| 1 | Growth inhibition and apoptosis induction Sulindac on Human gastric cancer cells显示文摘AIM: To evaluate the effects of sulindac in inducing growth inhibition and apoptosis of human gastric cancer cells in comparison with human hepatocellular carcinoma (HCC)cells.METHODS: The human gastric cancer cell lines MKN45 and MKN28 and human hepatocellular carcinoma cell lines HepG2and SMMC7721 were used for the study. Anti-proliferative effect was measured by MTT assay, and apoptosis was determined by Hoechst-33258 staining, electronography and DNA fragmentation. The protein of cyclooxygenase-2 (COX(2) and Bcl-2 were detected by Westem dot blotting.RESULTS: Sulindac could initiate growth inhibition and apoptosis of MKN45, MKN28, HepG2 and SMMC7721 cells in a dose-and time-dependent manner. Growth inhibitory activity and apoptosis were more sensitive in HepG2 cells than in SMMC7721 cells, MKN45 and MKN28 cells. After 24hours incubation with sulindac at 2mmol. L-1 and 4mmol.L-1, the level of COX-2 and Bcl-2 protein were lowered in MKN45, SMMC7721 and HepG2 cells but not in MKN28 cells.CONCLUSION: Sulindac could inhibit the growth of gastric cancer cells and HCC cells effectively in vitro by apoptosis induction, which was associated with regression of COX-2and Bcl-2 expression. The growth inhibition and apoptosis of HCC cells were greater then that of human gastric cancer cells. | 2 | Treatment outcomes for different subgroups of nasopharyngeal carcinoma patients treated with intensity-modulated radiation therapy显示文摘Although many studies have investigated intensity-modulated radiation therapy (IMRT) for nasopharyngeal carcinoma (NPC),sample sizes in the reported studies are usually small and different in outcomes in different T and N subgroups are seldom analyzed.Herein,we evaluated the outcomes of NPC patients treated with IMRT and further explored treatment strategy to improve such outcome.We collected clinical data of 865 NPC patients treated with IMRT alone or in combination with chemotherapy,and classified all cases into the following prognostic categories according to different TNM stages:early stage group (T1-2N0-1M0),advanced local disease group (T3-4N0-1M0),advanced nodal disease group (T1-2N2-3M0),and advanced locoregional disease group (T3-4N2-3M0).The 5-year overall survival (OS),local relapse-free survival (LRFS),and distant metastases-free survival (DMFS) were 83.0%,90.4%,and 84.0%,respectively.The early disease group had the lowest treatment failure rate,with a 5-year OS of 95.6%.The advanced local disease group and advanced nodal disease group had similar failure pattern and treatment outcomes as well as similar hazard ratios for death (4.230 and 4.625,respectively).The advanced locoregional disease group had the highest incidence of relapse and death,with a 5-year DMFS and OS of 62.3% and 62.2%,respectively,and a hazard ratio for death of 10.402.Comparing with IMRT alone,IMRT in combination with chemotherapy provided no significant benefit to locoregionally advanced NPC.Our results suggest that the decision of treatment strategy for NPC patients should consider combinations of T and N stages,and that IMRT alone for early stage NPC patients can produce satisfactory results.However,for advanced local,nodal,and locoregional disease groups,a combination of chemotherapy and radiotherapy is recommended. | Sheng-Fa Su 1,2,3,Fei Han 1,2,Chong Zhao 1,2,Ying Huang 1,2,Chun-Yan Chen 1,2,Wei-Wei Xiao 1,2,Jia-Xin Li 1,2 and Tai-Xiang Lu 1,2 1 State Key Laboratory of Oncology in South China,Guangzhou,Guangdong 510060,P.R.China 2 Department of Radiation Oncology,Sun Yat-sen University Cancer Center,Guangzhou,Guangdong 510060,P.R.China 3 Department of Oncology,Guiyang Medical College Hospital,Tumor Hospital of Guizhou,Guiyang,Guizhou 550003,P.R.China. | 2011 | Chinese Journal of Cancer2011,30,8: | 40 |
| 3 | Sr-Nd isotopic compositions of the Changjiang sediments: Implications for tracing sediment sources显示文摘The suspended particulate and fine-grained floodplain sediments were collected from the main stream and tributaries of the Changjiang River for Sr-Nd isotopic measurements. The εNd(0) values gradually decrease downstream from -10.8 on average in the upper reaches to -12.3 in the lower reaches, whereas the 87Sr/86Sr ratios increase correspondingly, averaging 0.721899 and 0.725826 respectively in the upper and middle-lower reaches. The compositional variations primarily reflect the complex con- trols of provenance rocks, chemical weathering, and sediment characters between different catchments, among which the abnormal Sr-Nd isotopic compositions of the Yalong, Fujiang, Tuojiang and Yuanjiang rivers indicate the sediment provenance contributions from the Emeishan Basalt in the upper reaches and the old metamorphic and siliceous rocks in the middle-lower reaches. The Sr-Nd isotopic ratios of the Changjiang sediments can better reflect the average composition of weathered continental crust compared to other