1Further support for a Cretaceous age for the feathered-dinosaur beds of Liaoning,China:New ^(40)Ar/^(39)Ar dating of the Yixian and Tuchengzi Formations显示文摘We report new 40Ar/39Ar dating results ob-tained from total fusion and incremental-heating analyses of sanidine and biotite from three tuffs found interbedded within the fossil-bearing deposits of Liaoning, northeast China. The first is a new sample of the Bed 6 Sihetun tuff from the Yixian Formation, previously dated by our team as middle Early Cretaceous, and recently considered by Lo et al., partially reset due to metamorphism from a nearby ba-saltic sill. The second is the Yixian Bed 9 tuff from Heng-daozi considered by Lo et al. to be unaffected by metamor-phism and whose age, based on total fusion 40Ar/39Ar dating of biotite, argues for a Jurassic age for the Yixian Formation. The third tuff is a previously undated tuff from the upper part of the underlying Tuchengzi Formation. Single crystal total fusion 40Ar/39Ar analyses of the Sihetun sanidine showed homogeneous radiogenic Ar, Ca/K ratios, excellent reproducibility and gave a mean age of 125.0 ± 0.18 (1SD) ± 0.04 (SE) Ma. 3Relationships between the North Pacific Oscillation and the typhoon/hurricane frequencies显示文摘Relationships between the North Pacific Oscillation (NPO) and the typhoon as well as hurricane fre-quencies are documented. The correlation between NPO index in June-July-August-September and the annual typhoon number in the western North Pacific is 0.37 for the period of 1949―1998. The NPO is correlated with the annual hurricane number in the tropical Atlantic at -0.28 for the same period. The variability of NPO is found to be concurrent with the changes of the magnitude of vertical zonal wind shear, sea-level pressure patterns, as well as the sea surface temperature, which are physically asso- ciated with the typhoons and hurricanes genesis. 4Potash-rich volcanic rocks and lamprophyres in western Shandong Province:^(40)Ar-^(39)Ar dating and source tracing显示文摘Highly precise 40Ar-39Ar dating results demon-strate that the ages of potash-rich volcanic rocks in western Shandong Province are 114.7-124.3 Ma, and that of the lamprophyres is 119.6 Ma. The potash-rich volcanic rocks have relatively high (87Sr/86Sr)i ratios ( 0.708715-0.711418) and distinctly negative εNd values ( -11.47--17.54), and are enriched in radiogenic lead (206Pb/204Pb=17.341-17.622, 207Pb/204Pb=15.525-15.538, 208Pb/204Pb=37.563-37.684). Similarly, the lamprophyres also have quite low εNd values (-11.57 - -19.64). 5Relationship between the Asian-Pacific oscillation and the tropical cyclone frequency in the western North Pacific显示文摘The relationship between the Asian-Pacific oscillation (APO) and the tropical cyclone frequency over the western North Pacific (WNP) in summer is preliminarily investigated through an analysis of ob- served data. The result has shown clearly that APO is significantly and positively correlated to the tropical cyclone frequency in the WNP. If APO is above (below) the normal in summer, more (less) tropical cyclones will tend to appear in the WNP. The present study also addresses the large-scale at- mospheric general circulation changes underlying the linkage between APO and the WNP tropical cy- clone frequency. 