Here, we report the generation of a cynomolgus monkey(Macaca fascicularis) carrying MECP2 mutations using transcription activator-like effector nucleases(TALENs)-mediated gene targeting. After injecting TALENs mRNA into monkey zygotes achieved by in vitro fertilization and embryo transplantation into surrogate monkeys, we obtained one male newborn monkey with an MECP2 deletion caused by frameshifting mutation in various tissues. The monkey carrying the MECP2 mutation failed to survive after birth, due to either the toxicity of TALENs or the critical requirement of MECP2 for neural development. The level of MeCP2 protein was essentially depleted in the monkey's brain. | 5 | Intraportal mesenchymal stem cell transplantation prevents acute liver failure through promoting cell proliferation and inhibiting apoptosis显示文摘BACKGROUND: Transplantation of mesenchymal stem cells(MSCs) has been regarded as a potential treatment for acute liver failure(ALF), but the optimal route was unknown. The present study aimed to explore the most effective MSCs transplantation route in a swine ALF model.METHODS: The swine ALF model induced by intravenous injection of D-Gal was treated by the transplantation of swine MSCs through four routes including intraportal injection(In P group), hepatic intra-arterial injection(AH group), peripheral intravenous injection(PV group) and intrahepatic injection(IH group). The living conditions and survival time were recorded. Blood samples before and after MSCs transplantation were collected for the analysis of hepatic function. The histology of liver injury was interpreted and scored in terminal samples. Hepatic apoptosis was detected by TUNEL assay. Apoptosis and proliferation related protein expressions including cleaved caspase-3, survivin, AKT, phospho-AKT(Ser473), ERK and phospho-ERK(Tyr204) were analyzed by Western blotting.RESULTS: The average survival time of each group was 10.7 ±1.6 days(In P), 6.0±0.9 days(AH), 4.7±1.4 days(PV), 4.3±0.8 days(IH), respectively, when compared with the average survival time of 3.8±0.8 days in the D-Gal group. The survival rates between the In P group and D-Gal group revealed a statistically significant difference(P<0.01). Pathological and biochemical analysis showed that liver damage was the worst in the D-Gal group, while less injury in the In P group. Histopathological scores revealed a significant decrease in the In P group(3.17±1.04, P<0.01) and AH group(8.17±0.76, P<0.05) as compared with that in the D-Gal group(11.50±1.32). The apoptosis rate in the In P group(25.0%±3.4%, P<0.01) and AH group(40.5%±1.0%, P<0.05) was lower than that in the D-Gal group(70.6%±8.5%). The expression of active caspase-3 was inhibited, while the expression of survivin, AKT, phosphoAKT(Ser473), ERK and phospho-ERK(Tyr204) was elevated in the In P group.CONCLUSIONS: Intraportal injection was superior to other pathways for MSC transplantation. Intraportal MSC transplantation could improve liver function, inhibit apoptosis and prolong the survival time of swine with ALF. | 7 | Timing of upper gastrointestinal endoscopy does not influence short-term outcomes in patients with acute variceal bleeding显示文摘AIM To examine the association between the timing of endoscopy and the short-term outcomes of acute variceal bleeding in cirrhotic patients.METHODS This retrospective study included 274 consecutive patients admitted with acute esophageal variceal bleeding of two tertiary hospitals in Korea. We adjusted confounding factors using the Cox proportional hazards model and the inverse probability weighting(IPW) method. The primary outcome was the mortality of patients within 6 wk.RESULTS A total of 173 patients received urgent endoscopy(i.e., ≤ 12 h after admission), and 101 patients received nonurgent endoscopy(> 12 h after admission). The 6-wk mortality rate was 22.5% in the urgent endoscopy group and 29.7% in the non-urgent endoscopy group, and there was no significant difference between the two groups before(P = 0.266) and after IPW(P = 0.639). The length of hospital stay was statistically different between the urgent group and non-urgent group(P = 0.033); however, there was no significant difference in the inhospital mortality rate between the two groups(8.1% vs 7.9%, P = 0.960). As the air flow approaches,local strong wind may appear around the buildings. The strong wind makes the pedestrians on sidewalks, entrances and terrace very uncomfortable and causes the