1Inhibiting effect of antisense oligonucleotides phosphorthioate on gene expression of TIMP-1 in rat liver fibrosis显示文摘AIM To observe the inhibition of antisenseoligonucleotides (asON) phosphorthioate to thetissue inhibitors metalloproteinase-1 (TIMP-1)gene and protein expression in the liver tissue ofimmunologically induced hepatic fibrosis rats.The possibility of reversing hepatic fibrosisthrough gene therapy was observed.METHODS Human serum albumin (HSA) wasused to attack rats, as hepatic fibrosis model, inwhich asONs were used to block the gene andprotein expressing TIMP-1. According to theanalysis of modulator, structure protein, codingseries of TIMP-1 genome, we designed fourdifferent asONs. These asONs were injected intothe hepatic fibrosis models through coccygealvein. The results was observed by RT-PCR formeasuring TIMP-1 mRNA expression,immunohistochemistry and in situ hybridizationfor collagen Ⅰ, Ⅲ, special staining of collagenfiber, and electron microscopic examination.RESULTS Hepatic fibrosis could last within 363days in our modified model. The expressinglevel of TIMP-1 was high during hepatic fibrosisprocess. It has been proved by theimmunohistochemical and the electronmicroscopic examination that the asONphosphorthioate of TIMP-1 could exactly expressin vivo. The effect of colchicine wasdemonstrated to inhibit the expressing level ofmRNA and the content of collagen Ⅰ, Ⅲ in theliver of experimental hepatic fibrosis rats.However, the electron microscopy research andthe pathologic grading of hepatic fibrosisshowed that there was no significant differencebetween the treatment group and the modelgroup (P>0.05).CONCLUSION The experimental rat model ofhepatic fibrosis is one of the preferable modelsto estimate the curative effect of anti-hepaticfibrosis drugs. The asON phosphorthioate ofTIMP-1 could block the gene and proteinexpression of TIMP-1 in the liver of experimentalhepatic fibrosis rats at the mRNA level. It ispossible to reverse hepatic fibrosis, and it isexpected to study a new drug of anti-hepaticfibrosis on the genetic level. 2Contamination of Fungi and Mycotoxins in Foodstuffs in High Risk Area of Esophageal Cancer显示文摘Fungal and mycotoxin contamination has been found in fordstuffs from Cixian County, an area with a high incidence of esophageal carcinoma (Eca). To set a scientific foundation for the prevention of Eca at the etiological level, fungal and mycotoxin contaminations of local foodstuffs in Cixian County were analyzed using classical fungal culture methods and HPLC. From 1990 to 1994, 220 corn/wheat samples and 34 corn samples were studied. As a control, 26 corn samples collected from a relatively low incidence area of Eca in Zanhuang County were analyzed for mycotoxins in 1990. The results showed that fungal contamination in corn and wheat was severe and that several of the predominant contaminating fungi such as Furasium moniliforme, etc. were carcinogenic. HPLC results showed that the detection rate and mean content of the mycotoxin sterigmatocystin (ST) in the mountainous area (5/8; 9. 14 μg/kg) were significantly higher than those in the hilly (0; 0) and plain areas (1/18; 1. 29 μg/kg) as well as in low risk area control samples (3/26; 0. 76 μg/kg). Detection rates of deoxynivalenol (DON) in mountainous and hilly areas (5/8, 4/8 respectively) were slightly higher than that in plain area (8/18), while the mean content of DON in the plain area was significantly higher (90. 45-170. 22 vs 50. 56 and 46. 45 μg/kg).Among the different aflatoxins, AFB1 was detected in samples from the two villages in the plain area for the subsequent two years. The mean content of AFB1 was 0. 0183μg/kg, and the highest level was 0. 0497 μg/kg. No AFB1 was detected in the samples from mountainous and hilly areas. AFG1 was detected in more than half of all the samples, and its concentration ranged from 8. 77 to 46. 51 ng/kg. No AFG2 was found in the samples. 