| 1 | Survival and prognostic factors in hepatitis B virus-related acute-on-chronic liver failure显示文摘AIM:To investigate the survival rates and prognostic factors in patients with hepatitis B virus-related acuteon-chronic liver failure(HBV-ACLF).METHODS:Clinical data in hospitalized patients with HBV-ACLF admitted from 2006 to 2009 were retrospectively analyzed.Their general conditions and survival were analyzed by survival analysis and Cox regression analysis.RESULTS:A total of 190 patients were included in this study.The overall 1-year survival rate was 57.6%.Patients not treated with antiviral drugs had a significantly higher mortality[relative risk(RR)=0.609,P=0.014].The highest risk of death in patients with ACLF was associated with hepatorenal syndrome(HRS)(RR=2.084,P=0.026),while other significant factors were electrolyte disturbances(RR=2.062,P=0.010),and hepatic encephalopathy(HE)(RR=1.879,P<0.001).CONCLUSION:Antiviral therapy has a strong effect on the prognosis of the patients with HBV-ACLF by improving their 1-year survival rate.HRS,electrolyte disturbances,and HE also affect patient survival. | Kun Huang Jin-Hua Hu Hui-Fen Wang Wei-Ping He Jing Chen XueZhang Duan Ai-Min Zhang Xiao-Yan Liu | 2011 | World Journal of Gastroenterology2011,17,29: | 32 |
| 2 | Expression of survivin in primary and metastatic gastric cancer cells obtained by laser capture microdissection显示文摘AIM:Survivin, a recently identified member of the inhibitor of apoptosis protein family, is expressed during development and in various human cancers. However, its expression in normal tissues and clinical relevance in cancers are still debated. In the present study, we analyzed the expression of the survivin gene in human primary and metastatic gastric cancer cells as well as in paired epithelial cells from normal gastric mucosa by means of a novel laser capture microdissection (LCM) technique coupled with reverse transcription -polymerase chain reaction (RT-PCR).METHODS: Thirty patients who had undergone gastrectomy with lymph node dissection for gastric cancer without preoperative treatments were included. Neoplastic tissue, metastatic lymph nodes, and apparently uninvolved normal tissue were collected from each patient. LCM-captured 'pure' cell groups were respectively subjected to RT-PCR analysis with primers specific for the survivin gene.RESULTS: Of the paired samples from 30 gastric cancer patients studied, 24 (80%) primary gastric cancer cell groups and 7 (23%) adjacent morphologically 'normal' gastric epithelial cell groups were shown to have a detectable survivin expression. There was a statistically significant difference in suvivin expression between these two groups (P<0.01). Meanwhile, 95% (19/20) of the metastatic gastric cancer cell groups from lymph nodes had a clear expression of the survivin gene. However, no significant correlation between survivin expression and clinicopathological features of gastric cancer was observed in the present study. CONCLUSION: Survivin expression is present in the majority of gastric cancer cell groups obtained by LCM techniques. The high expression rate in metastatic lesions suggests a possible role of survivin in cancer invasiveness and metastasis. It may contribute to the detection of gastric cancer micrometastasis as a potential molecular marker. | 4 | A COMPLETE SCREEN FOR MUTATIONS OF THE RHODOPSIN GENE IN A PANEL OF CHINESE PATIENTS WITH AUTOSOMAL DOMINANT RETINITIS PIGMENTOSA显示文摘Objective To evaluate the prevalence of rhodopsin (RHO) mutations and the genotype-phenotype relationships in Chinese patients with autosomal dominant retinitis pigmentosa (ADRP) by conformation sensitive gel electrophoresis (CSGE) and direct DNA sequencing. Methods We have screened the five coding exons and splice sites of RHO gene in 27 probands who had no relativity from Chinese ADRP families and 100 normal controls to identify disease-associated mutations, using CSGE and direct DNA sequencing. Family members of some probands with disease-associated mutations were also genotyped to determine whether the RHO mutations segregated with retinitis pigmentosa (RP) in their families. Results Two RHO mutations, Pro347Leu and Pro327 (1-bp del), were identified separately in two families, thus the frequency of RHO mutations among this set of Chinese ADRP families is about 