| 2 | Membrane fouling in ultrafiltration of natural water after pretreatment to different extents显示文摘The combined fouling during ultrafiltration(UF) of surface water pretreated to different extents was investigated to disclose the roles of polysaccharides, proteins, and inorganic particles in UF membrane fouling. Both reversible and irreversible fouling decreased with enhanced pretreatment(biologically active carbon(BAC) treatment and sand filtration). The sand filter effluent fouled the membrane very slowly. The UF membrane removed turbidity to less than 0.1 nephelometric turbidity unit(NTU), reduced polysaccharides by 25.4%–29.9%, but rejected few proteins. Both polysaccharides and inorganic particles were detected on the fouled membranes, but inorganic particles could be effectively removed by backwashing. The increase of turbidity in the sand filter effluent to 3.05 NTU did not significantly increase the fouling rate, but an increase in the turbidity in the BAC effluent to6.11 NTU increased the fouling rate by more than 100%. The results demonstrated that the polysaccharide, not the protein, constituents of biopolymers were responsible for membrane fouling. Membrane fouling was closely associated with a small fraction of polysaccharides in the feed water. Inorganic particles exacerbated membrane fouling only when the concentration of fouling–inducing polysaccharides in the feed water was relatively high. | 3 | Effects of multidrug resistance, antisense RNA on the chemosensitivity of hepatocellular carcinoma cells显示文摘BACKGROUND: Multidrug resistance is a major obstacle in cancer chemotherapy. We examined whether the antisense RNA of multidrug resistance gene 1 (mdr1) could reverse multidrug resistance in the human hepatocellular carcinoma (HCC) cell line SMMC7721/ADM. METHODS: The recombinant adenoviruses pAdEasy- GFP-ASmdr1 product was produced by the adenoviral vector AdEasy system, which can express antisense RNA against the mdr1 gene. Following that, the recombinant adenovirus was transfected into the P-glycoprotein- producing multidrug resistance cell line, SMMC7721/ADM human HCC cells resistant to adriamycin (ADM) and daunorubicin (DNR). In order to investigate the reversal of multidrug resistance phenotype, we measured the expression of mdr1 mRNA by RT-PCR and the production of P-glycoprotein by flow cytometry. The sensitivities for ADM and DNR SMMC7721/ADM cells were examined by [3-(4, 5-dimethylthi-azol-2-yl)-2,5 diphenyl-terazolium bromide] (MTT) analysis. RESULTS: The low-level expression of mdr1 mRNA and P-glycoprotein production were observed in parental sensitive cells SMMC/7721 in addition to the overexpressionof mdr1 mRNA and P-glycoprotein in SMMC7721/ADM cells. The transfection of antisense-RNA into SMMC7721/ ADM cells resulted in decreases of mdr1 mRNA and P-glycoprotein, but increase of drug sensitivities. The sensitivities of transfected SMMC7721/ADM cells to ADM and DNR in IC50 reduced by 31.25% and 62.96% respectively. CONCLUSIONS: Mdr1 antisense RNA can increase the sensitivities of SMMC7721/ADM cells to anticancer drug by decreasing the expression of the mdr1 gene and inhibiting P-glycoprotein expression. | 4 | Cytocompatibility with osteogenic cells and enhanced in vivo anti-infection potential of quaternized chitosan-loaded titania nanotubes显示文摘Infection is one of the major causes of failure of orthopedic implants. Our previous study demonstrated that nanotube modification of the implant surface, together with nanotubes loaded with quaternized chitosan(hydroxypropyltrimethyl ammonium chloride chitosan, HACC), could effectively inhibit bacterial adherence and biofilm formation in vitro. Therefore, the aim of this study was to further investigate the in vitro cytocompatibility with osteogenic cells and the in vivo anti-infection activity of titanium implants with HACC-loaded nanotubes(NT-H). The titanium implant(Ti), nanotubes without polymer loading(NT), and nanotubes loaded with chitosan(NT-C) were fabricated and served as controls. Firstly, we evaluated the cytocompatibility of these specimens with human bone marrow-derived mesenchymal stem cells in vitro.The observation of cell attachment, proliferation, spreading, and viability in vitro showed that NT-H has improved osteogenic activity compared with Ti and NT-C. A prophylaxis rat model