| 2 | Selective blockade of NF-kappa B by novel mutated I kappa B alpha suppresses CD3/CD28-induced activation of memory CD4(+) T cells in asthma显示文摘Background:Nuclear factor kappa B(NF-kappa B) overactivation plays a crucial role in T-helper 2 (Th2)-biased allergic airway inflammation by increased activation and decreased apoptosis of CD4(+) T cells. We have shown that targeted NF-kappa B suppression in dendritic cells by adenoviral gene transfer of a novel mutated inhibitor of NF-kappa B(I kappa B alpha) (AdI kappa B alpha M) contributes to T-cell tolerance, but the immunosuppressive action of AdI kappa B alpha M on memory(CD45RO(+)) CD4(+) T cells remains enigmatic. Methods:CD45RO(+) T cells from Dermatophagoides farinaei-sensitized asthmatic patients, untransfected or transfected with AdI kappa B alpha M or AdLacZ(beta-galactosidase) for 24 h, were stimulated with anti-CD3(1.0 mu g/ml) plus anti-CD28(0.5 mu g/ml) monoclonal antibody for an additional 24 h. I kappa B alpha M transgene expression and NF-kappa B activation were detected by polymerase chain reaction(PCR), reverse transcription-PCR(RT-PCR), Western blot analysis, and electrophoretic mobility shift assay. Phenotype and apoptosis were measured by flow cytometry, annexin V binding, and terminal deoxyribonucleotidyl transferase-mediated dUTP nick end labeling analyses. Cytokine production and cell proliferation were determined using enzyme-linked immunosorbent assay andH-3 thymidine incorporation. Results:A unique 801-bp I kappa B alpha M cDNA and a dose-dependent increase in I kappa B alpha M transgene expression were observed in AdI kappa B alpha M-transfected CD45RO(+) T cells. Significantly, AdI kappa B alpha M inhibited CD3/CD28-mediated NF-kappa B activation in CD45RO(+) T cells, leading to evident apoptosis, reduction of eotaxin, RANTES, Th1 interferon (IFN)-gamma and interleukin (IL)-2, and Th2 (IL-4, IL-5, and IL-13 despite a slight decrease in IL-10) cytokines and secondary proliferative response. AdI kappa B alpha M also upregulated cytotoxic T lymphocyte-associated antigen 4(CTLA-4) and downregulated CD69 besides no change in CD28. Conclusion:I kappa B alpha M might be beneficial to augment memory CD4(+) T-cell tolerance through modulating B7-CD28/CTLA-4 co-stimulatory pathways and NF-kappa B-dependent cytokine profiles in allergic inflammatory diseases including asthma. | Zhou,L. | 5 | Cadmium Accumulation and Translocation in Two Jerusalem Artichoke(Helianthus tuberosus L.) Cultivars显示文摘Jerusalem artichoke(Helianthus tuberosus L.) not just can be used for bioethanol production but may be potentially used in phytoremediation for the removal of heavy metal pollutants.Two Jerusalem artichoke cultivars,N2 and N5,were subjected to six cadmium(Cd) concentrations(0,5,25,50,100 and 200 mg L1) to investigate Cd tolerance and accumulation.After 21 days of growth,the effects of Cd on growth,chlorophyll content,net photosynthetic rate,intercellular CO2 concentration and malondialdehyde content were evaluated.Most growth parameters were reduced under Cd stress.The two Jerusalem artichoke cultivars had relatively high Cd tolerance and accumulation capacity(> 100 mg kg1),with N5 being more tolerant and having higher Cd accumulation than N2.Roots accumulated more Cd than stems and leaves.The bioconcentration factors(far higher than 1) and translocation factors(lower than 1) decreased with an increase in Cd applied.The results suggested that Jerusalem artichoke could be grown at relatively high Cd loads,and N5 could be an excellent candidate for phytoremediation of Cd-contaminated soils. | CHEN Liang LONG Xiao-Hua ZHANG Zhen-Hua ZHENG Xiao-Tao Z. | 7 | TiZr-base Bulk Metallic Glass with over 50 mm in Diameter显示文摘Low-cost TiZr-base bulk metallic glasses(BMGs)(Ti36.1Zr33.2Ni5.8Be24.9)100-xCux(x=5,7 and 9) with a maximum size of over 50 mm in diameter were developed by optimizing the alloy composition.The idea is initiated by selecting a particular microstructure comprising primary β-Ti dendrite and amorphous phase.Afterwards,based on this composition of amorphous phase,a class of TiZr-base bulk metallic glasses was designed step by step to reach the optimum composition range.The glass transition temperature(Tg),initial crystallization temperature(Tx) and width of supercooled region(△T) of(Ti36.1Zr33.2Ni5.8Be24.9)91Cu9 BMG are 611,655 and 44 K,respectively.The(Ti36.1Zr33.2Ni5.8Be24.9)91Cu9 BMG exhibits low density of 5.541 g·cm-3 and high compressive fracture strength of 1800 MPa,which promises the potential application as structural materials. | M. But