7.4% (2/27). Pro347Leu mutation was found in one ADRP proband as well as three her children who also had RP. She had relatively early onset at about 17 years. The only one child without this mutation had no symptom or sign of RP at age of 34. Pro327 (1-bp del) was identified in a late-onset ADRP patient, who appeared night blindness around 30 years old and in her fifties electroretinogram (ERG) has been flat in both scotopic and photopic phases. Family analysis showed that this mutation also existed in her younger dau-ghter and her elder sister, both of them also had RP. Three other family members were genotypically and phenotypically normal. Neither of the two mutations was detected in 100 normal controls.Conclusions The frequency of RHO mutations in Chinese patients was lower than that in Europe and North America. The phenotype of the patients with Pro347Leu corresponded to type 1 ADRP, with severe rod degeneration and some cone preservation later, while the phenotype of the patients carrying Pro327 (1-bp del) corresponded to type 2 ADRP, with a concomitant loss of rod and cone visual function. | 5 | Superior de/hydrogenation performances of MgH2 catalyzed by 3D flower-like TiO2@C nanostructures显示文摘Magnesium hydride has been seen as a potential material for solid state hydrogen storage,but the kinetics and thermodynamics obstacles have hindered its development and application.Three-dimensional flower-like TiO2@C and TiO2 were synthesized as the catalyst for MgH2 system and great catalytic activities are acquired in the hydrogen sorption properties.Experiments also show that the flower-like TiO2@C is superior to flower-like TiO2 in improving the hydrogen storage properties of MgH2.The hydrogen desorption onset and peak temperatures of flower-like TiO2 doped MgH2 is reduced to 199.2℃and 245.4℃,while the primitive MgH2 starts to release hydrogen at 294.6℃and the rapid dehydrogenation temperature is even as high as 362.6℃.The onset and peak temperatures of flower-like TiO2@C doped MgH2 are further reduced to 180.3℃and 233.0℃.The flower-like TiO2@C doped MgH2 composite can release6.0 wt%hydrogen at 250℃within 7 min,and 4.86 wt%hydrogen at 225℃within 60 min,while flowerlike TiO2 doped MgH2 can release 6.0 wt%hydrogen at 250℃within 8 min,and 3.89 wt%hydrogen at225℃within 60 min.Hydrogen absorption kinetics is also improved dramatically.Moreover,compared with primitive MgH2 and the flower-like TiO2 doped MgH2,the activation energy of flower-like TiO2@C doped MgH2 is significantly decreased to 67.10 kJ/mol.All the improvement of hydrogen sorption properties can be ascribed to the flower-like structure and the two-phase coexistence of TiO2 and amorphous carbon.Such phase composition and unique structure are proved to be the critical factor to improve the hydrogen sorption properties of MgH2,which can be considered as the new prospect for improving the kinetics of light-metal hydrogen storage materials. | Meng Zhang Xuezhang Xiao Bosang Luo Meijia Liu Man Chen Lixin Chen | 2020 | Journal of Energy Chemistry2020,29,7: | 5 |

| 7 | Somatic mutation analysis of p53 and ST7 tumor suppressor genes in gastric carcinoma by DHPLC显示文摘AIM: To verify the effectiveness of denaturing highperformance liquid chromatography (DHPLC) in detecting somatic mutation of p53 gene in gastric carcinoma tissues.The superiority of this method has been proved in the detection of germline mutations, but it was not very affirmative with respect to somatic mutations in tumor specimens. ST7 gene, a candidate tumor suppressor gene identified recently at human chromosome 7q31.1, was also detected because LOH at this site has also been widely reported in stomach cancer.METHODS: DNA was extracted from 39 cases of surgical gastric carcinoma specimen and their correspondent normal mucosa. Seven fragments spanning the 11 exons were used to detect the mutation of p53 gene and the four exons reported to have mutations in ST7 gene were amplified by PCR and directly analyzed by DHPLC without mixing with wild-type allele.RESULTS: In the analysis of p53 gene mutation, 9 aberrant DHPLC chromatographies were found in tumor tissues, while their normal-adjacent counterparts running in parallel showed a normal shape. Subsequent sequencing revealed nine sequence variations, 1 polymorphism and 8 mutations including 3 mutations not reported