3Effect of Cultivation on Spatial Distribution of Nematode Trophic Groups in Black Soil显示文摘Geostatistics combined with GIS was applied to assess the spatial distribution of nematode trophic groups following two contrasting soil uses in the black soil region of Northeast China. Two plots, one with fallow for 12 years and the other cultivated, were marked on regular square grids with 2-m spacing. Soil samples were collected from each sampling point, nematodes were extracted from these samples and classified into four trophic groups: bacterivores, fungivores, plant parasites, and omnivores/predators. The numbers of total nematodes and trophic groups analyzed had normal distributions on both fallow and cultivated plots. The absolute abundances of total nematodes and trophic groups were observed to be much more homogeneous on cultivated plot than on fallow one. Geostatistical analysis showed that the densities of total nematodes and trophic groups on both fallow and cultivated plots exhibited spatial dependence at the sampled scale and their experimental semivariograms were adjusted to a spherical or exponential model, except those of bacterivores and fungivores on cultivated plot. 4SF/HGF-c-Met autocrine and paracrine promote metastasis of hepatocellular carcinoma显示文摘AIM: To explore the role of SF/HGF-Met autocrine and parscrine in metastasis of hepatocellular carcinoma (HCC).METHODS: SF/HGF and c-met transcription and protein expression in HCC were examined by RT-PCR and Western Blot in 4 HCC cell lines, including HepG2, Hep3B,SMMC7721 and MHCC-1, the last cell line had a higher potential of metastasis. Sf/hgf cDNA was transfected by the method of Lipofectin into SMMC7721. SF/HGF and c-met antibody were used to stimulate and block SF/HGF-c-met signal transduction. Cell morphology, mobility, and proliferation were respectively compared by microscopic observation, wound healing assay and cell growth curve.RESULTS: HCC malignancy appeared to be relative to its met-SF/HGF expression. In MHCC-1, c-met expression was much stronger than that in other cell lines with lower potential of metastasis and only SF/HGF autocrine existed in MHCC-1. After sf/hgf cDNA transfection or conditioned medium of MHCC-1 stimulation, SMMC7721 changed into elongated morphology, and the abilities of proliferation ( P < 0.05) and mobility increased. Such bio-activity could he blocked by c-met antibody ( P< 0.05).CONCLUSION: The system of SF/HGF-c-met autocrine and paracrine played an important role in development and metastasis potential of HCC. 5Expression of calreticulin is associated with infiltration of T-cells in stage ⅢB colon cancer显示文摘AIM: To investigate the correlation between expression of calreticulin and infiltration of lymphocytes in stage ⅢB colon cancer. METHODS: Sixty-eight pathologically-confirmed speci-mens were obtained from stage ⅢB (T3N1M0) colon cancer patients who underwent radical resection between January 1999 and May 2002 at the Cancer Center of Sun Yat-Sen University, Guangzhou, China. Immuno-histochemical analysis was performed to show infiltration of lymphocytes and expression of calreticulin in colon cancer. Association between calreticulin expression, infiltration of lymphocytes, and 5-year survival rate of patients was assessed. RESULTS: The expression level of calreticulin was lower in cancer nest than in its adjacent normal epithelium since 61.8% (42/68) of the samples were stained with calreticulin in colon cancer. The expression of calreticulin in colon cancer was associated with the infi ltration of CD45RO+ cells rather than with that of CD3+ cells. In addition, the stronger expression of calreticulin and the higher infiltration of CD3+ and CD45RO+ cells in colon cancer were associated with the higher 5-year survival rate of patients. 6Spatial heterogeneity of DTPA-extractable zinc in cultivated soils induced by city pollution and land use显示文摘The spatial heterogeneity of DTPA-extractable zinc in the cultivated soils of Shenyang suburbs in Liaoning Province of China was investigated, and its map was drawn by the methods of geostatistics combined with geographic information system. The data of soil DTPA-extractable zinc fitted normal distribution after logarithm transformation, and its semivariogram fitted a spherical model. The semivariogram indicated that the spatial dependence of soil DTPA-extractable zinc content was moderate, with the spatial dependence range of 1.69 km and the fractal dimension of 1.96. Stochastic factors contributed to 49.9% of the spatial variability, while structural factors contributed to 50.1% of it. The spatial heterogeneity of soil DTPA-extractable zinc shown by a kriged interpolation map was deeply influenced by stochastic factors such as city pollution, land use pattern and crop distributions. For example, the average content of Zn in vegetable garden soils was 2.5-4 times as much as in their originated soils, and was lower in paddy soils than in their originated soils. The areas with a higher content of soil DTPA-extractable zinc appeared in the near suburbs and the riverside along Hunhe River and the wastewater drainage of Xihe River, and the extremely high values in the near suburb of the city's residential area were a striking feature, indicating