1Non-invasive assessment of liver fibrosis in chronic hepatitis B显示文摘The goal of this review is to provide a comprehensive picture of the role,clinical applications and future perspectives of the most widely used non-invasive techniques for the evaluation of hepatitis B virus(HBV)infection.During the past decade many non-invasive methods have been developed to reduce the need for liver biopsy in staging fibrosis and to overcome whenever possible its limitations,mainly:invasiveness,costs,low reproducibility,poor acceptance by patients.Elastographic techniques conceived to assess liver stiffness,in particular transient elastography,and the most commonly used biological markers will be assessed against their respective role and limitations in staging hepatic fibrosis.Recent evidence highlights that both liver stiffness and some bio-chemical markers correlatewith survival and major clinical end-points such as liver decompensation,development of hepatocellular carcinoma and portal hypertension.Thus the non-invasive techniques here discussed can play a major role in the management of patients with chronic HBV-related hepatitis.Given their prognostic value,transient elastography and some bio-chemical markers can be used to better categorize patients with advanced fibrosis and cirrhosis and assign them to different classes of risk for clinically relevant outcomes.Very recent data indicates that the combined measurements of liver and spleen stiffness enable the reliable prediction of portal hypertension and esophageal varices development.Federica Branchi Clara Benedetta Conti Alessandra Baccarin Pietro Lampertico Dario Conte Mirella Fraquelli 2014World Journal of Gastroenterology2014,20,40:20
2Low temperature,IBA concentrations and optimal time for adventitious rooting of Eucalyptus benthamii mini-cuttings显示文摘Eucalyptus benthamii is a forest species of economic interest that has difficulty with seed production and also is considered to have difficulty with adventitious rooting using propagation techniques,such as cutting or mini-cutting.We aimed to assess the adventitious rooting percentage under different storage times in low temperatures and at various IBA(indole-3-butyric acid) concentrations to determine the optimal time of permanence for rooting Eucalyptus benthamii minicuttings in a greenhouse.Shoots collected from mini-stumps cultivated in a semi-hydroponic system were used to obtain the mini-cuttings.For the first experiment,the mini-cuttings were stored at 4℃ for 0(immediate planting),24,48,72,96 and 120 h.The second experiment evaluated the rooting dynamic to determine the optimal time of permanence for minicuttings in a greenhouse.The basal region of the mini-cutting was treated with various IBA solutions:0(free of IBA),1,000,2,000,3,000 and 4,000 mg·L-1.Every seven days(0(immediate planting),7,14,21 and 28 days),destructive sampling of the mini-cuttings was performed to evaluate the histology of the adventitious rooting.Eucalyptus benthamii minicuttings should be rooted immediately after the collection of the shoots.The 2,000 mg·L-1 IBA concentration induced a greater speed and percentage of adventitious rooting,and an interval of 35 to 42 days was indicated for permanence of the mini-cuttings in the greenhouse.Exposure to low temperature induced adventitious root formation with diffuse vascular connections.Gilvano Ebling Brondani Francisco JoséBenedini Baccarin Heron Wilhelmus de Wit Ondas JoséLuiz Stape Antonio Natal Gonçalves Marcilio de Almeida 2012Journal of Forestry Research2012,23,4:5

10Different culture conditions applied to in vitro shoot multiplication of two Eucalyptus benthamii explant sources显示文摘Eucalyptus adult material requires more successive subcultures in the in vitro multiplication phase for increased vigor and cellular activity. This study evaluated the endophytic manifestation and shoot multiplication of one 13-year-old Eucalyptus benthamii clone under different culture conditions and used canopy branches(CB) and trunk base material as explant sources. The culture media were wood plant medium(WPM), Murashige and Skoog medium(MS) and JADS(Correia and co-authors medium).Based on the results of the initial multiplication experiment, further tests examined sucrose concentrations and p H. Morphophysiology, dry mass production, endophyticmanifestation and histochemical were determined. Explant sources responded differently to MS and JADS media, but the WPM medium promoted homogeneous development.The responses were similar for both explant sources when sucrose concentrations varied. Shoots died in the absence of sucrose, showed high oxidation at 60 g L-1 and optimal development at 30 g L-1. Endophytes were more evident for shoots from the CB origin. Explant sources responded distinctively to treatment due to physiological and intrinsic genetic factors. 11Chondrogenic potential of mesenchymal stem cells from horses using a magnetic 3D cell culture system显示文摘BACKGROUND Mesenchymal stem cells(MSCs)represent a promising therapy for the treatment of equine joint diseases,studied due to their possible immunomodulatory characteristics and regenerative capacity.However,the source of most suitable MSCs for producing cartilage for regenerative processes in conjunction with biomaterials for an enhanced function is yet to be established.AIM To compare the chondrogenicity of MSCs derived from synovial fluid,bone marrow,and adipose tissue of horses,using the aggrecan synthesis.METHODS MSCs from ten horses were cultured,phenotypic characterization was done with antibodies CD90,CD44 and CD34 and were differentiated into chondrocytes.The 3D cell culture system in which biocompatible nanoparticles consisting of gold,iron oxide,and poly-L-lysine were added to the cells,and they were forced by magnets to form one microspheroid.The microspheroids were exposed to a commercial culture medium for 4 d,7 d,14 d,and 21 d.Proteoglycan extraction was performed,and aggrecan was quantified by enzyme-linked immunosorbent assay.Keratan sulfate and aggrecan in the microspheroids were identified and localized by immunofluorescence.RESULTS All cultured cells showed fibroblast-like appearance,the ability to adhere to the plastic surface,and were positive for CD44 and CD90,thus confirming the characteristics and morphology of MSCs.The soluble protein concentrations were higher in the microspheroids derived from adipose tissue.The aggrecan concentration and the ratio of aggrecan to soluble proteins were higher in microspheroids derived from synovial fluid than in those derived from bone marrow,thereby showing chondrogenic superiority.Microspheroids from all sources expressed aggrecan and keratan sulfate when observed using confocal immunofluorescence microscopy.All sources of MSCs can synthesize aggrecan,however,MSCs from synovial fluid and adipose tissue have demonstrated better biocompatibility in a 3D environment,thus suggesting chondrogenic superiority.CONCLUSION All sources of MSCs produce hyaline cartilage;however,the use of synovial liquid or adipose tissue should be recommended when it is intended for use with biomaterials or scaffolds.Joice Fülber Fernanda R Agreste Sarah R T Seidel Eric D P Sotelo Ângela P Barbosa Yara M Michelacci Raquel Y A Baccarin 2021World Journal of Stem Cells2021,13,6:0
