| 1 | Blunted nitric oxide regulation in Tibetans under high-altitude hypoxia显示文摘Nitric oxide(NO) is an important molecule for vasomotor tone, and elevated NO signaling was previously hypothesized as a unique and adaptive physiological change in highland Tibetans. However, there has been lack of NO data from Tibetans living at low altitude and lowlander immigrants living at high altitude, which is crucial to test this hypothesis. Here, through cross-altitude(1990–5018 m) and cross-population(Tibetans and Han Chinese) analyses of serum NO metabolites(NOx) of 2086 individuals, we demonstrate that although Tibetans have a higher serum NOx level compared to lowlanders, Han Chinese immigrants living at high altitude show an even higher level than Tibetans. Consequently, our data contradict the previous proposal of increased NO signaling as the unique adaptive strategy in Tibetans.Instead, Tibetans have a relatively lower circulating NOx level at high altitude. This observation is further supported by data from the hypoxic experiments using human umbilical vein endothelial cells and gene knockout mice. No difference is detected between Tibetans and Han Chinese for endothelial nitric oxide synthase(e NOS), the key enzyme for circulating NO synthesis, suggesting that e NOS itself is unlikely to be the cause. We show that other NO synthesis-related genes(e.g. GCH1) carry Tibetan-enriched mutations significantly associated with the level of circulating NOx in Tibetans. | 2 | De novo assembly of a Tibetan genome and identification of novel structural variants associated with high-altitude adaptation显示文摘Structural variants(SVs) may play important roles in human adaptation to extreme environments such as high altitude but have been under-investigated. Here, combining long-read sequencing with multiple scaffolding techniques, we assembled a high-quality Tibetan genome(ZF1), with a contig N50 length of 24.57 mega-base pairs(Mb) and a scaffold N50 length of 58.80 Mb. The ZF1 assembly filled 80 remaining N-gaps(0.25 Mb in total length) in the reference human genome(GRCh38). Markedly, we detected17 900 SVs, among which the ZF1-specific SVs are enriched in GTPase activity that is required for activation of the hypoxic pathway. Further population analysis uncovered a 163-bp intronic deletion in the MKL1 gene showing large divergence between highland Tibetans and lowland Han Chinese. This deletion is significantly associated with lower systolic pulmonary arterial pressure, one of the key adaptive physiological traits in Tibetans. Moreover, with the use of the high-quality de novo assembly, we observed a much higher rate of genome-wide archaic hominid(Altai Neanderthal and Denisovan) shared non-reference sequences in ZF1(1.32%–1.53%) compared to other East Asian genomes(0.70%–0.98%),reflecting a unique genomic composition of Tibetans. One such archaic hominid shared sequence-a662-bp intronic insertion in the SCUBE2 gene-is enriched and associated with better lung function(the FEV1/FVC ratio) in Tibetans. | 3 | Dab2 attenuates brain injury in APP/PS1 mice via targeting transforming growth factor-beta/SMAD signaling显示文摘Transforming growth factor-beta(TGF-β)type II receptor(TβRII)levels are extremely low in the brain tissue of patients with Alzheimer’s disease.This receptor inhibits TGF-β1/SMAD signaling and thereby aggravates amyolid-beta deposition and neuronal injury.Dab2,a specific adapter protein,protects TβRII from degradation and ensures the effective conduction of TGF-β1/SMAD signaling.In this study,we used an adenoviral vector to overexpress the Dab2 gene in the mouse hippocampus and investigated the regulatory effect of Dab2 protein on TGF-β1/SMAD signaling in a mouse model of Alzheimer’s disease,and the potential neuroprotective effect.The results showed that the TβRII level was lower in APP/PS1 mouse hippocampus than in normal mouse hippocampus.After Dab2 expression,hippocampal TβRII and p-SMAD2/3 levels were significantly increased,while amyloid-beta deposition,microglia activation,tumor necrosis factor-βand interleulin-6 levels and neuronal loss were significantly attenuated in APP/PS1 mouse brain tissue.These results suggest that Dab2 can exhibit neuroprotective effects in Alzheimer’s disease by regulating TGF-β1/SMAD signaling. | Lei Song Yue Gu Jing Jie Xiaoxue Bai Ying Yang Chaoying Liu Qun Liu | 2014 | Neural Regeneration Research2014,9,1: | 4 |

