| 2 | Detection of serum tumor markers in the diagnosis and treatment of patients with pancreatic cancer显示文摘BACKGROUND: Although a variety of tumor markers areavailable for diagnosis of pancreatic cancer, their sensitivityand specificity have not yet been ideal. The aims of thisstudy was to detect a panel of serum tumor markers and toevaluate their significance in the diagnosis and prognosis ofpancreatic cancer patients.METHODS: Eight serum tumor markers including AFP,CEA, CA-50, CA72-4, CA-125, CA153, CA19-9 and CA242were detected in 129 patients with pancreatic cancer by usingchemiluminescence immunoassay, immunofluorescence as-say and immunoradiometric assay, respectively. The levelsof these markers were compared in 99 patients with non-pancreatic malignant tumor, 63 patients with other benigndiseases, and 27 patients with pancreatic cancer after pan-createctomy.RESULTS: Among the 8 tumor markers, CA19-9, CA242,CA-50, and CA72-4 were more sensitive in the diagnosis ofpancreatic cancer. Parallel combined testing could increasethe diagnostic sensitivity to 89.2%, and serial combined exa-mination could increase the diagnostic specificity to 92.3%.The serum tumor markers levels were decreased significant-ly after radical tumor resection.CONCLUSIONS: Serum CA19-9, CA242, CA-50, andCA72-4 are the preferred tumor markers to be used in thediagnosis and follow-up of operated cases of pancreaticcancer. | 4 | Early J_2 basalts in SE China:Incipience of large-scale late Mesozoic magmatism显示文摘Magmatism in SE China was dormant during 204―180 Ma, but was reactivated in 180― 170 Ma (early J2), and then became more and more intensive towards the end of early Cretaceous. The small-scale early J2 magmatism is the incipience to long-term and large-scale magmatism in this region. A near east-west (EW) trend volcanic belt was distributed across south Hunan, south Jiangxi and southwest Fujian was formed during early J2 time. Along this belt from the inland toward the coast, the lithology of basalts changes from alkali into tholeiite, and the amount of erupted volcanic rocks and the proportions of rhyolites coexisting with the basalts increase. On the basis of geochemical char- acteristics of these basalts, we infer that the melting degree of source rocks and the extent of frac- tional crystallization and crustal contamination all increased whereas the depth of mantle source de- creased from the inland to the coast, which led to the variations of geological characteristics of the volcanic belt. In early J2, the western spreading Pacific plate began to subduct underneath SE China continental block, reactivating near EW trend deep fault that was originally formed during the Indos- inian event. | 5 | Proliferative activity of bile from congenital choledochal cyst patients显示文摘AIM: To explore the potential carcinogenicity of bile from congenital choledochal cyst (CCC) patients and the mechanism of the carcinogenesis in congenital choledochal cyst patients.METHODS: 20 bile samples from congenital choledochalcyst patients and 10 normal control bile samples were usedfor this study. The proliferative effect of bile was measuredby using Methabenzthiazuron (MTT) assay; Cell cycle andapoptosis were analyzed by using flow cytometry (FCM),and the PGE2 levels in the supernatant of culturedcholangiocarcinoma cells were quantitated by enzyme-linkedimmunoabsordent assay (ELISA).RESULTS: CCC bile could significantly promote theproliferation of human cholangiocarcinoma QBC939 cellscompared with normal bile (P=0.001) and negative controlgroup (P=0.002), and the proliferative effect of CCC bilecould be abolished by addition of cyclooxygenase-2 specificinhibitor celecoxib (20 μM). | 6 | Celecoxib inhibits proliferation and induces apoptosis via prostaglandin E_2 pathway in human cholangiocarcinoma cell lines显示文摘AIM: To evaluate the roles and mechanisms of celecoxib in inducing proliferation inhibition and apoptosis of human cholangiocarcinoma cell lines.
METHODS: Cyclooxygenase-2-overexpressing human cholangiocarcinoma cell line QBC939 and cyclooxygenase2-deficient human cholangiocarcinoma cell line SK-CHA-1were used in the present study. The anti-proliferative effect was measured by methabenzthiazuron (MTT) assay;apoptosis was determined by transferase-mediated dUTP nick end labeling (TUNEL) detection and transmission electron microscopy (TEN). Cell cycle was analyzed by flow cytometry (FCM). The PGE2 levels in the supernatant of cultured cholangiocarcinoma cells were quantitated by enzyme-linked immunoabsordent assay (ELISA).