before. The mutation rate of p53 gene (21%) was consistent with that previously reported. Furthermore, no additional aberrant chromatography was found when wild-type DNA was added into the DNA of other 30 tumor samples that showed normal shapes previously. The positivity of p53 mutations was significantly higher in intestinal-type carcinomas (40 %) than that in diffuse-type (8.33 %) carcinomas of the stomach. No mutation of ST7 gene was found.CONCLUSION: DHPLC is a very convenient method for the detection of somatic mutations in gastric carcinoma. The amount of wild type alleles supplied by the non-tumorous cells in gastric tumor specimens is enough to form heteroduplex with mutant alleles for DHPLC detection. | 8 | Effect of cyclin G2 on proliferative ability of SGC-7901 cell显示文摘AIM: To study the effect of cyclin G2 on proliferation of gastric adenocarcinoma cell line-SGC-7901 cell in vitro.METHODS: By use of cation lipofectamine transfection reagent,the pIRES-G2 and pIRESneo plasmids were transferred into SGC-7901cell line. Anticlones were selected by G418. Positive clones were observed and counted using Giemsa staining.Cell proliferative ability was assayed by Ml-r.RESULTS: (1) The done number of pIRES-G2 group decreased,clone volume reduced. The number of cell clones in pIRESneo group was 87+3, that of pIRES-G2 group was 53+4,occupying 60.1% of pIRESneo group, there was significant difference obviously (P<0.01, t=15.45). | 10 | Extreme high reversible capacity with over 8.0 wt% and excellent hydrogen storage properties of MgH2 combined with LiBH4 and Li3AlH6显示文摘Magnesium hydride has attracted great attention because of its high theoretical capacity and outstanding reversibility, nevertheless, its practical applications have been restricted by the disadvantages of the sluggish kinetics and high thermodynamic stability. In this work, an unexpected high reversible hydrogen capacity over 8.0 wt% has been achieved from MgH2 metal hydride composited with small amounts of LiBH4 and Li3AlH6 complex hydrides, which begins to release hydrogen at 276 ℃ and then completely dehydrogenates at 360 ℃. The dehydrogenated MgH2+LiBH4/Li3AlH6 composite can fully reabsorb hydrogen below 300 ℃ with an excellent cycling stability. The composite exhibits a significant reduction of dehydrogenation activation energy from 279.7 kJ/mol(primitive MgH2) to 139.3 kJ/mol(MgH2+LiBH4/Li3AlH6),as well as a remarkable reduction of dehydrogenation enthalpy change from 75.1 k J/mol H2(primitive MgH2) to 62.8 kJ/mol H2(MgH2+LiBH4/Li3AlH6). The additives of LiBH4 and Li3AlH6 not only enhance the cycling hydrogen capacity, but also simultaneously improve the reversible de/rehydrogenation kinetics, as well as the dehydrogenation thermodynamics. This notable improvement on the hydrogen absorption/desorption behaviors of the MgH2+LiBH4/Li3AlH6 composite could be attributed to the dehydrogenated products including Li3Mg7, Mg17Al12 and MgAlB4, which play a key role on reducing the dehydrogenation activation energy and increasing diffusion rate of hydrogen. Meanwhile, the LiBH4 and Li3AlH6 effectively destabilize MgH2 with a remarkable reduction on dehydrogenation enthalpy change in terms of thermodynamics. | 16 | Significantly enhanced hydrogen desorption properties of Mg(AlH4)2 nanoparticles synthesized using solvent free strategy显示文摘Mg(AlH_4)_2 nanoparticles with a particle size less than 10 nm have been successfully synthesized by mechanochemical method using LiAlH_4 and MgCl_2 as raw materials together with Li Cl buffering additive. In comparison to Mg(AlH_4)_2 microparticles, Mg(AlH_4)_2 nanoparticles exhibit a faster hydrogen desorption kinetics and lower desorption temperature. The hydrogen desorption temperatures of the first and second dehydrogenation steps are 80 and 220 °C for the Mg(AlH_4)_2 nanoparticles, which are about 65 and 60 °C, respectively, lower than those of Mg(AlH_4)_2 microparticles. The decomposition activation energy is reduced from 135 k J/mol for Mg(AlH_4)_2 microparticles to 105.3 k J/mol for Mg(AlH_4)_2 nanoparticles. It is proposed that the shortened diffusion distance and enhanced diffusivity of Mg(AlH_4)_2/MgH_2 nanoparticles provide an energy destabilization for lowering the dehydrogenation temperature, and thus being the key factor for promoting the hydrogen desorption kinetics.
