| 1 | 8-bromo-7-methoxychrysin-induced apoptosis of hepatocellular carcinoma cells involves ROS and JNK显示文摘AIM:To investigate whether the apoptotic activities of 8-bromo-7-methoxychrysin(BrMC) involve reactive oxygen species(ROS) generation and c-Jun N-terminal kinase(JNK) activation in human hepatocellular carcinoma cells(HCC).METHODS:HepG2,Bel-7402 and L-02 cell lines were cultured in vitro and the apoptotic effects of BrMC were evaluated by flow cytometry(FCM) after propidium iodide(PI) staining,caspase-3 activity using enzymelinked immunosorbent assay(ELISA),and DNA agarose gel electrophoresis.ROS production was evaluated by FCM after dichlorodihydrofluorescein diacetate(DCHFDA) probe labeling.The phosphorylation level of JNK and c-Jun protein was analyzed by Western blotting.RESULTS:FCM after PI staining showed a dose-dependent increase in the percentage of the sub-G1 cell pop-ulation(P < 0.05),reaching 39.0% ± 2.8% of HepG2 cells after 48 h of treatment with BrMC at 10 μmol/L.The potency of BrMC to HepG2 and Bel-7402(32.1% ± 2.6%) cells was found to be more effective than the lead compound,chrysin(16.2% ± 1.6% for HepG2 cells and 11.0% ± 1.3% for Bel-7402 cell) at 40 μmol/L and similar to 5-flurouracil(33.0% ± 2.1% for HepG2 cells and 29.3% ± 2.3% for Bel-7402 cells) at 10 μmol/L.BrMC had little effect on human embryo liver L-02 cells,with the percentage of sub-G1 cell population 5.4% ± 1.8%.Treatment of HepG2 cells with BrMC for 48 h also increased the levels of active caspase-3,in a concentration-dependent manner.z-DEVD-fmk,a caspase-3specific inhibitor,prevented the activation of caspase-3.Treatment with BrMC at 10 μmol/L for 48 h resulted in the formation of a DNA ladder.Treatment of cells with BrMC(10 μmol/L) increased mean fluorescence intensity of DCHF-DA in HepG2 cells from 7.2 ± 1.12 at 0 h to 79.8 ± 3.9 at 3 h and 89.7 ± 4.7 at 6 h.BrMC did not affect ROS generation in L-02 cells.BrMC treatment failed to induce cell death and caspase-3 activation in HepG2 cells pretreated with N-acetylcysteine(10 mmol/L).In addition,in HepG2 cells treated with BrMC(2.5,5.0,10.0 μmol/L) for 12 h,JNK activation was observed.Peak JNK activation occurred at 12 h post-treatment and this activation persisted for up to 24 h.The expression of phosphorylated JNK and c-Jun protein after 12 h with BrMC-treated cells was inhibited by N-acetylcysteine and SP600125 pre-treatment,but GW9662 had no effect.SP600125 substantially reduced BrMC-induced cell death and caspase-3 activation of HepG2 cells.N-acetylcysteine and GW9662 also attenuated induction of cell death and caspase-3 activation in HepG2 cells treated with BrMC.CONCLUSION:BrMC induces apoptosis of HCC cells by ROS generation and sustained JNK activation. | Xiao-Hong Yang Jian-Guo Cao Hong-Lin Xiang Fei Liu Yuan Lv Hunan Province China Xing Zheng | 2010 | World Journal of Gastroenterology2010,16,27: | 38 |
| 2 | Transient Macular Edema after Laser In-Situ Keratomileusis显示文摘Purpose: To investigate the macular changes induced by laser in situ keratomileusisc(LASIK) procedure.Methods: Optical Coherence Tomography(OCT) was used to examine 27 eyes of 15LASIK patients before surgery, and 1 day, 1 week, 1 month after surgery.Results: The mean thick iness of neuroepithelial layer of macula was (195±24)microns, ( 178 ± 16) microns 1 day and 1 month after surgery, respectively, while thebaseline was (174 ± 12)microns. The increase in macular neuroepithelial thickness wascorrelated with the degree of myopia, but not with the duration of suction or thepostoperative BCVA.Conclusion: Mild macular edema is common in the first month following LASIK,especially in high myopia. | 3 | Time-frequency characteristics based motion estimation and imaging for high speed spinning targets via narrowband waveforms显示文摘This paper focuses on the motion parameters estimation and inversed synthetic aperture radar (ISAR) imaging for the high-speed spinning targets via the time-frequency characteristics. Assuming that the translational motion is compensated with the available motion compensation techniques, then the residual part of translational motion and the spinning motion are estimated with high precision based on the timefrequency characteristics. Combining with the procedure of the motion estimation, a novel coherent timefrequency spectrum intensity redistribution method, coherent single range Doppler interferometry (CSRDI), is proposed, which provides higher resolution and sustain lower signal to noise rate (SNR) than the current approach. | 4 | Novel functional proteins interact with midkine in hepatic cancer cells显示文摘BACKGROUND: Midkine is a heparin-binding growth factor that promotes the proliferation, survival, migration and differentiation of various target cells. Midkine plays an important role in tumorigenesis and tumor progression, and is overexpressed in many human malignant tumors. Patients with high tumor midkine expression frequently have a worse prognosis than those with low expression. The present study was designed to investigate the interaction network of midkine in hepatic cancer cells, and to elucidate its role in hepatocellular carcinoma. METHODS: DNA encoding full-length midkine was cloned into pDBLeu vector to serve as bait in yeast two-hybrid screening to identify interacting proteins. Candidate proteins were examined on SC-Leu-Trp-His+3-AT (20 mmol/L) plates and assayed for X-gal activity, then sequenced and classified according to the GenBank. Finally, identified proteins were expressed by the in vitro expression system pCMVTnT, and protein interactions were confirmed by co-immunoprecipitation. RESULTS: Using the yeast two-hybrid system, we found 6 proteins that interacted with midkine: NK-kappa-B inhibitor alpha (I-κ-B-α), Dvl-binding protein naked cuticle 2, granulin, latent active TGF-β binding protein 3, latent active TGF-β binding protein 4, and phospholipid scramblase 1. | 6 | Trend and seasonal variations of atmospheric CH_4 in Beijing显示文摘The atmospheric CH\-4 in Beijing is still increasing, even though its increasing rate has significantly decreased from 1.76%/a during 1985—1989 to 0.50%/a during 1990—1997. The seasonal variation of CH\-4 concentration showed a double\|peak pattern, one peak appearing in winter and the other in summer. It is evident that the annually seasonal variations of atmospheric CH\-4 in Beijing are different. From 1986 to 1997, the atmospheric CH\-4 increased by 185 ppbv, 37% and 21% of which were due to the increase in winter and in summer, respectively. After 1993, the annually seasonal increasing rate of CH\-4 concentration in summer (due to emission from biogenic sources) is negative while the increasing rate in winter (due to emission from non\|biogenic sources) is positive about 25 ppbv/a. As a result, the increase of CH\-4 emission from non\|biogenic sources in winter is the major reason that caused the annually seasonal increasing rate from 1993 to 1997.
