| 1 | Overexpression of an ABA biosynthesis gene using a stress-inducible promoter enhances drought resistance in petunia显示文摘The response of plants to drought stress includes reduced transpiration as stomates close in response to increased abscisic acid(ABA)concentrations.Constitutive overexpression of 9-cis-epoxycarotenoid dioxygenase(NCED),a key enzyme in ABA biosynthesis,increases drought resistance,but causes negative pleiotropic effects on plant growth and development.We overexpressed the tomato NCED(LeNCED1)in petunia plants under the control of a stress-inducible promoter,rd29A.Under water stress,the transgenic plants had increased transcripts of NCED mRNA,elevated leaf ABA concentrations,increased concentrations of proline,and a significant increase in drought resistance.The transgenic plants also displayed the expected decreases in stomatal conductance,transpiration,and photosynthesis.After 14 days without water,the control plants were dead,but the transgenic plants,though wilted,recovered fully when re-watered.Well-watered transgenic plants grew like non-transformed control plants and there was no effect of the transgene on seed dormancy. | Alejandro C Estrada-Melo Chao Ma Michael S Reid Cai-Zhong Jiang | 2015 | Horticulture Research2015，2，1： | 12 |
| 2 | Inhibition of apoptosis by oncogenic hepatitis B virus X protein: Implications for the treatment of hepatocellular carcinoma显示文摘Hepatitis B virus X protein(HBx) plays an important role in the development of hepatocellular carcinoma(HCC). In addition, hepatoma upregulated protein(HURP) is a cellular oncogene that is upregulated in a majority of HCC cases. We highlight here recent findings demonstrating a link between HBx, HURP and anti-apoptosis effects observed in cisplatin-treated HCC cells. We observed that Hep3B cells overexpressing HBx display increased HURP mRNA and protein levels, and show resistance to cisplatin-induced apoptosis. Knockdown of HURP in HBx-expressing cells reverses this effect, and sensitizes cells to cisplatin. The anti-apoptotic effect of HBx requires activation of the p38/MAPK pathway as well as expression of SATB1, survivin and HURP. Furthermore, silencing of HURP using short-hairpin RNA promotes accumulation of p53 and reduces cell proliferation in SK-Hep-1 cells(p53^(+/–)), whereas these effects are not observed in p53-mutant Mahlavu cells. Similarly, HURP silencing does not affect the proliferation of H1299 lung carcinoma cells or Hep3 B HCC cells which lack p53. Silencing of HURP sensitizes SK-Hep-1 cells to cisplatin. While HURP overexpression promotes p53 ubiquitination and degradation by the proteasome, HURP silencing reverses these effects. Inoculation of SK-Hep-1 cancer cells in which HURP has been silenced produces smaller tumors than control in nude mice. Besides, gankyrin, a positive regulator of the E3 ubiquitin ligase MDM2, is upregulated following HURP expression, and silencing of gankyrin reduces HURP-mediated downregulation of p53. In addition, we observed a positive correlation between HURP and gankyrin protein levels in HCC patients(r^2 = 0.778; n = 9). | 3 | Cervical inlet patch-optical coherence tomography imaging and clinical significance显示文摘AIM:To demonstrate the feasibility of optical coherence tomography(OCT)imaging in differentiating cervical inlet patch(CIP)from normal esophagus,Barrett'sesophagus(BE),normal stomach and duodenum. METHODS:This study was conducted at the Veterans Affairs Boston Healthcare System(VABHS).Patients undergoing standard esophagogastroduodenoscopy at VABHS,including one patient with CIP,one representative patient with BE and three representative normal subjects were included.White light video endoscopy was performed and endoscopic 3D-OCT images were obtained in each patient using a prototype OCT system.The OCT imaging probe passes through the working channel of the endoscope to enable simultaneous video endoscopy and 3D-OCT examination of the human gastrointestinal(GI)tract.Standard hematoxylin and eosin(H and E)histology was performed on biopsy or endoscopic mucosal resection specimens in order to compare and validate the 3D-OCT data. RESULTS:CIP was observed from a 68-year old male with gastroesophageal