the key role of city pollution in the spatial heterogeneity of soil DTPA-extractable zinc. 7Report of 16 kindreds and one kindred with hMLH1 germline mutation显示文摘AIM: To analyze the diagnosis and treatment of 16 hereditarynonpolyposis colorectal cancer (HNPCC) kindreds, and toreport the first kindred with hMLH1 germline mutation inChina's Mainland.METHODS: The diagnosis, treatment and follow-up study of16 HNPCC kindreds were retrospectively reviewed. Dataconceming site of the malignant tumor, age st the diagnosis,history of synchronous and/ or metachronous cancer, andhistopathology of tumors were recorded. All treatments hadwon formal consent. PCR and SSCP were used to screen thecoding region of hMLH1 and hMSH2 genes. Variant bandswere sequenced by a 377 DNA sequencer.RESULTS: Among sixteen kindreds, sixty-eight patients hada mean age of 50.8 years, including twenty-one multiplecancer patients and forty-six colorectal cancer patients(metachronous colorectal cancers in sixteen). A total of onehundred and one malignant neoplasms were found in thesesixty-eight patients, including 50 colonic, 17 rectal, 11gastric, 7 endometrial, and 4 esophageal cancers. 39.5%colorectal patients had metachronous cancers within tenyears who needed reopeertions. A germline G265T nonsensemutation was found in the third exon of hMLH1, resulting in astop codon and truncated protein. Three phenotypically normalfamily members were also found to carry the mutated gene.ONCLUSION: HNPCC is a typical auto-dominant hereditaryisease, the main characteristics include early onset andfrequency of cancers; predominance of colorectal,especially right-sided colon cancers; frequency of multipleprimary cancers (especially colorectal cancers). Segmentalresection for colorectal cancers is not eligible for colorectalcancer patient in HNPCC kindreds. Intensive follow-up isessential for all patients and possible gene carriers. The firstHNPCC kindred with hMUH1 gene germline mutation wasidentified in China's Mainland, and three phenotypicallynormal family members were found to be carriers of themutated gene. 8KAI1 inhibits HGF-induced invasion of pancreatic cancer by sphingosine kinase activity显示文摘BACKGROUND:KAI1/CD82 has been reported to attenuate the process of metastases in a variety of tumors;however,its mechanism of action in invasion has not been fully elucidated. The present study aimed to investigate the importance of KAI1 in invasion and its correlation with activation of sphingosine kinase(SPK)in human pancreatic cancer PANC1 and Miapaca-2 cell lines. METHODS:The expression of KAI1 in PANC1 and Miapaca-2 cells,which was mediated by recombinant adenovirus(Ad-KAI1), was assessed by a flow cytometer and Western blotting.After successful infection was established,in vitro growth curve and invasive ability in Boyden Chamber assay were studied.The presence of KAI1 correlating with c-Met and SPK was detected by co-immunoprecipitation and[γ-32P]ATP incorporation. RESULTS:KAI1 genes had no significant effects on the curve representing cell growth.After infection with the KAI1 gene,decreased invasive ability in the Boyden Chamber assay was observed in PANC1 and Miapaca-2 cells that were induced by hepatocyte growth factor.Over-expression of KAI1 in the cells led to the deactivation of SPK and a decreased level of intracellular sphingosine-1-phosphate.No correlation was observed between c-Met and KAI1 during co-immunoprecipitation. 10Moleucle action mechunisms of NM-3 on human gastric cancer SGC-7901 Cells in vivo or in vitro显示文摘AIM: To study the moleucle action mechunisms of NM-3 on the growth of human gastric cancer SGC-7901 cells in vivo or in vitro.METHODS: SGC-7901 from human non-differentiated gastric cancer cell line was cultured with NM-3 at 100 mg/ml for 24 h. We observed its inhibitory rate and the density of micro-vascular growth in grafted mice with human gastric cancer SGC-7901. The apoptosis of human gastric cancer SGC-7901 was revealed in NM-3 treatment group by using terminal deoxynucleotidyl transferase-mediated deoxyuridine triphosphate-fluorescene nick end labeling (TUNEL)method and flow cytometry analysis.RESULTS: The growth of SGC-7901 cells was markedly inhibited compared with control growp, which was smaller than that in normal saline control group (4.17 g±0.22 g VS 9.45 g±1.38 g, P<0.01). The level of apoptosis of human gastric cell line SGC-7901 was obviously increased in NM-3treatment group at 1 mg.L-1 for 24 h. NM-3 inducing apoptotic index in NM-3 plus carboplatin group was 3.5 times that of carboplatin control group (TUNEL: 27.98±6.12 % VS 12.94±2.12 %, FACScan: 26.86±5.69 % VS11.86±1.09 %,P<0.01). Western blot analysis showed that the apoptotic index of human gastric cancer was elevated for 12, 24 and 36 h with an evident time-effect relationship in groups at 100 mg.L-L. NM-3 enhanced the inhibitive effects and sensitivity of chemotherapy for human gastric cancer in nude mice. 13Effects of slow-release urea fertilizers on urease activity, microbial biomass, and nematode communities in an aquic brown soil显示文摘A field experiment was carried out at the Shenyang Experimental Station of Ecology (CAS) in order to study the effects of slow-release urea fertilizers high polymer-coated urea (SRU1), SRU1 mixed with dicyandiamide DCD (SRU2), and SRU1 mixed with calcium carbide CaC2 (SRU3) on urease activity, microbial biomass C and N, and nematode communities in an aquic brown soil during the maize growth period. The results demonstrated that the application of slow-release urea fertilizers inhibits soil urease activity and increases the soil NH4+-N content.Soil available N increment could promote its immobilization by microorganisms. Determination of soil microbial biomass N indicated that a combined application of coated urea and nitrification inhibitors increased the soil active N pool. 14AN EXPERIMENTAL STUDY OF INFILTRATION AND EROSION UNDER SLOPE GRADIENTS AND VEGETAL COVERS显示文摘The effect of slope gradient and vegetal cover on soil infiltration and erosion were studied with field simulator. Results showed that infiltration decreases with slope gradients (especially for gradient less than 10°) and increases with vegetal covers, while soil erosion increases with slope gradients and decreases with vegetal covers. Fittlng the data by Philip’s infiltration equation it was found that in the equation, gravitational conductivity decreases with gradient and increases with vegetation, while diffusion decreases with vegetal cover and does not vary with slope gradient In the erosion process, the formation of a layer of thin water is crucial in dctermining the rainsplash and sheetwash. Recently, more attention has been paid to bio-diversity and complexity. According to the study on M-CSF and its receptor for years, the author suggests that, the multi-level of bio-diversity also appears at the bio-macromolecular level. Probability of bio-diversity is one of the bases for bio-complexity. Cellular sociology and topobiology are important aspects in bio-complexity, and should be developed. 16Thermal Decomposition Kinetics and Mechanism of Tb(Ⅲ) m-Methylbenzoate Complex with 1,10-Phenanthroline in Static Air Atmosphere显示文摘The thermal behavior of [Tb2( m -MBA)6(phen)2](H2O)2( m -MBA=C8H7O2, methoxybenzoate; phen=C 12 H8N2, 1,10-phenanthroline) in static air atmosphere was investigated by means of TG-DTG and DTA methods. The thermal decomposition of the title compound takes place mainly in two steps. The intermediate and the residue for each decomposition were identified by the TG curve. 17Characterization of SoxB2 and SoxC genes in Amphi-oxus (Branchiostoma belcheri):Implications for their evolutionary conservation显示文摘Most Sox genes directly affect cell fate determination and differentiation. In this study,we isolated two Sox genes:SoxB2 and SoxC from amphioxus (Branchiostoma belcheri),the closest living invertebrate relative of the vertebrates. Alignments of SoxB2 and SoxC protein sequences and their vertebrate homologs show high conservation of their HMG domains. Phylogenic analysis shows that amphioxus SoxB2 and SoxC fall out of the vertebrate branches,suggesting that vertebrate homologs might arise from gene duplications during evolution. The two genes possess similar spatial and temporal expression patterns during embryogenesis and in adults. They are both maternally inherited. During neurulation,they are expressed in the neural ectoderm and archenterons. In adults,they are expressed not only in the nerve cord,but also in the gut,midgut diverticulum,gill and oocytes. 19Chemiluminescence Determination of Molybdenum by on-Line Reduction with a Flow Injection System显示文摘The chemiluminescence (CL) reaction between lucigenin (Lu) and molybdenum (III) produced by a Jones rcductor was investigated using a flow injection system. On the basis of this, a novel method for the determination of trace amount of molybdenum has been established. The emission intensity was linear with molybdenum concentration in the range of 0.1-1000ng/mL; the detection limit was 0.02 ng/mL molybdenum, the relative standard deviation was less than 2% for the determination of 0.1 ng/mL molybdenum (n=11).