with implantation in the femoral medullary cavity and inoculation with methicillin-resistant Staphylococcus aureus was established and evaluated by radiographical, microbiological, and histopathological assessments. Our in vivo study demonstrated that NT-H coatings exhibited significant anti-infection capability compared with the Ti and NT-C groups. | 7 | Rational use of mesenchymal stem cells in the treatment of autism spectrum disorders显示文摘Autism and autism spectrum disorders(ASD) refer to a range of conditions characterized by impaired social and communication skills and repetitive behaviors caused by different combinations of genetic and environmental influences. Although the pathophysiology underlying ASD is still unclear, recent evidence suggests that immune dysregulation and neuroinflammation play a role in the etiology of ASD. In particular, there is direct evidence supporting a role for maternal immune activation during prenatal life in neurodevelopmental conditions. Currently, the available options of behavioral therapies and pharmacological and supportive nutritional treatments in ASD are only symptomatic. Given the disturbing rise in the incidence of ASD, and the fact that there is no effective pharmacological therapy for ASD, there is an urgent need for new therapeutic options. Mesenchymal stem cells(MSCs) possess immunomodulatory properties that make them relevant to several diseases associated with inflammation and tissue damage. The paracrine regenerative mechanisms of MSCs are also suggested to be therapeutically beneficial for ASD.Thus the underlying pathology in ASD, including immune system dysregulation and inflammation, represent potential targets for MSC therapy. | 15 | Genome-wide association study and genomic prediction of Fusarium ear rot resistance in tropical maize germplasm显示文摘Fusarium ear rot(FER)is a destructive maize fungal disease worldwide.In this study,three tropical maize populations consisting of 874 inbred lines were used to perform genomewide association study(GWAS)and genomic prediction(GP)analyses of FER resistance.Broad phenotypic variation and high heritability for FER were observed,although it was highly influenced by large genotype-by-environment interactions.In the 874 inbred lines,GWAS with general linear model(GLM)identified 3034 single-nucleotide polymorphisms(SNPs)significantly associated with FER resistance at the P-value threshold of 1×10^(-5),the average phenotypic variation explained(PVE)by these associations was 3%with a range from 2.33%to 6.92%,and 49 of these associations had PVE values greater than 5%.The GWAS analysis with mixed linear model(MLM)identified 19 significantly associated SNPs at the P-value threshold of 1×10^(-4),the average PVE of these associations was 1.60%with a range from 1.39%to 2.04%.Within each of the three populations,the number of significantly associated SNPs identified by GLM and MLM ranged from 25 to 41,and from 5 to 22,respectively.Overlapping SNP associations across populations were rare.A few stable genomic regions conferring FER resistance were identified,which located in bins 3.04/05,7.02/04,9.00/01,9.04,9.06/07,and 10.03/04.The genomic regions in bins 9.00/01 and 9.04 are new.GP produced moderate accuracies with genome-wide markers,and relatively high accuracies with SNP associations detected from GWAS.Moderate prediction accuracies were observed when the training and validation sets were closely related.These results implied that FER resistance in maize is controlled by minor QTL with small effects,and highly influenced by the genetic background of the populations studied.Genomic selection(GS)by incorporating SNP associations detected from GWAS is a promising tool for improving FER resistance in maize. | Yubo Liu Guanghui Hu Ao Zhang Alexander Loladze Yingxiong Hu Hui Wang Jingtao Qu Xuecai Zhang Michael Olsen Felix San Vicente Jose Crossa Feng Lin Boddupalli M.Prasanna | 2021 | The Crop Journal2021,9,2: | 3 |

