1Zika virus infection induces RNAi-mediated antiviral immunity in human neural progenitors and brain organoids显示文摘The re-emergence of Zika virus(ZIKV)in the Western Hemisphere has resulted in global public health crisis since 2015.ZIKV preferentially infects and targets human neural progenitor cells(hNPCs)and causes fetal microcephaly upon maternal infection.hNPCs not only play critical roles during fetal brain development,but also persist in adult brain throughout life.Yet the mechanism of innate antiviral immunity in hNPCs remains largely unknown.Here,we show that ZIKV infection triggers the abundant production of virus-derived small interfering RNAs in hNPCs,but not in the more differentiated progenies or somatic cells.Ablation of key RNAi machinery components significantly enhances ZIKV replication in hNPCs.Furthermore,enoxacin,a broad-spectrum antibiotic that is known as an RNAi enhancer,exerts potent anti-ZIKV activity in hNPCs and other RNAi-competent cells.Strikingly,enoxacin treatment completely prevents ZIKV infection and circumvents ZIKV-induced microcephalic phenotypes in brain organoid models that recapitulate human fetal brain development.Our findings highlight the physiological importance of RNAi-mediated antiviral immunity during the early stage of human brain development,uncovering a novel strategy to combat human congenital viral infections through enhancing RNAi.Yan-Peng Xu Yang Qiu Boya Zhang Guilai Chen Qi Chen Miao Wang Fan Mo Jiuyue Xu Jin Wu Rong-Rong Zhang Meng-Li Cheng Na-Na Zhang Bao Lyu Wen-Liang Zhu Meng-Hua Wu Qing Ye Da Zhang Jiang-Hong Man Xiao-Feng Li Jie Cui Zhiheng Xu Baoyang Hu Xi Zhou Cheng-Feng Qin 2019Cell Research2019,29,4:8
2Effects of microscopic pore structure heterogeneity on the distribution and morphology of remaining oil显示文摘Waterflooding experiments were performed using Micro-CT on four cores of different pore structures from Donghe sandstone reservoirs in the Tarim Basin. The water, oil and grains were accurately separated by the advanced image processing technology, the pore network model was established, and parameters such as the number of throats and the throat size distribution were calculated to characterize the microscopic heterogeneity of pore structure, the flow of oil phase during displacement, and the morphology and distribution of remaining oil after displacement. The cores with the same macroscopic porosity-permeability have great differences in microscopic heterogeneity of pore structure. Both macro porosity-permeability and micro heterogeneity of pore structure have an influence on the migration of oil phase and the morphology and distribution of remaining oil. When the heterogeneity is strong, the water phase will preferentially flow through the dominant paths and the remaining oil clusters will be formed in the small pores. The more the number of oil clusters(droplets) formed during displacement process, the smaller the average volume of cluster is, and the remaining oil is dominated by the cluster continuous phase with high saturation. The weaker the heterogeneity, the higher the pore sweep efficiency is, and the remaining oil clusters are mainly trapped in the form of non-continuous phase. The distribution and morphology of micro remaining oil are related to the absolute permeability, capillary number and micro-heterogeneity. 3Pore-scale investigation on occurrence characteristics and conformance control mechanisms of emulsion in porous media显示文摘Microscopic computed tomography(Micro-CT)is used to visualize microscopic flow in sandstone core samples during emulsion flooding.The images obtained during the experiment are processed quantitatively with a series of methods to evaluate the occurrence characteristics and oil recovery enhancement mechanisms of emulsion.(1)The emulsion is distributed in the cores in spherical shape,and its sphericity is significantly different from that of the remaining oil.Sphericity can be taken as a characteristic parameter to identify emulsion.(2)The emulsion with specific size prefers to stay in pores with matching sizes;when the emulsion volume is smaller than the product of the lower limit of pore occupancy and the corresponding pore vol-ume,it will not be able to effectively trap in the pore to achieve conformance control.In the process of emulsion displacement designing,we need to design emulsion with suitable particle size according to the pore distribution of the reservoir.(3)Mobi-lization ratio of the pores can be increased from 23.1%to 59.3%by emulsion flooding after subsequent water flooding,and the average oil displacement efficiency at the pore-scale can be increased from 22.9%to 75.8%under the test conditions;(4)After emulsion flooding,the clustered remaining oil and the oil phase in the trapped emulsion are the main targets for further EOR.SU Hang ZHOU Fujian LIU Yang GAO Yajun CHENG Baoyang DONG Rencheng LIANG Tianbo LI Junjian 2021Petroleum Exploration and Development2021,48,6:0
