2幽门螺杆菌OmpP1蛋白的生物信息学分析显示文摘目的应用生物学软件预测幽门螺杆菌(Helicobacter pylori,Hp)ompP1基因编码蛋白(OmpP1)的结构与功能。方法从NCBI数据库中获取ompP1基因的相关信息,应用软件MEGAX 10.0.5构建Hp进化树,应用生物信息学软件分析OmpP1蛋白的氨基酸序列、理化性质、跨膜区、信号肽、糖基化和磷酸化位点、空间结构以及抗原表位。结果 ompP1基因核苷酸序列编码区长1 764 bp,编码587个氨基酸;生物信息学分析显示ompP1基因在不同菌株间的同源性为96.2%~97.17%,其编码的OmpP1蛋白是一种性质稳定、偏碱性的亲水蛋白;该蛋白无跨膜区和信号肽,含有糖基化和磷酸化位点;OmpP1二级结构中含α-螺旋178个,延长链162个,β转角37个,无规则卷曲210个,分别占30.32%、27.60%、6.30%和35.78%;三级结构分析显示OmpP1含有OM_channels蛋白家族特征性结构域;BepiPred1.0和SYFPEITHI9预测该蛋白含有22个B淋巴细胞和28个CTL淋巴细胞相关抗原表位。结论 Hp OmpP1为碱性亲水性蛋白,含有T、B细胞抗原表位,可为其抗原性研究和高效表位疫苗的研发提供理论参考。王海燕 何梦雅 余飞艳 张柯 Wajid Ameen Mirza 陈帅印 张荣光 段广才 2020中国病原生物学杂志2020,15,1:6
3Early therapeutic effect of platelet-rich fibrin combined with allogeneic bone marrow-derived stem cells on rats’ critical-sized mandibular defects显示文摘BACKGROUND Critically sized bone defects represent a significant challenge to orthopaedic surgeons worldwide.These defects generally result from severe trauma or resection of a whole large tumour.Autologous bone grafts are the current gold standard for the reconstruction of such defects.However,due to increased patient morbidity and the need for a second operative site,other lines of treatment should be introduced.To find alternative unconventional therapies to manage such defects,bone tissue engineering using a combination of suitable bioactive factors,cells,and biocompatible scaffolds offers a promising new approach for bone regeneration.AIM To evaluate the healing capacity of platelet-rich fibrin(PRF)membranes seeded with allogeneic mesenchymal bone marrow-derived stem cells(BMSCs)on critically sized mandibular defects in a rat model.METHODS Sixty-three Sprague Dawley rats were subjected to bilateral bone defects of critical size in the mandibles created by a 5-mm diameter trephine bur.Rats were allocated to three equal groups of 21 rats each.Group I bone defects were irrigated with normal saline and designed as negative controls.Defects of group II were grafted with PRF membranes and served as positive controls,while defects of group III were grafted with PRF membranes seeded with allogeneic BMSCs.Seven rats from each group were killed at 1,2 and 4 wk.The mandibles were dissected and prepared for routine haematoxylin and eosin(HE)staining,Masson's trichrome staining and CD68 immunohistochemical staining.RESULTS Four weeks postoperatively,the percentage area of newly formed bone was significantly higher in group III(0.88±0.02)than in groups I(0.02±0.00)and II(0.60±0.02).The amount of granulation tissue formation was lower in group III(0.12±0.02)than in groups I(0.20±0.02)and II(0.40±0.02).The number of inflammatory cells was lower in group III(0.29±0.03)than in groups I(4.82±0.08)and II(3.09±0.07).CONCLUSION Bone regenerative quality of critically sized mandibular bone defects in rats was better promoted by PRF membranes seeded with BMSCs than with PRF membranes alone.Muhammad A Awadeen Fouad A Al-Belasy Laila E Ameen Mohamad E Helal Mohammed E Grawish 2020World Journal of Stem Cells2020,12,1:4
4Finite element analysis of dynamic stability of skeletal structures under periodic loading显示文摘This paper addresses the dynamic stability problem of columns and frames subjected to axially applied periodic loads. Such a structure can become unstable under certain combinations of amplitudes and frequencies of the imposed load acting on its columns/beams. These are usually shown in the form of plots which describe regions of instability. The finite element method (FEM) is used in this work to analyse dynamic stability problems of columns. Two-noded beam elements are used for this purpose. The periodic loading is decomposed into various harmonics using Fourier series expansion. Computer codes in C++ using object oriented concepts are developed to determine the stability regions of columns subjected to periodic loading. A number of nu-merical examples are presented to illustrate the working of the program. 