| 1 | Meso-scale circulation at the intermediate-depth east of Mindanao observed by Argo profiling floats显示文摘The meso-scale circulation at the intermediate depth east of Mindanao is studied using Argo profiling floats observations. The trajectories and the parking-depth velocities of Argo floats show that the intermediate-depth circulation east of Mindanao contains significant meso-scale features that are highly variable both in space and in time. Both cyclonic and anticyclonic eddies at the intermediate depth (1000–2000 m) are indicated by the trajectories east of Mindanao. The mean tangential velocities of these eddies are about 10 cm/s at 2000 m and over 20 cm/s at 1000 m, which indicates that the geostrophic calculation may contain large errors due to the vigorous eddy activity at the reference levels. | 4 | Community structure, dispersal ability and functional profiling of microbiome existing in fat body and ovary of the brown planthopper, Nilaparvata lugens显示文摘The endosymbionts play vital roles in growth, development and reproduction in insects. Yeast-like endosymbionts (YLSs) have been well studied in Nilaparvata lugens (N. lugens), but little is known about the tissue-specific bacterial microbiomes, especially on the microbial intersection among internal tissues. Here, the correlation of microbial composition, structure, dispersal ability and functional profiling were illuminated in two tissues, the fat body and ovary in N. lugens. A total of 11 phyla and 105 genera were captured from all samples;Firmicutes and Proteobacteria were the most predominant and accounted for more than 99% in all samples. However, the relative abundance ofFirmicutes and Proteobacteria was significantly different in ovary and fat body through Fisher's Least Significant Difference test. Microbial diversity but not the richness index in the two tissues exhibited significant difference. Furthermore, the microbial community structure of the ovary and fat body were primarily determined by tissue quality. Firmicutes showed strong dispersal ability between ovary and fat body based on the quantitative null model assessing, indicating the frequent interaction of these microbiomes in the two tissues. In addition, the Kyoto Encyclopedia of Genes and Genomes pathways of microbial participation were delineated. The ten most abundant pathways counted for over 46% of the annotation and were shared between the two tissues, mainly containing Energy Metabolism and Amino Acid Metabolism/Biosynthesis. | 6 | Urinary nuclear magnetic resonance spectroscopy of a Bangladeshi cohort with hepatitis-B hepatocellular carcinoma: A biomarker corroboration study显示文摘AIM: To establish if a distinct urinary metabolic profile could be identified in Bangladeshi hepatitis-B hepatocellular carcinoma(HCC) patients compared to cirrhosis patients and controls. METHODS: Urine samples from 42 Bangladeshi patients with HCC(39 patients with hepatitis-B HCC), 47 with cirrhosis on a background of hepatitis B, 46 with chronic hepatitis B, and seven ethnically-matched healthy controls were analyzed using nuclear magnetic resonance(NMR) spectroscopy. A full dietary and medication history was recorded for each subject. The urinary NMR data were analyzed using principal component analysis(PCA) and orthogonal partial leastsquared discriminant analysis(OPLS-DA) techniques. Differences in relative signal levels of the most discriminatory metabolites identified by PCA and OPLSDA were compared between subject groups using an independent samples Kruskal-Wallis one-way analysis of variance(ANOVA) test with all pairwise multiple comparisons. Within the patient subgroups, the MannWhitney U test was used to compare metabolite levels depending on hepatitis B e-antigen(HBe Ag) status and treatment with anti-viral therapy. A BenjaminiHochberg adjustment was applied to acquire the level of significance for multiple testing, with a declared level of statistical significance of P < 0.05.RESULTS: There were significant differences in age(P < 0.001), weight(P < 0.001), and body mass index(P < 0.001) across the four clinical subgroups. Serum alanine aminotransferase(ALT) was significantly higher in the HCC group compared to controls(P < 0.001); serum α-fetoprotein was generally