RESULTS: Celecoxib suppressed the production of PGE2and inhibited the growth of QBC939 cells. Celecoxib at 10,20, and 40 μmol/L inhibited PGE2 production by 26 %,58 %, and 74 % in QBC939 cells. The PGE2 level was much lower constitutively in SK-CHA-1 cells (18.6±3.2)compared with that in QBC939 (121.9±5.6) cells (P<0.01)and celecoxib had no significant influence on PGE2 level in the SK-CHA-1 cells. The PGE2 concentration in SK-CHA-1cells also reduced but not significantly after treatment with celecoxib. The PGE2 concentration in SK-CHA-1 cells was (16.5±2.9) ng/well, (14.8±3.4) ng/well, (13.2±2.0) ng/well and (12.6±3.1) ng/well respectively, when pre-treated with 1 Jmol/L, 10 Jmol/L, 20 Jmol/L and 40 Jmol/L of celecoxib for 48 h (P>0.05, vs control). The anti-proliferation effect of celecoxib (20 Jmol/L) on QBC939 cells was time-dependent,it was noticeable on day 2 (OD490=0.23±0.04) and became obvious on day 3 (OD490=0.31±0.07) to day 4 (OD490=0.25±0.06), and the OD490 in the control group (day 1)was 0.12±0.03 (P<0.01, vscontrol). The anti-proliferation effect of celecoxib could be abolished by the addition of 200 pg/mL PGE2. The proliferation of SK-CHA-1 cells was inhibited slightly by celecoxib, the cell density OD490 in the presence of celecoxib and in control group was 0.31±0.04 and 0.42±0.03 respectively on day 2 (P>0.05), 0.58±0.07 and 0.67±0.09 respectively on day 3 (P>0.05), and 0.71±0.08 and 0.78±0.06 respectively on day 4 (P>0.05). Celecoxib induced proliferation inhibition and apoptosis by G1-S cell cycle arrest: the percentage of QBC939 cells in G0-G1 phase after treatment with 40 Jmol/L (74.66±6.21) and 20 Jmol/L (68.63±4.36) celecoxib increased significantly compared with control cells (54.41±5.12, P<0.01). The percentage of SK-CHA-1 cells in G0-G1 phase after treatment with various concentrations of celecoxib didn't change significantly compared with control cells. The TUNEL index was much higher in QBC939 cells treated with 20 Jmol/L celecoxib for 2 d (0.063±0.018) and for 4 d (0.102±0.037) compared with control cells (0.017±0.004, P<0.01).
CONCLUSION: The currentin vitro study indicates that inhibition of proliferation and induction of apoptosis in human cholangiocarcinoma cells by cyclooxygenase-2 specific inhibitor celecoxib may involve in COX-dependent mechanisms and PGE2 pathway. | 7 | The global image of the Moon obtained by the Chang'E-1:Data processing and lunar cartography显示文摘The global lunar image of the first phase of Chinese Lunar Exploration Program is the first image that covered all over the surface of the Moon. It will serve as a critical foundation for succeeding exploration and scientific research. In this paper, the acquisition, characteristics, and data quality of Chang'E-1 CCD image data are described in detail. Also described are the methodology and procedure of data processing. According to rule of planetary cartography, the image data have been processed, geometrically corrected, and then mosaicked and merged in a scale of 1:2.5 million. The results of data processing and charting show that the image data of Chang'E-1 CCD and their geometric precision meet the demand of charting a map in the scale of 1:2.5 million. The relative geometric positioning precision of the global image is better than 240 m, and the absolute geometric positioning precision is slightly better than that of the ULCN2005 and Clementine lunar basemap (V2.0). The plane positioning precision is approximately 100-1500 m. | 9 | RPPK modulation with high data rates显示文摘With the development of communication technology, the precious frequency spectrum is becoming more and more crowded. Exploiting the particular filter with zero group delay, ultra narrow band (UNB) modulation is intended for acquiring the high frequency efficiency. Unfortunately, this UNB filter is confronted with great challenges from the classical communication theories. The validation on its realization is also difficult. This paper proposes a novel UNB modulation, namely random pulse position keying (RPPK). It is demonstrated from analysis that there is no inessential discrete spectral line in modulated signal’s power density spectrum (PDS), and the UNB filter has been successfully avoided. Consequently, RPPK is rather explicit in theory and simple in implementation. As is shown by the simulation, the frequency utilization of RPPK can even reach 