the noise signal is often difficult to be analyzed due to the complicated nature of the specific systems being investigated. In this paper the potential noise fluctuations during the bee corrosion of commercial aluminum alloy LY12 in sodium chloride solution was recorded and analyzed with different techniques. The typical results showed that the fractal dimension (Dn) obtained from spectral power density (SPD) is mainly directly proportional to the intensity of pitting corrosion and to the value of pitting parameter (SE) derived hem dimensional analysis, while the fractal dimension (DE) obtained from EIS is mainly related to the uniform corrosion. | J.Q. | 9 | Contamination by Organochlorine Pesticides in the Estuaries of Southeast China显示文摘Persistent organic pollutants(POPs) such as organochlorine pesticides are of global concern due to their widespread occurrence and persistence. This paper reports recent research studying the distribution and fate of organochlorine pesticides in the Jiulong River, the Minjiang River and the Pearl River estuaries in Southeast China. Eighteen organochlorine pesticides were extracted from water, pore water and sediment samples, followed by analysis by GC ECD. The results showed that the contamination levels were similar in these three estuaries. The levels of the total organochlorine pesticides in porewater were significantly higher than those in surface water, due to the high affinity of these hydrophobic compounds for sediment phase. Among the hexachlorocyclohexane(HCH) compounds, β HCH was found to be the most important isomer. The analysis of 1,1,1 trichloro 2,2 bis chlorophenyl ethane(DDT) and its metabolites showed that 1,1 dichloro 2[ o chlorophenyl] 2[ p chlorophenyl] ethylene(DDE) was dominant in the group. | Z. L. | 12 | Polymorphisms of MTHFD,plasma homocysteine levels, and risk of gastric cancer in ahigh-risk Chinese population显示文摘Purpose:Accumulative evidence suggests that folate has a protective effect on gastric cancer. The methylenetetrahyd-rofolate dlehydrogenase(MTHFD) plays an important role in folate and homocysteine metabolisms, and polymorphisms of MTHFD may result in disturbance of the folate-mediated homocysteine pathway. The aim of this study is to test the hypothesis that genetic variants of MTHFD and plasma homocysteine levels are associated with risk of gastric cancer and modulated by genotypes of methylenetetrahydrofolate reductase(MTHFR). Experimental Design: We genotyped G1958A and T401C in MTHFD and C677T in MTHFR and detected total plasma homocysteine(tHcy) levels in a case-control study of 589 gastric cancer cases and 635 cancer-free controls in a high-risk Chinese population. Results:The variant genotypes of MTHFD 1958AA and 401CC were associated with a significantly increased risk of gastric canceradjusted odds ratio(OR), 2.05; 95% confidence interval(95% CI),1.34-3.13 for 1958AA; adjusted OR,1.43; 95% CI,1.14-1.80 for 401CC compared with 1958GG/GA and 401TT/TC genotypes, respectively. Both of the effects were more evident in the subjects carrying MTHFR 677CT/TT genotypes. The average tHcy level was significantly higher in gastric cancer cases than in controls(P < 0.01), and the upper quartile of tHcy (> 13.6 mu mol/L) was associated with an 82% significantly increased risk of gastric cancer, compared with the lowest quartile of tHcy(<= 8.0 pmol/L;adjusted OR,1.82; 95% CI,1.20-2.75). Conclusions:The strong associations between MTHFD variants and the plasma tHcy levels and gastric cancer risk suggest, for the first time, a possible gene-environment interaction between genetic variants of folate-metabolizing genes and high tHcy levels in gastric carcinogenesis. | Wang, L. | 18 | ELECTROCHEMICAL NOISE ANALYSIS OF PURE ALUMINUM IN SODIUM CHLORIDE SOLUTION WITH WAVELET TRANSFORM TECHNIQUE显示文摘Wavelet transforms (WT) are proposed as an alternative tool to overcome the limitations of Fourier transforms (FFT) in the analysis of electrochemical noise (EN) data. The most relevant feature of this method of analysis is its capability of decomposing electrochemical noise records into different sets of wavelet coefficients, which contain information about the time scale characteristic of the associated corrosion event. In this context, the potential noise fluctuations during the free corrosion of pure aluminum in sodium chloride solution was recorded and analyzed with wavelet transform technique. The typical results showed that the EN signal is composed of distinct type of events, which can be classified according to their scales, i.e. their time constants.
