| 2 | EXPRESSION OF HUMAN BETA-DEFENSIN 3 IN COS-7 CELL显示文摘To establish a cell line for stable expression of human beta-defensin 3 (hBD3). Methods Full length cDNA of hBD3 was isolated from previously constructed pGEM-hBD3 and then inserted into pcDNA3. The recombinant vector identified carrying hBD3 with right direction was introduced into COS-7 cells by Lipofe-ctamine. Cell clones survived in G418-rich medium and with stable expression of hBD3 in both mRNA and protein levels were identified by RT-PCR and Western blot respectively. Genomic integration of the hBD3 gene with the COS-7 cells was confirmed by Southern dot blot and primary analysis. The antimicrobial activity of the secreted hBD3 was also evaluated. Results COS-7 cells transfected with pcDNA3-hBD3 expressed hBD3 stably in mRNA and protein level. Southern dot blot analysis showed successful integration of the hBD3 gene into the genome of COS-7 cell and the hBD-3 protein secreted into the culture medium showed antimicrobial activity. | 4 | KNEMAG:Key Node Estimation Mechanism Based on Attack Graph for IoT Security显示文摘With the rapid development and widespread application of the IoT,the at-tacks against IoT vulnerabilities have become more complex and diverse.Most of the previous research focused on node vulnerability and its risk analysis.There is little information available about the importance of the location of the node in the system.Therefore,an estimation mechanism is proposed to assess the key node of the IoT system.The estimation of the key node includes two parts:one is the utilization relationship between nodes,and the other is the impact on the system after the node is conquered.We use the node importance value and the node risk value to quantify these two parts.First,the node importance value is calculated by considering the attack path that pass through the node and the probability that the attacker will abandon the attack.Second,in addition to node vulnerabilities and the consequences of being attacked,two quantitative indicators are proposed to comprehensively assess the impact of nodes on the system security,and the node risk value is calculated based on the grey correlation analysis method.Third,the key node in the IoT system could be obtained by integrating the node importance value and risk value.Finally,the simulation experiment result shows that the presented method could find the key node of the system quickly and accurately. | Bichen Che Long Liu Huali Zhang | 2020 | Journal on Internet of Things2020,2,4: | 1 |
| 5 | Module Guard:A Gatekeeper for Dynamic Module Loading Against Malware显示文摘We analyze the attack steps of malware and focus on the malware loading. Our assumption is that a malware contains no less than one module, so monitoring module loading is indispensable to defeat malware. Moreover, we design security policies and employ these policies when a module is loaded by the operating system. These policies depend on properties of module, the connection to created modules, and the link to user intention. The properties of module and this connection can improve the accuracy of malware detection. User intention can be helpful to handle unknown module and enhances the flexibility of policy. Finally, ModuleGuard, a gatekeeper for dynamic module loading against malware, has been designed and implemented, which integrates these security policies. | 6 | Colony-stimulating factor 3 receptor (CSF3R) M696T mutation does not impact on clinical outcomes of a Ph+ acute lymphoblastic leukemia patient显示文摘Colony-stimulating factor 3 receptor(CSF3R)mutations have been identified in a variety of myeloid disorders.Although CSF3R point mutations(eg,T618I)are emerging as key players in chronic neutrophilic leukemia/atypical chronic myelogenous leukemia,the significance of rarer CSF3R mutations is unknown.Here,we report a 32-year-old female who was diagnosed as Philadelphia chromosome-positive acute lymphoblastic leukemia(Ph^(+)ALL)with the CSF3R M696T mutation and was undergone unrelated donor hematopoietic stem