5Occurrence and distribution of metals and porphyrins in Nigerian coal minerals显示文摘The metal contents of Nigerian coal minerals were analyzed using an atomic absorption spectrophotometer. Calcium, Na, and Fe occurred as the major elements with concentrations ranging from 9 782 μg/g for Ca to 432 μg/g for Na whereas K, Mg, Mn, Ni, Cr, Zn, Pb, and Cu, which occurred at trace levels ranged from 673.73 μg/g for Mg to 2.97 μg/g for Mn. The results of the quantitative analysis of porphyrins extracted from the coal minerals showed that Onyeama coal has the highest amount of porphyrins (ca~0.96 μg/g) while Okpara has the lowest (ca~0.30 μg/g). The porphyrins were qualitatively characterized by a combination of thin layer chromatography (TLC), infrared, and ultraviolet-visible spectrophotometers. The results of the mid infrared analysis (MIR) showed the presence of absorption bands at 3 440 cm~1~3 450 cm-1 and 1 640 cm-1~1 680 cm-1 , which are owing to the stretching vibrations of N ─ H and C─ C of aromatics, with C─ H out of plane (oop) bending vibrations at wavenumbers less than 900 cm-1, all of which are characteristic absorptions of porphyrin free base. The ultraviolet-visible data showed prominent peaks at ~400 nm(Soret band) and at wavelength ranges of 535 nm~550 nm(β-band) and 565 nm~ 600 nm (α-band) for the coal porphyrins analyzed. 7Differential expression of cell cycle regulators in HCV-infection and related hepatocellular carcinoma显示文摘AIM:To investigate cell cycle proteins in chronic hepatitis C virus infection in order to analyze their role in the process of hepatocyte transformation and to characterize their prognostic properties. METHODS:Subjects of the current study included 50 cases of chronic hepatitis C(CHC) without cirrhosis,30 cases of CHC with liver cirrhosis(LC) ,and 30 cases of hepatitis C-related hepatocellular carcinoma(HCC) admitted to the Department of Hepato-Gastroenterology,Theodor Bilharz Research Institute(TBRI) ,Giza,Egypt.Fifteen wedge liver biopsies,taken during laparoscopic cholecystectomy,were also included as normal controls.Laboratory investigations including urine and stool analysis,liver function tests and prothrombin concentration;serologic markers for viral hepatitis and ultrasonography were done for all cases of the study together with immunohistochemical analysis using primary antibodies against Cyclin D1,Cyclin E,p21,p27 and Rb/p105 proteins. RESULTS:Normal wedge liver biopsies didn't express Cyclin E or Rb/p105 immunostaining but show positive staining for Cyclin D1,p21 and p27.Cyclin D1 expressed nuclear staining that was sequentially increased from CHC to LC(P<0.01) to HCC(P<0.001) cases;meanwhile,Cyclin E revealed nuclear positivity only in the case of HCCs patients that was directly correlated to Rb/p105 immuno-reactivity.The expression of p21 and p27 was significantly increased in CHC and LC cases compared to normal controls and HCCs with no significant difference between well-and poorlydifferentiated tumors.p21 showed only a nuclear pattern of staining,while,p27 presented with either cytoplasmic and/or nuclear reactivity in all studied cases.Correlation analysis revealed a direct relation between Cyclin D1 and p21 in CHC cases(P<0.001) ,between Cyclin D1 and Cyclin E in HCCs(P<0.01);however,an inverserelationship was detected between Cyclin D1 and p21 or p27(P<0.001) and between p21 and Rb/p105(P<0.05) in HCCs. 8Predicting the presence of adenomatous polyps during colonoscopy with National Cancer Institute Colorectal Cancer Risk-Assessment Tool显示文摘AIM To evaluate the National Cancer Institute(NCI)Colorectal Cancer(CRC)Risk Assessment Tool as a predictor for the presence of adenomatous polyps(AP) found during screening or surveillance colonoscopy.METHODS This is a retrospective single center observational study.We collected data of adenomatous polyps in each colonoscopy and then evaluated the lifetime CRC risk.We calculated the AP prevalence across risk score quintiles,odds ratios of the prevalence of AP across risk score quintiles,area under curves(AUCs)and Youden’s indexes to assess the optimal risk score cut off value for AP prevalence status.RESULTS The prevalence of AP gradually increased throughout the five risk score quintiles:i.e.,27.63%in the first and 51.35%in the fifth quintile.The odd ratios of AP prevalence in the fifth quintile compared to the first and second quintile were 2.76[confidence interval(CI):1.71-4.47]and 2.09(CI:1.32-3.30).The AUC for all patients was 