| 1 | Impact of co-infections with enteric pathogens on children suffering from acute diarrhea in southwest China显示文摘Background:Acute diarrhea is a global health problem,resulting in high morbidity and mortality in children.It has been suggested that enteric pathogen co-infections play an important role in gastroenteritis,but most research efforts have only focused on a small range of species belonging to a few pathogen groups.This study aimed to assess the impact of co-infections with a broad range of enteric pathogens on children aged below five years who suffer from acute diarrhea in southwest China.Method:A total of 1020 subjects(850 diarrhea cases and 170 healthy controls)were selected from four sentinel hospitals in Kunming,Yunnan province,southwest China,from June 2014 to July 2015.Stool specimens were collected to detect five virus(rotavirus group A,RVA;norovirus,NoV;Sapovirus,SaV;astrovirus,As;and adenovirus,Ad),seven bacterial(diarrheagenic Escherichia coli,DEC;non-typhoidal Salmonella,NTS;Shigella spp.;Vibrio cholera;Vibrio parahaemolyticus;Aeromonas spp.;and Plesiomonas spp.),and three protozoan(Cryptosporidium spp.,Giardia lamblia,and Blastocystis hominis,B.hominis)species using standard microbiologic and molecular methods.Data were analyzed using the partial least square regression technique and chi-square test.Results:At least one enteric pathogen was detected in 46.7%(n=397)of acute gastroenteritis cases and 13.5%(n=23)of healthy controls(χ^(2)=64.4,P<0.05).Single infection with RVA was associated with acute diarrhea(26.5%vs.5.8%,P<0.05).The prevalence of a single infection with B.hominis in diarrhea cases was higher than in healthy controls(3.1%vs.0.5%,OR=4.7,95%CI:1.01–112.0).Single infection with NoV GII was not associated with diarrhea(4.4%vs.3.5%,OR=1.2,95%CI:0.5–3.3).Single infections with bacterial species were not observed.The prevalence of co-infections with two enteric pathogens in diarrhea cases was higher than in asymptomatic children(20.1%vs.5.3%,P<0.05).RVA-NoV GII was the most common co-infection in symptomatic children(4.4%),with it aggravating the severity of diarrhea.Conclusions:Although it is clear that RVA has an overwhelming impact on diarrhea illnesses in children,co-infection with other enteric pathogens appears to also aggravate diarrhea severity.These findings should serve as evidence for public health services when planning and developing intervention programs. | Shun-Xian Zhang Yong-Ming Zhou Wen Xu Li-Guang Tian Jia-Xu Chen Shao-Hong Chen Zhi-Sheng Dang Wen-Peng Gu Jian-Wen Yin Emmanuel Serrano Xiao-Nong Zhou | 2016 | Infectious Diseases of Poverty2016,5,1: | 19 |

| 3 | Consensus on the digestive endoscopic tunnel technique显示文摘With the digestive endoscopic tunnel technique(DETT), many diseases that previously would have been treated by surgery are now endoscopically curable by establishing a submucosal tunnel between the mucosa and muscularis propria(MP). Through the tunnel, endoscopic diagnosis or treatment is performed for lesions in the mucosa, in the MP, and even outside the gastrointestinal(GI) tract.At present, the tunnel technique application range covers the following:(1)Treatment of lesions originating from the mucosal layer, e.g., endoscopic submucosal tunnel dissection for oesophageal large or circular early-stage cancer or precancerosis;(2) treatment of lesions from the MP layer, per-oral endoscopic myotomy, submucosal tunnelling endoscopic resection, etc.; and(3) diagnosis and treatment of lesions outside the GI tract, such as resection of lymph nodes and benign tumour excision in the mediastinum or abdominal cavity. With the increasing number of DETTs performed worldwide, endoscopic tunnel therapeutics, which is based on DETT, has been gradually developed and optimized. However, there is not yet an expert consensus on DETT to regulate its indications, contraindications, surgical procedure, and postoperative treatment.The International DETT Alliance signed up this consensus to standardize the procedures of DETT. | 6 | Antiatherosclerotic effect of dehydrocorydaline on ApoE^(−/−) mice:inhibition of macrophage inflammation显示文摘Despite improvements in cardiovascular disease(CVD)outcomes by cholesterol-lowering statin therapy,the high rate of CVD is still a great concern worldwide.Dehydrocorydaline(DHC)is an alkaloidal compound isolated from the traditional Chinese herb Corydalis yanhusuo.Emerging evidence shows that DHC has anti-inflammatory and antithrombotic benefits,but