| 1 | Fatty acid content in epididymal fluid and spermatozoa during sperm maturation in dogs显示文摘Background: During sperm maturation, there is a reorganization of fatty acids from plasmatic membrane of the spermatozoa, which allows higher membrane integrity and acquisition of sperm motility. However, the fatty acid profile during sperm maturation remains unclear in dogs. Thus, the aim of this study was to identify the fatty acids from the epididymal spermatozoa and plasma during the sperm maturation, and observed changes in the motility and plasmatic membrane parameters. Twenty one adult dogs were used, subsequently to bilateral orchiectomy and epididymal storage, sperm samples were collected from the different segments of the epididymis. Samples were evaluated for conventional microscopy, computer-assisted motility analysis, sperm plasma membrane permeability and the fatty acid analysis(lipids were extracted, transmethylated and analyzed by chromatography).Results: Caput and corpus sperm showed lower values for the motility variables evaluated and plasmatic membrane integrity, indicating different levels of the fatty acids organization. Saturated, monounsaturated and polyunsaturated fatty acids were in higher concentrations in the spermatozoa from epididymis cauda. | 2 | Inhibition of BACE1, the β-secretase implicated in Alzheimer’s disease, by a chondroitin sulfate extract from Sardina pilchardus显示文摘The pharmaceutical and anticoagulant agent heparin,a member of the glycosaminoglycan family of carbohydrates,has previously been identified as a potent inhibitor of a key Alzheimer’s disease drug target,the primary neuronalβ-secretase,β-site amyloid precursor protein cleaving enzyme 1(BACE1).The anticoagulant activity of heparin has,however,precluded the repurposing of this widely used pharmaceutical as an Alzheimer’s disease therapeutic.Here,a glycosaminoglycan extract,composed predominantly of 4-sulfated chondroitin sulfate,has been isolated from Sardina pilchardus,which possess the ability to inhibit BACE1(IC50[half maximal inhibitory concentration]=4.8μg/mL),while displaying highly attenuated anticoagulant activities(activated partial thromboplastin time EC50[median effective concentration]=403.8μg/mL,prothrombin time EC50=1.3 mg/mL).The marine-derived,chondroitin sulfate extract destabilizes BACE1,determined via differential scanning fluorimetry(ΔTm–5°C),to a similar extent as heparin,suggesting that BACE1 inhibition by glycosaminoglycans may occur through a common mode of action,which may assist in the screening of glycan-based BACE1 inhibitors for Alzheimer’s disease. | Courtney J.Mycroft-West Anthony J.Devlin Lynsay C.Cooper Patricia Procter Gavin J.Miller David G.Fernig Marco Guerrini Scott E.Guimond Marcelo A.Lima Edwin A.Yates Mark Andrew Skidmore | 2020 | Neural Regeneration Research2020,15,8: | 2 |

| 5 | 膳食模式与动脉粥样硬化关系的多种族研究显示文摘目的人们对膳食模式、代谢紊乱与左心室功能的关系研究报道不多,本研究旨在探讨膳食模式和左心室功能之间的关系以及代谢紊乱与动脉粥样硬化的关系。方法膳食模式采用降秩回归(reduced rankregression,RRR)方法构建;左室重量、心搏量和左室射血分数采用磁共振成像测量;膳食模式和左室指数之间的关系采用横断面分析法。结果选用4 601例45~84岁、无临床心血管疾病受试者,RRR膳食模式得分数与高血糖指数、高脂肉类、奶酪、加工食品的摄入量呈正相关,而与蔬菜、大豆、水果、绿茶、黑茶、低脂甜点、种子、坚果和鱼肉的低摄入量呈负相关。多变量分析显示,RRR膳食模式得分增加1个单位,左室重量/体表面积就增加0.32g/m2,心博量减少0.43mL/m2,左室射血分数则减少0.21%。调整代谢综合征组分后,RRR膳食模式得分与左室重量和心搏指数之间的相关性减低,变为差异有统计学意义(P<0.05)。结论 RRR膳食模式、代谢紊乱与左室功能紊乱有关。本研究属于横断面研究,结果的可靠性有待前瞻性研究进一步证实。 | 刘隆健 孔翔瑜 Jennifer A.Nettleton Alain G.Bertoni David Bluemke Joo A.Lima Moyses Szklo 邹强 杨书 李茂全 陈建 | 2013 | 成都医学院学报2013,8,2: | 0 |