0.62(CI:0.58-0.66).Youden’s Index indicated the optimal risk score cutoff value discriminating AP prevalence status was 3.60.CONCLUSION Patients with the higher NCI risk score have higher risk of AP and subsequent CRC;therefore,measures to increase the effectiveness of CRC detection in these patients include longer withdrawal time,early surveillance colonoscopy,and choosing flexible colonoscopy over other CRC screening modalities.Hassan Tariq Muhammad Umar Kamal Harish Patel Ravi Patel Muhammad Ameen Shehi Elona Maram Khalifa Sara Azam Aiyi Zhang Kishore Kumar Ahmed Baiomi Danial Shaikh Jasbir Makker 2018World Journal of Gastroenterology2018,24,34:3
9Modeling,Parameter Measurement,and Control of PMSG-based Grid-connected Wind Energy Conversion System显示文摘The design of reliable controllers for wind energy conversion systems(WECSs)requires a dynamic model and accurate parameters of the wind generator.In this paper,a dynamic model and the parameter measurement and control of a direct-drive variable-speed WECS with a permanent magnet synchronous generator(PMSG)are presented.An experimental method is developed for measuring the key parameters of the PMSG.The measured parameters are used in the design of the controllers.The generator-side converter is controlled using a vector control scheme that maximizes the power extraction under varying wind speeds.A model predictive controller(MPC)is designed for the grid-side voltage source converter(VSC)to regulate the active and reactive power flows to the power grid by controlling the d-and q-axis currents in the synchronous reference frame.The MPC predicts the future values of the control variables and takes control actions based on the minimum value of the cost functions.To comply with the grid code requirement,a modified design approach for an LCL filter is presented and incorporated into the system.The design process is simple and incorporates significant filter parameters while avoiding iterative calculations.The comparative analysis of the designed filter with conventional L,LC,and iterative LCL filters demonstrates the effectiveness of the modified design approach.The proposed wind energy system with MPC and LCL filter is simulated in MATLAB/Simulink and experimentally implemented in the laboratory using the dSpace digital signal processor(DSP)system.The simulation and experimental results validate the efficacy of the designed controllers using the measured parameters and show dynamic and steady-state performance under varying wind speeds.Mohammad Kamruzzaman Khan Prince Graduate Mohammad TArif Ameen Gargoom Aman MTOo Md Enamul Haque 2021Journal of Modern Power Systems and Clean Energy2021,9,5:3

Further cyclization of 3a-c led to a fusion of a pyrimidine ring, yielding the tetracyclic products 6,7 and 8. 12为在尖锐的肝失败病人预言差的结果合并动力学显示文摘 Acute liver failure (ALF), also known as fulminant hepatic failure (FHF), is a devastating clinical syndrome with a high mortality of 60%-90%. An early and exact assessment of the severity of ALF together with prediction of its further development is critical in order to determine the further management of the patient. A number of prognostic models have been used for outcome prediction in ALF patients but they are mostly based on the variables measured at one time point, mostly at admission. ALF patients rarely show a static state: rapid progress to a life threatening situation occurs in many patients. Since ALF is a dynamic process, admission values of prognostic variables change over time during the clinical course of the patient. Kumar et al developed a prognostic model [ALF early dynamic (ALFED)] based on early changes in values of variables which predicted outcome. ALFED is a model which seems to be worthwhile to test in ALF patients in other parts of the world with different aetiologies. Since the exact pathophysiology of ALF is not fully known and is certainly complex, we believe that adding promising variables involved in the pathophysiology of ALF to the dynamic approach might even further improve prognostic performance. We agree with Kumar et al that an improved dynamic prognostic model should be based on simplicity (easily to be performed at the bedside) and accuracy.