6The^(40)Ar/^(39)Ar age record of formation and uplift of the blueschists and eclogites in the western Tianshan Mountains显示文摘The 40Ar/39Ar ages indicate that the eclogite facies rocks of the Hasiate slice in the western Tianshan Mountains were formed at the early stage of Devonian (401 Ma) and had been uplifted to the greenschist facies tectonic level in the middle stage of Devonian (381 Ma). The formation and uplift of the blueschists of the Akesayi slice are constrained to the late stage of Devonian (370-364 Ma). 7活血祛瘀法干预激素性股骨头坏死骨组织的修复显示文摘背景:激素性股骨头坏死的发病机制尚不明确,该病所伴随的塌陷破坏和重建性修复或与'骨形成-吸收'偶联失效所引起微观骨结构改变有关。活血祛瘀法经临床证实对该病有积极作用。但目前针对活血祛瘀法促进骨坏死修复的作用机制尚不清楚,相关国内外文献报道较少。目的:制备激素性股骨头坏死兔模型,探究活血祛瘀法对激素性股骨头坏死修复的干预作用。方法:50只新西兰兔随机分为3组:正常组10只、模型组和中药组(桃红四物汤组)各20只,采用内毒素联合甲强龙的方法制备激素性股骨头坏死模型,最后一次肌肉注射甲强龙后24 h,中药组给予0.3 g/kg桃红四物汤混悬液灌胃,正常组和模型组灌服等体积超纯水,连续8周。实验第8周分别进行高分辨率MRI检查和病理学组织检测以评估建模效果,采用Western Blot检测兔股骨头ABCB1、RUNX2、OPN、RANK、RANKL、PPAR、骨保护素、血管内皮生长因子的蛋白表达水平。结果和结论:(1)经苏木精-伊红染色显示:模型组可见部分骨小梁断裂,骨细胞核固缩,骨小梁空骨陷窝,骨髓细胞坏死,与正常组相比空骨陷窝率差异有显著性意义(P<0.05),中药组可见成骨活动仍较为活跃,骨小梁形态较好,说明活血祛瘀法可改善坏死区域组织修复。(2)高分辨率MRI显示:模型组股骨头外形基本正常,股骨头内可见低信号带;中药组股骨头外形趋于正常,股骨头内近骨骺处有小块低密度区,大部分骨组织灰度值正常,提示活血祛瘀法可改善坏死区域影像学改变。(3)Western Blot检测结果:模型组与正常组相比RUNX2、RANK、RANKL的蛋白表达升高,ABCB1、骨保护素、血管内皮生长因子蛋白表达下降;中药组与模型组相比,除RANK、RANKL表达下降外,其余均升高,差异有显著性意义(P<0.05)。(4)结果说明:活血祛瘀法促进兔激素性股骨头坏死的修复可能与调节ABCB1、RUNX2、RANK、RANKL、骨保护素、血管内皮生长因子蛋白的表达水平有关。陈雷雷 陈晓波 洪郭驹 陈达 杨鹏 何伟 2016中国组织工程研究2016,20,15:35
8Activating mechanism of transcriptor NF-kappaB regulated by hepatitis B virus X protein in hepatocellular carcinoma显示文摘AIM:To investigate the mechanism and significance of NF-κB activation regulated by hepatitis B virus X protein (HBx) in hepatitis B virus (HBV)-associated hepatocellular carcinoma (HCC).METHODS:The expression levels of HBx, p65,IκB-α and ubiquitin were detected by immunohistochemistry in HCC tissue microarrays (TMA) respectively, and IκB-α was detected by Western blot in HCC and corresponding liver tissues.RESULTS: The percentage of informative TMA samples was 98.8% in 186 cases with a total of 367 samples. Compared with corresponding liver tissues (60.0%),the HBx expression was obviously decreased in HBV-associated HCC (47.9%,u=2.24,P<0.05).On the contrary, the expressions of p65 (20.6% vs45.3%, u=4.85, P<0.01) and ubiquitin (8.9% vs 59.0%,u=9.68,P<0.01) were notably elevated in HCC.In addition, IκB-α had a tendency to go up. Importantly, positive relativity was observed between HBx and p65 (X^2=10.26,P<0.01), p65 and IκB-α (x^2=16.86,P<0.01), IκB-α and ubiquitin (x^2=8.90, P<0.01) in HCC, respectively.CONCLUSION:Both active and non-active forms of NF-κB are increased in HBV-associated HCC. Variant HBx is the major cause of the enhancement of NF-κB activity. 10黄芩苷对FM1肺炎小鼠肺损伤的作用机制研究显示文摘目的研究黄芩苷对甲型流感病毒性肺炎小鼠肺组织炎性损伤的作用机制。方法将96只ICR鼠随机分为正常对照组、模型组、利巴韦林组(100 mg.kg-1),黄芩苷组(1 500、750、375 mg.kg-1)[5],每组16只。用流感病毒亚甲型鼠肺适应株A/FM/1/47(H1N1)感染小鼠,制备小鼠流感病毒性肺炎模型,用不同剂量黄芩苷溶液灌胃治疗后,观察肺组织H-E染色切片病理变化,采用RT-PCR测定肺组织c-jun、c-fos mRNA表达,采用Western blot测定肺组织c-jun、磷酸化c-jun蛋白表达,应用ELISA检测肺匀浆炎性细胞因子TNF-α、IL-1β的含量。结果黄芩苷750、375 mg.kg-1剂量均能明显减轻肺组织炎性损伤;明显降低c-jun、c-fosmRNA的表达(P<0.05,P<0.01),明显降低c-jun、磷酸化c-jun蛋白表达(P<0.01);明显抑制TNF-α、IL-1β的分泌(P<0.01)。结论黄芩苷能够缓解甲型流感病毒诱导的炎性病理损伤,其可能通过抑制流感病毒感染引起的转录因子AP-1高表达而降低炎性细胞因子的分泌水平,从而发挥抗流感病毒感染的作用。万巧凤 顾立刚 殷胜骏 吴珺 邱泽计 2012中国药理学通报2012,28,2:33
