| 1 | Adenovirus-mediated gene delivery:Potential applications for gene and cell-based therapies in the new era of personalized medicine显示文摘With rapid advances in understanding molecular pathogenesis of human diseases in the era of genome sciences and systems biology,it is anticipated that increasing numbers of therapeutic genes or targets will become available for targeted therapies.Despite numerous setbacks,efficacious gene and/or cell-based therapies still hold the great promise to revolutionize the clinical management of human diseases.It is wildly recognized that poor gene delivery is the limiting factor for most in vivo gene therapies.There has been a long-lasting interest in using viral vectors,especially adenoviral vectors,to deliver therapeutic genes for the past two decades.Among all currently available viral vectors,adenovirus is the most efficient gene delivery system in a broad range of cell and tissue types.The applications of adenoviral vectors in gene delivery have greatly increased in number and efficiency since their initial development.In fact,among over 2000 gene therapy clinical trials approved worldwide since 1989,a significant portion of the trials have utilized adenoviral vectors.This review aims to provide a comprehensive overview on the characteristics of adenoviral vectors,including adenoviral biology,approaches to engineering adenoviral vectors,and their applications in clinical and preclinical studies with an emphasis in the areas of cancer treatment,vaccination and regenerative medicine.Current challenges and future directions regarding the use of adenoviral vectors are also discussed.It is expected that the continued improvements in adenoviral vectors should provide great opportunities for cell and gene therapies to live up to its enormous potential in personalized medicine. | Cody S.Lee Elliot S.Bishop Ruyi Zhang Xinyi Yu Evan M.Farina Shujuan Yan Chen Zhao Zongyue Zeng Yi Shu Xingye Wu Jiayan Lei Yasha Li Wenwen Zhang Chao Yang Ke Wu Ying Wu Sherwin Ho Aravind Athiviraham Michael J.Lee Jennifer Moriatis Wolf Russell R.Reid Tong-Chuan He | 2017 | Genes & Diseases2017,4,2: | 19 |

| 3 | Detecting the polymorphism sites of p53 and Fas genes of Han population in Zhejiang province显示文摘BACKGROUND: It is of significance for single nucleotide polymorphisms (SNPs), a difference of rank, which exists widely in biology, genetics and other fields. OBJECTIVE: To detect polymorphism sites in exon-4 of p53 gene, promotor of Fas gene and intron-7 of Fas gene of healthy people in Han nationality in Zhejiang province. DESIGN: Simple random sampling. SETTING: Department of Surgery of the 118 Hospital of Chinese PLA. PARTICIPANTS: A total of 80 healthy people in Han nationality were selected from hospitals in Zhejiang province from August 2005 to January 2006. There were 43 males and 37 females aged from 3 to 78 years with the mean age of 39.5 years, and all subjects were consent. DNA which was used in genetic analysis was selected from peripheral venous blood of all subjects and maintained at -20 ℃. METHODS: Polymorphism sites in exon-4 of p53 gene, promotor of Fas gene and intron-7 of Fas gene were detected with directly DNA sequencing technique. MAIN OUTCOME MEASURES: Polymorphism sites in exon-4 of p53 gene, promotor of Fas gene and intron-7 of Fas gene of healthy people in Han nationality in Zhejiang province. RESULTS: A total of 80 samples were involved in the final analysis. SNPs sites were found at the 119th base of exon-4 of p53 gene (the 72nd codon of p53 gene), the 670th base of upper start codon in promotor of Fas gene (Fas-670), and the 995th base of intron-7 of Fas gene, especially SNPs in the 995th base of intron-7 pf Fas gene, i.e. C→A transversion, was a new site. CONCLUSION: One unknown SNPs site is discovered in intron-7 of Fas gene of people in Han nationality in Zhejiang province. | 6 | Global gene expression profiles for the growth phases of Trichophyton rubrum显示文摘Trichophyton rubrum (T.rubrum) is a common superficial fungus.Molecular and genetic studies of T.rubrum are still limited.In this paper,we report the global analysis of gene expression profiles at different growth phases using cDNA microarray technology.A total of 2044 differentially expressed genes were obtained and clustered into three expression patterns.Our data confirmed previous results that many mRNAs were pre-stored in the conidia of T.rubrum.Transcriptional profiling and function analysis showed that some glycolytic enzymes share similar expression patterns and may be coregulated during the transition of growth phases.Some genes involved in small GTPase signaling pathways,and in cAMP-dependent and MAPK regulation pathways were induced in response to the growth dynamics of T.rubrum.Although the detailed biological roles of these T.rubrum genes are still unknown,our results suggest that these genes may be involved in regulation mechanisms in the life cycle of the fungus. | XU XingYe LIU Tao LENG WenChuan DONG Jie XUE Ying YANG HanChuna JIN Qi | 2011 | Science China(Life Sciences)2011,54,7: | 2 |

