| 8 | Differential seeding and propagating efficiency ofα-synuclein strains generated in different conditions显示文摘Background:Accumulation of alpha-synuclein(α-syn)is a main pathological hallmark of Parkinson’s and related diseases,which are collectively known as synucleinopathies.Growing evidence has supported that the same protein can induce remarkably distinct pathological progresses and disease phenotypes,suggesting the existence of strain difference amongα-syn fibrils.Previous studies have shown thatα-syn pathology can propagate from the peripheral nervous system(PNS)to the central nervous system(CNS)in a“prion-like”manner.However,the difference of the propagation potency from the periphery to CNS among differentα-syn strains remains unknown and the effect of different generation processes of these strains on the potency of seeding and propagation remains to be revealed in more detail.Methods:Three strains of preformedα-syn fibrils(PFFs)were generated in different buffer conditions which varied in pH and ionic concentrations.Theα-syn PFFs were intramuscularly(IM)injected into a novel bacterial artificial chromosome(BAC)transgenic mouse line that expresses wild-type humanα-syn,and the efficiency of seeding and propagation of these PFFs from the PNS to the CNS was evaluated.Results:The three strains ofα-syn PFFs triggered distinct propagation patterns.The fibrils generated in mildly acidic buffer led to the most severeα-syn pathology,degeneration of motor neurons and microgliosis in the spinal cord.Conclusions:The differentα-syn conformers generated in different conditions exhibited strain-specific pathology and propagation patterns from the periphery to the CNS,which further supports the view thatα-syn strains may be responsible for the heterogeneity of pathological features and disease progresses among synucleinopathies. | Di Liu Jian-Jun Guo Ji-Hui Su Alexander Svanbergsson Lin Yuan Caroline Haikal Wen Li Gunnar Gouras Jia-Yi Li | 2021 | Translational Neurodegeneration2021,10,2: | 0 |

| 10 | Collagen membrane functionalized with magnesium oxide via room-temperature atomic layer deposition promotes osteopromotive and antimicrobial properties显示文摘Artificial bone grafting materials such as collagen are gaining interest due to the ease of production and implantation.However,collagen must be supplemented with additional coating materials for improved osteointegration.Here,we report room-temperature atomic layer deposition(ALD)of MgO,a novel method to coat collagen membranes with MgO.Characterization techniques such as X-ray photoelectron spectroscopy,Raman spectroscopy,and electron beam dispersion mapping confirm the chemical nature of the film.Scanning electron and atomic force microscopies show the surface topography and morphology of the collagen fibers were not altered during the ALD of MgO.Slow release of magnesium ions promotes bone growth,and we show the deposited MgO film leaches trace amounts of Mg when incubated in phosphate-buffered saline at 37◦C.The coated collagen membrane had a superhydrophilic surface immediately after the deposition of MgO.The film was not toxic to human cells and demonstrated antibacterial properties against bacterial biofilms.Furthermore,in vivo studies performed on calvaria rats showed MgO-coated membranes(200 and 500 ALD)elicit a higher inflammatory response,leading to an increase in angiogenesis and a greater bone formation,mainly for Col-MgO500,compared to uncoated collagen.Based on the characterization of the MgO film and in vitro and in vivo data,the MgO-coated collagen membranes are excellent candidates for guided bone regeneration. | Soumya Saha Raphael Cavalcante Costa Mirela Caroline Silva Joao Matheus Fonseca-Santos Lin Chen Abhijit H.Phakatkar Harshdeep Bhatia Leonardo P.Faverani Valentim A.R.Barao Tolou Shokuhfar Cortino Sukotjo Christos Takoudis | 2023 | Bioactive Materials2023,,12: | 0 |

| 13 | Are University Rankings Statistically Significant?A Comparison among Chinese Universities and with the USA显示文摘Purpose:Building on Leydesdorff,Bornmann,and Mingers(2019),we elaborate the differences between Tsinghua and Zhejiang University as an empirical example.We address the question of whether differences are statistically significant in the rankings of Chinese universities.We propose methods for measuring statistical significance among different universities within or among countries.Design/methodology/approach:Based on z-testing and overlapping confidence intervals,and using data about 205 Chinese universities included in the Leiden Rankings 2020,we argue that three main groups of Chinese research universities can be distinguished(low,middle,and high).Findings:When the sample of 205 Chinese universities is merged with the 197 US universities included in Leiden Rankings 2020,the results similarly indicate three main groups:low,middle,and high.Using this data(Leiden Rankings and Web of Science),the z-scores of the Chinese universities are significantly below those of the US universities albeit with some overlap.Research limitations:We show empirically that differences in ranking may be due to changes in the data,the models,or the modeling effects on the data.The scientometric groupings are not always stable when we use different methods.Practical implications:Differences among universities can be tested for their statistical significance.The statistics relativize the values of decimals in the rankings.One can operate with a scheme of low/middle/high in policy debates and leave the more fine-grained rankings of individual universities to operational management and local settings.Originality/value:In the discussion about the rankings of universities,the question of whether differences are statistically significant,has,in our opinion,insufficiently been addressed in research evaluations. | Loet Leydesdorff Caroline SWagner Lin Zhang | 2021 | Journal of Data and Information Science2021,6,2: | 0 |
| 14 | Molecular signature of soil organic matter under different land uses in the Lake Chaohu Basin显示文摘The concentration and molecular composition of soil organic matter(SOM)are important factors in mitigation against climate change as well as providing other ecosystem services.Our quantitative understanding of how land use influences SOM molecular composition and associated turnover dynamics is limited,which underscores the need for high-throughput analytical approaches and molecular marker signatures to clarify this etiology.Combining a high-throughput untargeted mass spectrometry screening and molecular markers,we show that forest,farmland and urban land uses result in distinct molecular signatures of SOM in the Lake Chaohu Basin.Molecular markers indicate that forest SOM has abundant carbon contents from vegetation and condensed organic carbon,leading to high soil organic carbon(SOC)concentration.Farmland SOM has moderate carbon contents from vegetation,and limited content of condensed organic carbon,with SOC significantly lower than that of forest soils.Urban SOM has high abundance of condensed organic carbon markers due to anthropogenic activities but relatively low in markers from vegetation.Consistently,urban soils have the highest black carbon/SOC ratio among these land uses.Overall,our results suggested that the molecular signature of SOM varies significantly with land use in the Lake Chaohu Basin,influencing carbon dynamics.Our strategy of molecular fingerprinting and marker discovery is expected to enlighten further research on SOM molecular signatures and cycling dynamics. | Han Gao Huixin Li Chen Lin Pedro J.J.Alvarez Caroline A.Masiello Dongqiang Zhu Ao Kong Xiaolei Qu | 2022 | Eco-Environment & Health2022,1,4: | 0 |