11艾灸循经筋阿是穴对膝骨关节炎RANKL和OPG调控机制研究显示文摘目的:观察艾灸循经筋对膝骨性关节炎的镇痛疗效以及对破骨细胞分化因子(RANKL)及破骨细胞分化抑制因子(OPG)调控机制的表达影响。方法:采用随机原则将膝骨关节炎患者分为经穴组、阿是穴组以及循经筋规律性阿是穴组,共90例患者,连续治疗4周。分别于治疗前后采用疼痛VAS评分比较,抽取膝关节滑液分离细胞,检测RANKL及OPG表达变化,观察比较各疗法的镇痛疗效和对RANKL及OPG调控机制。结果:三组均有明显的镇痛效果,经穴组与阿是穴组治疗后RANKL及OPG显著降低,循经筋阿是穴组具有更强的抑制RANKL及OPG表达的作用,统计学有显著性差异。结论:艾灸对膝骨关节炎止痛效果良好,在抑制RANKL及OPG介导的炎症反应方面,经穴灸法与阿是穴灸法具有明显的抑制作用,且循经筋阿是穴灸法疗效更佳。董宝强 张书剑 李春日 韩煜 陈以国 2013辽宁中医药大学学报2013,15,10:31
12Analysis of gene expression profile induced by EMP-1 in esophageal cancer cells using cDNA Microarray显示文摘AIM: To obtain human esophageal cancer cell EC9706 stably expressed epithelial membrane protein-1 (EMP-1) with integrated eukaryotic plasmid harboring the open reading frame (ORF) of human EMP-1, and then to study the mechanism by which EMP-1 exerts its diverse cellular action on cell proliferation and altered gene profile by exploring the effect of EMP-1.METHODS: The authors first constructed pcDNA3.1/mychis expression vector harboring the ORF of EMP-1 and then transfected it into human esophageal carcinoma cell line EC9706. The positive clones were analyzed by Western blot and RT-PCR. Moreover, the cell growth curve was observed and the cell cycle was checked by FACS technique. Using cDNA microarray technology, the authors compared the gene expression pattern in positive clones with control. To confirm the gene expression profile, semi-quantitative RT-PCR was carried out for 4 of the randomly picked differentially expressed genes. For those differentially expressed genes,classification was performed according to their function and cellular component.RESULTS: Human EMP-1 gene can be stably expressed in ECg706 cell line transfected with human EMP-1. The authors found the cell growth decreased, among which S phase was arrested and G1 phase was prolonged in the transfected positive clones. By cDNA microarray analysis, 35 genes showed an over 2.0 fold change in expression level after transfection, with 28 genes being consistently up-regulated and 7 genes being down-regulated. Among the classified genes, almost half of the induced genes (13 out of 28 genes) were related to cell signaling, cell communication and particularly to adhesion.CONCLUSION: Overexpression of human EMP-1 gene can inhibit the proliferation of EC9706 cell with S phase arrested and G1 phase prolonged. 13New predictors and a new prediction model for the typhoon frequency over western North Pacific显示文摘In this paper, the impacts of the atmospheric circulation during boreal winter-spring on the western North Pacific (WNP) typhoon frequency (WNPTF) are studied. Several new factors in winter-spring in- fluencing the typhoon frequency were identified, including the sea ice cover in the North Pacific and the North Pacific oscillation. Based on these results, the multi-linear regression was applied to establishing a new forecast model for the typhoon frequency by using the datasets of 1965―1999. The forecast model shows a high correlation coefficient (0.79) between the model simulated and the actual typhoon frequencies in the period of 1965―1999. The forecast model also exhibits reasonable hindcasts for the typhoon frequencies for the years 2000―2006. Therefore, this work demonstrates that the new pre- dictors are significant for the prediction of the interannual variability of the WNPTF, which could be potentially used in the operational seasonal forecast of the typhoon frequency in the WNP to get a more physically based operational prediction model and higher forecast skill.FAN Ke 2007Science China Earth Sciences2007,50,9:27
