| 5 | Inhibition of Breast Cancer Resistance Protein(BCRP) by Ko143 Can Affect Pharmacokinetics of Enrofloxacin in Exopalaemon carinicauda显示文摘Adenosine triphosphate-binding cassette transporter breast cancer resistance protein(BCRP) exists highly in the apical membranes of epithelia, and is involved in drug availability. Ko143 is a typical inhibitor of BCRP in rodents. The synthetic antibacterial agent enrofloxacin(ENRO) is a fluoroquinolone employed as veterinary and aquatic medicine, and also a substrate for BCRP. BCRP gene highly expressed in the hepatopancreas and intestine of Exopalaemon carinicauda as was determined with real-time quantitative reverse transcription-polymerase chain reaction(RT-q PCR) method. The effects of Ko143 on the abundance of BCRP m RNA and ENRO pharmacokinetics in E. carinicauda were studied. The m RNA abundance of BCRP decreased significantly in hepatopancreas and intestine(P < 0.05) after Ko143 treatment. Co-administration of Ko143 significantly changed the pharmacokinetics of orally administered enrofloxacin, which was supported by higher distribution half-life(t_(1/2α)), elimination half-life(t_(1/2β)), area under the curve up to the last measurable concentration(AUC_(0-t)), peak concentration(C_(max)) and lower clearance(CL/F). These findings revealed that Ko143 downregulates BCRP expression in hepatopancreas and intestine, thus affects the pharmacokinetics of orally administered enrofloxacin in E. carinicauda. | 10 | Purification and Refolding of a Novel β-Agarase from Inclusion Body of E. coli显示文摘β-agarase AgaB appears to represent a new family of glycoside hydrolase; it is structurally and functionally different from other known agarases. In the present study, AgaB was expressed with a temperature-inducible expression system in E. coli BL21 (DE3) as a fusion protein bearing a C-terminal hexahistidine tag. The protein existed mainly in the form of inclusion body. After being washed and solubilized, AgaB in inclusion body was denatured and purified to electrophoretic purity by immobilized metal affinity chromatography. The purified AgaB was then refolded using a simple pulse dilution method, and the refolded AgaB showed a high specific hydrolysis activity of about 1600 units /mg protein. | 12 | Recent progress on the hydrogen storage properties of ZrCo-based alloys applied in International Thermonuclear Experimental Reactor(ITER)显示文摘Under the development of International Thermonuclear Experimental Reactor(ITER) system aimed at realizing the controllable fusion reaction to solve the energy crisis fundamentally, there is an urgent need to find an appropriate material for tritium handling. ZrCo alloy is considered to be a promising candidate for the storage and delivery of hydrogen isotopes due to the favorable characteristics such as low plateau pressure for absorption, high dissociation pressure at moderate temperature and better ability of trapping ~3He. However, the hydrogen induced disproportionation and the slower recovery/deliverty rate of ZrCo-based alloys have limited their further application in ITER system. This review summarizes the efforts towards enhancing the hydrogen storage properties of ZrCo-based alloys including element substitution, surface modification, disproportionation mechanism investigation and the isotope effect study. Element substitution and surface modification play positive role to improve the anti-disproportionation ability and kinetic property of the alloys. However, the ZrCo-based alloys require to be further modified by more attempts such as new composition, novelty modification method or catalyst addition in order to better satisfy the application demands for tritium handling. | 15 | Low-Temperature Synthesis of Crystalline Inorganic/Metallic Nanocrystal-Halloysite Composite Nanotubes显示文摘We have demonstrated a facile approach for the low-temperature synthesis of crystalline inorganic/metallic nanocrystal-halloysite composite nanotubes by employing the bulk controlled synthesis of inorganic/metallic nanocrystals on halloysite nanotubes.The halloysite clay nanotubes can adsorb the target precursor and induce inorganic/metallic nanocrystals to grow in situ.The crystalline phase and morphology of the composite clay nanotubes is tunable.By simply tuning the acidity of the titania sol,the crystalline titania-clay nanotubes with tunable crystalline phases of anatase,a mixture of anatase and rutile or rutile are achieved.The approach is general and has been extended to synthesize the representative perovskite oxide(barium and strontium titanate)-halloysite composite nanotubes.Metallic nickel nanocrystal can also be grown on the surface of halloysite nanotubes at low temperature.The traditional thermal treatment for crystallite transformation is not required,thus intact contour of halloysite nanotubes and the crystallinity structure of halloysite nanotubes can be guaranteed.The combined properties from inorganic/metallic nanocrystal(high refractive index,high dielectric constant and catalytic ability)and the halloysite clay nanotubes are promising for applications such as photonic crystals,high-k-gate dielectrics,photocatalysis and purification. | Cuiping Li Jiaqiang Wang Siquan Feng Zan Liu | 2014 | Chinese Journal of Chemistry2014,32,7: | 1 |
| 16 | 乳腺癌新辅助化疗前后HER-2表达的变化(英文)显示文摘Objective:The aim of this study was to study changes of HER-2 expression after neoadjuvant chemotherapy in the breast cancer cases.Methods:One hundred and thirty-seven female patients with primary breast cancers,who received neoadjuvant chemotherapy,underwent core needle puncture and Mammotome biopsy before chemotherapy,and the biopsy results were used as the basis of histological diagnosis,fluorescence in situ hybridization (FISH) was performed to test HER2 status of tumor tissues before and after chemotherapy.All patients underwent FEC,TE,or AC neoadjuvant chemotherapy of 2-6 cycles before surgery.Results:Twenty-two patients were positive according to FISH test among 137 preoperative patients,8 patients achieved pathological complete remission after chemotherapy (three HER-2 positive patients and five negative patients),91 patients achieved partial remission,24 patients were stable,and 14 cases were invalid.Twenty-two patients were positive according to FISH test (8 patients with pathological complete remission did not undergo test),and positive patients still expressed positively after chemotherapy before neoadjuvant chemotherapy.Three negative patients were converted to be positive,and changes before and after chemotherapy had no statistical difference (P>0.05).Conclusion:Neoadjuvant chemotherapy makes no influence on patients with HER-2 positive expression,while patients with negative expression can be converted to be positive,but without significant difference. | Yaojun Feng Xinhong Wu Cuiping Pan Juan Xu Wei Zhong Jun Shao Biao Ma | 2011 | The Chinese-German Journal of Clinical Oncology2011,10,8: | 1 |

| 18 | Study on Extraction and Properties of Gelatin from Squid Skin显示文摘A kind of aquatic gelatin was prepared from squid skin. The technological parameters for extracting gelatin were optimized by the method of orthogonal design. The fundamental properties of the raw material and gelatin were analyzed. The results showed that the optimal extraction conditions were as follows: pH value of 8,temperature at 70 ℃,material-to-liquid ratio at 1∶ 4,and extraction time of 1. 5 h. under such conditions,the highest yield of gelatin and the purity were 5. 94% and 96. 73%,respectively. The physicochemical properties of squid skin gelatin were coincided with standards of edible gelatin.
