1速尿、苯氧比酸、硫唑嘌呤对慢性炎症性肠病患者红细胞TPMT活性的影响显示文摘目的:研究利尿剂速尿、苯氧比酸及其硫唑嘌呤对慢性炎症性肠病患者红细胞硫嘌呤甲基转移酶(TPMT)活性的影响。方法:应用高效液相法(HPLC)测定慢性炎症性肠病患者红细胞TPMT活性,其中中等活性、正常活性、较高活性的患者各6例,依据浓度效应曲线计算每个药物的IC50。结果:速尿对TPMT活性的抑制作用较强,平均IC50为15~19μmol·L-1,在健康志愿者的正常血浆浓度范围内。苯氧比酸和硫唑嘌呤的平均IC50值分别为300~313μmol·L-1、430~532μmol·L-1,均远高于体内所能达到的血浆浓度。3种药物的IC50值在3个不同TPMT活性组间无明显差异(P>0.05)。结论:速尿可抑制红细胞TPMT活性,当速尿和硫嘌呤类药物合用时,应警惕发生不良相互作用。辛华雯 Christine Fischer Matthias Schwab Ulrich Klotz 2005中国临床药理学与治疗学2005,10,6:3

7Reduced expression of Ca^(2+)-regulating proteins in the upper gastrointestinal tract of patients with achalasia显示文摘AIM: To compare expression of Ca2+-regulating proteins in upper gastrointestinal (GI) tract of achalasia patients and healthy volunteers and to elucidate their role in achalasia. METHODS: Sarcoplasmic reticulum Ca2+ ATPase (SERCA) isoforms 2a and 2b, phospholamban (PLB), calsequestrin (CSQ), and calreticulin (CRT) were assessed by quantitative Western blotting in esophagus and heart of rats, rabbits, and humans. Furthermore, expression profi les of these proteins in biopsies of lower esophageal sphincter and esophagus from patients with achalasia and healthy volunteers were analyzed. RESULTS: SERCA 2a protein expression was much higher in human heart (cardiac ventricle) compared to esophagus. However, SERCA 2b was expressed predominantly in the esophagus. The highest CRT expression was noted in the human esophagus, while PLB, although highly expressed in the heart, was below our detection limit in upper GI tissue. Compared to healthy controls, CSQ and CRT expression in lower esophageal sphincter and distal esophageal body were signif icantly reduced in patients with achalasia (P < 0.05). CONCLUSION: PLB in the human esophagus mightbe of lesser importance for regulation of SERCA than in heart. 9Soil conditions and phylogenetic relatedness influence total community trait space during early plant succession显示文摘Aims the total space of traits covered by the members of plant com-munities is an important parameter of ecosystem functioning and complexity.We trace the variability of trait space during early plant succession and ask how trait space co-varies with phylogenetic community structure and soil conditions.Particularly,we are inter-ested in the small-scale variability in trait space and the influence of biotic and abiotic filters.Methods We use data on species richness and soil conditions from the first 7 years of initial succession of an artificial catchment in north-east-ern germany.total functional attribute diversity serves as a proxy to total trait space.Important Findingstotal trait space steadily increased during succession.We observed high small-scale variability in total trait space that was positively correlated with species richness and phylogenetic segregation and negatively correlated with total plant cover.trait space increased with soil carbonate content,while pH and the fraction of sandy material behaved indifferently.our results indicate that during early succession,habitat filtering processes gain importance leading to a lesser increase in trait space than expected from the increase in species richness alone.Werner Ulrich Marcin Piwczyński Markus K.Zaplata Susanne Winter Wolfgang Schaaf Anton Fischer 2014Journal of Plant Ecology2014,7,4:1