markedly elevated in HCC compared to controls; and serum creatinine levels were significantly reduced in the HCC group compared to the cirrhosis group(P = 0.004). A threefactor PCA scores plot showed clustering of the urinary NMR spectra from the four subgroups. Metabolites that contributed to the discrimination between the subgroups included acetate, creatine, creatinine, dimethyamine(DMA), formate, glycine, hippurate, and trimethylamine-N-oxide(TMAO). A comparison of relative metabolite levels confirmed that carnitine was significantly increased in HCC; and creatinine, hippurate, and TMAO were significantly reduced in HCC compared to the other subgroups. HBe Ag negative patients showed a significant increase in creatinine(P = 0.001) compared to HBe Ag positive patients in the chronic hepatitis B subgroup, whilst HBe Ag negative patients showed a significant decrease in DMA(P = 0.004) in the cirrhosis subgroup compared to HBe Ag positive patients. There were no differences in metabolite levels in HCC patients who did or did not receive antiviral treatment. CONCLUSION: Urinary NMR changes in Bangladeshi HCC were identified, corroborating previous findings from Egypt and West Africa. | 9 | Gut microbiome profiling and colorectal cancer in African Americans and Caucasian Americans显示文摘AIM To determine whether and to what extent the gut microbiome is involved in regulating racial disparity in colorectal cancer(CRC). METHODS All patients were recruited and experiments were performed in accordance with the relevant guidelines and regulations by the Institutional Review Boards (IRB), committees of the John D. Dingell VAMC and Wayne State University guidelines. African American (AA) and Caucasian American (CA) patients were scheduled for an outpatient screening for colonoscopy, and no active malignancy volunteer patients were doubly consented, initially by the gastroenterologist and later by the study coordinator, for participation in the study. The gut microbial communities in colonic effluents from AAs and CAs were examined using 16 sRNA profiling, and bacterial identifications were validated by performing SYBR-based Real Time PCR. For metagenomic analysis to characterize the microbial communities, multiple software/tools were used, including Metastats and R statistical software.RESULTS It is generally accepted that the incidence and mortality of CRC is higher in AAs than in CAs. However, the reason for this disparity is not well understood. We hypothesize that the gut microbiome plays a role in regulating this disparity. Indeed, we found significant differences in species richness and diversity between AAs and CAs. Bacteroidetes was more abundant in AAs than in CAs. In particular, the pro-inflammatory bacteria Fusobacterium nucleatum and Enterobacter species were significantly higher in AAs, whereas probiotic Akkermansia muciniphila and Bifidobacterium were higher in CAs. The polyphyletic Clostridia class showed a divergent pattern, with Clostridium XI elevated in AAs, and Clostridium IV, known for its beneficial function, higher in CAs. Lastly, the AA group had decreased microbial diversity overall in comparison to the CA group. | 10 | Updated therapeutic outcome for patients with periampullary and pancreatic cancer related to recent translational research显示文摘Chemotherapy with improved effect in patients with metastatic pancreatic cancer has recently been established, launching a new era for patients with this very aggressive disease. FOLFIRINOX and gemcitabine plus nab-paclitaxel are different regimens, both capable of stabilizing the disease, thus increasing the number of patients who can reach second line and even third line of treatment. Concurrently, new windows of opportunity open for nutritional support and other therapeutic interventions, improving quality of life. Also pancreatic surgery has changed significantly during the latest years. Extended operations, including vascular/multivisceral resections are frequently performed in specialized centers, pushing borders of resectability. Potentially curative treatment including neoadjuvant and adjuvant chemotherapy is offered new patient groups. Translational research is the basis for the essential understanding of the ongoing development. Even