pedestrian level wind environment problem. In this studies, hot-wire wind measurement, wind scouring in wind tunnel and numerical computation were carried out to evaluate the wind environment of tall buildings in the prevailing flow conditions in Beijing areas. The results obtained by three techniques were compared and mutually verified. The conclusions drawn from three approaches agree with each other. Also the advantages and limitations of each method were analyzed. | 11 | Population genetics and evolutionary history of Miscanthus species in China显示文摘Miscanthus species have received considerable attention as a potential biomass source for renewable energy production because of their ability to produce high yields of biomass and adapt to a wide range of climates and soils. To explore the genetic diversity and phylogenetic relationship of Miscanthus species in China, we used 24 simple sequence repeat markers to genotype 100 natural populations representing all four Chinese Miscanthus species (M. sinensis Andersson, M. floridulus (Lab.) Warb. ex K. Schum.& Lauterb., M. sacchariflorus (Maxim.) Hack., and M. lutarioriparius L. Liu ex Renvoize & S. L. Chen). Based on phylogenetic, principal coordinate, and STRUCTURE analyses, we found that the 100 populations formed two major groups corresponding to sect. Triarrhena and sect. Miscanthus. Group 1 (i.e., sect. Triarrhena) was further subdivided into two subgroups corresponding to M. sacchariflorus and M. lutarioriparius;Group 2 (i.e., sect. Miscanthus) was subdivided into three subgroups, group 2a (M. sinensis populations in southern China), group 2b (M. sinensis populations in northern China), and group 2c (all the M. floridulus populations). Population genetics analyses indicated high levels of the genetic diversity at both population (HE = 0.468–0.599) and species (HE = 0.559–0.708) levels, indicative of the potential of these wild resources in future breeding programs. The species distribution modeling showed that M. sacchariflorus and M. lutarioriparius have experienced population reductions during the last glacial maximum and population expansion afterward;in contrast, M. sinensis and M. floridulus both underwent gradual population expansions from the last interglaciation to the present. We also suggest that M. floridulus originated from M. sinensis in southeast China through ecological speciation. The purpose of this study was to investigate monthly litterfall production in three forests in Jeju Island differentiated by forest composition and precipitation: Cheongsu(Quercus glauca as the dominant species;low precipitation), Seonheulb(Q. glauca as the dominant species;high precipitation), and Seonheulm(Q. glauca and Pinus thunbergii as the dominant species;high precipitation). Litterfall was collected monthly from April to December 2015 and divided into leaf litter, twig, bark, seeds, and unidentified materials.Seasonal patterns of litterfall production varied across stands according to their species composition. However,the amount of leaf litterfall and total litterfall were comparable among stands, ranging from 362 to 375 g m-2 for leaf litter and 524 g m-2 to 580 g m-2 for total litterfall.Oak leaf litter in May was the highest in all stands, while needle litter was the highest in December in Seonheulm.High twig litterfall in July may be attributable to high rainfall with strong winds and storms during the rainy season. | 14 | Application of the health risk assessment of acetochlor in the development of water quality criteria显示文摘Acetochlor is a widely used herbicide in agricultural production. Studies have shown that acetochlor has obvious environmental hormone effects, and long-term exposure may pose a threat to human health. To quantify the hazards of acetochlor in drinking water, a health risk assessment of acetochlor was conducted in major cities of China based on the data of acetochlor residue concentrations in drinking water. The approach of the Species Sensitivity Distributions(SSD) method is used to extrapolate from animal testing data to reflect worst case human toxicity. Results show that hazard quotients related to acetochlor residues in drinking water for different age groups range from 1.94 × 10-4 to 6.13 × 10-4, so, there are no indication of human risk. Compared to the total estimated hazard quotient from oral intake of acetochlor, the chronic exposure imputed to acetochlor residues in drinking water in China accounts for 0.4%.