| 9 | Fast and accurate earthquake location within complex medium using a hybrid global-local inversion approach显示文摘A novel hybrid approach for earthquake location is proposed which uses a combined coarse global search and fine local inversion with a minimum search routine,plus an examination of the root mean squares(RMS) error distribution.The method exploits the advantages of network ray tracing and robust formulation of the Fréchet derivatives to simultaneously update all possible initial source parameters around most local minima(including the global minimum) in the solution space,and finally to determine the likely global solution.Several synthetic examples involving a 3-D complex velocity model and a challenging source-receiver layout are used to demonstrate the capability of the newly-developed method.This new global-local hybrid solution technique not only incorporates the significant benefits of our recently published hypocenter determination procedure for multiple earthquake parameters,but also offers the attractive features of global optimal searching in the RMS travel time error distribution.Unlike the traditional global search method,for example,the Monte Carlo approach,where millions of tests have to be done to find the final global solution,the new method only conducts a matrix inversion type local search but does it multiple times simultaneously throughout the model volume to seek a global solution.The search is aided by inspection of the RMS error distribution.Benchmark tests against two popular approaches,the direct grid search method and the oct-tree important sampling method,indicate that the hybrid global-local inversion yields comparable location accuracy and is not sensitive to modest level of noise data,but more importantly it offers two-order of magnitude speed-up in computational effort.Such an improvement,combined with high accuracy,make it a promising hypocenter determination scheme in earthquake early warning,tsunami early warning,rapid hazard assessment and emergency response after strong earthquake occurrence. | Chaoying Bai Rui Zhao Stewart Greenhalgh | 2009 | Earthquake Science2009,22,5: | 0 |

| 11 | 3-D multi-parameter type traveltime tomography in a spherical coordinate frame: comparison of double and triple class simultaneous inversions显示文摘It is now common practice to perform simultaneous traveltime inversion for the velocity field and the reflector geometry in reflection/refraction tomography, or the velocity field and the hypocenter locations in regional earthquake tomography, but seldom are all three classes of model parameters updated simultaneously. This is mainly due to the trade-off between the different types of model parameters and the lack of different seismic phases to constrain the model parameters. Using a spherical-coordinate ray tracing algorithm for first and later(primary reflected) arrival tracing algorithm in combination with a popular linearized inversion solver, it is possible to simultaneously recover the three classes of model parameters in regional or global tomographic studies. In this paper we incorporate the multistage irregular shortest-path ray tracing algorithm(in a spherical coordinate system) with a subspace inversion solver to formulate a simultaneous inversion algorithm for triple model parameters updating using direct and later arrival time information.Comparison tests for two sets of data(noise free and added noise) indicate that the new triple-class parameter inversion algorithm is capable of obtaining nearly the same results as the double-class parameter inversion scheme. Furthermore,the proposed multi-parameter type inversion method is not sensitive to a modest level of picking error in the traveltime data, and also performs well with a relatively large uncertainty in earthquake hypocentral locations. | 12 | A highly effective incubation strategy enhanced the urban bird hatch success显示文摘Urbanization is currently considered one of the most rapid types of global environmental change.Urban habitats are biotically and abiotically different from their rural areas,i.e.,the ambient temperature,predator,and food availability.These novel challenges create new selection pressures,which allow one to investigate ecoevolutionary responses to contemporary environmental change.A total of 118 breeding nests were monitored for nest predation in both urban and rural areas from 2018 to 2020.We used environmental factors from urban and rural areas and behavioral data from 439 Chinese Blackbird(Turdus mandarinus)valid incubation days to understand the impact of urbanization on the incubation behavior of blackbirds and its adaptation mechanism to the urban environment.Cities have warmer ambient temperatures and lower predation pressures than rural areas.Urban blackbirds chose the incubation strategy with shorter and more bouts,while rural blackbirds selected the incubation strategy with longer and fewer bouts.The plasticity of incubation behavior of urban blackbirds was higher than that of rural areas,and the range of egg temperature was also higher than that of rural areas.In addition,incubation temperature and the number of bouts per day were the key factors affecting the day survival rate of blackbirds,and the hatching rate of urban blackbirds was higher than that of rural blackbirds.Our results provide evidence for behavioral shifts in blackbirds during adaptation to urbanization and support the central role of behavioral adaptation in the successful colonization of new environments by wildlife.These help us understand the behavioral characteristics required for wildlife to live in cities and the urban adaptors faced environmental pressures. | Long Ma Yuancheng Liu Wenjing Lu Zheng Zhang Wanyou Li Ziwei Zhang Xueli Zhang Chaoying Zhu Junpeng Bai Zhifeng Xu Yuqing Han Luzhang Ruan | 2023 | Avian Research2023,14,1: | 0 |