thou biomarkers for clinical management of patients with periampullary tumors have almost been lacking, biomarker driven trials are now in progress. New insight is constantly made available for clinicians; one recent example is selection of patients for gemcitabine treatment based on the expression level of the human equilibrium nucleoside transporter 1. An example of new diagnostic tools is identification of early pancreatic cancer patients by a three-biomarker panel in urine: The proteins lymphatic vessel endothelial hyaluronan receptor 1, regenerating gene 1 alpha and translation elongation factor 1 alpha. Requirement of treatment guideline revisions is intensifying, as combined chemotherapy regimens result in unexpected advantages. The European Study Group for Pancreatic Cancer 4 trial outcome is an illustration: Addition of capecitabine in the adjuvant setting improved overall survival more than expected from the effect in advanced disease. | 11 | 以PLGA为佐剂的花粉过敏原疫苗特异性免疫治疗过敏性鼻炎的研究显示文摘目的探讨用聚乳酸-羟基乙酸共聚物(PLGA)包载鱼尾葵花粉过敏原profilin蛋白的纳米疫苗特异性免疫治疗小鼠过敏性鼻炎的机制及疗效。方法将30只BALB/c小鼠随机分为5组:正常组、模型组、纳米疫苗治疗组、空白纳米治疗组及蛋白治疗组,每组6只。分别在第0、7、14天致敏小鼠,除正常组外,其余4组小鼠每次经腹腔注射50μg鱼尾葵花粉提取液和2mg Al(OH)3,正常组用PBS代替。致敏结束后,纳米疫苗治疗组、空白纳米组及蛋白治疗组分别使用PLGA-CmP纳米疫苗(含50μg profilin蛋白)、空PLGA纳米和50μg重组蛋白profilin经小鼠腹部皮下注射进行免疫治疗,每隔3d注射1次,共治疗5次。末次治疗后,取花粉粗浸液200μg分别滴鼻激发小鼠,每天1次,连续3d,激发结束后处死小鼠。小鼠激发后的30min内,观察并记录各组小鼠鼻部症状(鼻分泌物量、喷嚏次数及搔鼻)评分情况;检测血清中过敏原特异性抗体(IgE和IgG2a)及细胞因子(IL-4、IL-10和INF-γ)水平;观察鼻黏膜组织形态学变化。结果与模型组比较,蛋白治疗组及纳米疫苗治疗组小鼠鼻部症状评分、血清特异性IgE及IL-4水平均明显降低,纳米疫苗治疗组降低更明显,IgG2a、IL-10和INF-γ显著增加,纳米疫苗治疗组增加更明显(P<0.05或P<0.01)。结论以PLGA为佐剂的鱼尾葵花粉过敏原纳米疫苗可有效地抑制小鼠过敏性鼻炎的炎症反应,大大提高过敏原疫苗的治疗效果。 | 肖小军 张慧云 刘晓宇 马莉 何韶衡 刘志刚 | 2015 | 南昌大学学报(医学版)2015,55,6: | 3 |

| 13 | Evaluation of neuroactive effects of ethanol extract of Schisandra chinensis, Schisandrin, and Schisandrin B and determination of underlying mechanisms by zebrafish behavioral profiling显示文摘Schisandra chinensis, a traditional Chinese medicine(TCM), has been used to treat sleep disorders. Zebrafish sleep/wake behavioral profiling provides a high-throughput platform to screen chemicals, but has never been used to study extracts and components from TCM. In the present study, the ethanol extract of Schisandra chinensis and its two main lignin components, schisandrin and schisandrin B, were studied in zebrafish. We found that the ethanol extract had bidirectional improvement in rest and activity in zebrafish. Schisandrin and schisandrin B were both sedative and active components. We predicted that schisandrin was related to serotonin pathway and the enthanol extract of Schisandra chinensis was related to seoronin and domapine pathways using a database of zebrafish behaviors. These predictions were confirmed in experiments using Caenorhabditis elegans. | 14 | Copy number alteration profiling facilitates differential diagnosis between ossifying fibroma and fibrous dysplasia of the jaws显示文摘Ossifying fibroma(OF)and fibrous dysplasia(FD)are two fibro-osseous lesions with overlapping clinicopathological features,making diagnosis challenging.In this study,we applied a whole-genome shallow sequencing approach to facilitate differential diagnosis via precise profiling of copy number alterations(CNAs)using minute amounts of DNA extracted from morphologically correlated microdissected tissue samples.Freshly frozen tissue specimens from OF(n=29)and FD(n=28)patients were obtained for analysis.Lesion fibrous tissues and surrounding normal tissues were obtained by laser capture microdissection(LCM),with~30–50 cells(5000–10000µm2)per sample.We found that the rate of recurrent CNAs in OF cases was much higher(44.8%,13 of 29)than that in FD cases(3.6%,1 of 28).Sixty-nine percent(9 of 13)of the CNA-containing OF cases involved segmental amplifications and deletions on Chrs 7 and 12.We also identified eight CNA-associated genes(HILPDA,CALD1,C1GALT1,MICALL2,PHF14,AIMP2,MDM2,and CDK4)with amplified expression,which was consistent with the copy number changes.We further confirmed a jaw lesion with a previous uncertain diagnosis due to its ambiguous morphological features and the absence of GNAS mutation as OF based on the typical Chr 12 amplification pattern in its CNA profile.Moreover,analysis of a set of longitudinal samples collected from an individual with a cellular lesion in suspicion of OF at the first surgery,recurrence and the latest malignant transformation revealed identical CNA patterns at the three time points,suggesting that copy number profiling can be used as an important tool to identify borderline lesions or lesions with malignant potential.Overall,CNA profiling of fibro-osseous lesions can greatly improve differential diagnosis between OF and FD and help predict disease progression. | Ming Ma Lu Liu Ruirui Shi Jianyun Zhang Xiaotian Li Xuefen Li Jiaying Bai Jianbin Wang Yanyi Huang Tiejun Li | 2021 | International Journal of Oral Science2021,13,2: | 3 |