| 17 | Wuzi Yanzong prescription(五子衍宗丸)from Traditional Chinese Medicine for male infertility:a narrative review显示文摘Wuzi Yanzong prescription(WYP),as a classical prescription for male infertility with kidney essence deficiency,is composed of Gouqizi(Fructus Lycii),Tusizi(Semen Cuscutae),Wuweizi(Fructus Schisandrae Chinensis),Fupenzi(Fructus Rubi Chingii)and Cheqianzi(Semen Plantaginis).It has been used for hundreds of years in the treatment of male infertility,known as“the first prescription of ancient and modern seeds”,with convincing clinical evidence.At present,more than 100 chemical compounds have been isolated from WYP,including polysaccharide,fatty acids,flavonoids,phenylpropanoids,organic acids,alkaloids,terpenoids,etc.Pharmacological and clinical studies show that WYP has an obvious effect on reproductive system diseases,especially male infertility,which has a very wide application prospect.It also has effects on the nervous system,inhibiting liver injury,lowering blood sugar and blood lipid,anti-aging,improving immunity,resisting hypoxia and fatigue effects.This study reviewed the chemical constituents,quality control,pharmacology,and clinical application of WYP.There is no doubt about the clinical value of WYP,but its quality control system is not perfect,pharmacological mechanism is not fully explained,and clinical applications need to be reevaluated.Therefore,the follow-up researches should proceed from the theory of Traditional Chinese Medicine(TCM)and clinical applications,further explain the theoretical connotation,reveal the mechanism of action,and provide the basis for the secondary development of classic famous prescriptions.In addition,WYP is mostly used in combination with western medicines besides being used alone.Whether it can improve the efficacy and reduce side effects will also be a meaningful research direction in the future. | CHEN Zhimin AO Mingyue LIAO Yujiao YU Lingying YANG Zhuo HU Lin LI Wenbing HU Changjiang GAO Yongxiang | 2023 | Journal of Traditional Chinese Medicine2023,43,2: | 3 |
| 18 | The clinical relevance of MBL2 gene polymorphism and sepsis显示文摘Objective:To detect the clinical relevance of mannose-binding lectin 2(MBL2) gene polymorphism and sepsis in Chinese lived in Hainan island.Methods: Blood samples from 57 patients with sepsis and 69 patients without sepsis were collected in the ICU of several large hospitals in Hainan province. Genomic DNA was extracted from whole blood and then PCR purification product was sequenced and typed by 3730 sequencing analyzer. The concentration of MBIL2 in serum was detected by ELISA.Results: We found that genotype and allele distributions in two groups were in accordance with the Hardy-Weinberg Equilibrium. The frequency of GA genotype was significantly higher than that in non-sepsis group(P=0.013). A allele frequency in sepsis group was also much higher than that in non-sepsis group(P=0.028).Logister regression analysis showed that the patients who carried A allele were more prone to get sepsis than G allele carrier(P=0.014, OR=2,550, 95%CI=1.207-5.386). The MBL2 level in serum of sepsis patients with genotype GG and GA was significantly lower than that in non-sepsis group(P<0.05). | 20 | Comparison of different microsurgery methods for trigeminal neuralgia显示文摘Objective: To study the influence of different microsurgical methods on surgical outcomes and complications, and to improve the surgical outcomes for trigeminal neuralgia. Methods: The clinical data of 109 patients with trigeminal neuralgia, who were treated with microsurgery, were analyzed retrospectively. All patients were divided into 3 groups according to surgical modality: the trigeminal neuralgia decompression group(TND group, 19 patients), the TND and rhizotomy group(rhizotomy group,55 patients), and the TND and selective lesioning group(lesioning group, 35 patients).The mid-term and short-term effects of microsurgery, and the occurrences of complications, were compared between the 3 groups. Results: There were no statistical differences in the frequency of complications between the 3 groups(P > 0.05). Eighty-four patients were followed up for 6 to 33 months. The rate of pain disappearance was found to be 94.4% in the TND group, and 100% in both the rhizotomy and lesioning groups; thus, no significant differences were found between these 3 groups(P > 0.05). Additionally, 50% of the patients in the rhizotomy group and 3.6% of the patients in the lesioning group had facial numbness while no patients were affected with facial numbness in the TND group, and the differences between these 3 groups were significant(P < 0.05). Conclusions: Microsurgery is effective and safe for trigeminal neuralgia.