14^(40)Ar/^(39)Ar age evidence for Altyn fault tectonic activities in western China显示文摘Four 40Ar/39Ar age groups of mica, hornblende and K-feldspar were obtained from Proterozoic and early Paleozoic metamorphic rocks in the Aksay-Dangjin Pass area, western China. The samples away from the middle shear zone of the Altyn fault belt yield two plateau age groups in the range of 461—445.2 Ma and 414.9—342.8 Ma, respec-tively. They represent the tectono-thermal events that had been recorded in the rocks that were displaced by the Altyn strike-slip fault in late Ordovician-early Silurian and Devo-nian, respectively. These two age groups should be related to the closures of Northern and Southern Qilian Oceans. The deformed granitic gneiss from the northern belt gives a pla-teau age group of 178.4—137.5 Ma, which is interpreted as the active age of the Altyn fault in the middle-late Jurassic- early Cretaceous and should be related to the accretion of Lhasa block to the north. The sample from the middle shear zone of the Altyn fault belt yields two plateau ages of 36.4 and 26.3 Ma, respectively, suggesting the strike-slip move-ment with strong metamorphism at greenschist facies along the Altyn fault in the late Eocene. This event occurred in the most areas of the northern Tibet Plateau and should be in response in the north to the collision between Indian and Eurasian continents. The present study demonstrates that the Altyn fault is characterized by multiple pulse-style activi-ties under the tectonic setting of convergence between the Indian and Eurasian continents.LIU Yongjiang1, J. 16The age and tectonic environment of the rhyolitic rocks on the western side of Wuyi Mountain,South China显示文摘During the geological survey of the metamorphic rocks in Xingning-Wuhua region on the western side of Wuyi Mountain, South China, we discovered the Neoproterozoic rhyolite and rhyolitic greywacke for the first time that outcrop in the Proterozoic metamorphic rocks near Jingnan Town of Xingning County, eastern Guangdong Province. A systematic research on petrology, geochemistry and geochronology of rhyolitic rocks was conducted to understand their tectonic setting and formation age. The Jingnan rhyolite is interbedded with a coeval greywacke, with a total thickness of 60 m; both rhyolite and greywacke display a similar folding and metamorphic pattern. Meta-rhyolite consists of groundmass and phenocrystals including sanidine, orthoclase, and quartz with distinct undulose extinction; the groundmass has been recrystallized into fine-grain feldspar, quartz and sericite aggregation. Meta-greywacke is composed of crystallinoclastic grains (sanidine, orthoclase, quartz and oligoclase) and clay groundmass. Zircon grains used for the SHRIMP U-Pb analysis are light brown-colored and euhedral or subeuhedral. Dating data suggest two age groups; eight grains of magmatype zircon with an idiomorphic form yield an age of 972±8 Ma, and the other seven weakly corroded grains of zircon with euhedral to subeuhedral shape construct an average age of 1097±11 Ma, which