1Exosomes derived from differentiated human ADMSC with the Schwann cell phenotype modulate peripheral nerve-related cellular functions显示文摘Peripheral nerve regeneration remains a significant clinical challenge due to the unsatisfactory functional recovery and public health burden.Exosomes,especially those derived from mesenchymal stem cells(MSCs),are promising as potential cell-free therapeutics and gene therapy vehicles for promoting neural regeneration.In this study,we reported the differentiation of human adipose derived MSCs(hADMSCs)towards the Schwann cell(SC)phenotype(hADMSC-SCs)and then isolated exosomes from hADMSCs with and without differentiation(i.e.,dExo vs uExo).We assessed and compared the effects of uExo and dExo on antioxidative,angiogenic,anti-inflammatory,and axon growth promoting properties by using various peripheral nerve-related cells.Our results demonstrated that hADMSC-SCs secreted more neurotrophic factors and other growth factors,compared to hADMSCs without differentiation.The dExo isolated from hADMSC-SCs protected rat SCs from oxidative stress and enhanced HUVEC migration and angiogenesis.Compared to uExo,dExo also had improved performances in downregulating pro-inflammatory gene expressions and cytokine secretions and promoting axonal growth of sensory neurons differentiated from human induced pluripotent stem cells.Furthermore,microRNA(miRNA)sequencing analysis revealed that exosomes and their parent cells shared some similarities in their miRNA profiles and exosomes displayed a distinct miRNA signature.Many more miRNAs were identified in dExo than in uExo.Several upregulated miRNAs,like miRNA-132-3p and miRNA-199b-5p,were highly related to neuroprotection,anti-inflammation,and angiogenesis.The dExo can effectively modulate various peripheral nerve-related cellular functions and is promising for cell-free biological therapeutics to enhance neural regeneration.Bo Liu Yunfan Kong Wen Shi Mitchell Kuss Ke Liao Guoku Hu Peng Xiao Jagadesan Sankarasubramanian Chittibabu Guda Xinglong Wang Yuguo Lei Bin Duan 2022Bioactive Materials2022,7,8:3
2Comparative Analysis of Protein-Protein Interactions in Cancer-Associated Genes显示文摘Protein-protein interactions (PPIs) have been widely studied to understand the biological processes or molecular functions associated with different disease systems like cancer. While focused studies on individual cancers have generated valuable information, global and comparative analysis of datasets from different cancer types has not been done. In this work, we carried out bioinformatic analysis of PPIs corresponding to differentially expressed genes from microarrays of various tumor tissues (belonging to bladder, colon, kidney and thyroid cancers) and compared their associated biological processes and molecular functions (based on Gene Ontology terms). We identified a set of processes or functions that are common to all these cancers, as well as those that are specific to only one or partial cancer types. Similarly, protein interaction networks in nucleic acid metabolism were compared to identify the common/specific clusters of proteins across different cancer types. Our results provide a basis for further experimental investigations to study protein interaction networks associated with cancer. 13Predicting the Subcellular Localization of Human Proteins Using Machine Learning and Exploratory Data Analysis显示文摘Identifying the subcellular localization of proteins is particularly helpful in the func- tional annotation of gene products. In this study, we use Machine Learning and Ex- ploratory Data Analysis (EDA) techniques to examine and characterize amino acid sequences of human proteins localized in nine cellular compartments. A dataset of 3,749 protein sequences representing human proteins was extracted from the SWISS-PROT database. Feature vectors were created to capture specific amino acid sequence characteristics. Relative to a Support Vector Machine, a Multi-layer Perceptron, and a Na¨?ve Bayes classifier, the C4.5 Decision Tree algorithm was the most consistent performer across all nine compartments in reliably predict- ing the subcellular localization of proteins based on their amino acid sequences (average Precision=0.88; average Sensitivity=0.86). Furthermore, EDA graphics characterized essential features of proteins in each compartment. As examples, proteins localized to the plasma membrane had higher proportions of hydrophobic amino acids; cytoplasmic proteins had higher proportions of neutral amino acids; and mitochondrial proteins had higher proportions of neutral amino acids and lower proportions of polar amino acids. These data showed that the C4.5 classifier and EDA tools can be effective for characterizing and predicting the subcellular localization of human proteins based on their amino acid sequences.George K.
