed in PSC research before these theoretical possibilities are translated into clinical applications.These obstacles are:(1) cell proliferation;(2) cell differentiation;(3) genetic integrity;(4) allogenicity;and(5) ethical issues.We discuss these issues and underline the fact that the answers to these questions lie in a better understanding of the biology of PSCs.To contribute to this aim,we have developed a free online expression atlas,Amazonia!,that displays for each human gene a virtual northern blot for PSC samples and adult tissues(http://gffzzecaaedce5cc8476fh59pfxv60oxkf600q.ffgz.tsg.suse.edu.cn).Jean-Marie Ramirez Marilyne Dijon-Grinand Said Assou Sabine Gerbal-Chaloin Samir Hamamah John De Vos 2010World Journal of Stem Cells2010,2,2:2
5Vector-borne disease and climate change adaptation in African dryland socialecological systems显示文摘Background:Drylands,which are among the biosphere's most naturally limiting and environmentally variable ecosystems,constitute three-quarters of the African continent.As a result,environmental sustainability and human development along with vector-borne disease(VBD)control historically have been especially challenging in Africa,particularly in the sub-Saharan and Sahelian drylands.Here,the VBD burden,food insecurity,environmental degradation,and social vulnerability are particularly severe.Changing climate can exacerbate the legion of environmental health threats in Africa,the social dimensions of which are now part of the international development agenda.Accordingly,the need to better understand the dynamics and complex coupling of populations and environments as exemplified by drylands is increasingly recognized as critical to the design of more sustainable interventions.Main body:This scoping review examines the challenge of vector-borne disease control in drylands with a focus on Africa,and the dramatic,ongoing environmental and social changes taking place.Dryland societies persisted and even flourished in the past despite changing climates,extreme and unpredictable weather,and marginal conditions for agriculture.Yet intrusive forces largely out of the control of traditional dryland societies,along with the negative impacts of globalization,have contributed to the erosion of dryland's cultural and natural resources.This has led to the loss of resilience underlying the adaptive capacity formerly widely exhibited among dryland societies.A growing body of evidence from studies of environmental and natural resource management demonstrates how,in light of dryland system's inherent complexity,these factors and top-down interventions can impede sustainable development and vector-borne disease control.Strengthening adaptive capacity through community-based,participatory methods that build on local knowledge and are tailored to local ecological conditions,hold the best promise of reversing current trends.Conclusions:A significant opportunity exists to simultaneously address the increasing threat of vector-borne diseases and climate change through methods aimed at strengthening adaptive capacity.The integrative framework and methods based on social-ecological systems and resilience theory offers a novel set of tools that allow multiple threats and sources of vulnerability to be addressed in combination.Integration of recent advances in vector borne disease ecology and wider deployment of these tools could help reverse the negative social and environmental trends currently seen in African drylands.Bruce A.Wilcox Pierre Echaubard Michel de Garine-Wichatitsky Bernadette Ramirez 2019Infectious Diseases of Poverty2019,8,3:2