reflux disease.The CIP region appeared as a pink circular lesion in the upper esophagus under white light endoscopy.OCT imaging over the CIP region showed columnar epithelium structure,which clearly contrasted the squamous epithelium structure from adjacent normal esophagus.3D-OCT images obtained from other representative patients demonstrated distinctive patterns of the normal esophagus,BE,normal stomach,and normal duodenum bulb.Microstructures,such as squamous epithelium,lamina propria, muscularis mucosa,muscularis propria,esophageal glands,Barrett's glands,gastric mucosa,gastric glands, and intestinal mucosal villi were clearly observed with OCT and matched with H and E histology.These results demonstrated the feasibility of using OCT to evaluate GI tissue morphology in situ and in real-time. | 4 | Adult metaplastic hutch diverticulum with robotic-assisted diverticulectomy and reconstruction:A case report显示文摘BACKGROUND Hutch diverticulum arises from the compromised muscular development at the ureteral orifice.It is a congenital disease and extremely rare in adult,only accounting for about 3%occurrence worldwide.It can be either symptomatic or asymptomatic,and relies on image tools for diagnosis and preoperative planning.Indications for surgery are dependent on the complications from the diverticulum.Metaplasia is about 10%among those with hutch diverticulum,and it still has chances turning into malignancy,especially urothelial cell carcinoma.CASE SUMMARY A 27-year-old man was presented with frequently recurrent urinary tract infection for one year,and had suffered from intermittent right flank pain for 3 mo.No past medical histories were recorded before.No obvious abnormalities on laboratory data and urine examination were found.Under ultrasound,right hydronephrosis was seen and an anatomical abnormality was observed on intravenous pyelography.Further computed tomography urogram showed one diverticulum seated at superolateral side of right ureteral orifice.Cystoscopy was done and biopsy results showed focal metaplasia.After discussing with him,roboticassisted diverticulectomy with reconstruction was performed.Right hydronephrosis was greatly improved after surgery.He has completed his 1.5-year follow-ups,and no malignancies were seen from urine cytology and image of intravenous pyelography.CONCLUSION Robotic-assisted diverticulectomy and reconstruction to hutch diverticulum is a safe and efficient operation,providing several advantages over open and laparoscopic ones. | Che H Yang Yi S Lin Yen C Ou Wei C Weng Li H Huang Chin H Lu Chao Y Hsu Min C Tung | 2020 | World Journal of Clinical Cases2020，8，20： | 2 |
| 5 | The importance of an indigenous tree to southern African communities with specific relevance to its domestication and commercialization: a case of the marula tree显示文摘Local communities in southern Africa derive many benefits from marula [Sclerocarya birrea(A.Rich.) Hochst.subsp.caffra(Sond.) Kokwaro].These include the contribution of this species towards health,nutrition,food security and conservation by sharing local skills and knowledge related to it.Marula fruits can be eaten fresh,squeezed to make juice,brewed in traditional beer or used to make jam and jelly.The kernels are also edible and can be pressed to extract oil for cooking and cosmetics,i.e.,for skin and hair application.The bark,roots,seeds and leaves are exploited for traditional medicinal purposes.Marula has acquired significant commercial value since its fruits and other products have entered local,regional and international trade in southern Africa.To diver-sify fruit production and satisfy the increasing demands for this resource,several domestication initiatives have been carried out at regional and international levels.The accumulated knowledge and skills relevant to the establishment and commercialization of marula therefore provide an effective guide in areas where marula remains undomesticated or underutilized.We discuss the great importance of marula in southern Africa to reveal its great potential to regions where it remains unexploited. | Neo C MOKGOLODI DING You-fang Moffat P SETSHOGO MA Chao LIU Yu-jun | 2011 | Forestry Studies in China2011，13，1： | 2 |