major rivers in the world because of the unique source rock types in the Changjiang drainage basin. | 4 | Chronological and geochemical studies of granite and enclave in Baimashan pluton, Hunan, South China显示文摘Zircon LA-ICP-MS U-Pb dating reveals that the Baimashan Pluton is composed mainly of late Indosinian (204.5±2.8 Ma-209.2±3.8 Ma) biotite granodiorites/monzonitic granites (LIGs) and early Yanshanian (176.7±1.7 Ma) two-micas monzonitic granites (EYGs), and the coeval (203.2±4.5 Ma-205.1±3.9 Ma) mafic microgranular enclaves (MMEs) are generally found in the former. In addition, the ages of cores within zircons from LIGs and MMEs ranging from 221.4±4.0 Ma to 226.5±4.1Ma provide evidence of multistage magma intrusion during Indosinian in the study area. Measured 3010±20.6 Ma of inherited zircon age suggests that there may be recycling Archaean curstal material in existence in this area. LIGs and EYGs share some similar geochemical features: subalkaline and peraluminous granites, enrichment of Th, U, K, Ta, Zr, Hf and LREE but depletion of Ba, Nb, P, Ti and Eu, low εNd(t) values but high (87Sr/86Sr)i ratios, and old T2DM (ca. 1.9-2.0 Ga). The behaviors of incompatible elements and REE are mainly dominated by fractional crystallization of plagioclase, K-feldspar, ilmenite and apatite, but that of Sr isotope mainly controlled by EC-AFC. They are crust-sourced and derived from partial melting of paleo-Proterozoic metagreywackes and related to biotite dehydration melting. LIGs are formed in post-collisional tectonic setting as crustal local extension and thinning during late Indosinian. But EYGs may be evolved products of congeneric granitic magma with LIGs formed in late Indoinian, which were emplaced again when crust underwent extensive thinning and extension in post-orogenic tectonic setting during Yanshanian in SC after undergoing EC-AFC. | 6 | Diversity and ecological distribution of endophytic fungi associated with medicinal plants显示文摘A total of 973 isolates of endophytic fungi were recovered from 1144 tissue fragments of the six me-dicinal plant species belonging to 4 families collected in the Beijing Botanical Garden. Of these isolates 778 sporulated and were identified into 21 taxa by morphological characteristics. Among the taxa 11 belonged to Coelomycetes, 6 to Ascomycetes, and 4 to Hyphomycetes. Various numbers of endophytic fungi (5―8 taxa) were obtained from each plant. Alternaria alternata was the dominant species in the 6 plants, and Microsphaeropsis conielloides was also dominant in Eucommia ulmoides. There were high colonization rates (47.9%―63.1%) and isolation rates (0.7―0.93) of endophytic fungi, and they were conspicuously higher in twigs than those in leaves in the 6 plants examined. The colonization and isolation rates of endophytic fungi increased with the twig age. | 10 | Prognosis of 980 patients with gastric cancer after surgical resection显示文摘Background and Objective: Although surgery is the only possible means to cure gastric cancer, the prognosis is often discrepant. The American Joint Committee on Cancer/International Union against Cancer (AJCC/UICC) published the TNM classification of Malignant Tumors (seventh edition) for gastric cancer recently. This study aimed to use this new edition staging system to investigate the prognostic factors for gastric cancer. Methods: The clinicopathologic data of 980 patients with gastric cancer treated by surgical resection in our hospital between January 2000 and December 2006 were analyzed retrospectively. The overall survival rate was determined by using Kaplan-Meier method and log-rank test was used to determine significance. The prognosis was analyzed using univariate analysis and multivariate analysis with the Cox proportional hazards model. The 6th and 7th edition AJCC/UICC TNM staging systems were used to compare the survival outcomes for the cohort of patients. Results: The overall 1-, 3-, 5-year survival rates for the whole group were 82.5%, 58.7%, and 52.6%. The 5-year survival rates for patients with pTNM stage I, II, III, and IV disease classified by the 7th edition staging system were 93.2%, 72.4%, 39.1%, and 5.2%, respectively. In both univariate analysis and Cox multivariate analysis, age, tumor site, tumor size, histological type, resection type, radical resection, lymphatic/venous invasion, depth of invasion, nodal status, metastasis, retrieved lymph nodes, metastatic lymph node ratio, and adjuvant chemotherapy were prognostic factors with these patients. | 12 | Ensemble projection of 1―3℃ warming in China显示文摘Studies on the influences of climate change on biogeochemical cycles and on the key vulnerabilities and the risk from climate change suggest that annual surface temperature rise of 1℃, 2℃ and 3℃ above the present level would lead to changes in extreme weather and climate events, food production, fresh water resources, biodiversity, human mortality, etc. Here two sets of simulations as performed with seventeen