1Simple and sensitive determination of sparfloxacin in pharmaceuticals and biological samples by immunoassay显示文摘Plasma quinolone concentrations are not routinely measured in clinical practice.However,in order to optimize quinolone treatment,monitoring of plasma concentrations could sometimes be useful particularly in critically ill patients.In this study,anti-sparfloxacin antibody was obtained by immunizing rabbits with sparfloxacin conjugated with bovine serum albumin using isobutyl chloroformate method.After the assay procedure was optimized,the standard curve of sparfloxacin was established.The practical measuring range of the competitive ELISA extended from 5 ng/mL to 2 mg/mL.The recovery rates and coefficients of variation for rat plasma,urine and tissues were 87.7-106.2% and 4.8-15.3%,respectively.To demonstrate the potential of the ELISA,a preliminary pharmacokinetics and tissue distribution study of sparfloxacin in rats and quantitative analysis of sparfloxacin in several pharmaceuticals were performed and compared with high-performance liquid chromatography(HPLC).The experimental data indicated that the proposed method would be a valuable tool in therapeutic drug monitoring(TDM) for sparfloxacin.Hua-Jin Zenga,Ran Yangb,Bing Liub,Li-Fang Leib,Jian-Jun Lib,Ling-Bo Qub,c,n aSchool of Pharmaceutical Sciences,Zhengzhou University,Zhengzhou 450001,China bDepartment of Chemistry,Zhengzhou University,Zhengzhou 450001,China cSchool of Chemistry & Chemical Engineering,Henan University of Technology,Zhengzhou 450001,China 2012Journal of Pharmaceutical Analysis2012,2,3:2

3Influence of prolonged optic flow stimuli on spontaneous activities of cat PMLS neurons显示文摘Changes in neuronal spontaneous activities after prolonged optic flow stimulation (using the three basic flow modes: translation, radiation and rotation) were inves-tigated by extracellular single-unit recording in cortical area PMLS of the cat. The results showed that the evoked re-sponses decreased with the prolongation of visual stimuli, and the spontaneous activities usually dropped to a lower level after the stimuli were withdrawn. Generally, the reduc-tion in spontaneous activities was larger after adaptation in the preferred direction than in the non-preferred direction. This difference was much pronounced to translation stimuli, but relatively insignificant to radiation and rotation. These points suggest that non-specific fatigue may act as the key factor in adaptation to simple translation, while some kinds of more complicated, direction-specific mechanism may be involved in adaptation to the complex optic flow patterns.
