| 2 | Bromodomain-containing proteins BRD1, BRD2, and BRD13 are core subunits of SWI/SNF complexes and vital for their genomic targeting in Arabidopsis显示文摘Switch defective/sucrose non-fermentable(SWI/SNF)chromatin remodeling complexes are multi-subunit machines that play vital roles in the regulation of chromatin structure and gene expression.However,the mechanisms by which SWI/SNF complexes recognize their target loci in plants are not fully understood.Here,we show that the Arabidopsis thaliana bromodomain-containing proteins BRD1,BRD2,and BRD13 are core subunits of SWI/SNF complexes and critical for SWI/SNF genomic targeting.These three BRDs interact directly with multiple SWI/SNF subunits,including the BRAHMA(BRM)catalytic subunit.Phenotypic and transcriptomic analyses of the brd1 brd2 brd13 triple mutant revealed that these BRDs act largely redundantly to control gene expression and developmental processes that are also regulated by BRM.Genome-wide occupancy profiling demonstrated that these three BRDs extensively colocalize with BRM on chromatin.Simultaneous loss of function of three BRD genes results in reduced BRM protein levels and decreased occupancy of BRM on chromatin across the genome.Furthermore,we demonstrated that the bromodomains of BRDs are essential for genomic targeting of the BRD subunits of SWI/SNF complexes to their target sites.Collectively,these results demonstrate that BRD1,BRD2,and BRD13 are core subunits of SWI/SNF complexes and reveal their biological roles in facilitating genomic targeting of BRM-containing SWI/SNF complexes in plants. | Yaoguang Yu Wei Fu Jianqu Xu Yawen Lei Xin Song Zhenwei Liang Tao Zhu Yuhui Liang Yuanhao Hao Liangbing Yuan Chenlong Li | 2021 | Molecular Plant2021,14,6: | 1 |
| 3 | Hard nanocrystalline gold materials prepared via high-pressure phase transformation显示文摘As one of the important materials,nanocrystalline Au(n-Au)has gained numerous interests in recent decades owing to its unique properties and promising applications.However,most of the current n-Au thin films are supported on substrates,limiting the study on their mechanical properties and applications.Therefore,it is urgently desired to develop a new strategy to prepare nAu materials with superior mechanical strength and hardness.Here,a hard n-Au material with an average grain size of~40 nm is prepared by cold-forging of the unique Au nanoribbons(NRBs)with unconventional 4H phase under high pressure.Systematic characterizations reveal the phase transformation from 4H to face-centered cubic(fcc)phase during the cold compression.Impressively,the compressive yield strength and Vickers hardness(HV)of the prepared n-Au material reach~140.2 MPa and~1.0 GPa,which are 4.2 and 2.2 times of the microcrystalline Au foil,respectively.This work demonstrates that the combination of high-pressure cold-forging and the in-situ 4H-to-fcc phase transformation can effectively inhibit the grain growth in the obtained n-Au materials,leading to the formation of novel hard n-Au materials.Our strategy opens up a new avenue for the preparation of nanocrystalline metals with superior mechanical property. | Chenlong Xie Wenxin Niu Penghui Li Yiyao Ge Jiawei Liu Zhanxi Fan Xiaoxiao Liu Ye Chen Ming Zhou Zihe Li Mengdong Ma Yonghai Yue Jing Wang Li Zhu Kun Luo Yang Zhang Yingju Wu Lin Wang Bo Xu Hua Zhang Zhisheng Zhao Yongjun Tian | 2022 | Nano Research2022,15,7: | 0 |
