2Study of penetration mechanism of labrasol on rabbit cornea by Ussing chamber, RT-PCR assay, Western blot and immunohistochemistry显示文摘Labrasol, as a non-ionic surfactant, can enhance the permeation and absorption of drugs, and is extensively used in topical, transdermal, and oral pharmaceutical preparations as an emulsifier and absorption enhancer. Recent studies in our laboratory have indicated that labrasol has a strong absorption enhancing effect on different types of drugs in vitro and in vivo. This study was performed to further elucidate the action mechanism of labrasol on the corneal penetration. In this research, the fluorescein sodium, a marker of passive paracellular transport of tight junction, was selected as the model drug to assess the effect of labrasol on in vitro corneal permeability. To investigate the continuous and real-time influence of labrasol on the membrane permeability and integrity, the Ussing chamber system was applied to monitor the electrophysiological parameters. And, furthermore, we elucidated the effect of labrasol on excised cornea at the molecular level by application of RT-PCR, Western blot, and immunohistochemical staining. 15Autoinhibitory structure of preligand association state implicates a new strategy to attain effective DR5 receptor activation显示文摘Members of the tumor necrosis factor receptor superfamily(TNFRSF)are important therapeutic targets that can be activated to induce death of cancer cells or stimulate proliferation of immune cells.Although it has long been implicated that these receptors assemble preligand associated states that are required for dominant interference in human disease,such states have so far eluded structural characterization.Here,we find that the ectodomain of death receptor 5(DR5-ECD),a representative member of TNFRSF,can specifically self-associate when anchored to lipid bilayer,and we report this self-association structure determined by nuclear magnetic resonance(NMR).Unexpectedly,two non-overlapping interaction interfaces are identified that could propagate to higher-order clusters.Structure-guided mutagenesis indicates that the observed preligand association structure is represented on DR5-expressing cells.The DR5 preligand association serves an autoinhibitory role as single-domain antibodies(sdAbs)that partially dissociate the preligand cluster can sensitize the receptor to its ligand TRAIL and even induce substantial receptor signaling in the absence of TRAIL.Unlike most agonistic antibodies that require multivalent binding to aggregate receptors for activation,these agonistic sdAbs are monovalent and act specifically on an oligomeric,autoinhibitory configuration of the receptor.Our data indicate that receptors such as DR5 can form structurally defined preclusters incompatible with signaling and that true agonists should disrupt the preligand cluster while converting it to signaling-productive cluster.This mechanism enhances our understanding of a long-standing question in TNFRSF signaling and suggests a new opportunity for developing agonistic molecules by targeting receptor preligand clustering.Gang Du Linlin Zhao Yumei Zheng Anissa Belfetmi Tiantian Cai Boying Xu Karen Heyninck Kim Van Den Heede Marie-Ange Buyse Pietro Fontana Michael Bowman Lih-Ling Lin Hao Wu James Jeiwen Chou 2023Cell Research2023,33,2:0
16A Local Discontinuous Galerkin Method with Generalized Alternating Fluxes for 2D Nonlinear Schrödinger Equations显示文摘In this paper,we consider the local discontinuous Galerkin method with generalized alter-nating numerical fluxes for two-dimensional nonlinear Schrödinger equations on Carte-sian meshes.The generalized fluxes not only lead to a smaller magnitude of the errors,but can guarantee an energy conservative property that is useful for long time simulations in resolving waves.By virtue of generalized skew-symmetry property of the discontinuous Galerkin spatial operators,two energy equations are established and stability results con-taining energy conservation of the prime variable as well as auxiliary variables are shown.To derive optimal error estimates for nonlinear Schrödinger equations,an additional energy equation is constructed and two a priori error assumptions are used.This,together with properties of some generalized Gauss-Radau projections and a suitable numerical initial condition,implies optimal order of k+1.Numerical experiments are given to demonstrate the theoretical results.Hongjuan Zhang Boying Wu Xiong Meng 2022Communications on Applied Mathematics and Computation2022,4,1:0

18Analysis of the local discontinuous Galerkin method with generalized fluxes for one-dimensional nonlinear convection-diffusion systems显示文摘In this paper,we present optimal error estimates of the local discontinuous Galerkin method with generalized numerical fluxes for one-dimensional nonlinear convection-diffusion systems.The upwind-biased flux with the adjustable numerical viscosity for the convective term is chosen based on the local characteristic decomposition,which is helpful in resolving discontinuities of degenerate parabolic equations without enforcing any limiting procedure.For the diffusive term,a pair of generalized alternating fluxes is considered.By constructing and analyzing generalized Gauss-Radau projections with respect to different convective or diffusive terms,we derive optimal error estimates for nonlinear convection-diffusion systems with the symmetrizable flux Jacobian and fully nonlinear diffusive problems.Numerical experiments including long time simulations,different boundary conditions and degenerate equations with discontinuous initial data are provided to demonstrate the sharpness of theoretical results.Hongjuan Zhang Boying Wu Xiong Meng 2023Science China Mathematics2023,66,11:0
