3Cholesterol homeostasis regulated by ABCA1 is critical for retinal ganglion cell survival显示文摘Genome-wide association studies have suggested a link between primary open-angle glaucoma and the function of ABCA1.ABCA1 is a key regulator of cholesterol efflux and the biogenesis of high-density lipoprotein(HDL) particles. Here, we showed that the POAG risk allele near ABCA1 attenuated ABCA1 expression in cultured cells. Consistently, POAG patients exhibited lower ABCA1 expression, reduced HDL, and higher cholesterol in white blood cells. Ablation of Abca1 in mice failed to form HDL, leading to elevated cholesterol levels in the retina. Counting retinal ganglion cells(RGCs) by using an artificial intelligence(AI) program revealed that Abca1-deficient mice progressively lost RGCs with age. Single-cell RNA sequencing(scRNA-seq) revealed aberrant oxidative phosphorylation in the Abca1-/-retina, as well as activation of the mTORC1 signaling pathway and suppression of autophagy. Treatment of Abca1-/-mice using atorvastatin reduced the cholesterol level in the retina,thereby improving metabolism and protecting RGCs from death. Collectively, we show that lower ABCA1 expression and lower HDL are risk factors for POAG. Accumulated cholesterol in the Abca1-/-retina causes profound aberrant metabolism, leading to a POAG-like phenotype that can be prevented by atorvastatin. 4Novel SFTSV Phylogeny Reveals New Reassortment Events and Migration Routes显示文摘Severe fever with thrombocytopenia syndrome virus(SFTSV),the causative agent of a febrile human disease,was first identified from central and eastern provinces in China,and later in Japan and South Korea.Hubei Province is one of the major SFTS epidemic areas in the central part of China.This study reported the isolation of 11 new SFTSV strains from patients in Hubei Province collected in 2017.Extensive phylogenetic analyses were conducted based on the complete coding sequences of SFTSV segments including the new strains.It was suggested that five different SFTSV genotypes were circulating in Hubei,and 15 reassortment patterns and migration pathways correlated with each genotype were identified,which was more than previously recognized.Hubei Province was more involved in the evolutionary events of SFTSV than that previously thought in which the evolutionary events of SFTSV were reported to be independent from those in other epidemic regions.Further divergence of SFTSV strains was suggested by pairwise comparison of SFTSV sequences from each genotype and sequence identity normalized to representative strain in genotype C1.Subsequently,amino acid variations specific for genotype(s),strain(s),or cluster(s)were inspected,which may be related to differential biological activity of SFTSV strains/genotypes.In conclusion,we analyzed the current status of SFTSV phylogeny in Hubei Province and discussed the possible events correlated to SFTSV evolution.It provided an in-depth insight into SFTSV evolution,raising concerns for the use of proper SFTSV strains in future studies.Xiaoli Wu Mingyue Li Yanfang Zhang Boyun Liang Junming Shi Yaohui Fang Zhengyuan Su Mengmeng Li Wenjing Zhang Ling Xu Jun Wang Qiaoli Wu Shuang Tang Hualin Wang Tao Zhang Cheng Peng Xin Zheng Fei Deng Shu Shen 2021Virologica Sinica2021,36,2:2
5Liver Function Derangement in Patients with Severe Fever and Thrombocytopenia Syndrome显示文摘Background and Aims:Patients with severe fever with thrombocytopenia syndrome(SFTS)commonly show liver function impairment.This study aimed to characterize the liver function indices in SFTS patients and investigate their association with mortality.Methods:Clinical information and laboratory results of 459 laboratory-confirmed SFTS patients,including 78 deceased and 381 surviving patients,were retrospectively analyzed.To explore the infectivity of SFTS caused by novel Bunyavirus(SFTSV)in hepatocytes,Huh7 human hepatoma cells were infected with various concentrations of SFTSV in vitro.Results:The proportion of SFTS patients developing liver injury during hospitalization was 73.2%(336/459);the hepatocellular injury was the predominant type.The median time to occurrence of liver injury from disease onset was 8 d.Liver injury in the deceased group occurred earlier than that in the surviving group.Alanine aminotransferase(ALT)level between 2–5 times upper limit of normal(ULN)at 4–6 d and between 5–15 ULN at 7–12 d of disease course were independent predictors of mortality.Alkaline phosphatase(ALP)>2 ULN at 7–9 d and elevated ALP at 10–12 days after disease onset were risk factors for death.ALT and aspartate transaminase(AST)levels were correlated with lymphocyte count and platelet-to-lymphocyte ratio(PLR).Total bilirubin(TB),ALT,AST levels showed positive correlation with viral load.In the in vitro experiment,SFTSV infected and replicated inside Huh7 cells.Conclusions:Liver injury is common in SFTS patients.ALT and ALP were independent predictors of SFTS-related mortality.Frequent monitoring and evaluation of liver function indices are needed for SFTS patients.Sihong Lu Ling Xu Boyun Liang Hua Wang Tong Wang Tiandan Xiang Sumeng Li Lei Fan Junyuan Li Cheng Peng Xin Zheng 2022Journal of Clinical and Translational Hepatology2022,10,5:1

8SARS-CoV-2-specific T cell responses wane profoundly in convalescent individuals 10 months after primary infection显示文摘A key question in the coronavirus disease 2019(COVID-19)pandemic is the duration of specific T cell responses against the severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)post primary infection,which is difficult to address due to the large-scale COVID-19 vaccination and re-exposure to the virus.Here,we conducted an analysis of the long-term SARS-CoV-2-specific T cell responses in a unique cohort of convalescent individuals(CIs)that were among the first to be infected worldwide and without any possible antigen re-exposure since then.The magnitude and breadth of SARS-CoV-2-specific T cell responses correlated inversely with the time that had elapsed from disease onset and the age of those CIs.The mean magnitude of SARS-CoV-2-specific CD4 and CD8 T cell responses decreased about 82%and 76%,respectively,over the time period of ten months after infection.Accordingly,the longitudinal analysis also demonstrated that SARS-CoV-2-specific T cell responses waned significantly in 75%of CIs during the follow-up.Collectively,we provide a comprehensive characterization of the long-term memory T cell response in CIs,suggesting that robust SARS-CoV-2-specific T cell immunity post primary infection may be less durable than previously expected.Ziwei Li Tiandan Xiang Boyun Liang Jing Liu Hui Deng Xuecheng Yang Hua Wang Xuemei Feng Gennadiy Zelinskyy Mirko Trilling Kathrin Sutter Mengji Lu Ulf Dittmer Baoju Wang Dongliang Yang Xin Zheng Jia Liu 2023Virologica Sinica2023,38,4:0
