1Clinical translation of bioartificial liver support systems with human pluripotent stem cell-derived hepatic cells显示文摘There is currently a pressing need for alternative the-rapies to liver transplantation. The number of patients waiting for a liver transplant is substantially higher than the number of transplantable donor livers, resulting in a long waiting time and a high waiting list mortality. An extracorporeal liver support system is one possible approach to overcome this problem. However, the ideal cell source for developing bioartificial liver(BAL) support systems has yet to be determined. Recent advancements in stem cell technology allow researchers to generate highly functional hepatocyte-like cells from human pluripotent stem cells(h PSCs). 3JAK2转基因骨髓造血干/祖细胞体外长期扩增调控及定向分化的研究显示文摘目的 探讨酪氨酸激酶JAK2转基因骨髓造血干 祖细胞体外长期扩增调控和定向分化潜能的可行性。方法 克隆了一个MSCV based逆转录病毒载体称作MGI F2Jak2 ,它编码一个绿色荧光蛋白 (GFP)和两个改进的FK5 0 6结合蛋白 (F36v)与JAK2组成的融合蛋白 ,F36v是二聚化化学诱导物AP2 0 187的结合位点。应用该载体转染GpE +86包装细胞株 ,将C5 7BL 6小鼠的骨髓细胞与照射过15 0 0cGy的GpE +86产毒细胞株共同培养实现基因转导。转导后的细胞在X VIVO 15培养体系中扩增 ,分为①空白对照组 ;②AP2 0 187组 ;③干细胞因子 (SCF)组 ;④AP2 0 187+SCF组。扩增的细胞用直接法标记藻红蛋白荧光素 (PE)结合的抗Sca1、c kit、CD34 、Gr1、CD1 1b、TER 119、CD4 1 、B2 2 0和CD3单克隆抗体以流式细胞仪检测 ,并进行定向分化、祖细胞集落培养和脾细胞集落形成单位 (CFU S)的研究。结果 只有AP2 0 187+SCF组可以持续地使转基因的骨髓造血干 祖细胞大量增殖 ,扩增 80d后细胞可达10 1 4倍 ,细胞倍增时间约 30h ,扩增细胞的表型为CD34 、c kit和Sca1等干细胞表面标志强阳性 ,其它细胞表面标志如Gr1、CD1 1b、TER119、CD4 1 、B2 2 0、CD3接近阴性。所扩增的细胞在不同的细胞因子组合下 ,可定向分化为粒细胞、巨噬细胞、红细胞、赵声明 顾惜春 常乃柏 Tim Clackson C Anthony Blau 2004中华血液学杂志2004,25,2:8

5The role of forensic anthropology in disaster victim identification(DVI):recent developments and future prospects显示文摘Forensic anthropological knowledge has been used in disaster victim identification(DVI)for over a century,but over the past decades,there have been a number of disaster events which have seen an increasing role for the forensic anthropologist.The experiences gained from some of the latest DVI operations have provided valuable lessons that have had an effect on the role and perceived value of the forensic anthropologist as part of the team managing the DVI process.This paper provides an overview of the ways in which forensic anthropologists may contribute to DVI with emphasis on how recent experiences and developments in forensic anthropology have augmented these contributions.Consequently,this paper reviews the value of forensic anthropological expertise at the disaster scene and in the mortuary,and discusses the way in which forensic anthropologists may use imaging in DVI efforts.Tissue-sampling strategies for DNA analysis,especially in the case of disasters with a large amount of fragmented remains,are also discussed.Additionally,consideration is given to the identification of survivors;the statistical basis of identification;the challenges related to some specific disaster scenarios;and education and training.Although forensic anthropologists can play a valuable role in different phases of a DVI operation,they never practice in isolation.The DVI process requires a multidisciplinary approach and,therefore,has a close collaboration with a range of forensic specialists.Hans H.de Boer Soren Blau Tania Delabarde Lucina Hackman 2019Forensic Sciences Research2019,4,4:3
6Disaster victim identification operations with fragmented,burnt,or commingled remains:experience-based recommendations显示文摘Human-made and natural disasters can result in severely fragmented,compromised,and commingled human remains.The related disaster victim identification(DVI)operations are invariably challenging,with the state of the remains potentially precluding some identifications.Practitioners involved in these DVI operations will routinely face logistical,practical,and ethical challenges.This review provides information and guidance derived from firsthand experiences to individuals tasked with managing DVI operations with fragmented human remains.We outline several key issues that should be addressed during disaster preparedness planning and at the outset of an operation,when incident-specific strategies are developed.Specific challenges during recovery and examination of fragmented remains are addressed,highlighting the importance of experienced specialists at the scene and in the mortuary.DNA sample selection and sampling techniques are reviewed,as well as downstream effects of commingling and contamination,which can complicate reconciliation and emphasise the need for rigorous quality control.We also touch on issues that may arise during communication with families.While recommendations are provided,they are not intended as proscriptive policy but rather as an addition to the general recommendations given in the International Criminal Police Organization(INTERPOL)DVI Guide,to inform preparative discussions between government officials,judiciary,police,and forensic specialists.Hans H.de Boer Julie Roberts Tania Delabarde Amy Z.Mundorff Soren Blau 2020Forensic Sciences Research2020,5,3:3
7Fabrication and near-infrared optical responses of 2D periodical Au/ITO nanocomposite arrays显示文摘Two-dimensional(2D) periodical Au and indium tin oxide(ITO) nanocomposite arrays have been fabricated based on a self-assembled nanosphere lithography technique. A button-shaped Au nanoparticle was formed on each hollow hemisphere-shaped ITO shell. Importantly, the underlying formation mechanism during the thermal treatment has been thoroughly explored by comparing structures resulting from different deposition conditions in detail. Compared to the Au nanoparticle arrays without ITO shells, the Au/ITO nanocomposite arrays showed a stronger localized surface plasmon resonance effect and higher absorption in the near-infrared(NIR) region, benefiting from the free-electron interaction enhancement between Au and ITO. The nonlinear optical properties were investigated using a modified femtosecond intensity-scan system, and the results demonstrated Au/ITO nanocomposite arrays with a remarkable two-photon absorption saturation effect for femtosecond pulses at 1030 nm.
