1Metabolic engineering and flux analysis of Corynebacterium glutamicum for L-serine production显示文摘L-Serine plays a critical role as a building block for cell growth, and thus it is difficult to achieve the direct fermentation of L-serine from glucose. In this study, Corynebacterium glutamicum ATCC 13032 was engineered de novo by blocking and attenuating the conversion of L-serine to pyruvate and glycine, releasing the feedback inhibition by L-serine to 3-phosphoglycerate dehydrogenase (PGDH), in combination with the co-expression of 3-phosphoglycerate kinase (PGK) and feedback-resistant PGDH (PGDHr). The resulting strain, SER-8, exhibited a lower specific growth rate and significant differences in L-serine levels from Phase I to Phase V as determined for fed-batch fermentation. The intracellular L-serine pool reached (14.22±1.41) μmol gCDM 1, which was higher than glycine pool, contrary to fermentation with the wild-type strain. Furthermore, metabolic flux analysis demonstrated that the over-expression of PGK directed the flux of the pentose phosphate pathway (PPP) towards the glycolysis pathway (EMP), and the expression of PGDHr improved the L-serine biosynthesis pathway. In addition, the flux from L-serine to glycine dropped by 24%, indicating that the deletion of the activator GlyR resulted in down-regulation of serine hydroxymethyltransferase (SHMT) expression. 2Oil accumulation related to migration of source kitchens in the Lukeqin structural belt, Turpan-Hami Basin, China显示文摘The Lukeqin structural belt is the main heavy oil accumulation zone in the Turpan-Hami Basin. The recent discovery of light oil in the Triassic indicates that there may be multiple source kitchens contributing to the oil accumulation. According to oil geochemical analysis and oil-source correlation, the oil in deep and shallow reservoirs of the Lukeqin Oilfield presents different physical and saturated hydrocarbon mass spectrum characteristics. The Triassic heavy oil is from the northern Upper Permian lacustrine source rocks, and the light oil represented by the Yudong-9 Well is from the northwestern Lower Jurassic coal-measure source rocks. The timing of oil charging was determined by K/Ar isotope dating, reservoir fluid inclusion analysis and the evolution history of different source rocks. In summary, the accumulation process consists of two stages. From the end of Triassic to early Jurassic, the northern Permian source kitchen generated a considerable amount of oil, which was finally degraded to heavy oil, migrated to the south and then accumulated. The northwestern Jurassic coal-measure source kitchen began to generate oil at the end of Cretaceous, while the northern source kitchen could only generate a little hydrocarbon. The heavy oil and the light oil have different source rock locations, migration directions and accumulation times. 3Trace elements in tree rings and their environmental effects: A case study in Xi’an City显示文摘Using wet digested method and ICP mass spectrometer, we analyzed the concentration of five trace elements (Cd, Mn, P, Zn and Pb) for the tree rings from both urban and suburbs of Xi'an. At the urban sampling site, one Chinese mahogany (Toona sinensis) disc and one phoenix tree (Firmiana simplex) disc were sampled from a steelworks in Xi'an City. At the suburb site, a Chinese mahogany disc was collected from a village in the south of the City. In addition, some soils near the roots of the sampled trees were collected. The analysis results indicate that the concentration of each of the five elements in annual rings has a positive correlation with the production of the steelworks. Statistical calculations show that Pb and Mn elements were stable without lag effects. That means these two elements do not move between rings. Cd displayed one year moving, P two years and Zn three years.