100 bits/s/Hz. Its optimum receiving performance is slightly inferior to BPSK, yet is much superior to that of other UNB modulations such as VMSK/2. | 10 | Postreperfusion syndrome during orthotopic liver transplantation:a single-center experience显示文摘BACKGROUND: Marked hemodynamic alteration, commonly referred to as postreperfusion syndrome (PRS), often occurs after revascularization of the donor organ during orthotopic liver transplantation (OLT) and is associated with poor outcomes. This study aimed to investigate the incidence, predictive factors and clinical outcomes of PRS in Chinese patients following OLT at a liver transplantation center in China. METHODS: Over a 5-year period, 330 consecutive patients who had undergone OLT for hepatocellular carcinoma or cirrhosis were included in this retrospective study. PRS was defined as a >30% decrease in the mean arterial pressure compared with that before revascularization for more than 1 minute during the first 5 minutes of graft reperfusion. The patients were divided into 2 groups according to the development of PRS: group 1 (patients with PRS, n=56) and group 2 (patients without PRS, n=274). The demographic characteristics, operative and postoperative courses, and outcomes of the patients were analyzed using SPSS version 18.0. RESULTS: Multivariate regression analysis showed that left ventricular diastolic dysfunction determined by echocardiography and prolonged cold ischemia time were the independent risk factors for PRS. More patients in group 1 showed postoperative renal dysfunction than those in group 2 (19.23% vs 8.4%). | 11 | Expression of cyclooxygenase-1 and -2 in extra-hepatic cholangiocarcinoma显示文摘Objective: To investigate the expression of cyclooxy-genase-1 (COX-1) and cyclooxygenase-2 (COX-2)in extra-hepatic cholangiocarcinoma and the relation-ship between their expression and clinicopathologicalparameters.Methods: COX-1 and COX-2 were detected in 56 ex-tra-hepatic cholangiocarcinomas, including 31matched tissues originating from non-tumorous bileductal tissue adjacent to tumours and 6 normal bileductal tissues, by immunohistochemistry strept avi-din-biotin complex using isozyme selective antibod-ies.Results: There was no difference in expression ofCOX-1 between carcinomas (96%, 54/56) and non-cancerous specimens (94%, 29/31, P>0.05) ornormal bile ductal tissues (100%, 6/6, P>0.05).The positive rate of COX-2 expression in extra-hepat-ic cholangiocarcinomas (86%, 48/56) was signifi-cantly higher than their matched tissues (39%, 12/31, P<0.01) and normal bile ductal tissues (0%,0/6, P<0.01). | 13 | Abnormal expression of HSP gp96 associated with HBV replication in human hepatocellular carcinoma显示文摘BACKGROUND: Heat shock protein (HSP) gp96 is a member of the HSP90 family and presumably overexpresses as a result of stimulation by mutated or abnormal proteins. Its abnormal expression correlates with carcinogenesis, progression and prognosis of hepatocellular carcinoma (HCC). In this study, we investigated the pathological characteristics of liver gp96 expression and its relationship with hepatitis B virus (HBV) replication in HCC patients. METHODS: Tumor specimens were prospectively collected from 30 HCC patients undergoing liver resection. Total RNAs were extracted from HCC or their noncancerous tissues. The distribution of gp96 expression in hepatocytes was investigated by streptavidin peroxidase (S-P) immunohistochemistry and tissue HBV-DNA was detected by the in situ molecular hybridization technique. The association of gp96 expression with HBV replication, and the histopathological characteristics of HCC were analyzed. RESULTS: The gp96 was strongly expressed in HCC (73.3%, 22 of 30) and weakly (46.7%, 14 of 30) in non-cancerous tissues. The gp96 expression in HCC tissues was correlated with degree of tumor differentiation and tumor size, but not with tumor number (P>0.05). Immunohistochemical analysis showed that 17 of 19 HCC patients with HBVDNA -positive were strongly expressed for gp96, whereas only 5 of 11 patients with HBV-DNA-negative were positive for gp96. A significant difference was found between the two groups (89.5% vs. 45.5%, P<0.05). CONCLUSIONS: The abnormal expressions of HSP gp96 in HCC tissues are associated with HBV replication. | 14 | FREQUENTIST MODEL AVERAGING ESTIMATION:A REVIEW显示文摘In applications, the traditional estimation procedure