whether DHC exerts any antiatherosclerotic effects remains unclear.Our study revealed that intraperitoneal(i.p.)injection of DHC in apolipoprotein E-deficient(ApoE^(-/-))mice not only inhibited atherosclerosis development but also improved aortic compliance and increased plaque stability.In addition,DHC attenuated systemic and vascular inflammation in ApoE^(-/-) mice.As macrophage inflammation plays an essential role in the pathogenesis of atherosclerosis,we next examined the direct effects of DHC on bone marrow-derived macrophages(BMDMs)in vitro.Our RNA-seq data revealed that DHC dramatically decreased the levels of proinflammatory gene clusters.We verified that DHC significantly downregulated proinflammatory interleukin(IL)-1βand IL-18 mRNA levels in a time-and concentration-dependent manner.Furthermore,DHC decreased lipopolysaccharide(LPS)-induced inflammation in BMDMs,as evidenced by the reduced protein levels of CD80,iNOS,NLRP3,IL-1β,and IL-18.Importantly,DHC attenuated LPS-induced activation of p65 and the extracellular signal-regulated kinase 1/2(ERK1/2)pathway.Thus,we conclude that DHC ameliorates atherosclerosis in ApoE^(-/-) mice by inhibiting inflammation,likely by targeting macrophage p65-and ERK1/2-mediated pathways. | Bin Wen Yuan-ye Dang Su-hua Wu Yi-min Huang Kong-yang Ma Yi-ming Xu Xi-Long Zheng Xiao-yan Dai | 2022 | Acta Pharmacologica Sinica2022,43,6: | 3 |

| 12 | Phylogeny and taxonomy of Afrocayratia,a new genus of Vitaceae from continental Africa and Madagascar显示文摘The genus Cayratia Juss.in the traditional sense(i.e.,Cayratia s.l.)of the grape family has been shown to be non-monophyletic.Previous studies supported the splitting of Cayratia s.l.into three genera,that is,Cayratia s.s.,Causonis Raf.,and a new genus representing the African Cayratia clade.However,the morphology of the African Cayratia clade has not been studied carefully and its phylogenetic position within Vitaceae remains unclear.Our study integrates molecular,distributional,and morphological data and supports the recognition of the new genus Afrocayratia from continental Africa and Madagascar.Phylogenetic analyses strongly support the monophyly of Afrocayratia and resolve it as a sister of Cayratia s.s.based on the chloroplast data,but it is placed sister to Cyphostemma based on the internal transcribed spacer dataset.Molecular dating suggests that Afrocayratia split with Cayratia s.s.during the Paleocene,but that the extant species of Afrocayratia did not diversify until the early Miocene.Afrocayratia differs from its allied genera in having short stigmas and seeds with subcircular ventral infold cavities in cross-section.Three clades are detected within Afrocayratia,with A.debilis(Baker)J.Wen&L.M.Lu as the first diverged lineage.The second diverged lineage includes A.delicatula(Willems)J.Wen&Z.D.Chen and A.gracilis(Guill.&Perr.)J.Wen&Z.D.Chen.The third diverged lineage includes A.imerinensis(Baker)J.Wen&L.M.Lu,A.longiflora(Desc.)J.Wen&Rabarijaona,and A.triternata(Baker)J.Wen&Rabarijaona from Madagascar,which form a monophyletic group that diverged from the second lineage in the middle Miocene.Combining the morphological and molecular evidence,we formally describe the new genus Afrocayratia,make seven new combinations,and provide a key to species of the genus. | Romer Narindra Rabarijaona Viet-Cuong Dang Gaurav Parmar Bing Liu Jun Wen Zhi-Duan Chen Li-Min Lu | 2020 | Journal of Systematics and Evolution2020,58,6: | 2 |
| 13 | A potent PGK1 antagonist reveals PGK1 regulates the production of IL-1βand IL-6显示文摘Glycolytic metabolism enzymes have been implicated in the immunometabolism field through changes in metabolic status. PGK1 is a catalytic enzyme in the glycolytic pathway. Here, we set up a high-throughput screen platform to identify PGK1 inhibitors. DC-PGKI is an ATPcompetitive inhibitor of PGK1 with an affinity of Kd= 99.08 nmol/L. DC-PGKI stabilizes PGK1in vitro and in vivo, and suppresses both glycolytic activity and the kinase function of PGK1. In addition,DC-PGKI unveils that PGK1 regulates production of IL-1β and IL-6 in LPS-stimulated macrophages.Mechanistically, inhibition of PGK1 with DC-PGKI results in NRF2(nuclear factor-erythroid factor 2-related factor 2, NFE2L2) accumulation, then NRF2 translocates to the nucleus and binds to the proximity region of Il-1β and Il-6 genes, and inhibits LPS-induced expression of these genes. DC-PGKI ameliorates colitis in the dextran sulfate sodium(DSS)-induced colitis mouse model.