atmosphere-ocean general circulation models (AOGCMs) for the Fourth Assessment Report of the Intergovernmental Panel on Climate Change (IPCC AR4), i.e. the model outputs from the 20th Century Climate in Coupled Models (20C3M) and from the Special Report on Emissions Scenarios (SRES) emission scenarios B1, A1B and A2, are used to analyze spatial and temporal characteristics of the above values in China over the 21st century. The results indicate that the rate of warming varies from region to region. The above values are reached much later (earlier) when emission amount is lower (higher), and spread of the time when the lower (higher) value is exceeded is narrower (wider) among the three scenarios. | 14 | APC and K-ras gene mutation in aberrant crypt foci of human colon显示文摘AIM To study the genetic alteration in ACF andto define the possibility that ACF may be a veryearly morphological lesion with molecularchanges, and to explore the relationshipbetween ACF and colorectal adenoma evencarcinoma.METHODS DNA from 35 CRC, 15 adenomas, 34ACF and 10 normal mucus was isolated by meansof microdissection. Direct gene sequencing of K-ras gene including codon 12, 13 and 61 as well asthe mutation cluster region (MCR) of APC genewas performed.RESULTS K-ras gene mutation frequency inACF, adenoma and carcinoma was 17.6% (6/34), 13.3% (2/ 15), and 14.3% (5/ 35)respectively, showing no difference ( P > 0.05)in K-fas gene mutation among three pathologicprocedures. The K-ras gene mutation inadenoma, carcinoma and 4 ACF restricted incodon 12 (GGT→GAT), but the other 2 mutationsfrom ACF located in codon 13 (GGC→GAC). K-res gene mutation was found more frequently inolder patients and patients with polypoidcancer. No mutation in codon 61 was found in thethree tissue types. Mutation rate of APO gene inadenoma and carcinoma was 22.9% (8/35) and26.7% (4/ 15), which was higher than ACF(2.9%) (P < 0.05). APC gene mutation incarcinoma was not correlated with age ofpatients, location, size and differentiation oftumor.CONCLUSION ACF might be a very earlymorphological lesion in the tumorogenesis ofcolorectal tumor. The morphological feature andgene mutation status was different in ACF andadenoma. ACF is possibly putative'microadenoma' that might be the precursor ofadenoma. | 17 | Timing and geochemical characters of the Sanchazi magmatic arc in Mianlüe tectonic zone,South Qinling显示文摘The Sanchazi mafic-ultramafic complex in Mianlüe tectonic zone,South Qinling can be subdivided into two blocks,I.e.Sanchazi paleo-magmatic arc and Zhuangkegou paleo-oceanic crust fragment (ophiolite).The Sanchazi paleo-magmatic arc is mainly composed of ande-site,basaltic and basalt-andesitic gabbro(or diorite),andesitic dyke,plagiogranite and minor ul-tramafric rocks,which have typical geochemical features of island arc volcanic rocks,such as high field strength element(e.g.Nb,Ti)depletions and lower Cr,Ni contents.The Light rare earth element (LREE) and K enrichments of these rocks and zircon xenocrystals of 900 Ma from plagiogranite suggest that this magmatic arc was developed on the South active continental margin of the South Qinling micro-continent.The U-Pb age of (300±61) Ma for zircons from plagiogranite in-dicates that the Mianlüe paleo-oceanic crust was probably subducted underneath the South Qinling micro-continent in Carboniferous.This is consistent with the formation time (309Ma) of the Huwan eclogite originating form oceanic subduction in Dabie Mountains,suggesting that the Mianlüe paleo-ocean probably extended eastward to the Dabie Mountains in Carboniferous.The high-Mg adakitic rocks in Sanchazi paleo-magmatic arc suggest thet the subducted oceanic crust was relatively young (<25Ma) and hot. | LI Shuguang1, HOU Zhenhui1, YANG Yongcheng2, SUN Weidong1,1), ZHANG Guowei3, & LI Qiuli1 1. | 19 | Rb1 protects endothelial cells from hydrogen peroxide-induced cell senescence by modulating redox status显示文摘Objectives Endothelial senescence has been proposed to be involved in endothelial dysfunction and atherogenesis. This study investigates the effects of ginsenoside Rbl, a major constituent of ginseng,on H2O2-induced endothelial senescence.Methods Primary human umbilical vein endothelial cells(HUVECs) senescence was induced by H2O2 as judged by senescence-associated P-galactosidase assay (SA-P-gal).Fntracellur superoxide dismutase(S0D1) activity and malondialdehyde(MDA) level were determined by commercial kit.S0D1 mRNA and protein expression were analyzed by real time PCR and Western blot.Reactive oxygen species(ROS) were determined by flow cytometry.Results Rb1 was found to reverse endothelial senescence,as witnessed by a significant decrease of senescent cell numbers. Rbl could markedly increase intracellular SOD activity, decrease the MDA level,and suppress the generation of intracellular ROS in H2O2-treated HUVECs.Consistent with these findings,Rbl can effectively restore SOD1 mRNA and protein expression which decreased in H2O2 treated cells.
