7Platelet-rich plasma-induced bone marrow mesenchymal stem cells versus autologous nerve grafting for sciatic nerve repair显示文摘Autologous nerve grafting is the gold standard of peripheral nerve repair. We previously showed that autologous platelet-rich plasma (PRP) contains high concentrations of growth factors and can induce in vitro cultured bone marrow mesenchymal stem cells (BMSCs) to differentiate into Schwann cells. Here we used PRP-induced BMSCs combined with chemically extracted acellular nerves to repair sciatic nerve defects and compared the effect with autologous nerve grafting. The BMSCs and chemically extracted acellular nerve promoted target muscle wet weight restoration, motor nerve conduction velocity, and axonal and myelin sheath regeneration, with similar effectiveness to autologous nerve grafting. 10In vitro differentiation of rabbit bone marrow mesenchymal stem cells into Schwann cells Induction effect of platelet-rich plasma显示文摘BACKGROUND: Bone marrow mesenchymal stem cells (BMSCs) differentiate into Schwann cells via specific inducers. However, the induction and culture procedures are complicated. Various growth factors have been used for induction and culture, and are likely affected by their environment. OBJECTIVE: To explore the differentiation feasibility of platelet-rich plasma-induced rabbit BMSCs, supplemented with various growth factors, into Schwann-like cells in vitro, and to examine the secretory function of Schwann-like cells. DESIGN, TIME AND SETTING: This comparison study was performed at the Experimental Animal Center, Affiliated Hospital of Qingdao University Medical College, China in October 2008. MATERIALS: Platelet-rich plasma and BMSCs were respectively obtained from the femoral vein and bone marrow of 2-month old New Zealand rabbits. The rabbit nerve growth factor ELISA kit and rabbit nerve growth factor PCR kit (Jingmei Biotech, China), as well as anti-rabbit S-100 immunofluorescence staining kit (Boster, China) were used in the present study. METHODS: BMSCs at passage three were harvested and were induced by 1 mmol/L β-mercaptoethanol and Dulbecco's-modified eagle's medium complete medium containing 35 ng/mL retinoic acid and 10% fetal bovine serum for the combination induction group, followed by incubation in complete medium supplemented with platelet-rich plasma. BMSCs in the single induction group were induced by β-mercaptoethanol and retinoic acid, followed by incubation in L-DMEM complete medium. BMSCs in the control group were incubated in L-DMEM complete medium. MAIN OUTCOME MEASURES: Cell growth in each group was observed under an inverted microscope. Following induction, S-100 protein expression was identified by immunofluorescence staining. Nerve growth factor protein concentrations in BMSC supernatant were subsequently measured by ELISA. In addition, nerve growth factor mRNA expression in the BMSCs was determined by reverse transcription-polymerase chain reaction. RESULTS: Overall BMSC morphology was similar to Schwann cells in the combination induction group, and morphology of the majority of BMSCs was similar to Schwann cells in the single induction group 9 days after induction. In the control group, significant changes in cell morphology were not observed 9 days after induction. S-100 protein expression was detected in the combination induction and single induction groups following induction. At days 7, 9, and 11, the expression rate of S-100-positive cells was greater in the combination induction group (P < 0.05). At days 4, 7, 9, and 11, nerve growth factor levels were significantly greater in BMSC supernatant in the combination induction and single induction groups compared with the control group (P < 0.05). At days 7, 9, and 11, nerve growth factor levels were significantly greater in the combination induction group compared with the single induction group (P < 0.05). At day 11, nerve growth factor mRNA expression in BMSCs was significantly greater in the combination induction group compared with the single induction group (P < 0.05). 11MORPHOLOGICAL STUDY ON VERTEBARTE MELANOCYTES显示文摘In order to explore the development of melanocytes (MC) in comparative systematic biology, so as to make a further understanding of the function of MC and diseases related to MC, we made a comparative study through pathological and TEM technique, on morphology of MC in different vertebrate animals, such as fish class (finel eel), amphibious animal (matured and immatured bulldog and frog), reptilian (turtle, walzard), aves (chicken) and mammalia (white mice, human embryo and adult in formative process of vertibate evolution.). The results showed that histologically, from the fish to amphibious animal, reptilia, and mammalia. MC were increased in number gradually, forming from stellar, polygonal form to more regular form with cell body growing smaller and dendrites growing from pyknic to slender with a gradual increase of branches, MC also scattreed differently and the organells developed more with evolution. These suggest that the change of MC in the evolution process of vertebrates may be related to the gradual adapte to alteration of living environment from water to land. As to the sunlight and ultraviolet ray increases. 13Hydrochemical Characterization,Controlling Factors and Water Quality of Surface Water in Shandong Province,North China显示文摘In this study,90 surface water samples were collected from Shandong Province(SDP)in the dry and wet seasons and analyzed using statistical analysis,hydrochemical methods and water quality index(WQI).The content of main ions showed seasonal variations,with a higher average value in the dry season than in the wet season.Sampling points exhibiting high contents of the main chemical components were mainly distributed in areas southwest of SDP,north of Weifang,and around Jinan.The saturation index of carbonate minerals was greater than zero,while that of evaporite minerals was less than zero.The hydrochemical characteristics of surface water are mainly dominated by rock weathering as well as cation exchange.In addition,surface water in SDP has significantly been affected by anthropogenic factors.Most of the surface water could be classified as weakly alkaline soft-fresh water,with the hydrochemical types of SO_(4)·Cl-Ca·Mg and SO_(4)·Cl-Na.In terms of WQI,47.88% and 37.88% of the water samples in SDP were classified as poor water in the dry and wet seasons,respectively.On the whole,the water quality is higher in the wet season than in the dry season,and surface water in SDP is generally suitable for agricultural irrigation.Ran An Jiutan Liu Zongjun Gao Changsuo Li Yuming Peng 2024Journal of Earth Science2024,35,1:0