generally begins with model selection.Once a specific model is selected, subsequent estimation is conducted under the selected model withoutconsideration of the uncertainty from the selection process. This often leads to the underreportingof variability and too optimistic confidence sets. Model averaging estimation is an alternative to thisprocedure, which incorporates model uncertainty into the estimation process. In recent years, therehas been a rising interest in model averaging from the frequentist perspective, and some importantprogresses have been made. In this paper, the theory and methods on frequentist model averagingestimation are surveyed. | 15 | Orogenic and Anorogenic Granitoids in the Altay Mountains of Xinjiang and Their Discrimination Criteria显示文摘On the basis of field geological studies of the granotoids in the region, mineralogical, petrological, geochemical(including stable isotope geochemical) and isotopic geochronological investigations were carried out on selected representative granitic bodies of various types. The authors have concluded that, apart from the bodies of the orogenic granitoid series, there also exist intrusions of the anorogenic granitoid series in the region. The intrusives of the two series were formed not only in different ages and tectonic environments, but also were derived from different sources of materials. Moreover, they are associated with different mineralizations, belonging to different minerologenetic series(Cheng et al., 1983). They show distinctly a series of discriminative criteria. | 17 | An analysis of 680 cases of cholangiocarcinoma from 8 hospitals显示文摘BACKGROUND: The outcome of patients with cholangio-carcinoma is poor. To evaluate the experience in the diagnosis and surgical treatment of cholangiocarcinoma, we investigated the status quo of diagnosis and treatment of cholangiocarcinoma in China. METHOD: The clinical data of 680 patients with cholangiocarcinoma treated at 8 hospitals from 1995 to 2001 were retrospectively analyzed with SPSS software package. RESULTS: The incidence of the tumor was the highest in the age group of 60-65 years. Meanwhile,the incidence was higher in aged men than in aged women, with a male to female ratio of 1.39:1. Proximal cholangiocarcinoma was the commonest ( 41.6% ) and distant cholangiocarcinoma the second(28.7% ) in the 680 patients. B-mode ultrasonogra-phy for cholangiocarcinoma was performed in 80.3% of the patients. Non-traumatic examinations such as computed tomography (CT), magnetic resonance image (MRI) and magnetic resonance cholangiopancreatography ( MRCP ) were more widely used than that of traumatic examinations such as percutaneous transhepatic cholangiography (PTC) and endoscopic retrograde cholangiopancreatography ( ER-CP). The low- and middle-differentiation cancer of the proximal bile duct accounted for about 50%. Most of the patients suffered from late-stage cholangiocarcinoma. The resection rate of the tumor was low, and the rate of radical operation was only 21.6% (147/680). CONCLUSIONS: Cholangiocarcinoma is common in the aged men. Its diagnosis and treatment have been improved, but little. Most patients are diagnosed as having late-stage cholangiocarcinoma at the time of outpatient clinic, and the rate of radical operation is low. | 20 | Primary scientific results of Chang'E-1 lunar mission显示文摘The strategic plan for the development of the unmanned Chinese Lunar Exploration Program is characterized by three distinct stages: 'orbiting around', 'landing on' and 'returning from' the Moon. The first Chinese lunar probe, Chang'E-1, which was successfully launched on October 24th, 2007 at Xichang Satellite Launch Center, and guided to crash on the Moon on March 1st, 2009, at 52.36°E, 1.50°S, in the north of Mare Fecunditatis, is the first step towards the 'orbiting around' stage. The Chang'E-1 mission lasted 495 days, exceeding the expected life-span by about four months. A total of 1.37 TB raw data was received from Chang'E-1. It was then processed into 4 TB scientific data products at various levels. Many scientific results have been obtained by analyzing these data, including especially the 'global lunar image from the first Chinese lunar explora- tion mission'. All scientific goals of Chang'E-1 have been achieved. It provides much useful materials for further advances of lunar sciences and planetary chemistry.