were captured from older rocks by an uplifting magma, implying that a late Mesoproterozoic basement exists in the Nanling region. In addition, one Paleoproterozoic age, 2035±11 Ma, is obtained from a rounded detrital zircon, indicating that a Paleoproterozoic thermal event took place in the South China. Geochemically, the Jingnan rhyolitic rocks are characterized by high K2O content, intermediate Al2O3 content, with the ACNK value 0.98―1.11, and belonging to high-K alkaline series. They are rich in ΣREE, Rb, Th and Ce, depleted in Ba, Sr, Eu, Ti, P and Nb-Ta, and with moderate negative Eu and Sr anomalies. 17The subducted slab of Yangtze Continental block beneath the Tethyan orogen in western Yunnan显示文摘The western Yunnan area is a natural laboratory with fully developed and best preserved Tethyan orogen in the world. Seismic tomography reveals a slab-like high velocity anomaly down to 250 km beneath the western Yunnan Tethyan orogen, to its west there is a low-velocity column about 300 km wide. in the region from Lancangjiang to Mojiang an obvious low velocity in the lower crust and uppermost mantle overlies on the slab. Synthesizing the available geological and geochemical results, the present paper demonstrates that this slab-like high velocity anomaly is a part of the subducted plate of Yangtze Continental segment after the closure of Paleotethys. 18香鱼凝血因子X基因表达与鳗利斯顿氏菌感染的相关性显示文摘凝血途径的关键因子--凝血因子X(coagulation factor X,FX),是一种与免疫调控密切相关的维生素K依赖型丝氨酸蛋白酶。该研究克隆了香鱼(Plecoglossus altivelis)FX基因全长cDNA序列,它由1817个核苷酸组成,包含一个大的开放阅读框,编码一个由453个氨基酸组成的相对分子质量为5.07×104的蛋白。香鱼FX与已知的哺乳动物FX的结构相似,N端24个残基为信号肽序列。序列比较表明,香鱼FX与斑马鱼FX的氨基酸同一性最高,为53%。在健康香鱼中,FX基因mRNA主要在肝组织中表达,脑和鳃中也有少量表达。实时荧光定量PCR分析揭示,鳗利斯顿氏菌感染后香鱼肝组织中FX基因mRNA表达显著上调,16h时达到5.43倍。通过原核表达系统表达了香鱼FX丝氨酸蛋白酶结构域,并制备了抗血清。Western blotting分析显示,鳗利斯顿氏菌感染后香鱼血清中FX含量显著增加,并随着时间延长上调倍数不断增大,在36h时达到3.68倍。据此,FX基因mRNA及蛋白的表达与鳗利斯顿氏菌感染香鱼的过程紧密相关,揭示它可能在鱼类抗细菌感染的免疫反应中起重要作用。黄左安 陈炯 陆新江 史雨红 李明云 2011Zoological Research2011,32,5:25
19榄香烯对大鼠胶质瘤C6细胞Bcl-2家族基因及蛋白表达的影响显示文摘目的探讨莪术提取物-榄香烯诱导大鼠胶质瘤C6细胞系凋亡机制及其对Bcl2家族基因及蛋白质表达的影响。方法应用RTPCR、Westernblot方法检测榄香烯在0、20、40、60、80μg/ml浓度及作用时间为12、24、36、48、72h时处理的大鼠胶质瘤C6细胞系,利用流式细胞仪观察细胞的凋亡、细胞增殖的变化和Bcl2家族基因及蛋白质表达的变化。结果经榄香烯20、40、60、80μg/ml处理大鼠胶质瘤C6细胞48h后,单视野细胞计数分别为(536±9)个、(375±10)个、(246±9)个、(112±10)个,与浓度为0μg/ml时单视野细胞计数(625±12)个相比体外增殖抑制效应比差异有统计学意义(F=1292.416,P<0.05)。经不同浓度榄香烯处理后C6细胞凋亡率分别增加到(27±2)%、(29±4)%、(32±3)%、(35±5)%,同时有亚二倍体凋亡峰,即Ap峰出现。榄香烯可明显下调Bcl2/Bclx/l基因及蛋白质表达,且该作用呈浓度、时间依赖性,而对Bax基因及蛋白质无明显影响。结论下调Bcl2/Bclx/l基因及蛋白质表达可能是榄香烯诱导凋亡、抑制C6细胞增殖的主要机制。徐英辉 董斌 罗其中 周洪语 贾宜昌 杨友峰 王以政 2005中华医学杂志2005,85,24:24
20凋亡相关基因Livin、Smac/DIABLO在胃癌组织中的表达及其意义显示文摘背景与目的:作为一种促凋亡基因,Smac/DIABLO与凋亡抑制基因Livin相互作用,共同参与调控细胞癌变的起始及发展。本研究检测凋亡抑制蛋白Livin及线粒体促凋亡蛋白Smac/DIABLO在胃癌组织中的表达,观察其表达水平与胃癌各临床病理因素的关系。方法:应用实时荧光定量PCR SYBR Green荧光染料法检测75例手术切除的胃癌组织、20例癌旁组织及正常胃组织中Livin mRNA和Smac/DIABLO mRNA的表达,Western blot结合及免疫组化SP法检测两种蛋白的表达及组织学定位。结果:正常胃组织及癌旁组织中均无Livin mRNA表达,胃癌组织中Livin mRNA表达显著上调,相对表达量为6.374±4.759,低分化胃腺癌组织及有淋巴结转移组的相对表达量显著高于中高分化胃腺癌组织(P<0.05),其表达与肿瘤大小、浸润深度及TNM分期无关,Livin蛋白的表达结果与之相符;胃癌组织中Smac/DIABLO mRNA的表达(0.731±0.420)低于正常胃组织(1.104±0.276)及癌旁组织(1.061±0.737),但差异无统计学意义(P>0.05),Smac/DIABLO mRNA表达水平与胃癌各临床病理因素无关;Smac/DIABLO蛋白表达与mRNA表达存在差异。Smac/DIABLO在肠型胃癌(84.38%)与弥漫型胃癌(60.65%)中的表达有一定差异(P<0.05)。结论:Livin与Smac/DIABLO在不同阶段和不同病理类型胃癌中的表达及相关性存在差异,Livin基因高表达与胃癌的发生及分化转移密切相关。赵毅 邓鑫 王强 2009癌症2009,28,6:24
