| 1 | 抗狂犬病病毒中和抗体效价快速荧光灶抑制试验(RFFIT)的建立及应用显示文摘目的建立抗狂犬病病毒中和抗体效价的快速荧光灶抑制试验(RFFIT),验证其与小鼠脑内中和法(MNT)的相关性,并用于马抗狂犬病病毒血清(ERIG)和人狂犬病免疫球蛋白(HRIG)及狂犬病疫苗免疫后血清中和抗体效价的测定。方法待测血清或免疫球蛋白在96孔板中作3倍系列稀释,与能感染80%~95%细胞量的攻击病毒混合后,37℃体外中和1h,接种BSR细胞,培养24h后荧光染色,荧光显微镜下读取结果。用建立的RFFIT法与MNT法同时标定狂犬病免疫球蛋白国家标准品,并检测ERIG和HRIG以及疫苗免疫后血清样品的抗狂犬病病毒中和抗体效价,以比较两种方法的精密度和相关性。结果采用MNT法和RFFIT法检测我国抗狂犬病免疫球蛋白国家标准品的GMT分别为21.4和23.8IU/ml,两种方法的变异系数分别为62.3%和13.3%;两个实验室用RFFIT法测定我国国家标准品的结果分别为21.71和23.81IU/ml。两种方法测定ERIG和HRIG及疫苗免疫后血清中和抗体效价的结果差异无统计学意义,相关系数为0.976,两种方法的检测结果之间呈良好的正相关性。结论RFFIT法可替代MNT法检测抗狂犬病病毒中和抗体效价。 | 吴小红 李加 唐建蓉 Hervé Bourhy 刘景华 曲效民 曹守春 董关木 | 2009 | 中国生物制品学杂志2009，22，11： | 19 |