cell transplantation.The patient achieved complete remission with chemotherapy in combination with tyrosine kinase inhibitor(TKI)and long-term survival by unrelated donor transplantation.Meanwhile,we performed a series of experiments using murine interleukin 3(IL-3)-dependent Ba/F3 cell line to evaluate the transforming capacity of the CSF3R M696T mutation.We confirmed the presence of a CSF3R M696T germline mutation in this patient which was inherited from her mother.The in vitro experiment results showed that the CSF3R M696T mutation contributes marginally to the tumor transformation of Ba/F3 cells,indicating that CSF3R M696T mutation was neutral in tumor transformation ability.We concluded that TKI is effective in patients with the CSF3R M696T mutation in Ph+ALL and donors with CSF3R M696T mutation might still be selected as the candidate for transplantation. | Xin Chen Bichen Wang Aiming Pang Weiping Yuan Erlie Jiang Yajing Chu Sizhou Feng Mingzhe Han | 2021 | Blood Science2021,3,3: | 0 |

| 8 | L-type calcium channel blockers enhance 5-HTP-induced antinociception in mice显示文摘AIM: To investigate the involvement of L-type Ca2+ channels in antinociceptive action induced by the 5-HT precursor,5-hydroxytryptophan (5-HTP). METHODS: Female Kunming mice were treated with either 5-HTP (20-80 mg/kg,ip) alone, or the combination of 5-HTP and fluoxetine (2-8 mg/kg, ip), pargyline (15-60 mg/kg, ip), nimodipine (2.5-10 mg/kg, ip), nifedipine (2.5-10 mg/kg, ip), verapamil (2.5-10 mg/kg, ip), CaCl2 (5-20 mmol/L, icv), or EGTA(0.5-3 mmol/L, icv) prior to the hot-plate test (55 C, hind-paw licking latency). In addition, locomotor activity inmice treated with 5-HTP alone was measured using an ambulometer with five activity boxes. RESULTS: Ipinjection of 5-HTP alone had no influence on the spontaneous locomotor activity, whereas dose-dependently in-creased the latency to licking hind-paw in the hot-plate test in mice. The inhibitory effects of 5-HTP on nociceptiveresponse were significantly enhanced by fluoxetine in the mouse hot-plate test. At a sub-effective dose, pargylinecould cause a leftward shift in the dose-response curve of 5-HTP-induced antinociception. Co-administration with5-HTP and nimodipine, nifedipine, or verapamil obviously potentiated the antinociceptive effects elicited by 5-HTP.Interestingly, 5-HTP-induced antinociception was antagonized by CaCl2 and enhanced by EGTA injected icv in themouse hot-plate test. | 9 | Loss of Tet2 affects platelet function but not coagulation in mice显示文摘Ten-eleven translocation 2(TET2)functions as a methylcytosine dioxygenase that catalyzes the iterative oxidation of 5-methylcytosine to 5-hydroxymethylcytosine,5-formylcytosine and 5-carboxylcytosine.TET2 has been shown to be crucial for the maintenance and differentiation of hematopoietic stem cells,and its deletion and/or mutations results in the expansion of HSPCs,and leads to hematological malignancies.TET2 mutations were found in a variety of hematological disorders such as CMML(60%),MDS(30%),MPN(13%)and AML(20%).Interestingly,it was shown that CMML patients with TET2 mutation exhibited fewer platelets than CMML patients without TET2 mutation.However,the role and function of TET2 in platelet hemostasis and thrombogenesis is not well defined.Here in this study,using a genetically engineered Tet2 deletion mouse model,we found that the absence of Tet2 caused a decrease in the proportion of MEP cells and hyperploid megakaryocytes.Additionally,Tet2-deficient mice displayed impaired platelet activation and aggregation under stimulation of ADP and low concentrations of thrombin,although the modestly compromised platelet function and MEP differentiation in Tet2-deficient mice could be compensated without affecting blood coagulation function.Our study indicate that Tet2 deficiency leads to mild impairment of platelet function and thrombopoiesis in mice. | Bichen Wang Meijuan Xia Ting Chen Mengke Li Deyang Shi Xiaomin Wang Aiming Pang Jiaxi Zhou Weiping Yuan Yajing Chu | 2020 | Blood Science2020,2,4: | 0 |