| 17 | 建立对生药进行准确鉴定和全面质量评价的二元条形码系统显示文摘由于很多常用生药来源繁多,导致市场上品种混乱,药材质量参差,需要准确、快速、可靠的鉴定与质量评价体系。本文提出建立生药鉴定与质量评价的二元条形码系统的解决方法,即基于小片段基因序列的DNA条形码和基于代谢谱的化学条形码数据库:分子条形码可准确鉴定物种基原,化学条形码可弥补其在质量评价方面的欠缺;待检测生药分别提取分子条形码和化学条形码,输入数据库进行数据处理,即可获得其准确的基原、产地、质量等全面信息。按DNA条形码技术规程采集植物标本和生药样品;测定matK、trnH-psbA、ITS等序列,确定适合作为条形码的区域,在BOLD平台建立DNA条形码数据库;以UPLC-MS检测样品的代谢谱,建立可用MZmine或CASE等程序进行分析的化学条形码数据库。二元条形码系统可望为生药的准确鉴定与全面质量评价提供新途径,并可用于探讨植物间的亲缘关系及系统发育。 | 晁志 慈薇 颜刚 谭小丹 | 2009 | 世界科学技术-中医药现代化2009,11,1: | 2 |
| 18 | polyketide and benzopyran compounds of an endophytic fungus isolated from Cinnamomum mollissimum:biological activity and structure显示文摘Objective:To study bioactivity and compounds produced by an endophytic Phoma sp.fungus isolated from the medicinal plant Cinnamomum mollissimum.Methods:Compounds produced by the fungus were extracted from fungal broth culture with ethyl acetate.This was followed by hioaclivity profiling of the crude extract fractions obtained via high performance liquid chromatography.The fractions were tested for cytotoxicity to P388 murine leukemic cells and antimicrobial activity against bacteria and pathogenic fungi.Compounds purified from active fractions which showed antibacterial,antifungal and cytotoxic activities were identified using capillary nuclear magnetic resonance analysis,mass spectrometry and admission to AntiMarin database.Results:Three known compounds,namely 4—hydroxymellein,4,8—dihydroxy—6—melhoxy—3—methyl—3,4-dihydro—1H-isochromen-1—one and 1—(2,6-dihydroxyphenyl) ethanone,were isolated from the fungus.The polyketide compound 4—hydroxymellein showed high inhibitory activity against P388 murine leukemic cells(94.6%) and the bacteria Bacillus sublilis(97.3%).Meanwhile.4,8—dihydroxy-6—melhoxy—3—meth) 1—3,4-dihydro—1H—isochromen—1-one,a benzopyran compound,demonstrated moderate inhibitory activity against P388 murine leukemic cells(48.8%)and the fungus Aspergillus niger(56.1%).The second polyketide compound.1(2,6—dihydroxyphenyl)ethanone was inactive against the tested targets.Conclusions:These findings demonstrate the potential of endophytes as producers ol pharmacologically important compounds,including polyketides which are major secondary metabolites in fungi. | Carolina Santiago Lin Sun Murray Herbert Gibson Munro Jacinta Santhanam | 2014 | Asian Pacific Journal of Tropical Biomedicine2014,4,8: | 2 |

| 20 | Proteomic analysis of insulin secretory granules in INS-1 cells by protein correlation profiling显示文摘Insulin secretory granules (ISGs),a group of distinguishing organelles in pancreatic β cells,are responsible for the storage and secretion of insulin to maintain blood glucose homeostasis.The molecular mechanisms of ISG biogenesis,maturation,transportation,and exocytosis are still largely unknown because the proteins involved in these distinct steps have not been fully identified.Subcellular fractionation by density gradient centrifugation has been successfully employed to analyze the proteomes of numerous organelles.However,use of this method to elucidate the ISG proteome is limited by co-fractionated contaminants because 1SGs are very dynamic and have abundant exchanges or contacts with other organelles,such as the Golgi apparatus,lysosomes,and endosomes.In this study, we developed a new strategy for identifying ISG proteins by protein correlation profiling (PCP)-based proteomics,which included ISG purification by OptiPrep density gradient centrifugation,label-free quantitative proteome,and identification of ISG proteins by correlating fractionation profiles between candidates and known ISG markers.Using this approach,we were able to identify 81 ISG proteins.