| 4 | Mapping the distributions of blood-sucking mites and mite-borne agents in China:a modeling study显示文摘Background:Emerging mite-borne pathogens and associated disease burdens in recent decades are raising serious public health concerns,yet their distributions and ecology remain under-investigated.We aim to describe the geographical distributions of blood-sucking mites and mite-borne agents and to assess their ecological niches in China.Methods:We mapped 549 species of blood-sucking mites belonging to 100 genera at the county level and eight mite-associated agents detected from 36 species of blood-sucking mites in China during 1978–2020.Impacts of climatic and environmental factors on the ecology of 21 predominant vector mites and a leading pathogen,Orientia tsutsugamushi,were assessed using boosted regression tree(BRT)models,and model-predicted risks were mapped.We also estimated the model-predicted number,area and population size of afected counties for each of the 21 mite species in China.Results:Laelaps echidninus is the leading mite species that potentially afects 744 million people,followed by La.jettmari(517 million)and Eulaelaps stabularis(452 million).Leptotrombidium scutellare is the mite species harboring the highest variety of mite-borne agents including four Rickettsia species and two viruses,followed by Eu.stabularis(2 agents),L.palpale(2)and La.echidninus(2).The top two agents that parasitize the largest number of mite species are O.tsutsugamushi(28 species)and hantavirus(8).Mammalian richness,annual mean temperature and precipitation of the driest quarter jointly determine the ecology of the mites,forming four clusters of major mite species with distinct geographic distributions.High-risk areas of O.tsutsugamushi are mainly distributed in southern and eastern coastal provinces where 71.5 million people live.Conclusions:Ecological niches of major mite species and mite-borne pathogens are much more extensive than what have been observed,necessitating expansion of current fled surveillance. | Tao Wang Fanfei Meng Tianle Che Jinjin Chen Haiyang Zhang Yang Ji Zhengwei Fan Guoping Zhao Wenhui Zhang Baogui Jiang Qiang Xu Chenlong Lv Taoxing Shi Shiman Ruan Lanzheng Liu Wei Liu Yang Yang Liqun Fang | 2022 | Infectious Diseases of Poverty2022,11,2: | 0 |

| 6 | Identification and validation of key biomarkers and potential therapeutic targets for primary open-angle glaucoma显示文摘Primary open-angle glaucoma(POAG)is a prevalent cause of blindness worldwide,resulting in degeneration of retinal ganglion cells and permanent damage to the optic nerve.However,the underlying pathogenetic mechanisms of POAG are currently indistinct,and there has been no effective nonsurgical treatment regimen.The objective of this study is to identify novel biomarkers and potential therapeutic targets for POAG.The mRNA expression microarray datasets GSE27276 and GSE138125,as well as the single-cell high-throughput RNA sequencing(sc RNA-seq)dataset GSE148371 were utilized to screen POAGrelated differentially expressed genes(DEGs).Functional enrichment analyses,protein-protein interaction(PPI)analysis,and weighted gene co-expression network analysis(WGCNA)of the DEGs were performed.Subsequently,the hub genes were validated at a single-cell level,where trabecular cells were annotated,and the mRNA expression levels of target genes in different cell clusters were analyzed.Immunofluorescence and quantitative real-time PCR(q PCR)were performed for further validation.DEGs analysis identified 43 downregulated and 32 upregulated genes in POAG,which were mainly enriched in immune-related pathways,oxidative stress,and endoplasmic reticulum(ER)stress.PPI networks showed that FN1 and DUSP1 were the central hub nodes,while GPX3 and VAV3 were screened out as hub genes through WGCNA and subsequently validated by q PCR.Finally,FN1,GPX3,and VAV3 were determined to be pivotal core genes via single-cell validation.The relevant biomarkers involved in the pathogenesis of POAG,may serve as potential therapeutic targets.Further studies are necessary to unveil the mechanisms underlying the expression variations of these genes in POAG. | Jian Wu Caixia Lin Chenlong Yang Lijie Pan Hongyi Liu Sirui Zhu Shuwen Wei Xu Jia Qi Zhang Ziyu Yu Xiaofang Zhao Weihai Liu Yehong Zhuo Ningli Wang | 2023 | Science China(Life Sciences)2023,66,12: | 0 |