| 8 | Review on constitutive models of road materials显示文摘Road material constitutive model is the essential condition of pavement structure design,calculation,and maintenance decision.Experts have conducted in-depth studies on the problems and proposed many meaningful constitutive models.However,these constitutive models have not been summarized and no uniform conclusion on the applicability of them is reported in the academic circle.The linear elastic model based on Hooke's law is still used in pavement structure design.This paper systematically reviewed the existing constitutive models that reflect the materials'mechanical properties and used for pavement structure calculation.These constitutive models were divided into three categories:linear,nonlinear,and damage models under repeated loads.The applicable conditions,parameter determination methods,advantages,and disadvantages of each model were introduced.Moreover,the model's feasibility in structural design and calculation was analyzed.Follow-up studies should focus on the parameter determination standard of viscoelastic constitutive models,the extension of the 1D constitutive model to 3D state,and the nonlinear constitutive models related to stress-strain state to drive the application of the constitutive models in pavement structure calculation and design.Furthermore,to evaluate the life cycle service performance of road materials and structures,establishing the pavement damage model from the structure perspective will be the future direction. | Qian Li Xudong Wang Xu Liu Xingye Zhou | 2022 | Journal of Road Engineering2022,2,1: | 2 |

| 10 | Source-field-plated Ga_2O_3 MOSFET with a breakdown voltage of 550 V显示文摘Ga_2O_3 metal–oxide–semiconductor field-effect transistors(MOSFETs) with high-breakdown characteristics were fabricated on a homoepitaxial n-typed β-Ga_2O_3 film, which was grown by metal organic chemical vapor deposition(MOCVD) on an Fedoped semi-insulating(010) Ga_2O_3 substrate. The structure consisted of a 400 nm unintentionally doped(UID) Ga_2O_3 buffer layer and an 80 nm Si-doped channel layer. A high k HfO_2 gate dielectric film formed by atomic layer deposition was employed to reduce the gate leakage. Moreover, a source-connected field plate was introduced to enhance the breakdown characteristics. The drain saturation current density of the fabricated device reached 101 mA/mm at V_(gs) of 3 V. The off-state current was as low as 7.1 ×10^(-11) A/mm, and the drain current I_(ON)/I_(OFF) ratio reached 10~9. | 12 | Comparison of droplet distributions from fluidic and impact sprinklers显示文摘To adapt to the trend toward low-energy precision irrigation, the droplet distributions for two new prototype sprinklers, outside signal sprinkler(OS) and fluidic sprinkler(FS), were compared with impact sprinkler(IS). A laser precipitation monitor was used to measure the droplet distributions. Droplet size and velocity distributions were tested under four operating pressures for nozzles 1.5 m above the ground. For the operating pressures tested, the mean OS, FS and IS droplet diameters ranged from 0 to 3.4, 0 to 3.5, 0 to 4.0 mm, respectively.The mean OS and FS droplet velocities ranged from 0 to6.3 m$s–1, whereas IS ranged from 0 to 6.3 m$s–1. Being gas-liquid fluidic sprinklers, droplet distributions of the OS and FS were similar, although not identical. IS mostly produced a 0.5 mm larger droplet diameter and a 0.5 m$s–1greater velocity than OS and FS. A new empirical equation is proposed for determination of droplet size for OS and FS, which is sufficiently accurate and simple to use. Basic statistics for droplet size and velocity were performed on data obtained by the photographic methods. | 18 | Effect of selective uterine artery embolization on symptomatic uterine fibroids显示文摘Objective: To evaluate the outcomes of selective uterine artery embolization (SUAE) in treatment of symptomatic uterine fibroids using Color Doppler ultrasonography (CDUS). Methods: As the procedure SUAE for fibroids, prospective data of the initial 65 consecutive women treated from March 2007 to September 2009 were collected. The follow-up period from 1 week to 30 months, using questionnaires, we investigated the fibroid-related physical and psychological symptoms, and the cumulative rates of symptom control, gynecologic interventions, and complications. The volumes of uterine and uterine fibroids were calculated by CDUS pre- and post-SUAE for 6 months. And we tested the levels of internal hormone including FSH, LH and E2 before and after SUAE for 3, 6 months. Results: SUAE was performed successfully in all patients. There was no peri-operative morbidity in all procedures. Compared with pre-SUAE volumes of uterine [(322±112) cm3] and uterine fibroids [(125±46) cm3], their volumes in post-SUAE were (144±72) cm3 and (51±11) cm3 (P<0.01), and reduction rate was 58% and 61%, respectively. At 3, 6 months after SUAE, levels of FSH, LH, and E2 were not decreased than that of in pre-SUAE (P>0.05). After SUAE for 24 months, the fibroids related physical and psychological symptoms such as bleeding, pain and quality of life were significantly improved (P<0.01). 90.8% (59/65) women had resolution of symptoms and no significant post-procedural symptoms. Parts of patients discharge necrotic fibroids. 86% (34/40) complications were minor, requiring no therapy. 9.2% (6/65) had significant post-procedural symptoms, requiring therapy in hospital or out-patient department. Conclusion: SUAE contributes to a long-term significant improvement of all investigated uterine fibroid-related symptoms and markedly improves women's health-related quality of life. SUAE that no impair on changes of internal hormone, under suitable conditions, represents an attractive alternative to hysterectomy for the treatment of symptomatic fibroids and is a minimally invasive, new method of good clinical prognosis.
